ABSTRACT

The first high-protein rice variety of India, CR Dhan 310, developed at ICAR-NRRI, Cuttack is being selected for the study. It contains 10.1% protein in milled rice as compared to 6-7% protein content in the milled rice of any other normal variety. It has intermediate amylose content (25.1%), medium bold grains rich in protein (10.1%) The significant changes in properties of raw and parboiled rice on processing were studied at statistical differences of p ≤ 0.05. These properties included physical, optical, antioxidant and rheological properties which changed with different processing techniques. All the three processes namely, puffing, popping and flaking increased the dimensions as compared to the raw rice. Peak viscosity measurements demonstrated the breakdown of starch molecules, with white rice having the greatest value (4145 cP) and popped rice having the lowest value (2017 cP) as a result of the starch granules being gelatinized during the production of popped rice. Highest anthocyanin content (2.93 mg/100 g) was observed in puffed rice, phenolic content (347.93 mg/100 g) was highest in popped rice and flaked/flattened rice showed highest flavonoid content (127.12 mg/100 g) indicating that tertiary processing of rice obtained higher values of phytochemicals when compared to the plain high-protein rice. This indicates that the processed products of rice can be consumed directly as ready-to-eat or can be used in preparation of other functional foods to combat malnutrition and build nutritional security. The study indicates that processing could improve the nutritional quality of the rice products.

ABSTRACT

Reeta is a popular late-maturing high-yielding rice variety recommended for cultivation in the eastern Indian states. The cultivar is highly sensitive to submergence stress. Phosphorus deficiency is an additional constraint for realizing high yield. The quantitative trait loci (QTLs), Sub1, for submergence and Pup1 for low phosphorus stress tolerance along with narrow-grained trait, GW5 were introgressed into the variety from the donor parent, Swarna-Sub1 through marker-assisted breeding. In addition, phenotypic selections for higher panicle weight, grain number, and spikelet fertility were performed in each segregating generation. Foreground selection detected the 3 target QTLs in 9, 8 and 7 progenies in the BC1F1, BC2F1, and BC3F1 generation, respectively. Recurrent parent's genome recovery was analyzed using 168 SSR polymorphic markers. The foreground analysis in 452 BC3F2 progenies showed five pyramided lines in homozygous condition for the target QTLs. No donor fragment drag was noticed in the Sub1 and GW5 QTLs carrier while a segmentwas observed in the Pup1 carrier chromosome. The developed lines were higher yielding, had submergence, and had low phosphorus stress-tolerance alongwith similar to the recipient parent in the studied morpho-quality traits. A promising pyramided line is released in the name of Reeta-Panidhan (CR Dhan 413) for the flood-prone areas of Odisha state.

ABSTRACT

Seed vigor is the key performance parameter of good quality seed. A panel was prepared by shortlisting genotypes from all the phenotypic groups representing seedling growth parameters from a total of 278 germplasm lines. A wide variation was observed for the traits in the population. The panel was classified into four genetic structure groups. Fixation indices indicated the existence of linkage disequilibrium in the population. A moderate to high level of diversity parameters was assessed using 143 SSR markers. Principal component, coordinate, neighbor-joining tree and cluster analyses showed subpopulations with a fair degree of correspondence with the growth parameters. Marker-trait association analysis detected eight novel QTLs, namely qAGR4.1, qAGR6.1, qAGR6.2 and qAGR8.1 for absolute growth rate (AGR); qRSG6.1, qRSG7.1 and qRSG8.1 for relative shoot growth (RSG); and qRGR11.1 for relative growth rate (RGR), as analyzed by GLM and MLM. The reported QTL for germination rate (GR), qGR4-1, was validated in this population. Additionally, QTLs present on chromosome 6 controlling RSG and AGR at 221 cM and RSG and AGR on chromosome 8 at 27 cM were detected as genetic hotspots for the parameters. The QTLs identified in the study will be useful for improvement of the seed vigor trait in rice.

