ABSTRACT

Lipid analysis at the molecular species level represents a valuable opportunity for clinical applications due to the essential roles that lipids play in metabolic health. However, a comprehensive and high-throughput lipid profiling remains challenging given the lipid structural complexity and exceptional diversity. Herein, we present an 'omic-scale targeted LC-MS/MS approach for the straightforward and high-throughput quantification of a broad panel of complex lipid species across 26 lipid (sub)classes. The workflow involves an automated single-step extraction with 2-propanol, followed by lipid analysis using hydrophilic interaction liquid chromatography in a dual-column setup coupled to tandem mass spectrometry with data acquisition in the timed-selective reaction monitoring mode (12 min total run time). The analysis pipeline consists of an initial screen of 1903 lipid species, followed by high-throughput quantification of robustly detected species. Lipid quantification is achieved by a single-point calibration with 75 isotopically labeled standards representative of different lipid classes, covering lipid species with diverse acyl/alkyl chain lengths and unsaturation degrees. When applied to human plasma, 795 lipid species were measured with median intra- and inter-day precisions of 8.5 and 10.9%, respectively, evaluated within a single and across multiple batches. The concentration ranges measured in NIST plasma were in accordance with the consensus intervals determined in previous ring-trials. Finally, to benchmark our workflow, we characterized NIST plasma materials with different clinical and ethnic backgrounds and analyzed a sub-set of sera (n = 81) from a clinically healthy elderly population. Our quantitative lipidomic platform allowed for a clear distinction between different NIST materials and revealed the sex-specificity of the serum lipidome, highlighting numerous statistically significant sex differences.

ABSTRACT

Although acoustic-emission (AE) piezoelectric transducers have distinctive variations in sensitivity, depending on frequency, propagation medium, and coupling, the vast majority of AE research is conducted by utilizing uncalibrated AE transducers. As a consequence, most results obtained by different groups are not comparable among each other. In this work, primary calibration by the method of reciprocity is shown. Rayleigh and longitudinal wave calibration curves are presented for piezoelectric high-frequency broadband transducer, mounted on steel and aluminium, in the frequency range 300 kHz-4 MHz. Influences on primary calibration of AE transducers, namely, by coupling medium, contact pressure, and propagation medium, are investigated.

ABSTRACT

We present Brownian motion-based sizing of individual submicron and nanoparticles in liquid samples. The advantage of our approach is that particles can freely diffuse in a 10 µm thin liquid film and are therefore always within the focal depth of a low numerical aperture objective. Particles are visualized with dark-field microscopy, and the resulting diffraction-limited spots are tracked over a wide field of view of several hundred micrometers. Consequently, it is ascertained that long 2D trajectories are acquired, which leads to significantly increased particle sizing precision. The hydrodynamic diameters of metal particles with nominal sizes ranging from 70 to 200 nm in aqueous solution were determined by tracking for up to 2 min, and it was investigated if the diffusion characteristics were influenced by the proximity of substrates. This was not the case, and the estimated diameters were in good agreement with the values obtained by electron microscopy, thus validating the particle sizing principle. Furthermore, we measured a sample mixture to demonstrate the distinction of close particle sizes and performed the conjugation of a model protein (BSA) on the nanoparticle surface. An average increase in the radius of 9 nm was determined, which corresponds to the size of the BSA protein.

ABSTRACT

A total of 19 pesticide preparations were used according to agricultural practice in six trials in apple orchards. Using gas chromatography-mass spectrometry (GC-MS) and liquid chromatography-tandem mass spectrometry (LC-MS/MS), premature Golden Delicious apples collected 64, 50, 36 days before harvest and mature fruit were examined for residues of active ingredients. No residues of triflumuron, triazamate, chlorpyrifos, etofenprox, fenoxycarb, kresoxim-methyl, cyprodinyl, difenoconazole or thiram were detected in the first sampling. Also, the levels of chlorpyrifos-methyl, penconazole, tebuconazole and tolylfluanid dropped during the pre-harvest interval. Detectable residues of pyridaben, thiacloprid, trifloxystrobin and tetraconazole in harvested fruits were below 0.01 mg kg(-1), which is the maximum concentration of residues acceptable by baby-food producers in any raw material. The only residues exceeding this concentration were captan and teflubenzuron. Based on the data, farmers can choose pesticides for optimal treatment of plants, while enabling growth of a safe crop suitable for baby-food production.

ABSTRACT

Potential of ultra-performance liquid chromatography (UPLC) separation strategy coupled with tandem (in space) mass spectrometric detection (MS/MS) in multiresidue pesticide analysis was critically assessed. Performance parameters such as number of theoretical plates, height of theoretical plate, peak symmetry and peak capacity were measured/calculated on the basis of data generated by analysis of apple extracts containing 17 (semi)polar pesticides representing various classes of active ingredients of widely used crop protective preparations. Ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) procedure provided improved chromatographic parameters resulting in significantly increased sample throughput including lower solvent consumption and lower limits of quantitation (LOQs) for most of target analytes compared to common method employing conventional high-performance liquid chromatography (HPLC) separation.
