ABSTRACT

BACKGROUND: Several studies have reported the potential prognostic significance of tumor volume reduction ratio (VRR) induced by radiotherapy (RT) in patients with non-small-cell lung cancer. However, there are no data yet on the prognostic significance of volumetric shrinkage in patients with small-cell lung cancer (SCLC). This study aimed to demonstrate the correlation between tumor volume reduction ratio and treatment outcomes. MATERIALS AND METHODS: The study included 61 patients with SCLC treated with fractionated RT of the primary tumor at our institution between 2013 and 2020. The relationship between volumetric changes in gross tumor volume (GTV) during radiotherapy and outcomes were analyzed and reported. RESULTS: The median radiation dose was 59.4 Gy (median fraction dose was 1.8 Gy). The median GTV before radiotherapy was 74 cm3, with a median GTV reduction of 48%. There was a higher VRR in patients receiving concurrent radiochemotherapy (p = 0.05). No volumetric parameters were identified as relevant predictors of outcome in the entire cohort. In multivariate analysis, only age had an impact on survival, while prophylactic whole-brain radiation influenced the progression-free survival significantly. CONCLUSION: Concurrent chemotherapy was associated with a higher VRR than sequential chemotherapy. No significant impact of VRR on patients' outcome or survival was detected.

ABSTRACT

OBJECTIVE: Several personality theories distinguish between rather genetically rooted, universal dispositional traits (DTs) and rather environmentally shaped, more contextualized characteristic adaptations (CAs). However, no study so far has compared different measures of theoretically postulated DTs and CAs regarding their environmental and genetic components while considering differences in measurement abstraction and reliability. This study aims to bridge this gap by testing the assumed differences in the sensitivity to environmental influences based on representative sets of DTs (Big Five and HEXACO domains and facets) and CAs (goals, interests, value priorities, religiousness, and self-schemas). METHOD: Using intra-class correlations and running extended twin family and spouses-of-twins model analyses, we analyzed a large data set (N = 1967) encompassing 636 twin pairs, 787 parent-offspring dyads, and 325 spouses/partners. RESULTS: Findings consistently support lower environmentality of DTs compared to CAs. On average, more than half of reliable variance in DTs was genetic, whereas the reverse was found for CAs. Larger environmental components in CAs were primarily attributable to larger individual-specific effects (beyond error of measurement) and factors shared by spouses. CONCLUSIONS: Findings are discussed against the background of the definitional distinction between DTs and CAs and the value of extended twin family data.

ABSTRACT

Comparing twins from same- and opposite-sex pairs can provide information on potential sex differences in a variety of outcomes, including socioeconomic-related outcomes such as educational attainment. It has been suggested that this design can be applied to examine the putative role of intrauterine exposure to testosterone for educational attainment, but the evidence is still disputed. Thus, we established an international database of twin data from 11 countries with 88,290 individual dizygotic twins born over 100 years and tested for differences between twins from same- and opposite-sex dizygotic pairs in educational attainment. Effect sizes with 95% confidence intervals (CI) were estimated by linear regression models after adjusting for birth year and twin study cohort. In contrast to the hypothesis, no difference was found in women (ß = -0.05 educational years, 95% CI -0.11, 0.02). However, men with a same-sex co-twin were slightly more educated than men having an opposite-sex co-twin (ß = 0.14 educational years, 95% CI 0.07, 0.21). No consistent differences in effect sizes were found between individual twin study cohorts representing Europe, the USA, and Australia or over the cohorts born during the 20th century, during which period the sex differences in education reversed favoring women in the latest birth cohorts. Further, no interaction was found with maternal or paternal education. Our results contradict the hypothesis that there would be differences in the intrauterine testosterone levels between same-sex and opposite-sex female twins affecting education. Our findings in men may point to social dynamics within same-sex twin pairs that may benefit men in their educational careers.

