ABSTRACT

Despite the fact that pneumonia remains a leading cause of mortality and morbidity in pre-weaned calves, relatively little is known regarding the effects of the concurrent administration of intranasal pneumonia virus vaccines, particularly in calves with high levels of maternally derived antibodies. The objective of this study was to use a cohort of 40 dairy and dairy-beef female and male calves (27 females and 13 males) to determine serological responses to concurrent administration at 3 weeks of age (22 ± 4.85 days) of two commercially available intranasal (IN) vaccines for the viruses: bovine respiratory syncytial virus (BRSV), bovine herpes virus 1 (BoHV-1), and parainfluenza-3-virus (PI3-V). The study groups were as follows: (i) Bovilis IBR Marker Live only® (IO), (ii) Bovilis INtranasal RSP Live® only (RPO), (iii) Concurrent vaccination with Bovilis IBR Marker Live® & Bovilis Intranasal RSP Live® (CV), and (iv) a control group of non-vaccinated calves (CONT). The calves' serological response post-IN vaccination, clinical health scores, rectal temperatures, and weights were measured. Data were analyzed in SAS using mixed models and logistic regression. The CV calves had an average daily weight gain (ADG) of 0.74 (±0.02) kg, which was similar to CONT (0.77 ± 0.02 kg). Despite no significant differences in the antibody levels between study groups 3 weeks post-IN vaccination, following the administration of subsequent parenteral injections in the form of Bovilis Bovipast RSP®(antigens; inactivated BRSV, inactivated PI3-V, inactivated Mannheimia haemolytica) and Bovilis IBR Marker Live®, the antibody levels of the BRSV and PI3-V increased in both the CV and RPO study groups. Concurrent vaccination resulted in no increase in fever and no difference in health scores when compared to CONT.

ABSTRACT

Globally, the majority of dairy cows are milked twice a day (TAD); however, in pasture-based production systems, such as in Ireland, the idea of milking once a day (OAD) is being considered for reasons such as improved work-life balance. The immediate effects within a lactation, as well as the multilactation consequences of OAD, compared with TAD milking, require understanding. The objective of this randomized experiment was to compare OAD and TAD milking, over a 3-yr period, by examining the differences in milk production and composition, body weight (BW), body condition score (BCS), dry matter intake (DMI), udder characteristics, locomotion score, and milking time. Over the 3-yr period, 83 cows were enrolled in the experiment; 32, 44, and 48 cows in yr 1, 2, and 3 of the experiment, respectively. Each year, 23% of the herds were primiparous animals, while the remainder were second lactation or greater in parity. All cows were milked in the morning at 0700 h; only cows milked TAD were milked a second time each day at 1600 h. Cows rotationally grazed pastures for the duration of the lactating period and were housed during the nonlactating period. Milking cows OAD reduced cumulative milk yield by 26%, and milk solids yield (kg of fat + kg of protein) by 21%, across the 3 yr of the experiment when compared with cows milked TAD which produced 4,126 and 365 kg/cow, respectively. A contributory factor to the reduced production was a shorter lactation length (9.7 d) of the cows milked OAD compared with TAD (294 d). Milk fat percent of cows milked TAD was similar for all 3 yr of the study (5.05%), whereas milk fat percent of the cows milked OAD increased year on year, with each year being greater than the previous year (5.02%, 5.32%, and 5.70% for yr 1, 2, and 3; respectively). Milk protein percent was greater (+0.19%) for cows milked OAD compared with TAD which was 3.78%. Compared with cows milked TAD, total DMI for cows milked OAD was 22% less at the start of lactation (<167 d), but as the lactation progressed (>167 d) we observed no difference in DMI between treatments. Similar to the literature, milking cows OAD significantly increased average somatic cell score, both during (+16%) and at the end of lactation (+19%), compared with milking cows TAD which were 4.69 and 4.79, respectively. We detected positive aspects associated with OAD milking such as greater BW, BCS, and fertility performance. Milking OAD reduced both milking time per cow per day (reductions ranged from 34% in the first 4 mo of lactation to 43% during mo 5-9 of lactation) and milking time per liter of milk (-3.5 s/L) throughout lactation, leading to less labor inputs on-farm which can have positive implications for farmer work-life balance. The significant time saving and potential savings in costs (e.g., electricity) need to be considered in conjunction with the milk production reduction when considering OAD milking for the entire lactation.