ABSTRACT

The commercial significance of accurate and simple quantification of 2-Acetyl-1-pyrroline (2-AP) cannot be overstated. Present study was carried out to standardize a method for extraction and accurate quantitation of 2-AP from rice grain using GC-MS/MS equipped with HS-SPME auto sampler. The effect of sample quantity, addition of solvent, grinding process, sample particle size, head space parameters and SPME fiber incubation parameters, were optimized in the developed method. Dehusked rice powder (2 g) prepared under liquid nitrogen, and passed through the 80-mesh sieve, incubated for 40 min at 80 °C in headspace, followed by fiber (DVB/Carbon WR/PDMS) saturation time of 15 min, could produce the maximum response. The recovery of 2-AP from fortified sample ranged between 7.02 and 9.02% at 50-200 ng g-1 fortification irrespective of the grain matrices used. Standard addition method was appropriate to overcome the matrix effect and recovery of 2-AP was more than 90% using this method. The developed method was further utilized for quantification of 2-AP in four Basmati and two non-Basmati aromatic rice samples. The content of 2-AP ranged between 57.17 and 147.10 ng g-1 of rice and varied with geographical location. This fully automated method could improve the work efficiency and reduce error during the volatile extraction and adsorption phase. ABSTRACT

Because it is rich in antioxidant compounds, the staple food of rice provides many health benefits. Four antioxidant traits in rice grain, viz., catalase, CUPRAC, DPPH, FRAP and peroxidase, were mapped in a representative panel population containing 117 germplasm lines using 131 SSR markers through association mapping. Donor lines rich in multiple antioxidant properties were identified from the mapping population. The population was classified into three genetic groups and each group showed reasonable correspondence with the antioxidant traits. The presence of linkage disequilibrium in the population was confirmed from the estimated Fst values. A strong positive correlation of DPPH was established with TPC, FRAP and CUPRAC. A moderate to high mean gene diversity was observed in the panel population. Eleven significant marker-trait associations for antioxidant traits were mapped, namely, qACD2.1, qACD11.1 and qACD12.2 for DPPH; qCAT8.1 and qCAT11.1 for catalase; qFRAP11.1, qFRAP12.1 and qFRAP12.2 for FRAP; and qCUPRAC3.1, qCUPRAC11.1 and qCUPRA12.1 regulating CUPRAC. Co-localization of the QTLs for qACD11.1, qFRAP11.1 and qCUPRAC11.1 were detected, which may act as antioxidant hotspots regulating DPPH, FRAP and CUPRAC activities, respectively, while qACD12.2 and qFRAP12.1 remained close on the chromosome 12. These detected QTLs will be useful in antioxidant improvement programs in rice.

ABSTRACT

Assessing genetic variability of micronutrient content in association with qualitative and quantitative traits in germplasm is prerequisite for effective biofortification programme. Odisha, a state of eastern India is considered as one of the most potential hot spot of diversity of cultivated rice for grain yield and nutritional traits. Significant variability for most of the qualitative and quantitative traits including Fe and Zn content was observed in a set of 293 germplasm with varying kernel colour encompassing 14 districts of Odisha. Mostly these landraces were low yielding with some exception (Haldigundi: AC 36454, 50.08 g/plant). These landraces were mostly represented by medium Fe (10-20 ppm)-medium Zn group (20-30 ppm). Fe and Zn content had positive association with each other and also with grain size. Landraces with red kernel colour were observed to have slightly higher average Zn content (26.30 ppm) as compared to white (25.87 ppm) grains. Diversity analysis of 14 districts revealed that Nayagarh, a south-eastern district was rich in Fe content while Deogarh, Keonjhar and Mayurbhanj, all north-western districts were rich in Zn content. This study identified 10 superior micronutrient dense genotypes with medium to high Fe and Zn content. This set of donors for micronutrient content was validated in another year. Champeisiali (AC 43368) and Gedemalati (AC 34306) with highest Fe (44.1 ppm) and Zn (40.48 ppm) content, respectively were detected over the environments. Identified donors and associated traits could be utilized in biofortificaion programme using appropriate breeding methodologies for enhancing micronutrients in high yielding background. ABSTRACT