BACKGROUND/OBJECTIVES: Many personality traits correlate with BMI, but the existence and direction of causal links between them are unclear. If personality influences BMI, knowing this causal direction could inform weight management strategies. Knowing that BMI instead influences personality would contribute to a better understanding of the mechanisms of personality development and the possible psychological effects of weight change. We tested the existence and direction of causal links between BMI and personality. SUBJECTS/METHODS: We employed two genetically informed methods. In Mendelian randomization, allele scores were calculated to summarize genetic propensity for the personality traits neuroticism, worry, and depressive affect and used to predict BMI in an independent sample (N = 3 541). Similarly, an allele score for BMI was used to predict eating-specific and domain-general phenotypic personality scores (PPSs; aggregate scores of personality traits weighted by BMI). In a direction of causation (DoC) analysis, twin data from five countries (N = 5424) were used to assess the fit of four alternative models: PPSs influencing BMI, BMI influencing PPSs, reciprocal causation, and no causation. RESULTS: In Mendelian randomization, the allele score for BMI predicted domain-general (ß = 0.05; 95% CI: 0.02, 0.08; P = 0.003) and eating-specific PPS (ß = 0.06; 95% CI: 0.03, 0.09; P < 0.001). The allele score for worry also predicted BMI (ß = -0.05; 95% CI: -0.08, -0.02; P < 0.001), while those for neuroticism and depressive affect did not (P ≥ 0.459). In DoC, BMI similarly predicted domain-general (ß = 0.21; 95% CI:, 0.18, 0.24; P < 0.001) and eating-specific personality traits (ß = 0.19; 95% CI:, 0.16, 0.22; P < 0.001), suggesting causality from BMI to personality traits. In exploratory analyses, links between BMI and domain-general personality traits appeared reciprocal for higher-weight individuals (BMI > ~25). CONCLUSIONS: Although both genetic analyses suggested an influence of BMI on personality traits, it is not yet known if weight management interventions could influence personality. Personality traits may influence BMI in turn, but effects in this direction appeared weaker.

ABSTRACT

Behavior genetic research yielded that affiliation to a specific religion is primarily environmental, whereas religious beliefs and practices irrespective of a specific religion have been found to be heritable to some degree. This review synthesizes the literature and provides a meta-analytic overview on all identified behavior genetic studies on religiousness since 1999. This analysis allows new insight on the nature-nurture interplay in the development of religiousness: Genetic differences in religiousness and extra-familial environmental influences increased with age, whereas shared environmental influences within families declined - in particular in the first half of life. This age trend is in line with the interpretation of an increasing importance of active gene-environment transactions and accumulating extra-familial environmental factors across the lifespan.

ABSTRACT

Decades of research have shown that about half of individual differences in personality traits is heritable. Recent studies have reported that heritability is not fixed, but instead decreases across the life span. However, findings are inconsistent and it is yet unclear whether these trends are because of a waning importance of heritable tendencies, attributable to cumulative experiential influences with age, or because of nonlinear patterns suggesting Gene × Environment interplay. We combined four twin samples (N = 7,026) from Croatia, Finland, Germany, and the United Kingdom, and we examined age trends in genetic and environmental variance in the six HEXACO personality traits: Honesty-Humility, Emotionality, Extraversion, Agreeableness, Conscientiousness, and Openness. The cross-national sample ranges in age from 14 to 90 years, allowing analyses of linear and nonlinear age differences in genetic and environmental components of trait variance, after controlling for gender and national differences. The amount of genetic variance in Extraversion, Agreeableness, and Openness followed a reversed U-shaped pattern across age, showed a declining trend for Honesty-Humility and Conscientiousness, and was stable for Emotionality. For most traits, findings provided evidence for an increasing relative importance of life experiences contributing to personality differences across the life span. ABSTRACT

McCrae (2020) argues that it is premature to explore interventions focused on personality change. In his commentary, he suggests that interventions should be promoted only if their effects in self-report data are confirmed by the additional opinion of informants. We agree with the essence of his position and would go further by envisioning a new framework for rigorous collaborative research on personality change (Bleidorn et al., 2020). ABSTRACT