ABSTRACT

The aim of this research paper was to evaluate the effect of a slow-release milk replacer on health and behaviour of neonatal dairy calves. This was done with the potential benefits to welfare during transport in mind. A total of 15 calves were randomly divided into three groups of 5, namely, a control group fed twice in 24 h with 3 l of a conventional milk replacer, a slow-release group fed once in 24 h with 2 l of conventional milk replacer and 1 litre of a specialised micro-encapsulated feed and an enriched-replacer group fed once in 24 h with 3 l of milk replacer enriched with micellar casein. Blood samples were taken before feeding and 6, 12, 18 and 24 h after and analysed for acid-base parameters, electrolytes, glucose, haemoglobin, cortisol, insulin, cholecystokinin and adiponectin. Calf behaviour was recorded between 6 and 14 h after feeding. There was a significant increase in blood pH 6 h after feeding in all groups, but the glucose, HCO3 - and base excess increased significantly in the slow-release group only, whereas sodium increased significantly in the enriched group only. Glucose levels remained significantly higher in the slow-release group, relative to the control, at 6, 12, and 18 h after feeding. Insulin levels changed significantly over time in the enriched and control group but remained constant in the slow-release group. Insulin levels were significantly higher in the control group when compared to the slow-release group after feeding. Adiponectin changed significantly over time after feeding in the control group only, but no significant changes were observed between the feeding groups. Behavioural patterns were similar in control and slow release groups but less favourable (less lying time, more vocalisations) in the enriched group. In conclusion, once-daily feeding of slow-release milk replacer demonstrated favourable patterns of blood variables related to satiety and hunger as well as behavioural patterns that did not differ from conventional twice-daily feeding.

ABSTRACT

Vitamin D deficiency at birth, followed by prolonged insufficiency in early life may predispose bovine calves to infection and disease. However, the effects of vitamin D levels on innate immunity are unclear due to the lack of long-term supplementation trials in vivo and reliable approaches for reproducibly assessing immune function. Here, a standardized whole blood immunophenotyping assay was used to compare innate immune responses to infection relevant ligands (LPS, Pam3CSK4 and R848) between Holstein-Friesian calves supplemented with vitamin D (n = 12) from birth until 7 months of age and control calves (n = 10) raised on an industry standard diet. Transcriptomic analysis in unstimulated whole blood cells revealed increased expression of type I interferons and chemokines in vitamin D supplemented calves, while IL-1 and inflammasome gene expression was decreased. In response to stimulation with the bacterial ligand LPS, supplemented calves had significantly increased expression of CASP1, CX3CR1, CAT, whereas STAT1 was decreased. Stimulation with the bacterial ligand Pam3CSK4 revealed increased expression of IL1A, IL1B and CAT genes; and decreased C5AR1 expression. In response to the viral ligand R848, STAT1 and S100A8 expression was significantly decreased. An increased IL-1 and inflammasome gene expression signature in vitamin D supplemented calves in response to LPS and Pam3CSK4 was also found, with ELISA confirming increased IL-1ß protein production. In contrast, a decreased chemokine gene expression signature was found in response to R848 in supplemented animals, with decreased IL-8 protein expression exhibited in response to all PAMPs also found. These results demonstrated expression of several cytokine, chemokine and inflammasome genes were impacted by vitamin D supplementation in the first 7 months of life, with IL-8 expression particularly responsive to vitamin D. Overall, vitamin D supplementation induced differential innate immune responses of blood immune cells that could have important implications for disease susceptibility in cattle.

