ABSTRACT

Evaluating persistent trace organic chemicals (TOrCs) and transformation products (TPs) in membrane bioreactors (MBRs) is essential, given that MBRs are now widely implemented for wastewater treatment and water reuse. This research applied comprehensive two-dimensional gas chromatography coupled to time-of-flight mass spectrometry (GC×GC/TOF-MS)-based nontargeted analysis to compare the effectiveness of parallel aerobic and anaerobic MBRs (AeMBRs and AnMBRs, respectively), treating the same municipal wastewater. The average total chromatographic feature peak area abundances were significantly reduced by 84% and 72% from influent to membrane permeate in both the AeMBR and AnMBR (p < 0.05), respectively. However, the reduction of the average number of chromatographic features was significant for only AeMBR treatment (p = 0.006). A similar number of TPs were generated during both AeMBR and AnMBR treatments (165 vs 171 compounds, respectively). The overall results suggest that the AeMBR was more effective for reducing the diversity of TOrCs than the AnMBR, but both aerobic and anaerobic processes had a similar reduction of TOrC abundance. Suspect screening analysis using GC×GC/TOF-MS, which resulted in the tentative identification of 351 TOrCs, proved to be a powerful approach for uncovering compounds previously unreported in wastewater, including many fragrances and personal care products.

ABSTRACT

Coastal reintroduction sites for California condors (Gymnogyps californianus) can lead to elevated halogenated organic compound (HOC) exposure and potential health impacts due to the consumption of scavenged marine mammals. Using nontargeted analysis based on comprehensive two-dimensional gas chromatography coupled to time-of-flight mass spectrometry (GC×GC/TOF-MS), we compared HOC profiles of plasma from inland and coastal scavenging California condors from the state of California (CA), and marine mammal blubber from CA and the Gulf of California off Baja California (BC), Mexico. We detected more HOCs in coastal condors (32 ± 5, mean number of HOCs ± SD, n = 7) than in inland condors (8 ± 1, n = 10) and in CA marine mammals (136 ± 87, n = 25) than in BC marine mammals (55 ± 46, n = 8). ∑DDT-related compounds, ∑PCBs, and total tris(chlorophenyl)methane (∑TCPM) were, respectively, ∼7, ∼3.5, and ∼148 times more abundant in CA than in BC marine mammals. The endocrine-disrupting potential of selected polychlorinated biphenyls (PCB) congeners, TCPM, and TCPMOH was determined by in vitro California condor estrogen receptor (ER) activation. The higher levels of HOCs in coastal condors compared to those in inland condors and lower levels of HOC contamination in Baja California marine mammals compared to those from the state of California are factors to consider in condor reintroduction efforts.

ABSTRACT

The persistence of trace organic chemicals in treated effluent derived from both centralized wastewater treatment plants (WWTPs) and decentralized wastewater treatment systems (DEWATS) is of concern due to their potential impacts on human and ecosystem health. Here, we utilize non-targeted analysis (NTA) with comprehensive two-dimensional gas chromatography coupled with time of flight mass spectrometry (GC × GC/TOF-MS) to conduct an evaluation of the common persistent and removed compounds found in two centralized WWTPs in the USA and South Africa and one DEWATS in South Africa. Overall, removal efficiencies of chemicals were similar between the treatment plants when they were compared according to the number of chemical features detected in the influents and effluents of each treatment plant. However, the DEWATS treatment train, which has longer solids retention and hydraulic residence times than both of the centralized WWTPs and utilizes primarily anaerobic treatment processes, was able to remove 13 additional compounds and showed a greater decrease in normalized peak areas compared to the centralized WWTPs. Of the 111 common compounds tentatively identified in all three influents, 11 compounds were persistent in all replicates, including 5 compounds not previously reported in effluents of WWTPs or water reuse systems. There were no significant differences among the physico-chemical properties of persistent and removed compounds, but significant differences were observed among some of the molecular descriptors. These results have important implications for the treatment of trace organic chemicals in centralized and decentralized WWTPs and the monitoring of new compounds in WWTP effluent.

