ABSTRACT

OBJECTIVE: To find out the optimum condition for the germination of seed of Dendranthema indicum by studying the effects of pretreatment,phytohormone and temperature on it, and offer the basis for its standardized culture. METHODS: The seed purity, weight per 1000 seeds, seed moisture content and seed viability were determined. The germination of D. indicum seed was tested under following conditions: pretreatment (acid, base, warm water, boiling water), phytohormone (IBA, 6-BA, NAA) and treatments under different temperature (10, 20, 25, 30 degrees C). RESULTS: The seed purity was 99.4%; The weight per 1000 seeds was 0.2941 g; The seed moisture content was 4.39%; The seed viability was 85.3%; The tests of pretreatment couldnt increase the germination of D. indicum seed; Phytohormone had limited effect on the germination of the seed; Temperature condition showed significant effects on the germination. CONCLUSION: The optimum condition for the germination of the seed of D. indicum is 25 degrees C on filter paper.

ABSTRACT

OBJECTIVE: To study the genetic polymorphism and intraspecific genetic differentiation of different populations of Pogostemon cablin, and find out the effective method to distinguish DNA fingerprint of different populations of P. cablin. METHOD: Five plant populations of P. cablin were analyzed by RAPD markers. PopGen 32 software for clustering analysis and calculating. Fourteen of the 80 random primers were tested to possess the stronger detecting effect of polymorphous character. RESULT: A total of 84 bands was amplified by the 10 primers, among them 17 bands were monomorphic. 67 of them were polymorphic. The results indicated that the genetic variations existed within the different plant populations of the same species. CONCLUSION: It is feasible by RAPD technique with specifically primer to analyze the genetic diversity and identify 5 plant populations of P. cablin. RAPD technique has provided a new path for identification and classification of P. cablin genetic germplasm.

ABSTRACT

OBJECTIVE: To provide scientific basis for quality evaluation by investigating the influential factors of the volatile oil and main constituent content in Pogostemon cablin. METHOD: Several experiments were carried out to test different habitats, collection periods, processing methods, the level of spreading manure and using agricultural chemical with the volatile oil assay of pharmacopoeia and GC-MS method. RESULT: The effect of different habitats, collection periods, processing methods on volatile oil and main constituent content was shown significantly. CONCLUSION: Such factors as different habitats, collection periods, processing methods should be taken into account when the quality standard of medicinal materials was made.
