ABSTRACT

Circular RNAs (ciRNAs) are emerging as new players in the regulation of gene expression. However, how ciRNAs are involved in neuropathic pain is poorly understood. Here, we identify the nervous-tissue-specific ciRNA-Fmn1 and report that changes in ciRNA-Fmn1 expression in spinal cord dorsal horn neurons play a key role in neuropathic pain after nerve injury. ciRNA-Fmn1 was significantly downregulated in ipsilateral dorsal horn neurons after peripheral nerve injury, at least in part because of a decrease in DNA helicase 9 (DHX9), which regulates production of ciRNA-Fmn1 by binding to DNA-tandem repeats. Blocking ciRNA-Fmn1 downregulation reversed nerve-injury-induced reductions in both the binding of ciRNA-Fmn1 to the ubiquitin ligase UBR5 and the level of ubiquitination of albumin (ALB), thereby abrogating the nerve-injury-induced increase of ALB expression in the dorsal horn and attenuating the associated pain hypersensitivities. Conversely, mimicking downregulation of ciRNA-Fmn1 in naïve mice reduced the UBR5-controlled ubiquitination of ALB, leading to increased expression of ALB in the dorsal horn and induction of neuropathic-pain-like behaviors in naïve mice. Thus, ciRNA-Fmn1 downregulation caused by changes in binding of DHX9 to DNA-tandem repeats contributes to the genesis of neuropathic pain by negatively modulating UBR5-controlled ALB expression in the dorsal horn.

ABSTRACT

OBJECTIVE: To observe the effect of Chinese herbal therapy on T-lymphocyte subsets in patients with IgA nephropathy (IgAN). METHODS: Totally 36 inpatients and outpatients at Department of Nephropathy, Xiyuan Hospital, China Academy of Chinese Medical Sciences, from June 2011 to June 2013 were recruited in the treatment group, while 20 volunteers were recruited as the healthy control group. Patients in the IgAN group only took Chinese herbal decoctions by syndrome typing for 3 months (except those accompanied with hypertension additionally took antihypertensive agents such as angiotensin-converting enzyme inhibitor and/or dihydropyridines calcium antagonist). No intervention was performed in the healthy control group. The values of Th1, Th2, and CD4+ CD25+ Treg, and red blood cell number in urine were detected using flow cytometry before and after treatment. 24 h urine protein was detected using inmmunoturbidimetry. RESULTS: Compared with the healthy control group, the CD4+ CD25+ Treg level obviously decreased in the IgAN group, showing statistical difference (P < 0.01). In the IgAN group, Th1, 24 h urine protein, and urine red blood cell counts were obviously lower after treatment, showing statistical difference when compared with before treatment (all P < 0.05). CONCLUSION: Chinese herbal therapy could reduce urine erythrocyte number and 24 h urine protein of IgAN patients, and down-regulating Th1 expression might be its mechanism.

ABSTRACT

Visible near infrared reflectance spectra in the range of 350 nm to 1700 nm were collected from 98 pork samples to develop online, rapid and nondestructive detection system for water content in fresh pork Median smoothing filter (M-filter), multiplication scatter correlation (MSC) and first derivative (FD) were used as compound preprocessing method to reduce noise present in the original spectrum. Seventy four samples were randomly selected to develop training model and remaining 24 samples were used to test the model. The optimal punishment parameters for the support vector machine (SVM) were determined by using cross--validation and grid--search in the training set. SVM prediction model was developed with the radial basis function (RBF) and the developed model was compared with the model developed by partial least squares regression (PLSR) method. SVM prediction model based on RBF had the correlation coefficient and root mean standard error of 0.96 and 0.32 respectively in the training set. The model obtained correlation coefficient of 0.87 and root mean square error of 0.67 in the test set. The result thus obtained demonstrates the applicability of SVM model for rapid, nondestructive detection of water content in pork.

ABSTRACT

To investigate the relationship between structure and function of the deferens ducts in the Chinese rat snake (Zaocys dhumnades), morphological changes within an annual cycle were observed by routine histological techniques. Also, the correlation of androgen receptor (AR), estrogen receptor (ER), progesterone receptor (PR) and aromatase (Ar) expressions in the vas deferens and testis were studied immunohistochemically. To confirm that the sperm and the spherical structure existed in deferens ducts, we also used routine histological technique observed deferens ducts in the Striped-tailed rat-snake (Elaphe taeniura), Red-banded snake (Dinodon rufozonatum), and Tiger-spotted neck-troughed snake (Rhabdophis tigrina lateralis). The results showed that the deferens ducts of the Chinese Rat Snake were composed of efferent duct, epididymal duct and vas deferens. Efferent duct contained sperm from August-October, and the sperm were observed in the epididymal duct from August-the following January. Throughout the year (except July) a large number of sperm were present in the vas deferens where a previously unreported spherical structure formed by spermatids was observed, which showed no significant differences in the IOD values of AR-, ER-, PR- and Ar-immunoreactivities. Since the spermatids in the spherical structure were undergoing spermatogenesis and this phenomenon also existed in the Striped-tailed rat-snake and Red-banded snake, the term, seminiferous spherule, was named for this spherical structure This study demonstrated that the testis was the main site for snake spermiogenesis, and the seminiferous spherule in vas deferens was the other Both the epididymis and vas deferens stored sperm; however, the vas deferens was the main organ for sperm storage.
