ABSTRACT

Using long-read sequencing, we assembled and unzipped the polyploid genomes of Meloidogyne incognita, M. javanica and M. arenaria, three of the most devastating plant-parasitic nematodes. We found the canonical nematode telomeric repeat to be missing in these and other Meloidogyne genomes. In addition, we find no evidence for the enzyme telomerase or for orthologs of C. elegans telomere-associated proteins, suggesting alternative lengthening of telomeres. Instead, analyzing our assembled genomes, we identify species-specific composite repeats enriched mostly at one extremity of contigs. These repeats are G-rich, oriented, and transcribed, similarly to canonical telomeric repeats. We confirm them as telomeric using fluorescent in situ hybridization. These repeats are mostly found at one single end of chromosomes in these species. The discovery of unusual and specific complex telomeric repeats opens a plethora of perspectives and highlights the evolutionary diversity of telomeres despite their central roles in senescence, aging, and chromosome integrity.

ABSTRACT

Previous research on the use of peripheral vision to identify two spatially separated stimuli simultaneously has led to the conclusion that the focus of attention has the form of a symmetric ellipse with a broader expansion along the horizontal compared to the vertical meridian. However, research on pseudoneglect has indicated that attention is not symmetrically distributed to the whole visual field. Here, we test if the attention window is indeed symmetrical with regard to its shape and resolution during peripheral vision. The results indicate that the position of those stimuli relative to the focus of attention influences the ability to identify a given set of stimuli. Specifically, stimuli presented to the left and top of the fixation point were more frequently identified correctly compared to those presented to the right bottom. That is, the attention window is rather not symmetric, which must be considered in future studies on the nature of the focus of attention.

ABSTRACT

Heat waves are causing declines in coral reefs globally. Coral thermal responses depend on multiple, interacting drivers, such as past thermal exposure, endosymbiont community composition, and host genotype. This makes the understanding of their relative roles in adaptive and/or plastic responses crucial for anticipating impacts of future warming. Here, we extracted DNA and RNA from 102 Pocillopora colonies collected from 32 sites on 11 islands across the Pacific Ocean to characterize host-photosymbiont fidelity and to investigate patterns of gene expression across a historical thermal gradient. We report high host-photosymbiont fidelity and show that coral and microalgal gene expression respond to different drivers. Differences in photosymbiotic association had only weak impacts on host gene expression, which was more strongly correlated with the historical thermal environment, whereas, photosymbiont gene expression was largely determined by microalgal lineage. Overall, our results reveal a three-tiered strategy of thermal acclimatization in Pocillopora underpinned by host-photosymbiont specificity, host transcriptomic plasticity, and differential photosymbiotic association under extreme warming.

ABSTRACT

BACKGROUND: Over the last decade, several coral genomes have been sequenced allowing a better understanding of these symbiotic organisms threatened by climate change. Scleractinian corals are reef builders and are central to coral reef ecosystems, providing habitat to a great diversity of species. RESULTS: In the frame of the Tara Pacific expedition, we assemble two coral genomes, Porites lobata and Pocillopora cf. effusa, with vastly improved contiguity that allows us to study the functional organization of these genomes. We annotate their gene catalog and report a relatively higher gene number than that found in other public coral genome sequences, 43,000 and 32,000 genes, respectively. This finding is explained by a high number of tandemly duplicated genes, accounting for almost a third of the predicted genes. We show that these duplicated genes originate from multiple and distinct duplication events throughout the coral lineage. They contribute to the amplification of gene families, mostly related to the immune system and disease resistance, which we suggest to be functionally linked to coral host resilience. CONCLUSIONS: At large, we show the importance of duplicated genes to inform the biology of reef-building corals and provide novel avenues to understand and screen for differences in stress resilience.

ABSTRACT

Sea urchins are emblematic models in developmental biology and display several characteristics that set them apart from other deuterostomes. To uncover the genomic cues that may underlie these specificities, we generated a chromosome-scale genome assembly for the sea urchin Paracentrotus lividus and an extensive gene expression and epigenetic profiles of its embryonic development. We found that, unlike vertebrates, sea urchins retained ancestral chromosomal linkages but underwent very fast intrachromosomal gene order mixing. We identified a burst of gene duplication in the echinoid lineage and showed that some of these expanded genes have been recruited in novel structures (water vascular system, Aristotle's lantern, and skeletogenic micromere lineage). Finally, we identified gene-regulatory modules conserved between sea urchins and chordates. Our results suggest that gene-regulatory networks controlling development can be conserved despite extensive gene order rearrangement.

