ABSTRACT

This study aimed to evaluate the effects of replacing soyhulls (SH) for ground Rhodes grass hay (RGH) in a pelleted diet on ingestive behavior, digestibility, blood metabolites, growth performance, and economic viability of fattening Lohi lambs. A total of thirty male lambs (age: 5 months; body weight 20.4 ± 0.24 kg) were allotted to one of the three diets (n = 10 lambs/diet) under a completely randomized design. The diets were with 25% RGH inclusion (control), replacing 15% RGH with 15% SH as a fiber source (SH-15), and containing only 25% SH inclusion on a dry basis (SH-25). Ingestive behavior parameters like time spent (min/day), bouts frequency (number/day), and bout length (min/bout) for feeding, drinking, rumination, chewing, standing, and lying were not influenced (P > 0.05) by replacing RGH with SH. The dry matter (DM) and neutral detergent fiber (NDF) chewing rate, rumination rate, and feeding efficiency were also not changed (P > 0.05) by dietary treatments, whereas total dry matter and NDF intakes, and their rumination efficiencies were lower (P < 0.05) for the SH-25 than for the rest of the treatments. The digestibility of NDF and acid detergent fiber (ADF) was higher (P < 0.05) for the SH-25 than for the rest of the treatments. The concentrations of blood metabolites like glucose, blood nitrogen, and cholesterol were not changed (P > 0.05) among the treatment groups. Similarly, performance parameters like dry matter intake, initial and final body weight, average daily gain, and feed conversion ratio were similar (P > 0.05) across the treatments. However, the incidence of loose fecal consistency was higher (P < 0.05) for SH-25 than for the control. The values of economic efficiency were better for SH-25-fed lambs than those fed the rest of the treatments. Based on the results, substituting SH for RGH in a pelleted diet improved the digestibility of fiber fractions, and economics without affecting the growth performance, and blood metabolites of fattening lambs. However, lower rumination efficiency and loos fecal consistency are evidencing less effectiveness of SH fiber.

ABSTRACT

This study aimed to evaluate the influence of different physical forms of complete diets on performance, feeding behaviour, digestibility, ruminal health, blood and carcass indices in fattening lambs. A randomised complete block design was used to assign thirty male Lohi lambs (300 ± 15 d old) with an initial body weight of 33 ± 1.4 kg in ten replications to one of three physical forms of the diet. For different treatments, the dietary ingredients were ground and mixed as (I) ground conventional mash (CM), (II) whole corn grains were mixed with the remaining pelleted ingredients as a texturised diet (TX), and (III) whole corn grains and the remaining ingredients were mixed as an unprocessed diet (UP). During the 60-d growth trial and 7-d digestibility experiment, individually housed lambs were fed ad libitum. Feeding diet UP improved (p < 0.05) dry matter intake, average daily gain and feed-to-gain ratio of fattening lambs. The ruminal pH tended to be lower in group TX compared with the other groups. The incidence of loose faeces consistency was 3.5 times higher (p < 0.05) in group TX compared to group UP. The daily intakes of dry matter (DM) and neutral detergent fibre (NDF), the rumination time and chewing activities were highest (p < 0.05) for lambs fed on the UP diet. The digestibility of DM, NDF and ether extract were greater (p < 0.05) for diet UP as compared to diet TX. The chilled and hot carcass weights were highest (p < 0.05) for group UP. The papillae density tended to be greater for group UP. However, blood metabolites, intestinal morphology, carcass marbling, tenderness, meat pH, cooking loss, and meat composition were similar across the treatments. It can be concluded that the unprocessed diet based on whole corn grain and soybean hulls improved growth performance, feeding behaviour and carcass yield through better nutrient utilisation and a stable ruminal environment.