ABSTRACT

It is unknown whether calf rearing facilities in the Republic of Ireland are fit for purpose, or if facilities sufficiently consider calf and farmer welfare. The aim of this study was to review current calf housing facilities and management practices on Irish farms to determine if calves are reared in structurally appropriate facilities with management decisions that safeguard calf and farmer welfare. Fifty-one farms located in the Munster region in the Republic of Ireland were visited twice: (1) Pre-calving (December-January) and (2) During peak calving (January-March). During visit one, herd owners completed a questionnaire regarding calf housing and management practices on-farm and each facility used to rear calves was measured (measurement of cubic air capacity, ventilation, pen area, drainage etc.) without calves being present. Visit two consisted of a short interview with the principal calf manager to validate previously asked questions and environmental based measurements of each calf house that had been recorded, with any deviation from the first visit noted (measurements of temperature, wind speed, light intensity, facility provisions in-house and in-pen; calves present). Average herd size was 254, operating a spring calving system with a median calving season length of 11.6 weeks. While most farms expanded (88%; N = 51), this did not appear to have negatively affected calf space allowances (9.9% houses overcrowded at a space allowance of 1.5 m2/calf; N = 121). Calves were most commonly housed in group sizes of <12 (71.6% of all groupings; N = 394), with farmers moving away from individual housing for a period immediately post-birth, to grouping them immediately instead (58.8%; N = 51). The number of farmers testing colostrum was 31.4% (N = 51). Although the calving season was compact, most farmers were unconcerned about the upcoming spring workload (58.8%; N = 51). Farms appeared sufficiently prepared for spring, with most using the same number or less sheds during visit two than declared in visit one (76.5%; N = 51). To conclude, farmers made sufficient provision for calf housing and space allowances for calves that facilitated group housing post-birth. While structural and management components of rearing systems appear in line with sectoral recommendations, certain areas require attention on many farms (e.g., colostrum testing) to safeguard calf welfare and reduce the workload associated with calf rearing for farmers.

ABSTRACT

Housing and feeding are integral to calf rearing, and must meet calf needs while remaining functional for the farmer. This study compared health, behavior, growth, and labor requirements of calves housed in groups indoors and fed via an automatic or manual milk feeding system compared to calves manually fed in individual or group hutches outdoors. Seventy-six (49 Holstein Friesian [HF] and 27 HF × Jersey) dairy heifer calves were balanced for birth weight (35.2 ± 4.95 kg), birth date (1 February ± 7.2 d) and breed. The experiment was a randomized block design with four treatments; 1) indoor group housing with automated feeding (IN_AUTO; 12 calves per pen), 2) indoor group housing with manual feeding (IN_MAN; 12 calves per pen), 3) outdoor group hutch with manual feeding (OUT_G_MAN; 8 calves per pen), and 4) outdoor individual hutch with manual feeding (OUT_I_MAN; 6 calves: 1 per pen). Calves in OUT_treatments moved outdoors at 18 d (± 5.9 d). Each treatment was replicated once. Milk allowance increased gradually from 6 to 8 L/day (15% reconstitution rate) with ad libitum fresh water, concentrates, and hay offered from 3 d old. Gradual weaning occurred at 8 wk old. Measurements were divided into period 1; before movement outdoors, and period 2; after movement outdoors. Health was similar among treatments, regardless of period, with the most frequent score being zero (i.e., healthy). Summarized, standing and lying were observed 24.3% and 29.8%, respectively, in OUT_I_MAN calves, compared to 8.0% and 49.1%, for the other systems, which were similar. No difference in bodyweight (BW) existed between treatments, except at weaning where BW was lower for OUT_I_MAN (67.4 ± 2.84 kg) compared to IN_MAN (74.2 ± 2.01 kg), and day 102 where OUT_I_MAN (94.1 ± 2.85 kg) were lighter than IN_AUTO (101.1 ± 2.10 kg) (P = 0.047). Total labor input was greatest for OUT_I_MAN (00:02:02 per calf per day; hh:mm:ss) and least for IN_AUTO (00:00:21 per calf per day) (P < 0.001). The labor for feeding (00:00:29 per calf per day), feeding inspection (00:00:10 per calf per day), and cleaning equipment (00:00:30 per calf per day) was greatest for OUT_I_MAN. All calves showed good health and growth patterns. Differences in behavior expressed by calves in the OUT_I_MAN, compared to other treatments may indicate compromised welfare. Thus, although outdoor group hutches do not negatively impact calves, indoor housing, particularly using automated feeders, can improve labor efficiency.