ABSTRACT

Supramolecular hydrogels formed by noncovalent self-assembly of low molecular weight (LMW) agents are promising next-generation biomaterials. Thixotropic shear response and mechanical stability are two emergent properties of hydrogels that are critical for biomedical applications including drug delivery and tissue engineering in which injection of the hydrogel will be necessary. Herein, we demonstrate that the emergent thixotropic properties of supramolecular phenylalanine-derived hydrogels are dependent on the conditions in which they are formulated. Specifically, hydrogels formed from fluorenylmethoxycarbonyl (Fmoc) modified phenylalanine derivatives, 3-fluorophenylalanine (Fmoc-3F-Phe) and pentafluorophenylalanine (Fmoc-F5-Phe), were characterized as a function of gelation conditions to examine how shear response and mechanical stability properties correlate to mode of gelation. Two distinct methods of gelation were compared. First, spontaneous self-assembly and gelation was triggered by a solvent exchange method in which a concentrated solution of the gelator in dimethylsulfoxide was diluted in water. Second, gelation was promoted by dissolution of the gelator in water at basic pH followed by gradual pH adjustment from basic to mildly acidic by the hydrolysis of glucono-delta-lactone. Hydrogels formed under solvent exchange conditions were mechanically unstable and poorly shear-responsive whereas hydrogels formed by gradual acidification were temporally stable and had highly shear-responsive viscoelastic character. These studies confirm that gelation environment and mechanism have a significant influence on the emergent properties of supramolecular hydrogels and offer insight into how gelation conditions can be used to tune hydrogel properties for specific applications.

ABSTRACT

BACKGROUND: Psychological stress is recognized as a major risk factor for a range of non-communicable diseases and possibly mortality. The extent to which the type and timing of stress exposure influences mortality, and potential differences between genders, remains unknown. OBJECTIVE: To examine the association between early-life and recent stressful experiences and mortality risk in later life, and to determine possible gender differences in these associations. METHOD: Data were obtained from 2152 French community-dwelling participants (aged ≥65). Questionnaires were used to evaluate recent stress, as well as retrospective reporting of childhood adversity. Mortality status was determined through death registries. Adjusted Cox proportional hazards models were used to determine the association between stress and 16-year mortality risk. RESULTS: Over a mean 12.9 years, 850 people died. Having a childhood home environment with very serious conflicts was associated with a 54% increased mortality risk (95%CI:1.21-1.96), and childhood abuse/maltreatment with a 34% increased risk (95% CI:1.05-1.70). For females, specific childhood events (serious illness HR:1.91, 95%CI:1.40-2.60; war/natural disaster HR:1.47, 95%CI:1.14-1.88) and the number of events (≥5 adverse events HR:1.91, 95%CI:1.25-2.32), also increased mortality risk. In terms of recent events, mortality risk increased by 66% (95%CI:1.39-2.00) in participants reporting a recent serious illness or physical trauma and by 86% for those reporting problems with the police/justice (95%CI:1.05-3.30). Among males specifically, mortality risk also increased with major financial problems (HR:1.92, 95%CI:1.14-3.21), and when they had a relative with a serious illness (HR:1.26, 95%CI:1.01-1.55). CONCLUSIONS: Stressful life experiences are associated with all-cause mortality however the associations varied between early-life adversities and recent stress, and were different across the genders. Among females, certain types of childhood adversity continue to predict mortality risk in later life, while in males specific recent stress significantly increased mortality risk.

