ABSTRACT

The thermal stability of newborns is an essential parameter that can be recorded to evaluate neonatal care. Knowing the thermal windows to evaluate and maintain a constant temperature helps significantly reduce neonatal mortality. This study aimed to assess the superficial temperature alterations in the distinct thermal windows of puppies with mothers of diverse weights and their repercussions. We evaluated the superficial temperature using infrared thermography at eight thermal windows and seven different times: when wet due to the fetal fluid immediately after birth until 24 h of life in newborn puppies from bitches divided into four bodyweight groups. The results revealed a positive correlation between the dam's weight and the ability to achieve thermostability in the newborn puppies in all the evaluated thermal windows. The time effect showed the lowest temperatures when the puppies were still wet, a gradual increase, and the highest temperature at 24 h after birth. The thermal windows with the highest temperatures were abdominal, thoracic, nasal, and upper left palpebral, and those with the lowest were the thoracic limb brachial biceps, thoracic limb elbow, metacarpal, and femoral pelvic limb. A significant increase in the temperatures in the thermal windows of the abdominal, thoracic, and upper left palpebral immediately after ingesting colostrum was observed. The weight of the dams is an important factor that positively intervenes in the thermoregulatory capacity of the puppies, especially when newborns are dry and have been passed 24 h after birth.

ABSTRACT

Cocoa (Theobroma cacao L.) is the only tree that can produce cocoa. Cocoa beans are highly sought after by chocolate makers to produce chocolate. Cocoa can be fine aromatic, characterized by floral and fruity notes, or it can be described as standard cocoa with a more pronounced cocoa aroma and bitterness. In this study, the genetic and biochemical determinants of sensorial notes and nonvolatile compounds related to bitterness, astringency, fat content, and protein content will be investigated in two populations: a cultivated modern Nacional population and a population of cocoa accessions collected recently in the Ecuadorian South Amazonia area of origin of the Nacional ancestral variety. For this purpose, a genome-wide association study (GWAS) was carried out on both populations, with results of biochemical compounds evaluated by near-infrared spectroscopy (NIRS) assays and with sensory evaluations. Twenty areas of associations were detected for sensorial data especially bitterness and astringency. Fifty-three areas of associations were detected linked to nonvolatile compounds. A total of 81 candidate genes could be identified in the areas of the association.

ABSTRACT

Multiple paternity (MP) in the brown smooth-hound shark (Mustelus henlei) was assessed in 15 litters (15 mothers and 97 embryos) collected in the northern Gulf of California of which 86.7% were sired by more than one male (i.e., from 2 to 4 sires). When taken together with results from previous studies, this record indicates that there is regional variation in MP in M. henlei in the northeastern Pacific. This pattern is associated with variations in the reproductive traits of each population (e.g., female size and litter size). In the Gulf of California, the results of a generalized linear model (GLZ) indicated that the litters of larger females had a higher probability of MP compared to those of smaller females.

ABSTRACT

The path to sustainable small-scale fisheries (SSF) is based on multiple learning processes that must transcend generational changes. To understand young leaders from communities with sustainable SSF management practices in Mexico, we used in-depth interviews to identify their shared motivations and perceptions for accepting their fishing heritage. These possible future decision-makers act as agents of change due to their organizational and technological abilities. However, young people are currently at a crossroads. Many inherited a passion for the sea and want to improve and diversify the fishing sector, yet young leaders do not want to accept a legacy of complicated socioenvironmental conditions that can limit their futures. These future leaders are especially concerned by the uncertainty caused by climate change. If fishing and generational change are not valued in planning processes, the continuity of fisheries, the success of conservation actions, and the lifestyles of young fishers will remain uncertain.

ABSTRACT

We report for the first time a highly divergent lineage in the Caribbean Sea for the bull shark (Carcharhinus leucas) based on the analysis of 51 mitochondrial DNA genomes of individuals collected in the western North Atlantic. When comparing the mtDNA control region obtained from the mitogenomes to sequences reported previously for Brazil, the Caribbean lineage remained highly divergent. These results support the existence of a discrete population in Central America due to a phylogeographic break separating the Caribbean Sea from the western North Atlantic, Gulf of Mexico and South America.

ABSTRACT

Since the 1980s, there has been growing concern in the Mexican Atlantic regarding high catches of neonate and juvenile sharks in small-scale fisheries. Fishery-dependent data from 1993 to 1994 and 2007 to 2017 and fishers' ecological knowledge from 2017 were used to identify nursery areas for scalloped hammerhead, Sphyrna lewini, in the southern Gulf of Mexico. Catch records and fishing areas of neonates, YOYs, juveniles and adults of S. lewini (N = 1885) were obtained from calcareous and terrigenous regions in the western Yucatan Peninsula. The results suggest that a nursery for scalloped hammerhead is found in the terrigenous region, characterized by relatively shallow and turbid waters due to rivers' discharges. Neonates and YOYs (96% and 86% of their total records, respectively) were commonly found there over the years in May-August in multiple fishing areas identified by fishers, although mainly between isobaths 10-30 m. The enforcement of management measures is necessary because the nursery is located in a region with intense fishing effort.

