ABSTRACT

A given dose of hypoxia causes a greater increase in pulmonary ventilation during physical exercise than during rest, representing an exercise-induced potentiation of the acute hypoxic ventilatory response (HVR). This phenomenon occurs independently from hypoxic blood entering the contracting skeletal muscle circulation or metabolic byproducts leaving skeletal muscles, supporting the contention that neural mechanisms per se can mediate the HVR when humoral mechanisms are not at play. However, multiple neural mechanisms might be interacting intricately. First, we discuss the neural mechanisms involved in the ventilatory response to hypoxic exercise and their potential interactions. Current evidence does not support an interaction between the carotid chemoreflex and central command. In contrast, findings from some studies support synergistic interactions between the carotid chemoreflex and the muscle mechano- and metaboreflexes. Second, we propose hypotheses about potential mechanisms underlying neural interactions, including spatial and temporal summation of afferent signals into the medulla, short-term potentiation and sympathetically induced activation of the carotid chemoreceptors. Lastly, we ponder how exercise-induced potentiation of the HVR results in hyperventilation-induced hypocapnia, which influences cerebral blood flow regulation, with multifaceted potential consequences, including deleterious (increased central fatigue and impaired cognitive performance), inert (unchanged exercise) and beneficial effects (protection against excessive cerebral perfusion).

ABSTRACT

Query (Q) fever is a worldwide infectious disease with acute and chronic manifestations caused by Coxiella burnetii. The clinical manifestations are so variable that the disease is often only diagnosed if systematically considered as a differential diagnosis. Here, we present a case of a 39-year-old man who lived in a countryside house, with cattle and sheep in his field, with acute Q fever hepatitis with the typical granulomatous arrangement in the liver biopsy. The diagnosis was confirmed by polymerase chain reaction (PCR) assay in a serum sample and the presence of phase II antibodies. Anticardiolipin antibody (aCL) determination at diagnosis of acute Q fever and during follow-up was made to persecute early identification and to guide the treatment and prophylaxis of possible complications, such as endocarditis.

ABSTRACT

Hypoxia plays a significant role in the development of various cerebral diseases, many of which are associated with the potential risk of recurrence due to mitochondrial damage. Conventional drug treatments are not always effective for hypoxia-related brain diseases, necessitating the exploration of alternative compounds. In this study, we investigated the potential of diphenyl diselenide [(PhSe)2] to ameliorate locomotor impairments and mitigate brain mitochondrial dysfunction in zebrafish subjected to hypoxia. Additionally, we explored whether these improvements could confer resistance to recurrent hypoxia. Through a screening process, an appropriate dose of (PhSe)2 was determined, and animals exposed to hypoxia received a single intraperitoneal injection of 100 mg/kg of the compound or vehicle. After 1 h from the injection, evaluations were conducted on locomotor deficits, (PhSe)2 content, mitochondrial electron transport system, and mitochondrial viability in the brain. The animals were subsequently exposed to recurrent hypoxia to assess the latency time to hypoxia symptoms. The findings revealed that (PhSe)2 effectively crossed the blood-brain barrier, attenuated locomotor deficits induced by hypoxia, and improved brain mitochondrial respiration by modulating complex III. Furthermore, it enhanced mitochondrial viability in the telencephalon, contributing to greater resistance to recurrent hypoxia. These results demonstrate the beneficial effects of (PhSe)2 on both hypoxia and recurrent hypoxia, with cerebral mitochondria being a critical target of its action. Considering the involvement of brain hypoxia in numerous pathologies, (PhSe)2 should be further tested to determine its effectiveness as a potential treatment for hypoxia-related brain diseases.

ABSTRACT

Climate change increasingly influences the loss of biodiversity, especially in ectothermic organisms, which depend on environmental temperatures to obtain heat and regulate their life cycle. Studies that aim to understand the impact of temperature variation are important to better understand the possible impacts generated on the homeostasis of ectothermic organisms. Our objective was to characterize the responses of juvenile Liolaemus arambarensis lizards to abrupt changes in temperature, quantifying markers of body condition, intermediary and hormonal metabolism and oxidative balance. We collected 45 juvenile individuals of L. arambarensis (winter: 20 and summer: 25) in Barra do Ribeiro, Brazil. We transported the animals to the laboratory, where they were acclimatized for five days at a temperature of 20 °C, then divided and exposed to temperatures of 10 °C, 20 °C, 30 °C and 40 °C for 24 h. After exposure, the animals were euthanized and the brain, caudal muscle, thigh, and liver tissues were extracted for quantification of biomarkers of metabolism (glycogen and total proteins) and oxidative balance (acetylcholinesterase, superoxide dismutase, catalase, glutathione-S-transferase and lipoperoxidation) and plasma for corticosterone quantification. The results show that L. arambarensis is susceptible to sudden temperature variations, where higher temperatures caused greater activity of antioxidant enzymes, increased lipoperoxidation and higher plasma levels of corticosterone in animals eliminated in winter. The present study demonstrated that abrupt changes in temperature could significantly modify the homeostatic mechanisms of animals, which could lead to oxidative stress and a potential trade-off between survival and growth/reproduction. In this context, the organism mobilizes energy resources for survival, with possible damage to growth and reproduction. Demonstrate that a change in temperature can be a potential factor in extinction for a species given the profile of global climate change.