In seasonal calving dairy systems, cows calve in a period of approximately 12 wk. Demand for calf accommodation and labor is high during this time. Outdoor housing structures, such as robust plastic calf hutches, may offer an alternative to permanent indoor facilities. In this study, we compared indoor housing systems using automated and manual feeding methods and outdoor calf housing systems using manual feeding methods, to examine their effect on calf health, behavior, growth, and labor. Moving calves to their respective outdoor system commenced at approximately 18 d. This reflected a housing system with limited indoor availability, where older calves would be moved outdoors (allowing young calves to remain indoors). The most labor-efficient method of rearing was group housing calves indoors feeding via automatic feeder, followed by group housing indoors feeding via manual feeders, outdoor in group hutches, and outdoor in individual hutches with manual feeding. Calves in all systems showed health and growth patterns consistent with positive development. Calf behavior in the individual hutches outdoor may indicate compromised well-being, compared to all other systems. Thus, although outdoor group hutches do not negatively impact the calf, indoor housing, particularly when using automatic feeders, can provide improved labor efficiency.

ABSTRACT

To increase the dry matter and metabolisable energy intake of cows, dairy farmers often supplement pasture with concentrates and conserved fodder. Feeding large amounts of highly fermentable concentrates to cows can result in metabolic issues, such as ruminal acidosis, and thus safer but more efficient introduction strategies are desirable. We assessed the role that forages play in ruminal, behavioural and production responses to a wheat grain challenge in dairy cows with no previous wheat adaptation. Multiparous lactating Holstein dairy cows (n = 16) were fed a forage-only diet of either lucerne (Medicago sativa) hay, perennial ryegrass (Lolium perenne L.) hay or one of two cultivars of zero-grazing fresh perennial ryegrass herbage (Bealey or Base), for 3 weeks. The forage diet was then supplemented with crushed wheat grain at 8 kg dry matter/cow day-1, with no adaptation period. Wheat comprised between 32 and 43% of total dry matter intake. Cows fed hay maintained a higher mean ruminal fluid pH than those fed herbage, on both the forage-only diet (6.43 vs. 6.17) and the forage plus wheat diet (6.03 vs. 5.58). Following supplementation of wheat, cows fed herbage exhibited minimum ruminal fluid pH levels indicative of acute ruminal acidosis, at 5.15 and 5.06 for cultivars Bealey and Base, respectively. Furthermore, for both herbage cultivars, adding wheat resulted in a ruminal fluid pH under 6 for >20 h/day. The ruminal environment of cows fed lucerne hay remained most stable throughout the grain challenge, spending the least amount of time below pH 6.0 (9.0 h/day). Hay created a ruminal environment that was better able to cope with the accumulation of acid as wheat was digested. A combination of increased ruminating time and a slower rate of fermentation, due to higher neutral detergent fiber and lower metabolisable energy concentrations in the hays, is likely responsible for the higher ruminal fluid pH values. Forage plays a critical role in wheat introduction strategies; aggressive adaptation strategies could be implemented when a hay such as lucerne is used as the base forage.

ABSTRACT

An observational study was conducted to comparatively assess the efficacy of three different oral rehydration and buffering solutions, differentiated by their strong ion difference (SID) concentration, for treatment of neonatal calves with naturally acquired diarrhea. The SID concentrations tested were 100 mM, 170 mM and 230 mM for treatments SID100, SID170 and SID230, respectively. Clinical assessment and blood gas analysis were completed for 18 diarrheic calves once pre- and twice post- (6 and 24 hr after) oral administration with one of the three treatments. A repeated measure mixed model approach was used to analyze (a) the within-group efficacy of each treatment over time and (b) the between-group comparison at each timepoint. SID230 treatment resulted in a significant increase in blood pH, HCO3-, BE, SID and Na+ at 6 and 24 hr after treatment, and a significant decrease in AG and K+ by 24 hr after treatment. There were no significant changes in any of the blood gas parameters after treatment with SID100 and SID170. SID230 treatment also resulted in blood gas parameter changes that were significantly different to the other two groups. These results suggest that the optimum SID concentration for the treatment of calves with diarrhea is likely to be higher than current recommendations.

