ABSTRACT

Seasonality of reproductive activity in rams and bucks is the major constraint in temperate and subtropical zones. Rapid alternation between 1 month of short days and 1 month of long days (LD) over three years in lightproof buildings eliminates this seasonality. We examined if this would also work in open barns, using only supplementary light. Over two years, one group of bucks (n = 7) was subjected to alternate 1 month of LD and 1 month of permanent light (LD-LL) and another group (n = 7) to alternate 1 month of LD and 1 month of natural light (LD-NL). A simultaneous control group, used for both experiments (CG1, n = 6; CG2, n = 6), remained under natural photoperiod. BW, testis weight (TW), plasma testosterone (T) and cortisol (C) were evaluated in all bucks. CG1 and CG2 bucks showed identical dramatic seasonal variations in BW (stable or decreasing in summer), TW (from 85 ± 12 g in February to 127 ± 7 g in July) and T (from 2.7 ± 1.2 ng/mL in January-April to 24.3 ± 3.2 ng/mL in June-October). By contrast, BW of LD-LL and LD-NL bucks increased regularly during the experiment. From 5 and 9 months after the experiment onset, LD-LL and LD-NL bucks, respectively, maintained constant TW of 115 ± 5 g until the experiment end. After the first 3 months <5 ng/mL, T of LD-LL bucks remained constant (5-10 ng/mL) until the experiment end. By contrast, T of LD-NL bucks showed four periods of low (<5 ng/mL) and two periods of high concentrations (18.1 ± 2.6 and 11.9 ± 3.4 ng/mL). Plasma C remained low (5-8 ng/mL) and did not change with group or light treatment. These results show for the first time in any seasonal photoperiodic species that it is possible to maintain the sexual activity of males all year round in open buildings using alternating periods of LD and LL. By contrast, return to NL instead of LL every other month does not prevent seasonality in T concentration. These results raise interesting questions about the photoperiodic control of neuroendocrine regulation of seasonal sexual activity and suggest that these treatments can be used to manage males in open barns in farms and in artificial insemination centres. ABSTRACT

The resting sites of tropical American mosquitoes are poorly documented, and the few reports that do exist are largely from opportunistic collections. Since blood-engorged females (used in determining host associations) are more efficiently collected from resting sites than attractive traps, information on resting site utilization has practical value. To investigate differences in the resting sites utilized by tropical mosquitoes, we collected and identified female mosquitoes from one man-made (resting shelter) and three natural (buttress tree roots, hollow trees, and understory vegetation) resting environments at a tropical dry forest location in western Costa Rica. All of the most common species collected demonstrated associations with one or more resting environments. Females of five species (blood-engorged Anopheles albimanus, Uranotaenia apicalis, Uranotaenia lowii, Uranotaenia orthodoxa, and blood-engorged Mansonia titillans) were collected in significantly greater numbers from understory vegetation than other resting environments. Culex erraticus and other members of the subgenus Melanoconion were encountered more often in resting shelters, hollow trees, and buttress roots, while Culex restrictor (blood-engorged) females were associated with hollow trees. Similarity indices indicate that buttress tree roots, hollow trees, and resting shelters are similar with respect to the mosquito communities that utilize them as resting sites, while understory vegetation has a resting fauna that is different than the other environments surveyed here. These results add to the body of information regarding resting sites utilized by tropical American mosquitoes.

The study aimed to determine the antimicrobial resistance patterns and to identify molecular resistance markers in Staphylococcus spp. (n=210) isolated from small ruminant mastitis in Brazil. The antimicrobial resistance patterns were evaluated by the disk diffusion test and by detection of the presence of mecA, blaZ, ermA, ermB, ermC and msrA genes by PCR. The efflux pump test was performed using ethidium bromide and biofilm production was determined by Congo red agar test along with PCR for detection of the icaD gene. The isolates were most resistant to amoxicillin (50.0%), streptomycin (42.8%), tetracycline (40.4%), lincomycin (39.0%) and erythromycin (33.8%). Pan-susceptibility to all tested drugs was observed in 71 (33.8%) isolates and 41 Staphylococcus isolates were positive for the efflux pump. Although phenotypic resistance to oxacillin was observed in 12.8% of the isolates, none harbored the mecA gene. However, 45.7% of the isolates harbored blaZ indicating that beta-lactamase production was the main mechanism associated with staphylococci resistance to beta-lactams in the present study. The other determinants of resistance to antimicrobial agents ermA, ermB, ermC, and msrA were observed in 1.4%, 10.4%, 16.2%, and 0.9% of the isolates, respectively. (AU)