ABSTRACT

This study evaluated the impact of long-term storage conditions (medium and time) on the development of experimental dentinal microcracks through micro-computed tomography. Sixty freshly extracted premolars were stored in formalin, water, or dry conditions (n = 20) and scanned after 72 h, 30 days, 6 months and 3 years of extraction. The effect of the storage medium and time on the occurrence of dentinal defects was statistically evaluated. A total of 211 000 images were screened revealing the existence of 11 519 slices with dentinal defects. Dry conditions significantly contributed to the development of new defects in all time points. During the 3-year follow-up period, no new defects were detected in the teeth that were stored in water and, in a single tooth, in the formalin group, after 6 months of storage.

ABSTRACT

As SARS-CoV-2 continues to produce new variants, the demand for diagnostics and a better understanding of COVID-19 remain key topics in healthcare. Skin manifestations have been widely reported in cases of COVID-19, but the mechanisms and markers of these symptoms are poorly described. In this cross-sectional study, 101 patients (64 COVID-19 positive patients and 37 controls) were enrolled between April and June 2020, during the first wave of COVID-19, in São Paulo, Brazil. Enrolled patients had skin imprints sampled non-invasively using silica plates; plasma samples were also collected. Samples were used for untargeted lipidomics/metabolomics through high-resolution mass spectrometry. We identified 558 molecular ions, with lipids comprising most of them. We found 245 plasma ions that were significant for COVID-19 diagnosis, compared to 61 from the skin imprints. Plasma samples outperformed skin imprints in distinguishing patients with COVID-19 from controls, with F1-scores of 91.9% and 84.3%, respectively. Skin imprints were excellent for assessing disease severity, exhibiting an F1-score of 93.5% when discriminating between patient hospitalization and home care statuses. Specifically, oleamide and linoleamide were the most discriminative biomarkers for identifying hospitalized patients through skin imprinting, and palmitic amides and N-acylethanolamine 18:0 were also identified as significant biomarkers. These observations underscore the importance of primary fatty acid amides and N-acylethanolamines in immunomodulatory processes and metabolic disorders. These findings confirm the potential utility of skin imprinting as a valuable non-invasive sampling method for COVID-19 screening; a method that may also be applied in the evaluation of other medical conditions. KEY MESSAGES: Skin imprints complement plasma in disease metabolomics. The annotated markers have a role in immunomodulation and metabolic diseases. Skin imprints outperformed plasma samples at assessing disease severity. Skin imprints have potential as non-invasive sampling strategy for COVID-19.

ABSTRACT

O objetivo do artigo é refletir sobre a geração de valor de troca a partir do consumo de sites de redes sociais. A partir do conceito de indústria cultural, proposto por Adorno e Horkheimer (1985), é possível inferir que a teoria da escola de Frankfurt dialoga com a monetização de produtos culturais e a produção e consumo destes de maneira alienada. Por meio do método do materialismo histórico dialético de Karl Marx -o qual permite a compreensão de uma síntese de ideia apenas após o confronto de uma tese concreta com a sua negação abstrata- o trabalho conclui que é necessária uma regulamentação na obtenção, uso e venda de dados dos usuários nessa sociedade em contexto de plataformas digitais.


El propósito del artículo es reflexionar sobre la generación de valor de cambio a partir del consumo de sitios de redes sociales. A partir del concepto de industria cultural, propuesto por Adorno y Horkheimer (1985), es posible inferir que la teoría de la escuela de Frankfurt dialoga con la monetización de los productos culturales y su producción y consumo de forma alienada. A través del método del materialismo histórico dialéctico de Karl Marx -que permite la comprensión de una síntesis de idea sólo después de la confrontación de una tesis concreta con su negación abstracta- el trabajo concluye que es necesario regular la adquisición, uso y venta de datos de los usuarios de esta sociedad en el contexto de las plataformas digitales.