ABSTRACT

Cattle vary in their susceptibility to infection and immunopathology, but our ability to measure and longitudinally profile immune response variation is limited by the lack of standardized immune phenotyping assays for high-throughput analysis. Here we report longitudinal innate immune response profiles in cattle using a low-blood volume, whole blood stimulation system-the ImmunoChek (IChek) assay. By minimizing cell manipulation, our standardized system minimizes the potential for artefactual results and enables repeatable temporal comparative analysis in cattle. IChek successfully captured biological variation in innate cytokine (IL-1ß and IL-6) and chemokine (IL-8) responses to 24-hr stimulation with either Gram-negative (LPS), Gram-positive (PamCSK4) bacterial or viral (R848) pathogen-associated molecular patterns (PAMPs) across a 4-month time window. Significant and repeatable patterns of inter-individual variation in cytokine and chemokine responses, as well as consistent high innate immune responder individuals were identified at both baseline and induced levels. Correlation coefficients between immune response read-outs (IL-1ß, IL-6 and IL-8) varied according to PAMP. Strong significant positive correlations were observed between circulating monocytes and IL-6 levels for null and induced responses (0.49-0.61) and between neutrophils and cytokine responses to R848 (0.38-0.47). The standardized assay facilitates high-throughput bovine innate immune response profiling to identify phenotypes associated with disease susceptibility and responses to vaccination.

ABSTRACT

Global food production needs to increase in order to meet the demands of an ever growing global population. As resources are finite, the most feasible way to meet this demand is to minimize losses and improve efficiency. Regular monitoring of factors like animal health, soil and water quality for example, can ensure that the resources are being used to their maximum efficiency. Existing monitoring techniques however have limitations, such as portability, turnaround time and requirement for additional reagents. In this work, we explore the use of micro- and nano-scale electrode devices, for the development of an electrochemical sensing platform to digitalize a wide range of applications within the agri-food sector. With this platform, we demonstrate the direct electrochemical detection of pesticides, specifically clothianidin and imidacloprid, with detection limits of 0.22 ng/mL and 2.14 ng/mL respectively, and nitrates with a detection limit of 0.2 µM. In addition, interdigitated electrode structures also enable an in-situ pH control technique to mitigate pH as an interference and modify analyte response. This technique is applied to the analysis of monochloramine, a common water disinfectant. Concerning biosensing, the sensors are modified with bio-molecular probes for the detection of both bovine viral diarrhea virus species and antibodies, over a range of 1 ng/mL to 10 µg/mL. Finally, a portable analogue front end electronic reader is developed to allow portable sensing, with control and readout undertaken using a smart phone application. Finally, the sensor chip platform is integrated with these electronics to provide a fully functional end-to-end smart sensor system compatible with emerging Agri-Food digital decision support tools.

ABSTRACT

The different periods characterizing spring-calving, pasture-based dairy systems common in Ireland have seldom been the focus of large-scale dairy cow welfare research. Thus, the aim of this study was to devise and conduct an animal-based welfare assessment during both the grazing and housing periods on spring-calving, pasture-based dairy farms, to identify areas for improvement and establish benchmarks for indicators of good welfare. Assessment of seven animal-based welfare indicators was conducted during two visits (one each at grazing and housing) to 82 commercial dairy farms in southern Ireland. Herd-level descriptive statistics were performed for all welfare indicators at each visit, and differences between visits were analyzed using paired t-tests and Wilcoxon signed-rank tests. A mean of 9% and 10% clinically lame cows (mobility scores 2 and 3) were observed at housing and grazing, respectively. Recommended body condition scores (BCS) were not met for a mean of 13% of cows at grazing and 23% at housing, with more over-conditioned cows present at housing than grazing (P < 0.001). Ocular discharge was uncommon in both periods. Prevalence of moderate and severe nasal discharge combined was lower during housing (5%) than grazing (7%). In both periods, similar mean levels of tail injury were observed: 2% to 3% of cows with tail lacerations, 9% with broken tails, and 8% (measured at housing only) with docked tails. Integument alterations involved primarily hair loss and were most prevalent on the hindquarters (26%) during grazing and on the head-neck-back (66%) and the hindquarter (32%) regions during housing. Cows displayed an avoidance distance of >1 m (indicative of a fearful response) from an approaching human in an average of 82% of grazing cows and 42% to 75% of housed cows, dependent on test location. Opportunities to improve welfare in this system were identified in the areas of tail injury prevention, nasal health, and the management of indoor housing and feeding. The performance of the top 20% of farms for each welfare indicator was used to establish benchmarks of: 0% to 5% clinical lameness, 0% to 12% of cows outside recommended BCS, 0% to 27% ocular discharge, 2% to 16% nasal discharge, 0% tail lacerations and docked tails, 0% to 3% tail breaks, 0% to 14% integument alterations, and 4% to 74% for avoidance distance of >1 m. These represent attainable targets for spring-calving pasture-based farms to promote good dairy cow welfare.