ABSTRACT

The objectives of the experiment were to determine the effect of two doses of equine chorionic gonadotropin (eCG) in a standard synchronization protocol based on a short-term progesterone (P4 ) priming on ovarian structures and haemodynamics, concentrations of steroid hormones and prolificacy rate when oestrus was induced during low-breeding season (LBS) in Beetal dairy goats. We hypothesized that inclusion of eCG in a short-term P4 priming-based synchronization protocol would increase the blood perfusion to ovarian structures leading to enhance oestrous and ovulatory responses and prolificacy rate in goats. Forty-two multiparous acyclic goats were blocked by body condition and, within block, assigned randomly to receive saline as control (CON), low eCG (L-eCG; 300 IU) or high eCG (H-eCG; 600 IU) dose. Initially, a controlled internal drug release (CIDR) device was placed in the anterior vagina on d -8, followed by removal of CIDR on d -3, concurrent with the administration of PGF2α and eCG according to their respective treatments. Goats were monitored for oestrous response. B-mode and Doppler ultrasonography was performed with 12-h interval, starting from day -3 until natural breeding (day 0), and then on days 5, 10, 15 and 20 post-breeding to monitor follicular and luteal dynamics and blood flow, respectively. Blood was sampled at 0, 12, 24, 36 and 60 h after CIDR removal to quantify plasma concentrations of estradiol-17ß (E2 ), whereas plasma concentrations of P4 were assayed at days 5, 10, 15 and 20 after breeding. Pregnancy and prolificacy rates were determined at day 30 and 150 after breeding, respectively. Data were analysed with mixed-effects models, and orthogonal contrasts were used to evaluate the effect of treatment [Con vs. (½ L-eCG + ½ H-eCG)] and dose of eCG (L-eCG vs. H-eCG). Data are presented in sequence as CON, L-eCG, H-eCG (LSM ± SEM). The oestrous intensity score (152.9 vs. 182.7 vs. 186.5 ± 15.1; p = .02) was greater in eCG-treated goats as compared to CON. Administration of eCG reduced the intervals to standing oestrus (66.2 vs. 41.8 vs. 48.9 h ± 5.5; p = .05), breeding (70.2 vs. 44.4 vs. 45.4 h ± 4.5; p = .03) and ovulation (84.5 vs. 61.2 vs. 63.4 h ± 6.2; p = .05) compared with CON goats. The mean growth rate of pre-ovulatory follicle was greater (1.11 vs. 1.49 vs. 1.45 mm ± 0.08; p = .01) in eCG-treated goats resulting in an increased diameter of pre-ovulatory follicle (6.27 vs. 7.20 vs. 7.31 mm ± 0.07; p < .01) and corpora lutea (6.75 vs. 8.26 vs. 8.07 mm ± 0.42; p = .04) than CON. The mean follicular blood flow did not differ among treatments; however, the mean luteal blood flow was greater in L-eCG-treated goats (0.81 vs. 1.61 vs. 1.07 cm2 ± 0.12; p = .001). The mean concentrations of E2 (4.03 vs. 5.21 vs. 4.78 pg/ml ± 0.42; p = .04) and P4 (4.85 vs. 6.39 vs. 6.22 ng/ml ± 0.34; p = .04) were greater in eCG-treated goats. The twinning rate did not differ between treatments; nevertheless, prolificacy rate was greater (p = .04) in L-eCG-treated goats. Collectively, our data suggest that the administration of eCG improves the induction of oestrous and ovarian dynamics. Administration of L-eCG enhances prolificacy rate, therefore, a low dose of eCG might be practically beneficial to improve reproduction during LBS in acyclic Beetal dairy goats.

