ABSTRACT

A pure water soluble xylan (PS-I) with an average molecular weight ~1.1 × 105 Da was isolated from the hot water extraction of fresh leaves of Piper betle (paan). The xylan was found to be composed of xylose, galactose and methyl galacturonate in a molar ratio of nearly 3:1:1. The repeating unit was composed of a backbone containing three (1 â†’ 4)-α-D-Xylp residues, one of which was branched at O-2 position with the side chain consisting of (1 â†’ 4)-α-D-GalpA6Me and terminal ß-D-Galp residues. This xylan exhibited macrophage, splenocyte, and thymocyte stimulatory activities. In vitro antioxidant studies demonstrated that the xylan has DPPH radical scavenging potential (EC50 = 148 µg/mL), ABTS radical quenching activity (EC50 = 188 µg/mL) and ferrous ions chelating activity (EC50 = 370 µg/mL). These findings elicit the need for further exploration of the xylan as a natural antioxidant and a potent immunostimulating agent.

ABSTRACT

A water soluble polysaccharide (PGPS) with molecular weight ~ 1.4 × 105 Da was isolated by alkali treatment from an edible mushroom Polyporus grammocephalus and purified by gel chromatography using sepharose-6B column. Monosaccharide analysis revealed that PGPS was made up of glucose only. PGPS contained (1 â†’ 3)-α-D-Glcp and (1 â†’ 4)-α-D-Glcp moieties in a molar ratio of nearly 1:2. Through a series of chemical and spectroscopic (1D/2D NMR) investigations, the repeating unit of the glucan was established as: →3)-α-D-Glcp(1 â†’ [4)-α-D-Glcp(1]2→ This α-glucan was observed to stimulate some prime components of immune system, namely, macrophages, splenocytes, and thymocytes.

ABSTRACT

Nanocarrier mediated targeted delivery and biosensing in reproductive health care is a major exploratory domain. This work demonstrates the loading of silver nanoparticle (AgNP) inside the multiwalled carbon nanotube (MWCNT) and their targetability to the intracellular part of the sperm cell for its further application in biosensing based infertility diagnosis. Ocimum tenuiflorum (tulsi extract) mediated photosynthesized AgNP exhibited spherical shape, 5-40 nm size and surface plasmonic resonance at 430 nm. After loading of freshly prepared AgNP into emulsified MWCNT, the loading was confirmed with spectroscopic and microscopic methods. FTIR analysis displayed significant shifting at 3450 cm-1 (-OH stretching) and 1615 cm-1 (CNT back bone) which validated the binding of AgNP with MWCNT and interestingly heat flow analysis revealed that Ag loaded MWCNT has greater stability than AgNP. Moreover, AFM based surface profile height analysis clearly showed the loading of AgNP inside MWCNT as surface height of MWCNT increased from 22 to 32 nm, which in turn confirmed the encapsulation of 10 nm size of AgNP inside the tube. Furthermore, surface enhanced Raman spectroscopy (SERS) confirmed the homogeneous loading as there were no changes in D/G ratio. SERS analysis for the interaction of AgNP loaded MWCNT with freshly collected healthy, motile human spermatozoa showed a significant Raman shift at 800-780 cm-1 (NH2+ twist) and 1050-1060 cm-1 (vas PO3-) for change in DNA packaging process and its stabilizing protein polyamine respectively. Finally, DNA fragmentation and morphological examination confirmed the binding and targetability of AgNP to the sperm nucleus. Improved targeting efficiency and biosenssing ability make AgNP-MWCNT composite suitable in fertility diagnosis.

ABSTRACT

A water-soluble glucan was isolated from the alkaline extract of an ectomycorrhizal fungus, Ramaria botrytis. On the basis of sugar analysis, methylation analysis, Smith degradation, partial hydrolysis, and 1D/2D NMR studies, the structure of the repeating unit of the glucan was established as: [structure: see text]. This glucan showed immunostimulating activity by NO production on RAW 264.7, a murine macrophage cell line. Splenocyte and thymocyte proliferation were measured using, respectively, single cell suspensions of spleen and thymus obtained from normal mice.

A water-insoluble glucan, PFPSIN, has been isolated from the aqueous extract of an edible mushroom Pleurotus florida. On the basis of total acid hydrolysis, methylation analysis, periodate oxidation, Smith degradation, and (13)C NMR experiments, the repeating unit of the polysaccharide was established as Conformational analysis revealed the triple helical conformation of this glucan.

ABSTRACT

A polysaccharide fraction (AQS-II) has been isolated from the hot aqueous extract of the fruits of an ectomycorrhizal fungus Astraeus hygrometricus. It was found to contain 63% polysaccharide and 35% protein. The polysaccharide part contains glucose, galactose, and fucose in a 2:1:1 molar ratio. On the basis of total acid hydrolysis, methylation analysis, periodate oxidation, and NMR studies ((1)H, (13)C, DQF-COSY, TOCSY, NOESY, HMBC, and HSQC) the structure of the repeating unit of the polysaccharide was established as [structure: see text].

ABSTRACT

A water-insoluble glucan, TEINS has been isolated from the hot alkaline extract of an edible mushroom Termitomyces eurhizus. The total carbohydrate content of the polysaccharide fraction was found to be 98.4%, and it was found to contain only glucose as the monosaccharide constituent. On the basis of total acid hydrolysis, a methylation experiment, periodate oxidation and (13)C NMR experiment, the repeating unit of the polysaccharide was established as: -->3)-beta-D-Glcp-(1-->.

ABSTRACT

The structure of two polysaccharides isolated from the hot aqueous extract of fruiting bodies of the mushroom, Termitomyces eurhizus, have been reinvestigated. These consist of two homogeneous fractions PS-I and PS-II. PS-I contains only D-glucose as the monosaccharide constituent. From methylation analysis and periodate oxidation studies, followed by GLC-MS analysis the linkages, the sugar units in PS-I were identified as (1-->3)-D-Glcp and (1-->6)-D-Glcp. PS-II contains D-glucose, and the mode of linkage of d-glucose was identified as (1-->6)-D-Glcp. Finally, the following possible structures of the polysaccharides were assigned using 1H, 2D-COSY, TOCSY, NOESY and 13C NMR spectral analysis: [carbohydrate structure: see text].