Eighteen physio-biochemical traits influencing seed vigour were studied for their association with molecular markers using a mini core set constituted from 120 germplasm lines. High genetic variation was detected in the parameters namely chlrophyll a, Chlrophyll b, total chlorophyll, carotenoids, total anthocyanin content, gamma-oryzanols, total phenolics content, superoxide dismutase, catalase, guaicol peroxidase, total soluble sugar, total protein, seed vigour index -I and seed vigour index -II. Strong positive correlation of seed vigour index II was observed with amylose content, total anthocyanin content, catalase, total phenolic content and total flavonoid content while a negative association was observed for gamma-oryzanol content. High gene diversity (0.7169) and informative markers value (0.6789) were estimated from the investigation. Three genetic structure groups were observed in the panel population and genotypes were grouped in the subpopulations based on the seed vigour trait. Differences in the fixation indices of the three sub populations indicated existence of linkage disequilibrium in the studied panel population. Association of the traits namely total flavonoids, superoxide dismutase, catalase, chlorophyll a, Chlorophyll b, total chlorophyll, carotenoids, starch, amylose, total anthocyanin, gamma-oryzanol, total phenolics with the molecular markers were detected by Generalized Linear Model and Mixed Linear Model showing > 0.10 R2 value. Association of the trait, total flavonoids with marker RM7364 located on chromosome 8 reported in earlier study was validated in this investigation. The validated markers and the novel markers detected showing higher R2 value will be useful for improvement of seed vigour in rice.

ABSTRACT

This study was aimed to evaluate the physicochemical, textural, sensory and antioxidative attributes of various rice products such as boiled rice, beaten rice, popped rice, puffed rice and raw milled rice, prepared from the Indian black rice cultivars Chakha (CH), Kalobhat (KB), Mamihunger (MA) and Manipuri Black (MN). A popular white rice variety Swarna Sub-1 (SW) was considered as control. Significant differences in most of the physicochemical and cooking parameters of raw rice were observed across different cultivars. The head rice recovery, amylose content, elongation ratio (ER) and kernel length after cooking of MA were most satisfactory among the black rice cultivars and are found to be 50.67%, 17.6%, 1.87 and 10.10 mm respectively, while popped rice of MA recorded highest length (10.83 mm) and elongation ratio (ER; 2.01). MA showed the highest adhesiveness (11.18 mJ) in boiled rice but hardness (183.53 N) was medium in raw rice. Other textural quality varied differentially according to the various products and cultivars. The highest a* value was obtained from puffed rice of MA (6.61) but L value was highest in raw rice of MN. Popped and boiled rice of MA displayed higher DPPH-antioxidant activity (88.74% and 84.74% respectively) as compared to all other products. The raw rice of KB registered higher anthocyanin (57.23 mg/100 g) content while boiled rice of SW recorded the least (0.21 mg/100 g). A survey on the consumer preference of these products indicated that boiled rice was usually preferred in lunch and dinner by most of the consumers whiles other products in breakfast. With respect to most of the traits, MA showed the good potential for rice Industry as well as for breeding material.

ABSTRACT

Eleven pigmented rice genotypes were evaluated to estimate genetic parameters, heritability and association. The results indicated that, genotypic variation was high among the lines. The distinct seasonal effect on plant performance for antioxidant capacity, anthocyanin, flavonoids, head rice recovery and test weights was also observed. Wet season favoured the crop performance in all genotypes as compared to drought conditions. The differential accumulation of different quality traits such as AOA, anthocyanin content, flavonoids content, etc showed high heritability, which would be transfer to high yeilding popular rice cultivars through conventional or geneticaly modification techniques. The line Mamihunger was chosen as donor of the high-quality rice grain and Annapurna for high yield. Further, Mamihunger are foreseen to be good in nutritional quality and industry use.