ABSTRACT

Pesticide usage has increased over the last decades, leading to concerns regarding its effects on non-target organisms, especially amphibians. Tadpoles of Rhinella icterica were collected in a pesticide-free place, acclimated in the laboratory (21 days) and exposed (7 days) to three herbicides (20 µg/L atrazine (A); 250 µg/L glyphosate (G); 20 µg/L quinclorac (Q)) and their mixtures. Only 2% mortality was observed over the 28 days of the study. Despite this, significant variations were observed for markers of oxidative balance and body condition when comparing all experimental groups. K and Kn factor showed the lowest values in the group A+G+Q, as well as the activity levels of GST and SOD. In contrast to this, the CAT activity was higher in the same group (A+G+Q). The mixture of the three herbicides proved to be more harmful, which points to the need for more restrictive laws for the use of mixed herbicides.

NEW FINDINGS: What is the central question of this study? What is the effect of peripheral chemoreflex and muscle metaboreflex integration on ventilation regulation, and what is the effect of integration on breathing-related sensations and emotions? What is the main finding and its importance? Peripheral chemoreflex and muscle metaboreflex coactivation during isocapnic static handgrip exercise appeared to elicit a hyperadditive effect with regard to ventilation and an additive effect with regard to breathing-related sensations and emotions. These findings reveal the nature of the integration between two neural mechanisms that operate during small-muscle static exercise performed under hypoxia. ABSTRACT: Exercise augments the hypoxia-induced ventilatory response in an exercise intensity-dependent manner. A mutual influence of hypoxia-induced peripheral chemoreflex activation and exercise-induced muscle metaboreflex activation might mediate the augmentation phenomenon. However, the nature of these reflexes' integration (i.e., hyperadditive, additive or hypoadditive) remains unclear, and the coactivation effect on breathing-related sensations and emotions has not been explored. Accordingly, we investigated the effect of peripheral chemoreflex and muscle metaboreflex coactivation on ventilatory variables and breathing-related sensations and emotions during exercise. Fourteen healthy adults performed 2-min isocapnic static handgrip, first with the non-dominant hand and immediately after with the dominant hand. During the dominant hand exercise, we (a) did not manipulate either reflex (control); (b) activated the peripheral chemoreflex by hypoxia; (c) activated the muscle metaboreflex in the non-dominant arm by post-exercise circulatory occlusion (PECO); or (d) coactivated both reflexes by simultaneous hypoxia and PECO use. Ventilation response to coactivation of reflexes (mean ± SD, 13 ± 6 l/min) was greater than the sum of responses to separated activations of reflexes (mean ± SD, 8 ± 8 l/min, P = 0.005). Breathing-related sensory and emotional responses were similar between coactivation of reflexes and the sum of separate activations of reflexes. Thus, the peripheral chemoreflex and muscle metaboreflex integration during exercise appeared to be hyperadditive with regard to ventilation and additive with regard to breathing-related sensations and emotions in healthy adults.

ABSTRACT

Food safety and quality assessment mechanisms are unmet needs that industries and countries have been continuously facing in recent years. Our study aimed at developing a platform using Machine Learning algorithms to analyze Mass Spectrometry data for classification of tomatoes on organic and non-organic. Tomato samples were analyzed using silica gel plates and direct-infusion electrospray-ionization mass spectrometry technique. Decision Tree algorithm was tailored for data analysis. This model achieved 92% accuracy, 94% sensitivity and 90% precision in determining to which group each fruit belonged. Potential biomarkers evidenced differences in treatment and production for each group.

O presente trabalho analisou os riscos envolvidos na utilização dos recursos de telessaúde e telemedicina, autorizados durante a pandemia de covid-19, sem um correspondente amadurecimento com relação aos requisitos necessários para garantir a segurança dos dados pessoais e dados pessoais sensíveis de seus usuários, seja pela recente entrada em vigor da Lei n. 13.709/2018, seja pela incipiente criação da Autoridade Nacional de Proteção de Dados, que ainda caminha no sentido de se estruturar organicamente. Sob o lume da metodologia civil-constitucional capitaneada por Perlingieri, o artigo destacou a necessidade de que os requisitos tecnológicos abarcados nas relações privadas sejam devidamente adequados aos valores intrínsecos àqueles delineados no texto constitucional, tendo as normas de direito civil como importante vetor na garantia de tal aplicação. A partir de pesquisa qualitativa, valendo-se de fontes indiretas, inclusive legislação estrangeira, e análise à luz da metodologia dedutiva, elencou-se uma série de considerações para a aplicação de recursos da telemedicina no Brasil de maneira adequada e em sintonia com a proteção de dados pessoais de seus cidadãos.