ABSTRACT

Introduction: The wingwave® method combining a muscular test and elements of eye movement desensitization and reprocessing has been found to successfully reduce anxiety and improve relaxation in people. However, it is still unclear how exactly its application improves individual wellbeing (though it is assumed to be related to changes in gaze patterns) and if it works for team sport athletes. Method: To test both, team sport athletes who had reported a problem with a team sport aspect were randomly assigned to an experimental group and a control group. The experimental group members were individually coached by a wingwave® coach once while the other participants watched a tennis match instead. Results: Results showed that athletes in the wingwave® group benefited from the coaching as their individual perception of their problems improved. These improvements were associated with a decrease of catch-up saccades in a visual object-tracking task conducted before and immediately after coaching. Discussion: This points to wingwave® interventions affecting gaze behavior and consequently wellbeing of team sport athletes.

ABSTRACT

Previous research has indicated that social interactions and gaze behavior analyses in a group setting could be essential tools in accomplishing group objectives. However, only a few studies have examined the impact of social interactions on group dynamics in team sports and their influence on team performance. This study aimed to investigate the effects of game performance pressure on the gaze behavior within social interactions between beach volleyball players during game-like situations. Therefore, 18 expert beach volleyball players conducted a high and a low game performance pressure condition while wearing an eye tracking system. The results indicate that higher game performance pressure leads to more and longer fixation on teammates' faces. A higher need for communication without misunderstandings could explain this adaptation. The longer and more frequent look at the face could improve the receiving of verbal and non-verbal information of the teammate's face. Further, players showed inter-individual strategies to cope with high game performance pressure regarding their gaze behavior, for example, increasing the number of fixations and the fixation duration on the teammate's face. Thereby, this study opens a new avenue for research on social interaction and how it is influenced in/through sport.

ABSTRACT

The smallest phytoplankton species are key actors in oceans biogeochemical cycling and their abundance and distribution are affected with global environmental changes. Among them, algae of the Pelagophyceae class encompass coastal species causative of harmful algal blooms while others are cosmopolitan and abundant. The lack of genomic reference in this lineage is a main limitation to study its ecological importance. Here, we analysed Pelagomonas calceolata relative abundance, ecological niche and potential for the adaptation in all oceans using a complete chromosome-scale assembled genome sequence. Our results show that P. calceolata is one of the most abundant eukaryotic species in the oceans with a relative abundance favoured by high temperature, low-light and iron-poor conditions. Climate change projections based on its relative abundance suggest an extension of the P. calceolata habitat toward the poles at the end of this century. Finally, we observed a specific gene repertoire and expression level variations potentially explaining its ecological success in low-iron and low-nitrate environments. Collectively, these findings reveal the ecological importance of P. calceolata and lay the foundation for a global scale analysis of the adaptation and acclimation strategies of this small phytoplankton in a changing environment.

ABSTRACT

BACKGROUND: Ticks represent a major health issue for humans and domesticated animals. Exploring the expression landscape of the tick's central nervous system (CNS), known as the synganglion, would be an important step in understanding tick physiology and in managing tick-borne diseases, but studies on that topic are still relatively scarce. Neuron-specific genes like the cys-loop ligand-gated ion channels (cys-loop LGICs, or cysLGICs) are important pharmacological targets of acaricides. To date their sequence have not been well catalogued for ticks, and their phylogeny has not been fully studied. RESULTS: We carried out the sequencing of transcriptomes of the I. ricinus synganglion, for adult ticks in different conditions (unfed males, unfed females, and partially-fed females). The de novo assembly of these transcriptomes allowed us to obtain a large collection of cys-loop LGICs sequences. A reference meta-transcriptome based on synganglion and whole body transcriptomes was then produced, showing high completeness and allowing differential expression analyses between synganglion and whole body. Many of the genes upregulated in the synganglion were associated with neurotransmission and/or localized in neurons or the synaptic membrane. As the first step of a functional study of cysLGICs, we cloned the predicted sequence of the resistance to dieldrin (RDL) subunit homolog, and functionally reconstituted the first GABA-gated receptor of Ixodes ricinus. A phylogenetic study was performed for the nicotinic acetylcholine receptors (nAChRs) and other cys-loop LGICs respectively, revealing tick-specific expansions of some types of receptors (especially for Histamine-like subunits and GluCls). CONCLUSIONS: We established a large catalogue of genes preferentially expressed in the tick CNS, including the cysLGICs. We discovered tick-specific gene family expansion of some types of cysLGIC receptors, and a case of intragenic duplication, suggesting a complex pattern of gene expression among different copies or different alternative transcripts of tick neuro-receptors.

