ABSTRACT

A interação com videogame aumentou significativamente na pandemia de COVID-19. Por outro lado, um aspecto atencional que pode apresentar alterações relacionadas à quantidade de interação com videogames de ação é o mindfulness. Portanto, o objetivo deste estudo foi verificar uma possível relação entre o uso de videogames de ação (em horas semanais), o mindfulness e o medo da COVID-19. Para tanto, solicitou-se a jogadores e não jogadores de videogames de ação com idades entre 18 e 29 anos (n = 384, M = 21,6, DP = 2,6) que respondessem à Escala de Medo da COVID-19 e à Escala de Facetas do Mindfulness. Os resultados não demonstraram relação significativa entre uso de videogames de ação e mindfulness, mas apontaram que jogadores de videogame de ação apresentaram menos medo da COVID-19. Discute-se como o escapismo proporcionado pelo jogo pode explicar, em parte, uma diminuição da atenção a informações relativas à doença


Interaction with video games increased significantly in the COVID-19 pandemic. On the other hand, an attentional aspect that may show changes related to the amount of interaction with action video games is mindfulness. Therefore, the objective of this study was to verify a possible relationship between the use of action video games (in weekly hours), mindfulness and fear of COVID-19. To this end, players and non-players of action video games aged between 18 and 29 years (n = 384, M = 21.6, SD = 2.6) were asked to respond to the Covid Fear Scale and the Facets of Mindfulness. Results did not demonstrate a significant relationship between the use of action video games and mindfulness, but showed that action video game players were less afraid of COVID-19. It is discussed how the escapism provided by the game can partly explain a decrease in attention to information related to the disease


La interacción con videojuegos aumentó en la pandemia de COVID-19. Por otro lado, un aspecto atencional que puede presentar cambios relacionados con la cantidad de interacción con los videojuegos de acción es el mindfulness. Luego, el objetivo de este estudio fue verificar una relación entre uso de video-juegos de acción, mindfulness y miedo al COVID-19. Se pidió a jugadores y no jugadores de videojuegos de acción de entre 18 y 29 años (n = 384, M = 21,6, SD = 2,6) que respondieran a la Escala de Miedo al COVID y la Escala de Facetas de Mindfulness. Los resultados no demostraron una relación significativa entre uso de videojuegos de acción y mindfulness, pero señalaron que jugadores de videojuegos de acción tenían menos miedo a la COVID-19. Se discute cómo el escapismo proporcionado por el juego explicaría en parte una disminución en la atención a la información relacionada con la enfermedad

ABSTRACT

The brain-derived neurotrophic factor (BDNF) is a neurotrophin recognized to play a major role in neuroplastic modifications associated to drug abuse, being involved in various behavioral changes found in drug use disorders, such as drug sensitization, craving and relapses. These neuroplastic changes were shown to affect the prefrontal cortex functions, which can be briefly measured through cognitive tests such as the Frontal Assessment Battery (FAB). In this study we investigated the BDNF mRNA expression in peripheral blood lymphocytes of crack-cocaine use disorder (CUD) and alcohol use disorder (AUD) patients, after drug detoxification treatment, using a real-time PCR approach and examining its association to FAB performance. BDNF mRNA expression was found to be higher by 2.25-fold in CUD patients and by 2-fold in the AUD patients when normalized to controls, and these values were found to be associated with FAB scores. This preliminary study evaluates, for the first time, BDNF mRNA expression in leukocytes and its relationship to FAB scores in crack-cocaine and alcohol use disorder patients.

ABSTRACT

FosB gene heterodimerizes with Jun family proteins to form activator protein 1 (AP-1) complexes that bind to AP-1 sites in responsive genes to regulate transcription in all cells. The genic expression of FosB seems to be modified after long time exposure to drugs of abuse and these changes may be involved in craving and addicted behavior. This study investigated the FosB mRNA expression in peripheral blood lymphocytes of drug addicted patients using real-time PCR approach. Thus, patients with crack-cocaine use disorder (CUD, n = 10), alcohol use disorder (AUD, n = 12), and healthy non-addicted subjects (CONT, n = 12) were assessed. FosB mRNA expression was reduced by 1.15-fold in CUD and 2.17-fold in AUD when compared to CONT. Hedge's effect size gs over log FosB/Act was of 0.66 for CUD and of 0.30 for AUD when compared to controls. This study showed that FosB mRNA expression was detected in lymphocytes from peripheral blood for the first time, and it was less expressed in drug addicted patients. This molecular technique may constitute a potential peripheral marker for substance use disorder.

