ABSTRACT

To analyze the effects of concurrent training (CT) on the muscle fibers of Wistar rats submitted to standard and hypercaloric diets. In total, 40 rats were used, divided into 4 groups: Sedentary Group (GS); Exercise Group (GE), Obese Sedentary Group (OS) and Obese Exercise Group (OE). The animals performed a CT protocol consisting of: muscle strength training and aerobic training, carried out 3 times a week for 45 days. The smallest diameter of muscle fibers (MDF) was analyzed to evaluate muscle hypertrophy. It was observed that the OE group presented a significant decrease in MDF, compared to the OS group (OE=77.41 µm vs. OS=98.58 µm). In addition, the animals that performed CT demonstrated muscle hypertrophy (GE=74.39 µm vs. GS=72.13 µm). In conclusion, the CT with a standard diet promoted an increase in MDF while CT with a hypercaloric diet resulted in a decrease.


El objetivo de este trabajo fue analizar el efecto del entrenamiento concurrente (EC) en las fibras musculares de ratas Wistar sometidas a dietas normal e hipercalórica. Fueron utilizadas 40 ratas Wistar, distribuidas en cuatro grupos de animales: grupo sedentario (GS); grupo ejercicio (GE); grupo obeso sedentario (OS) y grupo obeso ejercicio (OE). Los animales realizaron el protocolo del EC compuesto por entrenamiento de fuerza y aeróbico, tres veces en la semana y por 45 días. La media del menor diámetro (MMD) de las fibras musculares fue medida para verificar la hipertrofia muscular. Fue observado que el grupo OE presentó una significante diminución del MMD comparado al grupo OS (OE=77.41 µm vs. OS=98.58 µm). Además, los animales que fueron sometidos al protocolo del EC demostraron hipertrofia muscular (GE=74.39 µm vs. GS=72.13 µm). Se puede concluir que el protocolo del EC con dieta normal tiene como resultado un aumento del MMD, mientras que el EC con dieta hipercalórica tiene como resultado la diminución del MMD.

ABSTRACT

OBJECTIVE: Obesity is a strong predictor of some kinds of diseases. High intake of high-fat foods contributes significantly to the growth of the obese population globally. The aim of this study was to verify if consumption of a cafeteria diet for fourteen weeks could increase white fat mass, body weight and skeletal muscle mass and promote insulin resistance in male Wistar rats. METHODS: Twenty animals were divided into two groups: control and obese. Both were fed standard chow and water ad libitum. Additionally, a cafeteria diet consisting of bacon, bologna sausage, sandwich cookies and soft drink was given to the obese group. RESULTS: The obese group was significantly heavier (p<0.0001) than controls from the second week until the end of the cafeteria-diet intervention. Absolute and relative fat mass, liver weight and Lee Index increased significantly (p<0.05) in the obese group. Furthermore, the obese group had lower (p<0.05) insulin sensitivity than the control group. CONCLUSION: In conclusion, fourteen weeks of cafeteria diet promoted a progressive increase of fat mass and insulin resistance. Therefore, this is a great and inexpensive diet-induced insulin resistance model.


OBJETIVO: A obesidade é um forte preditor de alguns tipos de doenças. A grande oferta de comida e a predominância de ácidos graxos presentes na maioria dos alimentos contribuem diretamente para o aumento da população obesa no mundo. O objetivo do estudo foi verificar se uma dieta de cafeteria durante um período de catorze semanas pode causar aumento dos pesos de tecido adiposo branco, corporal e muscular e provocar um quadro de resistência insulínica em ratos machos Wistar. MÉTODOS: Vinte ratos machos Wistar foram divididos em dois grupos: controle e obeso. Os dois grupos foram alimentados com ração padrão e água ad libitum. Ao grupo obeso foi ofertada dieta ocidental, composta por salsicha, mortadela, bolacha recheada, bacon e refrigerante. RESULTADOS: Os animais do grupo obeso estavam significantemente mais pesados a partir da segunda semana de tratamento e essa diferença permaneceu até o final do estudo (p<0,0001). O peso absoluto e relativo do tecido adiposo branco e do fígado, e o Índice de Lee foram maiores no grupo obeso (p<0,05), que apresentou uma menor sensibilidade à insulina no final do estudo quando comparado ao grupo controle (p<0,05). Podemos observar que a dieta cafeteria promoveu um aumento progressivo e significativo da massa de gordura corporal associada à disfunção da ação da insulina. CONCLUSÃO: Conclui-se que uma dieta de cafeteria por um período de catorze semanas é capaz de promover aumento progressivo da massa adiposa associada à disfunção da insulina, sendo ótimo e fácil modelo de para induzir resistência à insulina.

OBJECTIVE: To determine if resistive exercise protocol can modulate Tnf-α, SOCS3 and glucose transporter GLUT4 genes expression in skeletal muscle, and peripheral insulin sensitivity in obese rats induced by hyperlipidic diet. MATERIALS AND METHODS: Wistar obese rats induced by hyperlipidic diet were subjected a resistive exercise protocol as jump squat. Insulin sensitivity and mRNA content of Tnf-α, SOCS3 and GLUT4 were assayed and compared among the groups: obese sedentary (OS) and exercised (OE), control sedentary (CS) and exercised (CE). RESULTS: The mRNA content of Tnf-α and SOCS3 has increased in skeletal muscle from OS and has decreased in OE group. The protein and GLUT4 mRNA contents were correlated but they did not change among the groups. Peripheral insulin sensitivity has increased in the OE compared to OS group. CONCLUSION: The resistive exercise reverses the peripheral insulin resistance and the inflammatory state in skeletal muscle from diet-induced obese rats.

ABSTRACT

OBJETIVO: Investigar em ratos obesos o efeito da prática de exercício resistido sobre a sensibilidade à insulina e sobre a expressão de citocinas pró-inflamatórias e de transportador de glicose em músculo solear. MATERIAIS E MÉTODOS: Ratos Wistar alimentados com dieta hiperlipídica (grupos obesos) foram submetidos ao protocolo de exercício tipo jump squat. A sensibilidade à insulina e a expressão gênica de Tnf-α, SOCS3 e GLUT4 foram comparadas entre os grupos obesos sedentários (OS) e exercitados (OE) e controles sedentários (CS) e exercitados (CE). RESULTADOS: A sensibilidade à insulina estava reduzida no grupo OS e elevada no OE. Os conteúdos de RNAm de Tnf-α e de SOCS3 estavam aumentados no músculo esquelético do grupo OS e reduzidos no OE. O conteúdo proteico e de RNAm de GLUT4 não diferiu entre os grupos. CONCLUSÃO: O exercício resistido reverte o quadro de resistência à insulina periférica e de inflamação no músculo esquelético de obesos induzidos por dieta.