O presente trabalho teve como objetivo determinar os padrões de resistência a agentes antimicrobianos e identificar marcadores moleculares de resistência em Staphylococcus spp. (n=210) isolados de mastite de pequenos ruminantes no Brasil. Os padrões de resistência a agentes antimicrobianos foram avaliados pelo teste de difusão em disco e pela detecção da presença dos genes mecA, blaZ, ermA, ermB, ermC e msrA via PCR. O teste da bomba de efluxo foi realizado utilizando brometo de etídio e a produção de biofime foi determinada pelo teste do vermelho congo em paralelo com o PCR para detecção do gene icaD. Os isolados foram mais resistentes a amoxicilina (50,0%), estreptomicina (42,8%), tetraciclina (40,4%), lincomicina (39,0%) e eritromicina (33,8%). Setenta e um (33,8%) isolados foram sensíveis a todas as drogas testadas e 41 foram positivos para a bomba de efluxo. Embora a resistência fenotípica a oxacilina tenha sido observada observada em 12,8% dos isolados, nenhum possuiu o gene mecA. Entretanto, 45,7% dos isolados continham a gene blaZ, indicando que a produção de beta-lactamases foi o principal mecanismo associado com a resistência dos Staphylococcus aos beta-lactâmicos. Os outros determinantes de resistência a agentes antimicrobianos ermA, ermb, ermC e msrA foram observados em 1,4%, 10,4%, 16,2% e 0,9% dos isolados respectivamente. (AU)

The data obtained in this study show the resistance of the isolates to beta-lactamics, which may be associated with the use of antimicrobial drugs without veterinary control.

A mastite é a inflamação da glândula mamária que acomete raças de aptidão leiteira como também aquelas voltadas para produção de carne. Esta enfermidade ocasiona sérias alterações na produção de leite e na sua qualidade, redução no ganho de peso e mortalidade de cordeiros. O presente estudo teve por objetivo conhecer os principais agentes causadores de mastite em ovinos e caprinos, bem como a sua susceptibilidade aos agentes antimicrobianos, além de avaliar o grau de concordância entre testes diagnósticos. Foram visitadas 25 propriedades durante a realização do experimento, sendo criatórios de caprinos, ovinos e rebanhos mistos, nos estados de Pernambuco e Bahia. Coletou-se leite de 439 caprinos e 76 ovinos. Foi realizada lactocultura, o California Mastitis Test (CMT) e o teste de sensibilidade aos antimicrobianos. Além disso, determinou-se o grau de concordância entre os testes diagnósticos empregados. Foi constatada uma maior freqüência de Staphylococcus spp. nos casos de mastite em caprinos e ovinos, sendo observado ainda, isolados de Streptococcus spp., Corynebacterium spp. e bacilos gram negativos (BGN). Os isolados apresentaram alta sensibilidade aos antimicrobianos testados, sendo o menor percentual de sensibilidade observado para o ácido nalidíxico. Em relação ao diagnóstico da mastite caprina, a análise comparativa entre o exame microbiológico e o CMT demonstrou um grau de concordância igual a K=0,17, enquanto que para a espécie ovina, este valor foi de K=0,22. A utilização do CMT para o diagnóstico da mastite subclínica em cabras e ovelhas deverá ser associado à técnica da lactocultura.(AU)

We visit 25 goat, sheep, and goat and sheep farms in Pernambuco and Bahia State, and a total of 439 goats and 76 sheep milk samples were collected. To diagnose of small ruminant mastitis were compared two tests: Milk culture and California Mastitis Test (CMT). The bacterial drug-resistance pattern was determined by Kirby Bauer test. Staphylococcus spp. was the most frequent bacteria isolated from goat and sheep mastitis cases. Streptococcus spp., Corynebacterium spp. and gram-negative bacilli were isolated. It was possible to observe the high sensitivity to antimicrobial drugs in all tested bacteria, being the lower sensitivity percentage determined to nalidixic acid. Considering caprine mastitis diagnostic the comparative analysis between microbiologic culture and shown a concordance degree of K=0,17, although to ovine species these value was K=0,22. ABSTRACT

Clostridium perfringens type D-producing epsilon toxin is a common cause of death in sheep and goats worldwide. Although anti-epsilon toxin serum antibodies have been detected in healthy non-vaccinated sheep, the information regarding naturally acquired antibodies in ruminants is scanty. The objective of the present report was to characterize the development of naturally acquired antibodies against C. perfringens epsilon toxin in goats. The levels of anti-epsilon toxin antibodies in blood serum of goat kids from two different herds were examined continuously for 14 months. Goats were not vaccinated against any clostridial disease and received heterologous colostrums from cows that were not vaccinated against any clostridial disease. During the survey one of these flocks suffered an unexpectedly severe C. perfringens type D enterotoxemia outbreak. The results showed that natural acquired antibodies against C. perfringens epsilon toxin can appear as early as 6 weeks in young goats and increase with the age without evidence of clinical disease. The enterotoxemia outbreak was coincident with a significant increase in the level of anti-epsilon toxin antibodies.

