ABSTRACT

Indigenous chicken breeds in developing countries have diverse benefits to rural economy as a source of high-quality animal protein. However, there are few reports on the evaluation of economic traits in Egyptian indigenous breeds. Hence, this study aimed to investigate growth performance, carcass characteristics, body measurements and meat quality traits in two indigenous breeds of chickens (Benha line and Golden Montazah) versus Rhode Island Red as a reference worldwide breed. Besides, a time series expression profile of somatotropic axis genes including GH and IGF-1 and their plasma level concentrations were investigated. Benha line chickens (BL) revealed the highest improved estimates of growth performance, carcass characteristics and meat quality traits. In the same manner, it displayed the highest levels of hepatic GH and IGF-1 and muscle IGF-1 gene expression compared to Rhode Island Red (RIR) and Golden Montazah (GM) chickens. Accordingly, BL exhibited the highest levels of plasma IGF-1 and the lowest levels of plasma GH. This result suggests the direct association between growth performance, carcass characteristics and levels of IGF-1 gene expression in the selected chicken breeds. BL is a superior Egyptian genotype with candidate productive traits and competing characteristics, it could be used widely as a proven ancestor of commercial hybrid breeds.

ABSTRACT

Two novel transitional organic Zn/Cu complexes based on a new biocompatible bidentate triazine-hydrazone ligand (Thz) was designed, synthesized, and evaluated in this study. This study evaluated the effects of injecting 60 mg of Zn and 40 mg of Cu in three different forms, twice per week, for eight weeks on growth performance, expression of growth factors and cytokine genes, carcass yield, blood biochemicals, and intestinal morphology in weaned rabbits. The tested complexes were sulfate (Cu/ZnSO4), montmorillonite (Cu/Zn-Mnt), and triazine hydrazone (Cu/Zn-Thz). A total of 60 V-line weaned rabbits at four weeks of age were assigned to four treatments (n = 15), which were intramuscularly injected with 0.5 mL of either (1) saline (control) or saline containing (2) Cu/ZnSO4, (3) Cu/Zn-Mnt, or (4) Cu/Zn-Thz. Compared to the controls, the rabbits injected with Cu/Zn-Thz showed a higher (p < 0.01) growth rate, carcass yield (p < 0.05), and liver expression of insulin like growth factor-1 (IGF-1), growth hormone receptor (GHR), fibroblast growth factor-1 (FGF1), and transforming growth factor beta-1 (TGFB1) (p < 0.05), as well as better jejunum morphometric variables (p < 0.05). On the other hand, mRNA of FGF1, TGF1, TCIRG1, and adenosine deaminase (ADA) were higher expressed (p < 0.05) in the spleen tissues of Cu/Zn-Mnt group. Collectively, the results indicated that our novel synthesized organic complexes of Zn/Cu-Thz proved to be a suitable feed supplement, as it increased rabbit productive performance through enhancing expression of peptide growth factors and cytokine genes.

ABSTRACT

Rapid identification of Campylobacter-positive flocks before slaughter, following freezing and heat treatment for the Campylobacter-positive carcasses at the slaughterhouses is an effective control strategy against foodborne campylobacteriosis. We evaluated a loop-mediated isothermal amplification (LAMP) assay for the direct screening of naturally contaminated chicken cloacal swabs for C. jejuni/C. coli to compare this assay with conventional quantitative culture methods. In a comparison study of 165 broilers, the LAMP assay showed 82.8% (48/58 by conventional culture) sensitivity, 100% (107/107) specificity, 100% (48/48) positive predictive value (PPV), and 91.5% (107/117) negative predictive value (NPV). In a comparison of 55 flocks, LAMP showed 90.5% (19/21) sensitivity, 100% (34/34) specificity, 100% (19/19) PPV, and 94.4% (34/36) NPV. In the cumulative total of 28 farm-level comparisons, LAMP showed 100% (12/12) sensitivity, 100% (16/16) specificity, 100% (12/12) PPV, and 100% (16/16) NPV. The LAMP assay required less than 90 min from the arrival of the fecal samples to final results in the laboratory. This suggests that the LAMP assay will facilitate the identification of C. jejuni/C. coli-positive broiler flocks at the farm level or in slaughterhouses before slaughtering, which would make it an effective tool in preventing the spread of Campylobacter contamination.

ABSTRACT

The growth kinetics of Salmonella Enteritidis in raw beef has been little studied so far. Thus, this study aimed to clarify the growth kinetics of the pathogen in ground beef using a growth model. When Salmonella cells inoculated at various initial doses into ground beef were incubated at a given temperature (24℃), the maximum population (Nmax) of the microbe at the stationary phase varied with the doses. This relationship was expressed with a polynomialequation for Nmax using the initial dose. The combination of the growth model and the polynomial equation successfully predicted Salmonella growth at a given initial dose. When Salmonella cells inoculated in ground beef were incubated at various constant temperatures, the growth curves of the pathogen and natural microflora (NM) were well described with the growth model. The rate constant of growth and the Nmax values for Salmonella and NM were then analyzed kinetically. From these results, growth curves of Salmonella and NM in ground beef stored at dynamic temperatures were successfully predicted. Competition between Salmonella and NM in ground beef was also found during the storage. This study could give usable information on the growth of Salmonella and NM in ground beef at various temperatures.

ABSTRACT

Recently we clarified the growth kinetics of Salmonella Enteritidis in raw ground beef at various temperatures with our growth model. Based on those results, this study aimed to build a new methodology to predict the growth of Salmonella in ground beef at given initial concentrations of the pathogen and temperatures. Namely, the maximum cell population of Salmonella at various combinations of its initial concentration and temperature was developed with a polynomial equation. The rate constants of Salmonella growth at various temperatures were estimated with the square root model studied in our recent study. A new system consisting of our growth model, the polynomial equation, and the square root model successfully predicted the growth of Salmonella inoculated at given concentrations in beef at constant and dynamic temperatures. The growth of natural microflora in beef at those temperature patterns were also successfully predicted with the growth model.