The present work analyzed the risks involved in the use of telehealth and telemedicine resources, authorized during the covid-19 pandemic, without a corresponding maturity in relation to the necessary requirements to guarantee the security of personal data and sensitive personal data of users, whether by the recent entry into force of Law no. 13,709/2018, or the incipient creation of the National Data Protection Authority, which is still moving towards an organic structure. Under the light of the civilconstitutional methodology led by Perlingieri, the article highlights the need for technological requirements encompassed in private relations to be duly adapted to the intrinsic values of those outlined in the constitutional text, with the norms of civil law as an important vector in guaranteeing such an application. Based on qualitative research, using indirect sources, including foreign legislation, and analysis in the light of deductive methodology, a series of considerations are listed for the application of telemedicine resources in Brazil in an adequate manner and in line with the protection of personal data of citizens.

ABSTRACT

Infertility is a worldwide concern, affecting one in six couples throughout their reproductive period. Therefore, enhancing the clinical tools available to identify the causes of infertility may save time, money, and emotional distress for the involved parties. This study aims to annotate potential biomarkers in follicular fluid that are negatively affecting pregnancy outcomes in women suffering infertility-related diseases such as endometriosis, tuboperitoneal factor, uterine factor, and unexplained infertility, using a metabolomics approach through high-resolution mass spectrometry. Follicular fluid samples collected from women who have the abovementioned diseases and managed to become pregnant after in vitro fertilization procedures [control group (CT)] were metabolically compared with those from women who suffer from the same diseases and could not get pregnant after the same treatment [infertile group (IF)]. Mass spectrometry analysis indicated 10 statistically relevant differential metabolites in the IF group, including phosphatidic acids, phosphatidylethanolamines, phosphatidylcholines, phosphatidylinositol, glucosylceramides, and 1-hydroxyvitamin D3 3-D-glucopyranoside. These metabolites are associated with cell signaling, cell proliferation, inflammation, oncogenesis, and apoptosis, and linked to infertility problems. Our results indicate that understanding the IF's metabolic profile may result in a faster and more assertive female infertility diagnosis, lowering the costs, and increasing the probability of a positive pregnancy outcome.

ABSTRACT

In a cyberphysical production system, the connectivity between the physical entities of a production system with the digital component that controls and monitors that system takes fundamental importance. This connectivity has been increasing from the transducers' side, through gathering new functionalities and operating increasingly independently, taking the role of smart transducers, and from the applications' side, by being developed in a distributed and decentralized paradigm. This work presents a plug-and-play solution capable of integrating smart transducers compliant with the IEEE 1451 standard in industrial applications based on the IEC 61499 standard. For this, we implemented the NCAP module of the smart transducer defined in IEEE 1451, which, when integrated with 4diac IDE and DINASORE (development and execution tools compliant with IEC 61499), enabled a solution that presented automatically the smart sensors and actuators in the IDE application and embedded their functionalities (access to data and processing functions) in the runtime environment. In this way, a complete plug-and-play solution was presented from the connection of the transducer to the network until its integration into the application.

ABSTRACT

The use of Sensors and Actuators is ubiquitous in an industrial environment. The advent of the Industrial Internet-of-Things (IIoT) and the 4th industrial revolution demands new, more intelligent and more efficient ways to be able to connect, read and control transducers at the plant floor level. Newer control and data science techniques also largely benefit from structured information endpoints available at the edge of the network. The IEEE 1451 standard presents architecture and methodology to solve these problems with the usage of smart transducers, introducing into edge devices concepts such as self-identification and standardization of data communication. In this work, a transducer interface module is developed using the IEEE 1451 standard focused on flexibility, ease of integration and plug-and-play features. Furthermore, a system architecture, based on IEEE 1451.0 is presented, and development and implementation features are explained. This system is then released as an open-source platform to help and motivate the usage of smart transducer systems. At last, the system is tested, results are collected, and a methodology and metrics are defined for comparison between different smart transducer systems.