ABSTRACT

This research aimed to assess whether rabbits having lop ears, an artificially selected conformation, compromises welfare. We investigated the occurrence of aural and dental pathology in lop-eared compared with erect-eared rabbits. Thirty rabbits (15 lop-eared and 15 erect-eared) from a rabbit-only rescue shelter were examined. An otoscope was used to visualise the ear canals and mouth. Samples were taken from each ear to examine for mites, bacteria and yeast. Medical records were also examined. Lop-eared rabbits showed statistically significantly more frequent ear canal stenosis, higher scores of cerumen and erythema and more frequent potential pain response during ear examination, compared with erect-eared rabbits. We also found statistically significantly more frequent incisor pathology, molar overgrowth, molar sharpness, molar spurs and history of veterinary dental treatment in lop-eared compared with erect-eared rabbits. The effect sizes were often large. Age was not statistically significant between the lop-eared and erect-eared rabbit groups. Thus, lop-eared rabbits were at an increased risk of aural and dental pathology in this study. This brings into debate the ethics of breeding and buying lop-eared rabbits, as they are more likely to suffer conditions that negatively impact welfare, such as pain, and potentially deafness and difficulty eating.

BACKGROUND: Taenia saginata cysticercus is the larval stage of the zoonotic parasite Taenia saginata, with a life-cycle involving both cattle and humans. The public health impact is considered low. The current surveillance system, based on post-mortem inspection of carcasses has low sensitivity and leads to considerable economic burden. Therefore, in the interests of public health and food production efficiency, this study aims to explore the potential of risk-based and cost-effective meat inspection activities for the detection and control of T. saginata cysticercus in low prevalence settings. METHODS: Building on the findings of a study on risk factors for T. saginata cysticercus infection in cattle in Great Britain, we simulated scenarios using a stochastic scenario tree model, where animals are allocated to different risk categories based on their age, sex and movement history. These animals underwent different types of meat inspection (alternative or current) depending on their risk category. Expert elicitation was conducted to assess feasibility of scenarios and provide data for economic analysis. The cost-effectiveness of these scenarios was calculated as an incremental cost-effectiveness ratio, using the number of infected carcasses detected as the technical outcome. RESULTS: Targeting the high-risk population with more incisions into the heart while abandoning incisions into the masseter muscles was found to reduce the total number of inspections and cost, while simultaneously increasing the number of infected carcasses found. CONCLUSIONS: The results suggest that, under reasonable assumptions regarding potential improvements to current inspection methods, a more efficient and sensitive meat inspection system could be used on animals categorised according to their risk of harbouring T. saginata cysticercus at slaughter. Such a system could reduce associated cost to the beef industry and lower microbial contamination of beef products, improving public health outcomes.

Cathepsins are powerful proteases, once referred to as the lysosomal cysteine proteases, that have been implicated in breast cancer invasion and metastasis, but pharmaceutical inhibitors have suffered failures in clinical trials due to adverse side effects. Scientific advancement from lysosomotropic to cell impermeable cathepsin inhibitors have improved efficacy in treating disease, but off-target effects have still been problematic, motivating a need to better understand cellular feedback and responses to treatment with cathepsin inhibitors. To address this need, we investigated effects of E-64 and cystatin C, two broad spectrum cathepsin inhibitors, on cathepsin levels intra- and extracellularly in MDA-MB-231 breast cancer cells. Cathepsins S and L had opposing responses to both E-64 and cystatin C inhibitor treatments with paradoxically elevated amounts of active cathepsin S, but decreased amounts of active cathepsin L, as determined by multiplex cathepsin zymography. This indicated cellular feedback to selectively sustain the amounts of active cathepsin S even in the presence of inhibitors with subnanomolar inhibitory constant values. These differences were identified in cellular locations of cathepsins L and S, trafficking for secretion, co-localization with endocytosed inhibitors, and longer protein turnover time for cathepsin S compared to cathepsin L. Together, this work demonstrates that previously underappreciated cellular compensation and compartmentalization mechanisms may sustain elevated amounts of some active cathepsins while diminishing others after inhibitor treatment. This can confound predictions based solely on inhibitor kinetics, and must be better understood to effectively deploy therapies and dosing strategies that target cathepsins to prevent cancer progression.