ABSTRACT

Mexico is home to a broad biodiversity of shark species, and more than half of the sharks in Mexican waters are distributed in the Mexican Pacific, with over 62 species recorded. This high biodiversity is the result of numerous and diverse marine and coastal environments, including the dynamic Mexican seas, where circulation and spatial variation of oceanic currents is complex, and the seasonal variation of isotherms can be substantial. In the Mexican Pacific we can distinguish some patterns of species distribution, with temperate water and subtropical species found in the northern regions, and tropical conditions and species found in the south. Due to the blending of cold and warm waters, however, we can find a mixture of subtropical and tropical sharks in northern regions seasonally, off the west coast of the Baja California Peninsula and in the Gulf of California, and these areas contain the highest shark species richness. In this chapter we described the shark species biodiversity occurring in the Mexican Pacific, review their conservation status in a regional and global context, and summarize the main conservation measures and issues associated with their management.

ABSTRACT

BACKGROUND: Escherichia coli W3110 and a group of six isogenic derivatives, each displaying distinct specific rates of glucose consumption were characterized to determine levels of GFP production and population heterogeneity. These strains have single or combinatory deletions in genes encoding phosphoenolpyruvate:sugar phosphotransferase system (PTS) permeases as PtsG and ManX, as well as common components EI, Hpr protein and EIIA, also the non-PTS Mgl galactose/glucose ABC transporter. They have been transformed for expressing GFP based on a lac-based expression vector, which is subject to bistability. RESULTS: These strains displayed specific glucose consumption and growth rates ranging from 1.75 to 0.45 g/g h and 0.54 to 0.16 h-1, respectively. The rate of acetate production was strongly reduced in all mutant strains when compared with W3110/pV21. In bioreactor cultures, wild type W3110/pV21 produced 50.51 mg/L GFP, whereas strains WG/pV21 with inactive PTS IICBGlc and WGM/pV21 with the additional inactivation of PTS IIABMan showed the highest titers of GFP, corresponding to 342 and 438 mg/L, respectively. Moreover, we showed experimentally that bistable expression systems, as lac-based ones, induce strong phenotypic segregation among microbial populations. CONCLUSIONS: We have demonstrated that reduction on glucose consumption rate in E. coli leads to an improvement of GFP production. Furthermore, from the perspective of phenotypic heterogeneity, we observed in this case that heterogeneous systems are also the ones leading to the highest performance. This observation suggests reconsidering the generally accepted proposition stating that phenotypic heterogeneity is generally unwanted in bioprocess applications.

ABSTRACT

ABSTRACT Cacao (Theobroma cacao L.) breeding programmes in Ecuador have focused on obtaining high-yield clones with improved disease resistance. Cacao clones should also have photosynthetic characteristics which support increased productivity. Regarding the weather conditions at the coast of Ecuador, where most of the year there are overcasts and low air evaporative demand, there is the possibility to grow cacao without overhead shade. This study focused on the photosynthetic response at two different photosynthetic photon flux densities (PPFD) of Ecuadorian cacao clones. Seven-year old cacao clones were evaluated: eight clones of Nacional type and two commercial clones (CCN 51 and EET 103), used as controls. All clones showed an increase of 35 % on average in net photosynthetic rate (A)with increasing PPFD from the light saturation point for cacao (i.e. 400 μmol m-2 s-1) to high values (1000 μmol 2 s-1). Such light responsiveness in A has not been reported before. Higher A was associated with higher apparent electron transport rate, while high stomatal conductance was maintained under both PPFD conditions. Under high PPFD, low non-photochemical quenching values were found, suggesting low energy dissipation. All clones showed high maximum quantum yields of PSII (Fv/Fm), suggesting the absence of damage of the photochemical system.

ABSTRACT

INTRODUCTION:: Interferon-γ (IFN-γ) plays a crucial role in resistance to mycobacterial diseases; accordingly, variants of the gene encoding this cytokine may be associated with elevated risk of contracting pulmonary tuberculosis (TB). METHODS:: Blood samples were collected from 135 Warao indigenous individuals with newly diagnosed sputum culture-positive TB. Of these, 24 were diagnosed with active tuberculosis (ATB). The study comprised 111 participants, who were grouped as follows: 1) 14 tuberculin skin test (TST)-positive Warao indigenous individuals and 4 that were QuantiFERON-TB?Gold In-Tube (QFT-IT) test-positive, collectively comprising the latent TB infection group (LTBI), n = 18), and 2) healthy controls who were QFT-IT- and TST-negative, comprising the control group (CTRL, n = 93). Detection of the IFN γ gene (IFNG) +874A/T polymorphism was performed via PCR and quantification of IFNG expression via qPCR. RESULTS:: Relative to indigenous and white Americans, ATB and CTRL groups had a higher frequency of the IFNG SNP (+874A): 23 (95.8%) and 108 (97.3%), respectively. Indigenous Warao individuals homozygous for the IFNG (+874) A allele exhibited 3.59-fold increased risk of developing TB (95% confidence interval, 2.60-4.96, p =0.0001). A decreased frequency of the AT genotype was observed in individuals with pulmonary TB (4.16%) and controls (0.90%). The frequency of the TT genotype was decreased among controls (1.80%); none of the patients with TB were found to have this genotype. The differences in IFNG expression between the groups, under unstimulated and stimulated conditions, were not statistically significant. CONCLUSIONS:: Preliminary results demonstrate concordance between IFNG +874 A/A genotype and low expression of IFNG.