ABSTRACT

An experiment was executed to determine the effects of replacing wheat straw with corn stover on growth performance, behavioral characteristics, blood metabolites, and nutrient digestibility in Beetal bucks (23.92 ± 0.79 kg; age = 10 ± 1 month). A total of twenty four Beetal bucks were assigned randomly to one of three treatment groups, having eight animals each, for 15-week experimental period excluding adaption period of 2 weeks. The dietary treatments included conventional (25% wheat straw and 75% concentrate), corn stover 50 (50% of wheat straw (12.5%) was replaced with corn stover), and corn stover 100 (wheat straw was completely replaced with corn stover). Parameters were evaluated regarding growth performance, behavioral recording, digestibility, chemical analysis of feed and fecal materials, rumen pH, fecal score, and blood metabolite measurements. Dietary replacement of wheat straw with corn stover has resulted in an increased dry matter intake and average daily gain significantly (P ≤ 0.05) by 10 and 26%, respectively in the bucks. Rumen pH and fecal score, however, remained unaffected by dietary replacement of wheat straw with corn stover in the bucks. The replacement of wheat straw with corn stover has resulted in an increased feeding and rumination time, improved lying time and length, and decreased number of bouts in the bucks. Blood glucose, urea, bilirubin, and calcium levels were remained unaffected by replacing wheat straw with corn stover. The blood phosphorous level, however, was lower in bucks fed corn stover-based ration. The bucks fed corn stover-based ration has resulted in an increased digestibility of organic matter, crude protein, and neutral and acid detergent fiber compared to those fed wheat straw-based total mixed ration. It was concluded that replacement of wheat straw with corn stover as fiber residue in the ration of bucks has resulted in an increased dry matter intake and higher average daily gain, improved behavioral characteristics, and higher nutrient digestibility.

BACKGROUND: Thermophilic fungi have recently emerged as a promising source of thermostable enzymes. Superoxide dismutases are key antioxidant metalloenzymes with promising therapeutic effects in various diseases, both acute and chronic. However, structural heterogeneity and low thermostability limit their therapeutic efficacy. OBJECTIVE: Although several studies from hypethermophilic superoxide dismutases (SODs) have been reported, information about Cu,Zn-SODs from thermophilic fungi is scarce. Chaetomium thermophilum is a thermophilic fungus that could provide proteins with thermophilic properties. METHODS: The enzyme was expressed in Pichia pastoris cells and crystallized using the vapor-diffusion method. X-ray data were collected, and the structure was determined and refined to 1.56 Å resolution. Structural analysis and comparisons were carried out. RESULTS: The presence of 8 molecules (A through H) in the asymmetric unit resulted in four different interfaces. Molecules A and F form the typical homodimer which is also found in other Cu,Zn- SODs. Zinc was present in all subunits of the structure while copper was found in only four subunits with reduced occupancy (C, D, E and F). CONCLUSION: The ability of the enzyme to form oligomers and the elevated Thr:Ser ratio may be contributing factors to its thermal stability. Two hydrophobic residues that participate in interface formation and are not present in other CuZn-SODs may play a role in the formation of new interfaces and the oligomerization process. The CtSOD crystal structure reported here is the first Cu,Zn-SOD structure from a thermophilic fungus.

ABSTRACT

The objectives of the study were to determine the dose-dependent effects of active immunization against inhibin α-subunit (AIINHA) on ovarian dynamics, concentrations of progesterone (P4), pregnancy rate (PR), embryonic and fetal losses (EFL), and prolificacy during the non-breeding season when there was imposing of a progestin-based treatment regimen to induce estrus in Beetal goats. Goats (n = 30) were randomly assigned into following groups: 1) saline (G-CON-0 mg; n = 10), 2) small dose (G-AIINHA-0.5 mg; n = 10), and 3) large dose (G-AIINHA-1 mg; n = 10). The primary administration of inhibin immunogen was administered at Day -48, followed by another administration at Day -20, and subsequently there was induction of estrus using a progestin based treatment regimen that included a single administration of progestin-containing sponge and PGF2α at Day -8. The sponge was removed, and GnRH was administered at Day -3 followed by breeding (Day 0) at standing estrus. Results indicated mean diameter of the follicles, size of pre-ovulatory follicles and corpora lutea, and post-breeding P4 concentrations were greater (P < 0.05) in the goat does of the G-AIINHA-0.5 than G-CON-0 group. The PR, and EFL, however, did not differ (P> 0.05) among groups, whereas prolificacy rate was greater (P = 0.04) in goat does of the G-AIINHA-0.5 than G-CON-0 groups. The data from this study indicate G-AIINHA-0.5 is the recommended dose of inhibin immunogen to enhance the reproductive performance during non-breeding season in Beetal goats when estrus is induced using a progestin-based treatment regimen.