ABSTRACT

Marine planktonic eukaryotes play critical roles in global biogeochemical cycles and climate. However, their poor representation in culture collections limits our understanding of the evolutionary history and genomic underpinnings of planktonic ecosystems. Here, we used 280 billion Tara Oceans metagenomic reads from polar, temperate, and tropical sunlit oceans to reconstruct and manually curate more than 700 abundant and widespread eukaryotic environmental genomes ranging from 10 Mbp to 1.3 Gbp. This genomic resource covers a wide range of poorly characterized eukaryotic lineages that complement long-standing contributions from culture collections while better representing plankton in the upper layer of the oceans. We performed the first, to our knowledge, comprehensive genome-wide functional classification of abundant unicellular eukaryotic plankton, revealing four major groups connecting distantly related lineages. Neither trophic modes of plankton nor its vertical evolutionary history could completely explain the functional repertoire convergence of major eukaryotic lineages that coexisted within oceanic currents for millions of years.

The current study was designed to assess the personality traits and emotion regulation styles of elite beach volleyball players. Intra-team differences were examined with three primary objectives: (i) to create a personality profile of elite beach volleyball players, (ii) to examine the relationship of this profile in relation to performance and satisfaction levels, and (iii) to highlight the similarities in personalities of members of successful teams. A total of 82 elite beach volleyball players were asked to fill out the Big Five Inventory, the Personality Adjective Scale, and the Affective Style Questionnaire. In addition to these, the overall satisfaction and performance level of these athletes were measured. Results indicated a higher manifestation of warmth, liveliness, emotional stability and reasoning, along with lower levels of neuroticism in successful athletes. The players used a variety of emotional regulation styles and reported being moderately to highly satisfied with their team. A repeated-measures MANCOVA revealed no significant differences in personality traits between the team members. This study generates valuable insights into the personality of elite beach volleyball players and can be useful for coaches, sport psychologists, and academics for practical application and further scientific research.

ABSTRACT

Long-read technologies hold the promise to obtain more complete genome assemblies and to make them easier. Coupled with long-range technologies, they can reveal the architecture of complex regions, like centromeres or rDNA clusters. These technologies also make it possible to know the complete organization of chromosomes, which remained complicated before even when using genetic maps. However, generating a gapless and telomere-to-telomere assembly is still not trivial, and requires a combination of several technologies and the choice of suitable software. Here, we report a chromosome-scale assembly of a banana genome (Musa acuminata) generated using Oxford Nanopore long-reads. We generated a genome coverage of 177X from a single PromethION flowcell with near 17X with reads longer than 75 kbp. From the 11 chromosomes, 5 were entirely reconstructed in a single contig from telomere to telomere, revealing for the first time the content of complex regions like centromeres or clusters of paralogous genes.

In recent years, studies have increasingly dealt with the interaction of gaze behavior and decision making of team sports athletes. However, there is still a variety of important game situations, for example, in the case of penalty corners in field hockey, in which this interaction has not been investigated in detail yet. Penalty corners present a meaningful goal scoring opportunity by providing a relatively free shot. This paper considers two studies. The first study investigated a possible connection between the gaze behavior and the quality of decisions of experienced field hockey players and evaluated the level of success of different gaze strategies. A preliminary study (Study 1) was designed as a survey questionnaire with the aim of preparing for the main study by obtaining subjective assessments of the individual gaze behavior and decision making of professional athletes. In the second and the main study (Study 2), the gaze behavior of experienced field hockey players was recorded using mobile eye-tracking systems to analyze different strategical approaches in associated gaze behavior and decision making. Study 1 showed that players consider reacting to the defenders' behavior during a penalty corner a promising avenue for improving success at penalty corner attempts. It also indicated that such defense-dependent strategies are currently only rarely employed. Study 2 demonstrated how gaze behavior differs between different strategical approaches of the offense. It was shown that the gaze direction on the ball, the stopper, and the goal area is important to allow for a more optimal adaptation to the tactical behavior of defense. It can be concluded that adaptive decision making (i.e., choosing which variation will be carried out just after the "injection" of the ball) seems promising but requires further training to improve the success rate of penalty corner.