ABSTRACT

Ascariasis is a disease caused by the nematode A. lumbricoides affecting 1 billion people worldwide. In this study the specific response for IgA antibodies against extracts of A. lumbricoides eggs in the serum of children in rural areas of Miranda state. One hundred children were evaluated at ages 3 and 14 and were taken serum samples and diagnosis of stools. According to the reported, were classified into four groups: infected by A. lumbricoides (Asc +), uninfected (NSOP), infected A. lumbricoides and other parasite (Asc +/OP +); and individuals with other parasitic infections without A. lumbricoides (ASC/ OP+). The extract was tested by ELISA, Western blotting and immunocytochemestry. The electrophoretic profile showed 7 bands of 102, 92, 85, 65, 50, 45 and 30kDa. By ELISA, the 26% of patients in group A. lumbricoides were positive, while the 56% in the NSOP group were seronegative. Immune assay by inhibition with A. lumbricoides extract allowed identify three bands for the IgA antibodies of 92, 85 and 65 kDa different to the bands recognized by adult extracts of A. lumbricoiedes. The levels of antibodies were significantly higher in infected than non-infected children. In addition, the protein component on the surface of eggs was recognized by sera of infected individuals. These results indicate that eggs of Ascaris induce specific antibodies against proteins that could be used as antigens in serological diagnosis for complementing the diagnosis for Ascariasis.

ABSTRACT

Giardiasis is an infection caused by Giardia duodenalis, transmitted by consuming water or food contaminated with Giardia cysts, mainly in children living in rural areas. Little is known about proteins associated to the specific antibody response, the identification of specific antigens is necessary for studies of seroprevalence. The aim of this study was to select antigenic proteins from soluble extract or fractions of the parasite for the design of specific diagnostic tests. Were evaluated eighty individuals between 4-80 years, from rural communities were classified according to the presence or absence of cysts in the stool. The soluble extract and the fractions were obtained from WB (ATCC) strain of Giardia sp by differential centrifugation, analyzed by polyacrylamide gel electrophoresis and transferred to nitrocellulose membranes and finally tested by Western Blots for the IgG antibody response. The general prevalence was between 9-12%, and 22% of them were positive for G. duodenalis, 46% of individuals were negative in feces; and 32% were positive for G. duodenalis and other parasites. The protein profile revealed several bands from 190-15kDa; in which were recognized mainly proteins of 190, 176 and 51kDa in the fraction 2, and a protein of 23kDa in the fraction 3. The infection by G. duodenalis represents a human public health, and the identification of reactive proteins for the IgG response in the fractions 2 and 3, would allow us the design of serological test more specific, in order to discriminate the seropositive individuals from those seronegative.

The present study aimed at measuring seropositivities for infection by Ascaris suum and Toxocara canis using the excretory/secretory (E/S) antigens from Ascaris suum (AES) and Toxocara canis (TES) within an indigenous population. In addition, quantification of cytokine expressions in peripheral blood cells was determined. A total of 50 Warao indigenous were included; of which 43 were adults and seven children. In adults, 44.1% were seropositive for both parasites; whereas children had only seropositivity to one or the other helminth. For ascariosis, the percentage of AES seropositivity in adults and children was high; 23.3% and 57.1%, respectively. While that for toxocariosis, the percentage of TES seropositivity in adults and children was low; 9.3% and 14.3%, respectively. The percentage of seronegativity was comparable for AES and TES antigens in adults (27.9%) and children (28.6%). When positive sera were analyzed by Western blotting technique using AES antigens; three bands of 97.2, 193.6 and 200.2 kDas were mostly recognized. When the TES antigens were used, nine major bands were mostly identified; 47.4, 52.2, 84.9, 98.2, 119.1, 131.3, 175.6, 184.4 and 193.6 kDas. Stool examinations showed that Blastocystis hominis, Hymenolepis nana and Entamoeba coli were the most commonly observed intestinal parasites. Quantification of cytokines IFN-γ, IL-2, IL-6, TGF-ß, TNF-α, IL-10 and IL-4 expressions showed that there was only a significant increased expression of IL-4 in indigenous with TES seropositivity (p < 0.002). Ascaris and Toxocara seropositivity was prevalent among Warao indigenous.

Quantification of cytokines IFN-γ, IL-2, IL-6, TGF-β, TNF-α, IL-10 and IL-4 expressions showed that there was only a significant increased expression of IL-4 in indigenous with TES seropositivity (p < 0.002).
