ABSTRACT

OBJECTIVE: To characterize the clinical phenotype, genetic origin, and muscle pathology of patients with the FKRP c.1387A>G mutation. METHODS: Standardized clinical data were collected for all patients known to the authors with c.1387A>G mutations in FKRP. Muscle biopsies were reviewed and used for histopathology, immunostaining, Western blotting, and DNA extraction. Genetic analysis was performed on extracted DNA. RESULTS: We report the clinical phenotypes of 6 patients homozygous for the c.1387A>G mutation in FKRP. Onset of symptoms was <2 years, and 5 of the 6 patients never learned to walk. Brain MRIs were normal. Cognition was normal to mildly impaired. Microarray analysis of 5 homozygous FKRP c.1387A>G patients revealed a 500-kb region of shared homozygosity at 19q13.32, including FKRP. All 4 muscle biopsies available for review showed end-stage dystrophic pathology, near absence of glycosylated α-dystroglycan (α-DG) by immunofluorescence, and reduced molecular weight of α-DG compared with controls and patients with homozygous FKRP c.826C>A limb-girdle muscular dystrophy. CONCLUSIONS: The clinical features and muscle pathology in these newly reported patients homozygous for FKRP c.1387A>G confirm that this mutation causes congenital muscular dystrophy. The clinical severity might be explained by the greater reduction in α-DG glycosylation compared with that seen with the c.826C>A mutation. The shared region of homozygosity at 19q13.32 indicates that FKRP c.1387A>G is a founder mutation with an estimated age of 60 generations (∼1,200-1,500 years).

No período de agosto de 2000 a julho de 2005 foram atendidos 43 casos de Pênfigo Foliáceo (PF) canino no Serviço de Dermatologia do Hospital Veterinária, Universidade de São Paulo. Com este estudo retrospectivo visou-se atualizar dados referentes à caracterização sexual, definição racial e raça, idade, tipo e topografia lesional, quadro sintomático e resposta aos tratamentos isolados com prednisona e com a associação desta à azatioprina, além de demonstrar o aumento na ocorrência do PF relativamente à série histórica pretérita (1986-2000) do mesmo Serviço.(AU)


From August 2000 to July 2005 were attended 43 cases of canine Pemphigus foliaceous (PF) by the Dermatology Service of the Veterinary Teaching Hospital, University of São Paulo. The aim of the present study was to update the records referred to sex, breed, age, type and location of the lesions, clinical signs, and response to treatments with prednisone or combination with prednisone and azathioprine, and also to demonstrate the increase of occurrence of PF compared with the former series (1986-2000) observed in the same Service.(AU)

ABSTRACT

Nos meses de janeiro a março de 1983 e de 1984 ocorreram sucessivamente duas epidemias de conjuntivite hemorrágica aguda (C.H.A.) no Estado de Säo Paulo, a primeira atingindo exclusivamente a área litorânea e a segunda, mais extensa, abrangendo também regiöes do planalto. A presente investigaçäo apresenta dados sobre aspectos etiológicos da epidemia de C.A.H. ocorrida em 1984. Foram examinadas amostras pareadas de soro de 73 pacientes para a pesquisa de anticorpos e amostras da secreçäo conjuntival de 179 casos com C.H.A., para a tentativa de isolamento de vírus provenientes de diferentes regiöes do Estado de Säo Paulo. Houve conversäo sorológica em 45,2% dos 73 pacientes examinados e presença de anticorpos específicos para EV-70 sem conversäo sorológica em outros 19,2%. As proporçöes mais elevadas de casos positivos situaram-se entre pacientes maiores de 20 anos, com taxas de 70,6% para o grupo etário de 20 a 29 anos, de 55,6% na faixa de 30 a 39 anos e de 72,7% em maiores de 40 anos. Foram isoladas de amostras de secreçäo conjuntival 7 cepas de adenovírus, 1 de herpes simples e 1 de EV-70. Nenhum dos soros pareados examinados foi positivo para adenovirus e, nos casos em que esses vírus foram isolados, näo foi possível obter soros pareados para a comprovaçäo sorológica de infecçäo. Os resultados obtidos confirmam a participaçäo de EV-70 nessa epidemia. Näo foram identificadas complicaçöes neurológicas associadas à infecçäo pelo EV-70