Foi avaliada a resposta sorológica de vacina experimental contra a enterotoxemia em quatro grupos de caprinos. O Grupo 1 recebeu colostro de vacas não vacinadas e nenhuma dose de vacina. Os Grupos 2, 3 e 4 receberam colostro de vacas vacinadas, e uma dose de vacina aos 80 dias de idade nos Grupos 3 e 4. O Grupo 4 recebeu a segunda dose de vacina aos 120 dias de idade. Os níveis de anticorpos séricos foram avaliados pelo ELISA nas vacas antes e depois do parto e nos caprinos aos 3, 80, 120 e 160 dias de idade. Não houve diferença significativa nos níveis de anticorpos séricos das vacas vacinadas e não vacinadas, assim como entre os quatro grupos de caprinos avaliados aos três dias de vida. Os Grupos 3 e 4 apresentaram títulos médios de anticorpos de 0,6 UI/mL e 1,1 UI/mL, respectivamente, aos 40 dias após a primovacinação. A resposta vacinal do grupo 4, 40 dias após o reforço, foi de 1,8 UI/mL, superior ao Grupo 3 que foi de 0,2 UI/mL. Portanto, no esquema proposto, o uso de colostro não induziu a imunização passiva dos cabritos. No entanto, a primovacinação e reforço 40 dias após desencadearam níveis de anticorpos considerados satisfatórios.(AU)


The serological response to an experimental vaccine against Clostridium perfringens type D enterotoxemia was evaluated in four groups of goats. Group 1 received colostrum from unvaccinated cows and no vaccine. Groups 2, 3 and 4 received colostrum from vaccinated cows. In addition, Groups 3 and 4 received a vaccine dose at 80 days of age, and Group 4 received a second vaccine dose at 120 days of age. Serum antibody levels were determined by ELISA in cows before and after calving, and in goats at 3, 80, 120 and 160 days of age. No significant difference in serum antibody levels was observed between vaccinated and unvaccinated cows, or between the four groups of goats evaluated at 3 days of life. Groups 3 and 4 presented mean antibody titers of 0.6 and 1.1 IU/ml, respectively, 40 days after first vaccination. The vaccine response of Group 4 was 1.8 IU/ml 40 days after the booster dose and was higher than that observed for Group 3 (0.2 IU/ml). Thus, in the proposed regimen the use of heterologous colostrum did not induce passive immunization in goat kids. However, first vaccination and a booster dose after 40 days triggered satisfactory antibody levels.(AU)

ABSTRACT

Foi avaliada a resposta sorológica de vacina experimental contra a enterotoxemia em quatro grupos de caprinos. O Grupo 1 recebeu colostro de vacas não vacinadas e nenhuma dose de vacina. Os Grupos 2, 3 e 4 receberam colostro de vacas vacinadas, e uma dose de vacina aos 80 dias de idade nos Grupos 3 e 4. O Grupo 4 recebeu a segunda dose de vacina aos 120 dias de idade. Os níveis de anticorpos séricos foram avaliados pelo ELISA nas vacas antes e depois do parto e nos caprinos aos 3, 80, 120 e 160 dias de idade. Não houve diferença significativa nos níveis de anticorpos séricos das vacas vacinadas e não vacinadas, assim como entre os quatro grupos de caprinos avaliados aos três dias de vida. Os Grupos 3 e 4 apresentaram títulos médios de anticorpos de 0,6 UI/mL e 1,1 UI/mL, respectivamente, aos 40 dias após a primovacinação. A resposta vacinal do grupo 4, 40 dias após o reforço, foi de 1,8 UI/mL, superior ao Grupo 3 que foi de 0,2 UI/mL. Portanto, no esquema proposto, o uso de colostro não induziu a imunização passiva dos cabritos. No entanto, a primovacinação e reforço 40 dias após desencadearam níveis de anticorpos considerados satisfatórios.