What basic personality traits characterize the psychologically healthy individual? The purpose of this article was to address this question by generating an expert-consensus model of the healthy person in the context of the 30 facets (and 5 domains) of the Revised NEO Personality Inventory (Costa & McCrae, 1992) system of traits. In a first set of studies, we found that the healthy personality can be described, with a high level of agreement, in terms of the 30 facets of the NEO-PI-R. High levels of openness to feelings, positive emotions, and straightforwardness, together with low levels on facets of neuroticism, were particularly indicative of healthy personality functioning. The expert-generated healthy personality profile was negatively correlated with profiles of pathological personality functioning and positively correlated with normative personality functioning. In a second set of studies, we matched the NEO-PI-R profiles of over 3,000 individuals from 7 different samples with the expert-generated healthy prototype to yield a healthy personality index. This index was characterized by good retest reliability and cross-rater agreement, high rank-order stability, and substantial heritability. Individuals with high scores on the healthy personality index were psychologically well-adjusted, had high self-esteem, good self-regulatory skills, an optimistic outlook on the world, and a clear and stable self-view. These individuals were low in aggression and meanness, unlikely to exploit others, and were relatively immune to stress and self-sufficient. ABSTRACT

Political orientation is often assumed to be shaped by socialization processes; however, previous studies have shown substantial genetic variance components in party affiliation, political attitudes and behaviors, or closely related personality traits. The majority of these studies have relied on the Classical Twin Design, which comes with restrictive assumptions, some of which are easily violated. Moreover, most analyses lack a perspective of age-group differences. In this study, we investigated political orientation in adolescents (age: 16-18) and young adults (age: 21-25) in a cross-sectional Nuclear Twin Family Design. We used data of the German TwinLife project, including data from same-sex twins reared together, their biological parents, and nontwin full siblings. We found genetic variation in political orientation, which was significant in the older cohort, possibly indicating an increasing importance of active gene-environment correlation from adolescence to adulthood. Individual differences in political orientation because of passive gene-environment correlation and shared environmental effects were larger in the younger cohort, substantiating the same theoretical consideration and the importance of shared socialization contexts for adolescents' political views. By running Nuclear Twin Family model analyses, and considering age-group differences, as well as the relationship of political orientation with the Big Five personality traits, our study extended previous work, and resulted in more robust and fine-grained estimates of genetic and environmental sources of variance in political orientation. Therefore, it contributed to a better understanding of the complex nature-nurture interplay that forms political orientation in emerging adulthood. ABSTRACT

Personality traits are powerful predictors of outcomes in the domains of education, work, relationships, health, and well-being. The recognized importance of personality traits has raised questions about their policy relevance, that is, their potential to inform policy actions designed to improve human welfare. Traditionally, the use of personality traits in applied settings has been predicated on their ability to predict valued outcomes, typically under the assumption that traits are functionally unchanging. This assumption, however, is both untrue and a limiting factor on using personality traits more widely in applied settings. In this article, we present the case that traits can serve both as relatively stable predictors of success and actionable targets for policy changes and interventions. Though trait change will likely prove a more difficult target than typical targets in applied interventions, it also may be a more fruitful one given the variety of life domains affected by personality traits. ABSTRACT

The Study of Personality Architecture and Dynamics (SPeADy) is a German research project that aims to investigate the sources of interindividual differences in intraindividual personality development. The main focus lies in the dynamic interplay between more stable core characteristics and more environmentally malleable surface characteristics, as well as between personality and life experiences over time. SPeADy includes a twin family study encompassing data from 1962 individuals (age: 14-94) of 682 families, including 570 complete twin pairs (plus 1 triplet set), 327 parents, 236 spouses and 145 children of twins. Data collection started in 2016 and data from the first wave are currently obtainable as open source. Available data comprise a broad range of personality variables, such as personality trait constructs, motives, interests, values, moral foundations, religiosity and self-related concepts. For the currently ongoing second wave of data collection, we added retrospective reports on major life events. Special features of this genetically informative study are the extended twin family data and its longitudinal design. Three assessment waves in 2 years' intervals are planned until 2022. In this article, we briefly describe the design and contents of the SPeADy twin family study as well as some recent findings, future plans and open science issues.