BACKGROUND AND AIMS: The sports betting market has been growing rapidly over the last years, as have reports of problematic gambling behavior associated with betting. Due to the well-documented gambling problems of famous athletes and the betting supportive nature of many sports-related environments, athletes have been highlighted as a potential group at-risk for problematic sports betting. However, there currently remains a lack of research on individual-level athlete-specific risk-factors or mechanisms that might contribute to the development and perpetuation of betting-related problems. Here, we examine the influence of two potential risk-factors on sports betting behavior and problems: erroneous beliefs and athletes' emotional involvement. METHODS: 201 athletes with different levels of expertise completed a newly developed scale to assess both factors. Participants were sampled from the general German population, predominantly male (83.08%) and on average 29.52 (SD = 11.05) years old. We use principal components analysis to detect patterns of covariation, potentially due to the proposed underlying latent factors, and regression analyses to test associations of these factors with betting behavior and problems. RESULTS: We find that athletes' emotional involvement was strongly associated with betting problems whereas erroneous beliefs were not. However, distorted cognitions/beliefs were associated with higher volumes and more frequent betting activities. DISCUSSION AND CONCLUSIONS: This might contribute to betting problems in the long run. These results highlight athletes' emotional involvement and erroneous beliefs as potential targets for future intervention and prevention efforts.

ABSTRACT

It has been evidenced that in attention-window tasks, the participants fixate on the center of a screen while inspecting two stimuli that appear at the same time in parafoveal vision. Such tasks have successfully been used to estimate a person's breadth of attention under various conditions. While behavioral investigations of visual attention have often made use of response accuracy, recent research has shown that the pupil size can also be used to track shifts of attention to the periphery. The main finding of previous studies is that the harder the evaluation of the stimuli becomes, e.g., because they appear farther away from the central fixation point, the stronger the pupils dilate. In this paper, we present experimental data suggesting that in an attention-window task, the pupil size can also be used to assess whether the participants attend to static, non-moving, or dynamic, moving stimuli. That is, regression models containing information on presentation mode (static vs. dynamic) and the visual angle between spatially separated stimuli better predict accuracy of perception and pupil dilation than model without these sources of information. This finding is useful for researchers who aim at understanding the human attentional system, including potential differences in its sensitivity to static and dynamic objects.

Most studies on offside decision making in soccer have not addressed rather simplistic situational probabilities like the number of players involved in an offside situation. In three studies (one observational and two experimental), the authors tried to assess whether the number of players close to the offside situation can predict the quality of offside decision making. In all three studies, they found that the presence of additional players negatively affected the percentage of correct decisions. The exact relationship between the number of players and the decrease in decision-making performance differed between the studies, though. Importantly, there was a negative influence of the number of players on decision-making quality in Studies 2 and 3, even though the authors tried to add players clearly farther away from the offside line than the relevant pair of players. This points to a crowding effect as a potential explanation for why decision-making quality decreases with an increasing number of players.

ABSTRACT

The performance and the success of a group working as a team on a common goal depends on the individuals' skills and the collective coordination of their abilities. On a perceptual level, individual gaze behavior is reasonably well investigated. However, the coordination of visual skills in a team has been investigated only in laboratory studies and the practical examination and knowledge transfer to field studies or the applicability in real-life situations have so far been neglected. This is mainly due to the fact that a methodological approach along with a suitable evaluation procedure to analyze the gaze coordination within a team in highly dynamic events outside the lab, is still missing. Thus, this study was conducted to develop a tool to investigate the coordinated gaze behavior within a team of three human beings acting with a common goal in a dynamic real-world scenario. This team was a (three-person) basketball referee team adjudicating a game. Using mobile eye-tracking devices and an indigenously designed software tool for the simultaneous analysis of the gaze data of three participants, allowed, for the first time, the simultaneous investigation of the coordinated gaze behavior of three people in a highly dynamic setting. Overall, the study provides a new and innovative method to investigate the coordinated gaze behavior of a three-person team in specific tasks. This method is also applicable to investigate research questions about teams in dynamic real-world scenarios and get a deeper look at interactions and behavior patterns of human beings in group settings (for example, in team sports).