BACKGROUND: The citrus greening disease or Huanglongbing (HLB) is the most devastating disease of citrus crops. Trioza erytreae is a vector of HLB. Since its introduction in Europe, the insect reached the northern region of Spain and the southern region of Portugal, threatening relevant citrus production areas. Limiting the spread of HLB vectors is mandatory to prevent this disease. In this work, we assessed the effect of kaolin, a white mineral clay, on the landing, settling behavior and feeding behavior of Trioza erytreae on lemon plants. RESULTS: After kaolin application, the number of plants on which the insect was found was significantly lower than on untreated plants in the laboratory and in the field. Moreover, there were significantly fewer T. erytreae and a shorter duration of phloem-related events on kaolin-treated than untreated plants. CONCLUSION: The use of kaolin could be a suitable and efficient tool for inclusion into integrated pest management programs or organic production to reduce populations of T. erytreae and subsequently limit the spread of HLB in citrus crops. © 2022 The Authors. BACKGROUND: HIV infection affects millions of people globally. Currently, although several drugs have brought an improvement in the quality and life expectancy of these individuals, they are accompanied by several adverse effects. OBJECTIVE: To conduct a systematic review of studies examining the relationship between antiretroviral therapy (ART) uses and secondary dyslipidemia. METHODS: The review followed the criteria defined by PRISMA. Only articles that completely evaluated the lipid profile were included, which consisted of total cholesterol (TC), triglycerides (TG), and LDL cholesterol (LDL-c), HDL cholesterol (HDL-c). RESULTS: It was observed that the use of nucleoside and non-nucleoside reverse transcriptase inhibitor (NNRTI and NNRTI respectively) drugs and protease inhibitors are the most used in ART and are associated with changes in lipid profiles. The main changes observed were increases in TC, TG, and LDL-c in addition to a decrease in HDL-c. These patients had a higher risk of developing cardiovascular disease not only due to the use of therapy, but also due to the presence of other comorbidities evaluated in these studies, such as obesity, diabetes, and hypertension. The increase in age, the difference between genders, CD4 T-cell count, and viral load, were observed as risk factors for worsening dyslipidemia. CONCLUSION: According to the findings of this study, anti-HIV therapy is linked to dyslipidemia, which may or may not be the primary cause, and is frequently connected with a number of metabolic problems that can exacerbate the illness.

Amphibians have suffered population decline due to several factors, including exposure to pesticides. In the south of Brazil, rice cultivations use herbicides based on atrazine, glyphosate and quinclorac as the commercial formulations Primoleo®, Roundup® and Facet®, respectively. Rhinella icterica was chosen to evaluate oxidative balance markers and body condition after exposure to three concentrations of herbicides (10, 20 and 40 µg/L of atrazine and quinclorac; 100, 250 and 500 µg/L of glyphosate). These xenobiotics, regardless of the concentrations used, accelerated the development process of animals and seemed to act as modulators of development. We observed no significant variations for any of the oxidative balance markers studied (superoxide dismutase, catalase, TBARS and carbonylated proteins); however, we cannot rule out that other antioxidant system components prevent oxidative stress. In general, atrazine and glyphosate accelerated the development of tadpoles, and quinclorac retards this process, which could impact the survival of these animals.

ABSTRACT

Ilex paraguariensis A. St. Hil. plants are used for the preparation of food and drinks which are widely consumed worldwide. During the harvest season of these plants, 2-5 ton hec-1 of agricultural residue is generated, which remains underutilized. Therefore, this study aimed to obtain an edible extract with high content of bioactive compounds and antimicrobial properties from the agricultural residue of I. paraguariensis for industrial use in food applications. The extraction conditions were optimized through a multivariate experimental design using ethanol:water. The extracted compounds were characterized by HPLC-ESY-QTOF-MS. In the optimal extraction conditions, 55 compounds were extracted, including 8 compounds that were not previously reported in I. paraguariensis. The method proved to be simple, fast, economical and environmentally friendly, with the use of green solvents. This optimization allowed for the extraction of 15.07 g of phenolic compounds per 100 g of residue. The extract showed high antioxidant activity and the capacity to inhibit Staphylococcus aureus. Results indicate that it is possible to obtain an edible extract with a high content of bioactive compounds, particularly phenolic compounds, from the I. paraguariensis residue, which has high prospects for the valorization of unexplored natural resources.

ABSTRACT

This study evaluated the efficacy of sodium thiosulphate (ST) as an intermediate irrigant between sodium hypochlorite (NaOCl) and chlorhexidine (CHX) to avoid the formation of the brown-coloured precipitate. The in vitro experiment used microtubes to test the mixture of 2.5% NaOCl, 2% CHX and 5% ST solutions, varying their order and time of addition. In the ex vivo experiment, 10 bovine teeth were divided into two groups according to the intermediate irrigant used: distilled water (control) and 5% ST (experimental). Colour change occurred in all microtubes after interaction between solutions. While the precipitate was not observed in the experimental group, it was seen in all specimens of the control group. The 5% ST was able to prevent the formation of the brown-coloured precipitate in the ex vivo experiment, whereas it only prevented its in vitro formation when the waiting time for adding CHX was 10, 15 and 20 min.