ABSTRACT

The consequences of exposure to environmental contaminants have shown significant effects on brain function and behavior in different experimental models. The endocrine-disrupting chemicals (EDC) present various classes of pollutants with potential neurotoxic actions, such as organotins (OTs). OTs have received special attention due to their toxic effects on the central nervous system, leading to abnormal mammalian neuroendocrine axis function. OTs are organometallic pollutants with a tin atom bound to one or more carbon atoms. OT exposure may occur through the food chain and/or contaminated water, since they have multiple applications in industry and agriculture. In addition, OTs have been used with few legal restrictions in the last decades, despite being highly toxic. In addition to their action as EDC, OTs can also cross the blood-brain barrier and show relevant neurotoxic effects, as observed in several animal model studies specifically involving the development of neurodegenerative processes, neuroinflammation, and oxidative stress. Thus, the aim of this short review is to summarize the toxic effects of the most common OT compounds, such as trimethyltin, tributyltin, triethyltin, and triphenyltin, on the brain with a focus on neuronal damage as a result of oxidative stress and neuroinflammation. We also aim to present evidence for the disruption of behavioral functions, neurotransmitters, and neuroendocrine pathways caused by OTs.

ABSTRACT

BACKGROUND: OxyElite Pro (OEP) is a dietary supplement to increase metabolism which contains as key stimulant the ingredient 1,3-dimethylamylamine (DMAA). Serious adverse effects have been reported after OEP consumption however, these effects are related to poisoning or overdose. To our knowledge, no one studied the effects of OEP at controlled doses. Thus, the aim of this study was to evaluate acute and chronic OEP affects, at controlled doses in Wistar rats, on physical performance, metabolic parameters, liver injury markers and oxidative stress markers and mitochondrial biogenesis in skeletal muscle. METHODS: Rats were divided in control, 4.3 mg OEP/kg, 12.9 mg OEP/kg and 25.8 mg OEP/kg. All groups were submitted to supplementation with OEP for 4 weeks and the experimental protocols were performed 30 min after the first OEP administration (acute response) and 30 min after the last OEP administration at the end of the forth week (chronic response). RESULTS: Running distance and running time increased after acute administration of 12.9 mg OEP/kg (2.6-fold) and 25.8 mg OEP/kg (2.8-fold). Since no effect on the exercise tolerance test was observed at the lower OEP dose (4.3 mg OEP/kg), this group was removed from further analyzes. On other hand, running distance and running time decreased after daily supplementation for 4 weeks also in both groups (64% in 12.9 mg OEP/kg and 72% in 25.8 mg OEP/kg). Chronic supplementation at both 12.9 and 25.8 mg OEP/kg decreased TBARS levels in soleus muscle (36 and 31%) and liver (43 and 25%). AOPP was also decreased by both doses in the liver (39 and 45%). Chronic administration of the highest dose, 25.8 mg OEP/kg, was able to reduce mRNA expression of PGC-1α in soleus muscle (25%). No effect was found in other analyses such as spontaneous physical activity, body weight, food and water intake, hepatic toxicity, cardiac oxidative stress and mitochondrial DNA amount. CONCLUSION: Maximum and not recommended doses of OEP ingested acutely presented stimulating effect on the ability to exercise. However, its daily consumption for 4 weeks showed antioxidant effects in soleus muscle and liver which may have decreased the PGC-1α mRNA expression on soleus muscle and contributed to the impaired performance in the exercise tolerance test.

ABSTRACT

Marijuana (Cannabis sativa) is one of the most widely used illicit drugs in the world. Its use is associated with impairments in cognitive function. We previously reported that Δ(9)-tetrahydrocannabinol (Δ(9)-THC), the primary psychoactive component of marijuana, impaired spatial working memory in the radial maze task when injected intracortically (IC) into the medial prefrontal cortex (mPFC) of rats. Here, we used this paradigm to evaluate the involvement of prefrontal dopamine receptors in working memory disruption induced by Δ(9)-THC. Intracortical pre-treatment of animals with either the D(1)- or D(2)-like dopamine receptor antagonists SCH 23390 or clozapine, respectively, significantly reduced the number of errors rats made in the radial maze following treatment with Δ(9)-THC also administered intracortically. These results were obtained in the absence of locomotor impairment, as evidenced by the time spent in each arm a rat visited. Our findings suggest that prefrontal dopamine receptors are involved in Δ(9)-THC-induced disruption of spatial working memory. This interaction between the cannabinoid system and dopamine release in the PFC contributes to new directions in research and to treatments for cognitive dysfunctions associated with drug abuse and dependence.