ABSTRACT

Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2), the causative agent of the Coronavirus disease (COVID-19) pandemic, has so far resulted in more than 1.1 M deaths and 40 M cases worldwide with no confirmed remedy yet available. Since the first outbreak in Wuhan, China in December 2019, researchers across the globe have been in a race to develop therapies and vaccines against the disease. SARS-CoV-2, similar to other previously identified Coronaviridae family members, encodes several structural proteins, such as spike, envelope, membrane, and nucleocapsid, that are responsible for host penetration, binding, recycling, and pathogenesis. Structural biology has been a key player in understanding the viral infection mechanism and in developing intervention strategies against the new coronavirus. The spike glycoprotein has drawn considerable attention as a means to block viral entry owing to its interactions with the human angiotensin-converting enzyme 2 (ACE2), which acts as a receptor. Here, we review the current knowledge of SARS-CoV-2 and its interactions with ACE2 and antibodies. Structural information of SARS-CoV-2 spike glycoprotein and its complexes with ACE2 and antibodies can provide key input for the development of therapies and vaccines against the new coronavirus.

ABSTRACT

The current study was conducted to determine the optimal timing of artificial insemination and vaginal mucous characteristics relative to the onset of estrus in goats. Does (n = 257) were estrous synchronized using hormonal treatments. Intracervical inseminations with fresh semen were performed at 0, 12, 24 and 36 h after the onset of estrus. Characteristics of vaginal mucus (i.e., color, consistency, and volume) were observed and graded at the time of AI to calculate cumulative mucous score. The vaginal electrical resistance (VER) was recorded (Draminski® detector) at the time of AI. Pregnancy rate was less (P < 0.05) when inseminations occurred at 0 h (28.6%) compared with 12 (58.3%) and 24 h (56.4%) after the onset of estrus; however, pregnancy rate at 36 h (54.5%) did not differ (P> 0.05) compared with inseminations at 0, 12 or 24 h after estrous onset. Relative odds for pregnancy rate at 12, 24 and 36 h were 5.24, 5.20 and 3.29 times greater compared with 0 h. Cumulative mucous score varied (P < 0.05) relative to the onset of estrus and correlated well (P < 0.05) with the color and consistency than volume of the mucus. The VER was less (P < 0.05) at 12 than 0 and 36 h after estrous onset. In conclusion, goats can be inseminated between 12-36 h after the onset of estrus; however, the chances of pregnancy are greater with inseminations at the 12 or 24 h time periods.

ABSTRACT

Beta-glucosidases (ß-glucosidases) have attracted considerable attention in recent years for use in various biotechnological applications. They are also essential enzymes for lignocellulose degradation in biofuel production. However, cost-effective biomass conversion requires the use of highly efficient enzymes. Thus, the search for new enzymes as better alternatives of the currently available enzyme preparations is highly important. Thermophilic fungi are nowadays considered as a promising source of enzymes with improved stability. Here, the crystal structure of a family GH3 ß-glucosidase from the thermophilic fungus Chaetomium thermophilum (CtBGL) was determined at a resolution of 2.99 Å. The structure showed the three-domain architecture found in other ß-glucosidases with variations in loops and linker regions. The active site catalytic residues in CtBGL were identified as Asp287 (nucleophile) and Glu517 (acid/base). Structural comparison of CtBGL with Protein Data Bank (PDB)-deposited structures revealed variations among glycosylated Asn residues. The enzyme displayed moderate glycosylation compared to other GH3 family ß-glucosidases with similar structure. A new glycosylation site at position Asn504 was identified in CtBGL. Moreover, comparison with respect to several thermostability parameters suggested that glycosylation and charged residues involved in electrostatic interactions may contribute to the stability of the enzyme at elevated temperatures. The reported CtBGL structure provides additional insights into the family GH3 enzymes and could offer new ideas for further improvements in ß-glucosidases for more efficient use in biotechnological applications regarding cellulose degradation.

