ABSTRACT

Breast cancer is the main cause of cancer-related death among women, with a 0.5% increase in incidence per year. Natural killer cells (NK) are part of the innate immune system recognizing class I HLA molecules on target cells through their membrane receptors, called killer cell immunoglobulin-like receptors (KIR). The aim of our study was to evaluate the association between the KIR genes and HLA alleles in patients with breast cancer and healthy controls. Two hundred thirty patients with breast cancer and 272 healthy controls were typed for HLA class I and KIR genes by PCR-SSO. When both groups were compared, the presence of inhibitory KIR2DL2 receptors was significantly higher in breast cancer patients than in healthy controls. No significant differences were found for HLA-C2 and HLA-Bw4. However, a higher frequency of HLA-C1 in breast cancer patients was observed. These findings suggest a potential role for the KIR gene system in breast cancer. Further studies to confirm this observation are warranted.

ABSTRACT

Type 1 diabetes mellitus (T1D) is a multifactorial and chronic autoimmune disease caused by the deficiency of insulin synthesis and or by its secretion or action defects. Genetic and environmental factors are known to be involved in its pathogenesis. The human leukocyte antigen complex (human leukocyte antigen (HLA)) constitutes the most relevant region contributing with 50% of the inherited risk for T1D. Natural killer cells (NK) are part of the innate immune system recognizing class I HLA molecules on target cells through their membrane receptors, called killer immunoglobulin-like receptors (KIR). The aim of our study is to evaluate the association between the KIR genes and HLA alleles in patients with T1D and healthy controls. Two hundred forty-eight T1D patients and 250 healthy controls were typed for HLA and KIR genes by PCR-SSP. Our results showed an increase of C2 in controls (p = 0.002). The genotype 2DL1/C2+ was also more common in controls (p = 0.001), as well as haplotype association KIR2DL2/DR3/DR4+ and the combination with only DR3+ (p < 0.001; p < 0.001). The maximum protection was seen when KIR2DL2/DR3-were absent when the combination of KIR2DL1/C2+ were present (p < 0.001) and the maximum risk was observed when KIR2DL2/DR3/DR4+ were present in the absence of KIR2DL1/C2- (p = 0.005). Our results confirmed the association of the KIR2DL2/DR3 increasing risk for T1D and suggest a protective role of KIR2DL1/C2.

ABSTRACT

A presença de anticorpos naturais contra células de porco (xeno-anticorpos) foi analisada em soros de indivíduos normais e de pacientes imunossuprimidos, imunodeficientes primários e renais crônicos hiperimunizados. Em indivíduos normais a média dos títulos observada foi de 1:664. Em 80,9 por cento de pacientes renais transplantados nao foi observado reduçao do título dos xeno-anticorpos após um ano de imunossupressao. Nos pacientes imunodeficientes foi observado um título mais reduzido, mas nunca inferior a 1:1. Após a eliminaçao dos anticorpos naturais do soro de pacientes renais crônicos hipersensibilizados com antígenos HLA, pela adsorçao dos soros com eritrócitos de porco seguido do tratamento com dithiotreitol (DTT), foi observado que os anticorpos IgG anti-HLA nao reagem com os antígenos de histocompatibilidade das células do porco. As conclusoes deste trabalho foram: o título dos xeno-anticorpos em indivíduos normais é muito elevado; a imunossupressao utilizada nos transplantes renais nao permite reduçao da barreira de anticorpos naturais; os imunodeficientes com títulos muito baixos destes anticorpos talvez possam usufruir de transplantes xenogênicos; o método de eliminaçao "in vitro" dos xeno-anticorpos é reproduzível; os soros dos pacientes renais crônicos hiperimunizados com antígenos HLA, nao reagem cruzadamente com linfócitos de porco.

ABSTRACT

O desenvolvimento da biologia molecular proporcionou recentemente novos metodos de investigacao de paternidade que foram incorporados a rotina de nosso laboratorio. Entre eles citaremos as "impressoes digitais do DNA" observadas pelo metodo do RFLP ou polimorfismo de tamanho de fragmentos de restricao ("restriction fragment lenght polymorphism") e a tipagem dos antigenos leucocitarios humanos (HLA) pela amplificacao alelo-especifica do DNA, pelo metodo da reacao em cadeia da polimerase (PCR). Tendo em vista o controle de qualidade obrigatorio no atendimento dos clientes, introduzimos em nosso trabalho a analise matematica computadorizada por intermedio de digitalizacao direta das bandas de DNA por programa desenvolvido para estes estudos, proporcionando resultados estatisticos perfeitos na analise de casos simples ou complexos, onde o possivel pai e falecido. A pesquisa estatistica com a formacao de um banco de dados de frequencias de alelos dos sistemas geneticos por nos utilizados em estudos de investigacao de paternidade na populacao brasileira sera resumidamente analisada junto com os resultados de 1000 pericias efetuadas