ABSTRACT

Bovine antibodies, such as immunoglobulin G (IgG), cannot pass the placental barrier and as such are not transferred from the mother to the foetus, in utero. Instead a calf must absorb antibodies following ingestion of colostrum postpartum. Failure of Passive Transfer (FPT) is a condition that predisposes calves to development of disease and increases the risk of mortality. Thus, continuous early monitoring of IgG absorption in a calf, within the first 6 to 12 hours of life, is imperative to allow faster treatment and prevent FPT. In this paper, we present the development of a label-free impedimetric immunosensor device for bovine IgG in serum and demonstrate its suitability to determine early FPT in new-born calves. The developed sensors were challenged to discriminate between new born calf sera, both pre- and post-colostrum feeding, and demonstrated efficient detection of IgG in under 15 minutes. Such a device could enable rapid determination of FPT, thereby improving calves' vitality and survival rate.

ABSTRACT

Neonatal calf diarrhea (NCD) is a major problem to calf health worldwide, in terms of both morbidity and mortality. A five-point ordinal scale clinical assessment scoring (CAS) chart was utilized to assess calves suffering from NCD-related clinical abnormalities (acidosis and dehydration) on commercial farms. The objective of this research was to determine the predictive capability of this CAS chart against gold standard blood gas parameters, designed to assist farmers in the accurate assessment of the clinical consequences of NCD. A total of 443 diarrheic and non-diarrheic calves were enrolled in the study. The CAS chart rated a calf's health from no clinical signs to varying degrees of clinical severity on a 0 (clinically normal) to 4 (grave) scale, based on clinical indicators including calf demeanour, ear position, mobility, suckle reflex, desire-to-feed, and enophthalmos. Blood gas analysis was conducted for individual calves, consisting of pH, base excess, Na+, K+, Ca2+, Cl-, glucose, total hemoglobin, bicarbonate, anion gap, and strong ion difference. Statistical evaluation was performed by comparison of the CAS score with blood gas profiles using ordinal logistic regression and a non-parametric estimation of the Receiver Operating Characteristics (ROC). The ROC analysis indicated that the CAS chart had acceptable specificity (>95%) with low sensitivity (<60%) in differentiating clinically normal from acidotic/dehydrated cases. Assessment of individual severity classes indicated that the chart can predict and differentiate both clinically normal and advanced cases from the other severity classes (peak estimations >80%) but had reduced accuracy in differentiating mild and moderate cases (peak estimations >50%). The chart, as presented, provides a simple tool to differentiate clinically normal from calves suffering the consequences of diarrhea, but fails to accurately differentiate severity for NCD related acidosis and dehydration. Further efforts are required to enhance the sensitivity and differential diagnostic value of this type of chart.

ABSTRACT

Pooling colostrum is commonly practiced on Irish dairy farms. Pooling can result in dilution when colostrums with high and low IgG concentrations are mixed, thereby predisposing calves to failure of passive immunity. The objectives of this study were to compare IgG concentrations in colostrum from individual cows with colostrum pooled from several cows, and assess serum IgG concentrations, morbidity, and mortality among calves fed colostrum from their own dam, from a different cow, or pooled from several cows. We hypothesized that pooling colostrum reduces IgG concentration due to dilution compared with colostrum from individual cows, and that calves fed pooled colostrum achieve lower serum IgG concentrations than calves fed colostrum from individual cows. Calves were randomly assigned to 1 of 3 groups: (1) fed colostrum from their own dam (n = 20); (2) fed colostrum from a different dam (n = 20); or (3) fed pooled colostrum (n = 18). A sample of colostrum fed to each calf was collected. Serum samples were collected from calves at birth (0 h) and at 24 h after colostrum feeding. Colostrum and serum IgG concentrations were measured by radial immunodiffusion. Calves were weighed at birth and at weaning, and the health status of each calf was assessed twice daily. Health assessment was based on general demeanor, rectal temperature, fecal consistency, respiratory rate, and the presence of cough, nasal, or ocular discharge. Colostrum and serum IgG concentrations, and weaning weights were compared using ANOVA. Associations between group and morbidity or mortality rates were compared using χ2 or Fisher's exact tests. Median and 95% confidence intervals (95% CI) of IgG concentrations of colostrum were 99.4 (81.8-111.5), 95.2 (84.1-107.2), and 100.7 (90.5-104.4) g/L for own dam, different dam, and pooled groups, respectively. We did not find any differences in colostrum IgG concentrations among the colostrum sources. Median (95% CI) serum IgG concentrations at 24 h were 52.0 (45.6-65.9), 55.7 (51.2-65.9), and 53.1 (46.2-63.7) g/L for calves that received colostrum from own dam, different dam, and pooled, respectively. All calves achieved adequate passive immunity. Serum IgG concentrations at 24 h, weaning weights, and proportions of morbidity and mortality were not different among the 3 groups. Our results suggest that on dairy farms where median colostrum IgG concentrations are high and colostrum management is optimal, pooling has a minimal effect on passive immunity and subsequent calf health.