INTRODUCTION: AIDS among older adults is a public health problem emerging. This study compared the demographic and epidemiological profile and the evolution and trend of the epidemic among men and women aged 50 years and older and 20 to 39 years affected by AIDS in the State of Espirito Santo, Brazil. METHODS: We conducted a time serie study with secondary data from SINAN/AIDS for the period January 1991 to December 2006. RESULTS: In this period were 3,382 reported cases of AIDS in individuals aged 20 to 39 years and 551 cases among individuals with 50 years or older. In both age groups most affected are men. There are differences related to race or color, where the majority of the older is white (45.3% - p-value = 0.044) and the young brown (44.7%, p = 0.003). Illiteracy prevails among the older (17.7% - p-value = 0.001). More than half (80%) of the notifications occurred in cities of medium to large. The main risk factor was heterosexual in both the age groups more frequently to the group of 50 or more (77.3% - p = 0.0001). The cumulative incidence is higher for the age group 20 to 39 years (R(2) = 0.68), but is increasing proportionally between the two bands over the years, with a significant upward trend for both (p<0.01). CONCLUSIONS: The AIDS epidemic among the elderly can be seen growing among older on the Espirito Santo State.

INTRODUÇÃO: A AIDS entre adultos mais velhos é um problema de saúde pública emergente. Comparamos o perfil epidemiológico e sociodemográfico, bem como a evolução e a tendência da epidemia entre homens e mulheres nas faixas etárias de 50 anos e mais e 20 a 39 anos acometidos pela AIDS, no Estado do Espírito Santo, Brasil. MÉTODOS: Realizamos um estudo de série temporal, com dados secundários do SINAN/AIDS, no período de Janeiro de 1991 a dezembro 2006. RESULTADOS: Neste período, foram notificados 3.382 casos de AIDS em indivíduos com idade entre 20 e 39 anos e 551 casos entre indivíduos com 50 anos e mais. Em ambas as faixas etárias, os mais acometidos são os homens. Há diferenças referentes à raça/cor, em que a maioria dos mais velhos são brancos (45,3 por cento - p-valor = 0,044) e os mais jovens pardos (44,7 por cento, p-valor = 0,003). O analfabetismo prevalece entre os mais velhos (17,7 por cento - p-valor = 0,001). Mais da metade (80 por cento) das notificações ocorreu em municípios de médio a grande porte. A principal categoria de exposição foi a heterossexual, em ambos as faixas etárias, com maior frequência para o grupo de 50 anos ou mais (77,3 por cento - p=0,0001). A incidência acumulada é maior para a faixa etária de 20 a 39 anos (R²=0,68); porém, vem aumentando proporcionalmente, entre as duas faixas no decorrer dos anos, com tendência de crescimento significativa para ambas (p <0,01). CONCLUSÕES: A epidemia de AIDS pode ser considerada em expansão entre mais velhos no Espirito Santo.

ABSTRACT

Dopamine (DA) and N-methyl-D-aspartate (NMDA) receptors seem to be critically involved in working memory processing in the medial prefrontal cortex (mPFC). Effects of NMDA receptors blockade on dopamine D1 receptors activation in the mPFC on spatial working memory was investigated. Adult male Wistar rats, well trained in an eight-arm radial maze and bilaterally cannulated in the mPFC, received intracortical administrations of saline (SAL) or SKF-38393 (DA D1 receptor agonist) followed, 10 min later, by MK-801 (non-competitive NMDA receptor antagonist). They were tested in 1 h delayed tasks after 5 min of the second administration. SKF-38393 (0.56 and 1.8 microg) was disruptive to working memory, increasing significantly the number of errors in the 1 h post-delay performance when administered into the mPFC. MK-801, at doses with no significant effects alone (0.32 or 1.0 microg), reduced significantly the disruptive effect of 0.56 microg SKF-38393. These results showed that the disruptive effect of DA D(1) receptors activation in the mPFC on working memory was significantly reduced by an open-channel NMDA receptor blockade, suggesting that the processing of working memory in the mPFC involving DA D1 receptors depend, at least in part, of NMDA receptors activity in this cortical area.