ABSTRACT

We investigate the link between genes, psychological traits, and political engagement using a new data set containing information on a large sample of young German twins. The TwinLife Study enables us to examine the predominant model of personality, the Big Five framework, as well as traits that fall outside the Big Five, such as cognitive ability, providing a more comprehensive understanding of the underpinnings of political engagement. Our results support previous work showing genetic overlap between some psychological traits and political engagement. More specifically, we find that cognitive ability and openness to experience are correlated with political engagement and that common genes can explain most of the relationship between these psychological traits and political engagement. Relationships between genes, psychological traits, and political engagement exist even at a fairly young age, which is an important finding given that previous work has relied heavily on older samples to study the link between genes, psychological traits, and political engagement.

ABSTRACT

Obesity has inconsistent associations with broad personality domains, possibly because the links pertain to only some facets of these domains. Collating published and unpublished studies (N = 14 848), we meta-analysed the associations between body mass index (BMI) and Five-Factor Model personality domains as well as 30 Five-Factor Model personality facets. At the domain level, BMI had a positive association with Neuroticism and a negative association with Conscientiousness domains. At the facet level, we found associations between BMI and 15 facets from all five personality domains, with only some Neuroticism and Conscientiousness facets among them. Certain personality-BMI associations were moderated by sample properties, such as proportions of women or participants with obesity; these moderation effects were replicated in the individual-level analysis. Finally, facet-based personality "risk" scores accounted for 2.3% of variance in BMI in a separate sample of individuals (N = 3569), 409% more than domain-based scores. Taken together, personality-BMI associations are facet specific, and delineating them may help to explain obesity-related behaviours and inform intervention designs. ABSTRACT

Mõttus and colleagues (2017) reported evidence that the unique variance in specific personality characteristics captured by single descriptive items often displayed trait-like properties of cross-rater agreement, rank-order stability, and heritability. They suggested that the personality hierarchy should be extended below facets to incorporate these specific characteristics, called personality nuances. The present study attempted to replicate these findings, employing data from 6,287 individuals from 6 countries (Australia, Canada, Czech Republic, Denmark, Japan, and United States). The same personality measure-240-item Revised NEO Personality Inventory-and statistical procedures were used. The present findings closely replicated the original results. When the original and current results were meta-analyzed, the unique variance of nearly all items (i.e., items' scores residualized for all broader personality traits) showed statistically significant cross-rater agreement (median = .12) and rank-order stability over an average of 12 years (median = .24), and the unique variance of the majority of items had a significant heritable component (median = .14). These 3 item properties were intercorrelated, suggesting that items systematically differed in the degree of reflecting valid unique variance. Also, associations of items' unique variance with age, gender, and body mass index (BMI) replicated across samples and tracked with the original findings. Moreover, associations between item residuals and BMI obtained from one group of people allowed for a significant incremental prediction of BMI in an independent sample. Overall, these findings reinforce the hypotheses that nuances constitute the building blocks of the personality trait hierarchy, their properties are robust and they can be useful. Despite the increasing acceptance of this paradigm, several aspects of the approach remain underdeveloped. Specifically, limitations arise from a reliance on a twins-only design, and all previous studies have used self-reports only. There are also questions about the extent to which existing findings generalize cross-culturally. To address those issues, this study examined individual differences in liberalism/conservatism in a German sample that included twins, their parents, and their spouses and incorporated both self- and peer reports. The self-report findings from this extended twin family design were largely consistent with previous research that used that rater perspective, but they provided higher estimates of heritability, shared parental environmental influences, assortative mating, and genotype-environment correlation than the results from peer reports. The implications of these findings for the measurement and understanding of sociopolitical attitudes are explored.

ABSTRACT

Political participation (POP), social participation (SOP), and political interest (PI) are important indicators of social status and social inequality. Previous studies on related trait differences yielded genetic and environmental contributions. However, focusing on adult samples, classical twin designs, and convenience samples often restricts parameter estimation and generalizability, and limits the understanding of age differences. We investigated sources of variance in POP, SOP, and PI in late adolescence and early adulthood with an extended twin family design (ETFD). We analyzed data from over 2,000 representative German twin families. Individual environments not shared by family members reflected the major source of variance for all variables, but genetic influences were also pronounced. Genetic effects were mostly higher for young adults, whereas effects of twins' shared environment were significant in adolescence. Our study deepens the understanding of the interplay between genetic and environmental factors in shaping differences in young persons' integration in society.