ABSTRACT

Sensor technologies that measure grazing and ruminating behaviour as well as physical activities of individual cows are intended to be included in precision pasture management. One of the advantages of sensor data is they can be analysed to support farmers in many decision-making processes. This article thus considers the performance of a set of RumiWatchSystem recorded variables in the prediction of insufficient herbage allowance for spring calving dairy cows. Several commonly used models in machine learning (ML) were applied to the binary classification problem, i.e., sufficient or insufficient herbage allowance, and the predictive performance was compared based on the classification evaluation metrics. Most of the ML models and generalised linear model (GLM) performed similarly in leave-out-one-animal (LOOA) approach to validation studies. However, cross validation (CV) studies, where a portion of features in the test and training data resulted from the same cows, revealed that support vector machine (SVM), random forest (RF) and extreme gradient boosting (XGBoost) performed relatively better than other candidate models. In general, these ML models attained 88% AUC (area under receiver operating characteristic curve) and around 80% sensitivity, specificity, accuracy, precision and F-score. This study further identified that number of rumination chews per day and grazing bites per minute were the most important predictors and examined the marginal effects of the variables on model prediction towards a decision support system.

Transition milk is a source of immunoglobulin G (IgG) and could potentially be used to provide calves with passive immunity, when the IgG concentration is ≥50 g/L. Assessment of IgG concentrations in transition milk would be required before feeding and could be conducted using cow-side tests such as refractometers. Currently, limited information is available on the ability of refractometers to assess transition milk quality. We hypothesized that digital refractometry could be used to provide an accurate cow-side assessment of IgG concentrations in colostrum and transition milk, and IgG concentration in colostrum and one or more transition milking in an Irish herd is >50 g/L. The objectives of this study were to determine the IgG concentrations in colostrum and first, second, third, fourth, and fifth transition milk, and determine the utility of a digital refractometer in assessing quality of colostrum and transition milk produced by cows in a pasture-based dairy production system. A convenient sample of 75 dairy cows were enrolled. Colostrum and transition milk IgG concentrations were determined by radial immunodiffusion and refractometry. Sensitivity and specificity of the refractometer were determined and cut-off points that maximized sensitivity and specificity were determined using receiver operating characteristic curves. Median (range) IgG concentrations in colostrum and first, second, third, fourth, and fifth milking were 99.6, 43.5, 12.5, 5.3, 1.9, and 1.8 g/L, respectively. The sensitivity (0.8-1) of digital refractometry in identifying samples with low IgG concentrations in colostrum, first, second, and third transition milk was acceptable. In contrast, digital refractometry was not useful for assessing IgG concentrations in the fourth and fifth milking due to low IgG concentrations.