ABSTRACT

This study compares the factors associated with variable interval to oestrus and ovulation between early versus late ovulating goats following PGF2α administration. The time of ovulation in Beetal goats (n = 38) was monitored through transrectal ultrasound at every 6 hr following a single dose of PGF2α (experiment 1). Variations in oestrus and ovulation times were further explored through the changes in follicular dynamics, endocrine profiles and behaviour in another set of goats (n = 13) following single PGF2α given randomly during the luteal phase (experiment 2). The ovulation time varied between 60 and 96 hr, and 57% of ovulations occurred by 72 hr following PGF2α (experiment 1). Accordingly, the goats (n = 13) in the second experiment were retrospectively divided either into early and/or late ovulating, that is, ≤72 and/or ≥84 hr following PGF2α . The onset of oestrus, peak estradiol-17ß concentration and LH surge after PGF2α was first observed in early than late ovulating goats (p < 0.05). The goats ovulating early had larger follicle and smaller CL in diameter at the time of PGF2α administration than those ovulating late (5.4 ± 0.2 vs. 4.3 ± 0.2 mm and 10 ± 0.6 vs. 11.8 ± 0.3 mm, respectively; p < 0.05). Likewise, plasma progesterone concentration tended to be lower (p = 0.087) in early than late ovulating goats. In conclusion, the size of dominant follicle and CL at the time of PGF2a determines the interval to ovulation following a single dose of PGF2a during the luteal phase.

ABSTRACT

In this article, a simple strategy was employed to fabricate bioinspired hybrid composite with carboxymethyl cellulose (CMC), graphene oxide, and reduced graphene oxide/alumina (rGO/Al) by a facile solution casting method. The tensile strength and toughness of rGO/Al-CMC-GO can reach 586.6 ± 12 MPa, 12.1 ± 0.44 MJm-3, respectively, due to the interface strengthening of alumina, which is 1.43 and 12 times higher than steel and about 4.3 and 6.7 times that of nature nacre. The artificial nacre hybrid composite is conductive due to the introduction of rGO/Al on the surface. Interestingly this structure can also be coated on the surface of cotton thread to give the thread good mechanical performance and conductivity. Additionally, the artificial nacre has better fire shielding and gas barrier properties. The oxygen permeability (OP) for 1% rGO/Al-CMC decreased from 0.0265 to 0.003 mLµm m-2 day-1 kpa-1, the water vapor permeability (WVP) decreased from 0.363 to 0.205 gmmm-2 day-1 kpa-1 when the concentration increased from 1% rGO/Al to 6% rGO/Al. It is believed this work provided a simple and feasible strategy to fabricate ultrastrong and ultratough graphene-based artificial nacre multifunctional materials.

ABSTRACT

Demands for high strength integrated materials have substantially increased across various kinds of industries. Inspired by the relationship of excellent integration of mechanical properties and hierarchical nano/microscale structure of the natural nacre, a simple and facile method to fabricate high strength integrated artificial nacre based on sodium carboxymethylcellulose (CMC) and borate cross-linked graphene oxide (GO) sheets has been developed. The tensile strength and toughness of cellulose-based hybrid material reached 480.5 ± 13.1 MPa and 11.8 ± 0.4 MJm-3 by a facile in situ reduction and cross-linking reaction between CMC and GO (0.7%), which are 3.55 and 6.55 times that of natural nacre. This hybrid film exhibits better thermal stability and flame retardancy. More interestingly, the hybrid material showed good water stability compared to that in the original water-soluble CMC. This type of hybrid has great potential applications in aerospace, artificial muscle, and tissue engineering.