ABSTRACT

The present twin study combined self- and peer assessments of twins' general homophobia targeting gay men in order to replicate previous behavior genetic findings across different rater perspectives and to disentangle self-rater-specific variance from common variance in self- and peer-reported homophobia (i.e., rater-consistent variance). We hypothesized rater-consistent variance in homophobia to be attributable to genetic and nonshared environmental effects, and self-rater-specific variance to be partially accounted for by genetic influences. A sample of 869 twins and 1329 peer raters completed a seven item scale containing cognitive, affective, and discriminatory homophobic tendencies. After correction for age and sex differences, we found most of the genetic contributions (62%) and significant nonshared environmental contributions (16%) to individual differences in self-reports on homophobia to be also reflected in peer-reported homophobia. A significant genetic component, however, was self-report-specific (38%), suggesting that self-assessments alone produce inflated heritability estimates to some degree. Different explanations are discussed.

ABSTRACT

Whether monozygotic (MZ) and dizygotic (DZ) twins differ from each other in a variety of phenotypes is important for genetic twin modeling and for inferences made from twin studies in general. We analyzed whether there were differences in individual, maternal and paternal education between MZ and DZ twins in a large pooled dataset. Information was gathered on individual education for 218,362 adult twins from 27 twin cohorts (53% females; 39% MZ twins), and on maternal and paternal education for 147,315 and 143,056 twins respectively, from 28 twin cohorts (52% females; 38% MZ twins). Together, we had information on individual or parental education from 42 twin cohorts representing 19 countries. The original education classifications were transformed to education years and analyzed using linear regression models. Overall, MZ males had 0.26 (95% CI [0.21, 0.31]) years and MZ females 0.17 (95% CI [0.12, 0.21]) years longer education than DZ twins. The zygosity difference became smaller in more recent birth cohorts for both males and females. Parental education was somewhat longer for fathers of DZ twins in cohorts born in 1990-1999 (0.16 years, 95% CI [0.08, 0.25]) and 2000 or later (0.11 years, 95% CI [0.00, 0.22]), compared with fathers of MZ twins. The results show that the years of both individual and parental education are largely similar in MZ and DZ twins. We suggest that the socio-economic differences between MZ and DZ twins are so small that inferences based upon genetic modeling of twin data are not affected.

Background: Genes and the environment contribute to variation in adult body mass index [BMI (in kg/m2)], but factors modifying these variance components are poorly understood.Objective: We analyzed genetic and environmental variation in BMI between men and women from young adulthood to old age from the 1940s to the 2000s and between cultural-geographic regions representing high (North America and Australia), moderate (Europe), and low (East Asia) prevalence of obesity.Design: We used genetic structural equation modeling to analyze BMI in twins ≥20 y of age from 40 cohorts representing 20 countries (140,379 complete twin pairs).Results: The heritability of BMI decreased from 0.77 (95% CI: 0.77, 0.78) and 0.75 (95% CI: 0.74, 0.75) in men and women 20-29 y of age to 0.57 (95% CI: 0.54, 0.60) and 0.59 (95% CI: 0.53, 0.65) in men 70-79 y of age and women 80 y of age, respectively. The relative influence of unique environmental factors correspondingly increased. Differences in the sets of genes affecting BMI in men and women increased from 20-29 to 60-69 y of age. Mean BMI and variances in BMI increased from the 1940s to the 2000s and were greatest in North America and Australia, followed by Europe and East Asia. However, heritability estimates were largely similar over measurement years and between regions. There was no evidence of environmental factors shared by co-twins affecting BMI.Conclusions: The heritability of BMI decreased and differences in the sets of genes affecting BMI in men and women increased from young adulthood to old age. The heritability of BMI was largely similar between cultural-geographic regions and measurement years, despite large differences in mean BMI and variances in BMI. Our results show a strong influence of genetic factors on BMI, especially in early adulthood, regardless of the obesity level in the population.