ABSTRACT

An observational study was conducted on dairy calves (51 healthy, 31 with neonatal diarrhea) during outbreaks of diarrhea on 4 dairy farms. Clinical assessment scores (CAS) were assigned to each healthy and diarrheic calf [from 0 (healthy) to 4 (marked illness)]. Blood gas analysis [pH, base excess (BE), Na(+), K(+), Ca(2+), Cl(-), glucose, total hemoglobin, standard HCO3(-), strong ion difference (SID), and anion gap (AG)] was completed for each calf. Repeated measurements were taken in healthy animals, and pre- and postintervention measurements were taken for diarrheic calves. The mean CAS of diarrheic calves was 1.7, with 51, 30, 17, and 2% of calves scoring 1, 2, 3, and 4, respectively. The mean value for blood pH, BE, AG, and SID was 7.26, -4.93mM, 16.3mM, and 38.59mM, respectively. Calves were administered an oral rehydration and buffering solution (ORBS; Vitalife for Calves, Epsilion Ltd., Cork, Ireland) and reassessed. The mean CAS decreased to 0.38 (65% of calves scored 0 and 35% scored 1) at 6 to 18h posttreatment and to 0.03 (98% of calves scored 0 and 2% scored 1) within 24 to 48h. Significant increases in mean value for pH, BE, HCO3(-), Na(+), and SID, and significant decreases in AG, K(+), Ca(2+), and total hemoglobin were recorded posttreatment. The correlation estimates indicated that pH, HCO3(-), and BE were strongly correlated with CAS, with values exceeding 0.60 in all cases. Administration of an ORBS with a high SID and bicarbonate buffer demonstrated rapid recovery from a diarrheic episode in dairy calves.

BACKGROUND: Failure of passive transfer of maternal immunity via colostrum can occur in the bovine, and a number of blood tests have been developed to test calves for this failure. It is not clear which test is most suitable for this purpose. The objective was to examine the most commonly used tests for failure of passive transfer and to decide which is most suitable for routine laboratory use. 126 serum samples were taken from calves of dairy cows after birth but prior to colostrum feeding, and at 48 h of age. Five different tests were compared against radial immunodiffusion which is considered the appropriate reference method. These tests were serum gamma-glutamyltransferase levels, serum protein levels, serum globulin levels, an enzyme linked immunosorbent assay and the zinc sulphate turbidity test. RESULTS: The tests examined displayed high sensitivity but widely varying specificity. Examination of the use of different cut-off points allowed some improvement in specificity at the expense of sensitivity, but the tests which had performed best at the original cut-off points still displayed the best performance. Gamma-glutamyltransferase levels as a measure of colostrum absorption returned, in this study, the best balance between sensitivity and specificity. The ELISA used in this study and serum globulin levels displayed performance similar to the gamma-glutamyltransferase levels. Serum total protein was less successful than others examined at providing both sensitivity and specificity but may, when performed via refractometer, be useful for on-farm testing. As currently performed the poor sensitivity for which the zinc sulphate turbidity test is most often criticized is evident. Modification of the cut-off point to increase specificity is less successful at balancing these parameters than the ELISA, gamma-glutamyltransferase levels, and globulin levels. CONCLUSIONS: Gamma-glutamyltransferase levels, ELISA testing and circulating globulin levels performed best in detecting failure of passive transfer in serum samples, although all three had some practical considerations.

ABSTRACT

Calfhood diseases have a major impact on the economic viability of cattle operations. A three part review series has been developed focusing on calf health from birth to weaning. In this paper, the last of the three part series, we review disease prevention and management with particular reference to pneumonia, focusing primarily on the pre-weaned calf. Pneumonia in recently weaned suckler calves is also considered, where the key risk factors are related to the time of weaning. Weaning of the suckler calf is often combined with additional stressors including a change in nutrition, environmental change, transport and painful husbandry procedures (castration, dehorning). The reduction of the cumulative effects of these multiple stressors around the time of weaning together with vaccination programmes (preconditioning) can reduce subsequent morbidity and mortality in the feedlot. In most studies, calves housed individually and calves housed outdoors with shelter, are associated with decreased risk of disease. Even though it poses greater management challenges, successful group housing of calves is possible. Special emphasis should be given to equal age groups and to keeping groups stable once they are formed. The management of pneumonia in calves is reliant on a sound understanding of aetiology, relevant risk factors, and of effective approaches to diagnosis and treatment. Early signs of pneumonia include increased respiratory rate and fever, followed by depression. The single most important factor determining the success of therapy in calves with pneumonia is early onset of treatment, and subsequent adequate duration of treatment. The efficacy and economical viability of vaccination against respiratory disease in calves remains unclear.
