ABSTRACT

This study aimed to perform in vitro antiparasitic and antimicrobial tests with the essential oil (EO) of Schinus terebinthifolius against of fish and shrimp. The chemical composition of the EO of S. terebinthifolius was determined by gas chromatography. For the antiparasitic test, the protozoan Epistylis sp. obtained from parasitized Oreochromis niloticus was used, and exposed to different concentrations of EO (2%, 1%, 0.5%, 0.25%), and control with 1% grain alcohol. The Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) test with EO of S. terebinthifolius evaluated the antimicrobial potential, with serial dilutions starting at 2% and control with 1% grain alcohol, using the strains of Aeromonas hydrophila (2.2 × 108 CFU mL-1), Edwardsiella tarda, Vibrio parahaemolyticus, V. harveyi, and V. alginolyticus (2.0 × 108 CFU mL-1). Chemical analysis revealed that the major EO compounds of S. terebinthifolius were δ-3-Carene (56.00%) and α-Pinene (16.89%). In the antiparasitic test, the concentration of 2% EO showed 100% efficacy against Epistylis sp. within 5 min. In the antimicrobial tests, the concentration of 2% EO was effective against all bacteria tested. The EO of S. terebinthifolius demonstrated antiparasitic and antimicrobial activity at a concentration of 2%, standing out as an alternative to conventional antibiotics.

ABSTRACT

This study investigated the in vitro antimicrobial and anthelmintic effect of copper nanoparticles (CuNPs) against the bacterium Aeromonas hydrophila, the monogeneans Dactylogyrus minutus, Dactylogyrus extensus, Gyrodactylus cyprini, and the cestode Schyzocotyle acheilognathi, as well as their toxicity to Cyprinus carpio Koi. In the antimicrobial in vitro test, the inhibition zone method and minimum inhibitory concentration (MIC) were performed. In order to determine the time and efficacy of monogenean parasite mortality, the parasites were exposed to CuNP concentrations of 20, 50, 100, 150, 200, and 300 mg L-1, and a control group with tank water and one with copper sulphate pentahydrate (CuSO4.5H2O) at a concentration of 0.3 mg L-1, performed in triplicate. The parasites were observed every 10 min for 300 min, and mortality was recorded. For the cestodes, parasites were immersed in CuNP concentrations of 50, 100, 150, and 300 mg L-1. At the end of the in vitro tests, the anthelmintic efficacy of each treatment was calculated. To assess the tolerance and toxicity in fish, they were exposed to CuNP concentrations of 0.6, 1.25, 2.5, 5, 10, 20, and 50 mg L-1 for 12 h. The MIC demonstrated that CuNPs effectively inhibited the growth of A. hydrophila up to a dilution of 12,500 mg L-1 and showed an inhibition zone of 14.0 ± 1.6 mm for CuNPs. The results of anthelmintic activity showed a dose-dependent effect of concentration for both groups of parasites, with the most effective concentration being 300 mg L-1 in 120 min. In the toxicity test, the carps showed tolerance to lower concentrations. The study indicated that CuNPs were effective against the studied pathogens. However, it proved to be toxic to fish at high concentrations. The use of low concentrations is recommended still requires further investigation.

ABSTRACT

Zinc is one of the essential microelements for the metabolism of animals. Zinc nanoparticles may have higher bioavailability due to their low specific surface area, facilitating absorption by fish. The present study aimed to evaluate the effects of supplementation with different zinc-based products on the growth and health of Nile tilapia Oreochromis niloticus. Zinc, in different sizes (nanoparticles or bulk) and forms (inorganic or organic), were used as a supplement in the tilapia diet at a dose of 15 mg kg feed-1 for 60 days. At the end of the feeding trial, production performance, hemato-immunological parameters, activity of antioxidant system enzymes, exposure to Streptococcus agalactiae and zinc concentration in the muscle were examined. After the bacterial challenge, the mean corpuscular hemoglobin concentration (MCHC) significantly increased in the fish treated with organic zinc, inorganic nano zinc, and organic nano zinc, while in the control group (inorganic zinc), MCHC remained unchanged. Regarding defense cells, dietary inorganic nano zinc increased the number of basophils (1.50 ± 1.10) compared to organic zinc (0.80 ± 0.90). Lymphocyte count increased after the challenge only in the organic zinc treatments (bulk and nanoparticles). Neutrophils decreased in the control (inorganic zinc) (2.20 ± 1.70) and inorganic nano zinc (2.60 ± 2.70) treatments after the challenge. When compared before and after the bacterial challenge, the plasma antimicrobial titer significantly increased after the bacterial challenge in all treatments. No significant differences were observed for total proteins, enzymes (SOD and CAT), cumulative survival and zinc deposition on fillet. In conclusion, organic zinc in nanoparticles or bulk size increased Nile tilapia innate defense during bacterial infection. However, the other parameters evaluated were not affected by zinc particle size or form (organic or inorganic), indicating that further evaluations should be conducted with organic zinc in nanoparticles or bulk size in the tilapia diet.

Background: The resurgence of Mycobacterium tuberculosis (Mtb) strains that resist anti-tuberculosis (anti-TB) drugs used currently stresses the search for more effective low-toxicity drugs against new targets. Due to their role in ion homeostasis and virulence, Mtb plasma membrane P-type ATPases are interesting anti-TB targets, in particular, the Ca2+ transporting P2-type ATPase CtpF which is involved in oxidative stress response and persistence. Methods: In this study, the effect on the transcription level of the ctpF gene and other Mtb P2-type ATPases of two anti-Mtb hits was assessed by quantitative reverse transcription polymerase chain reaction (qRT-PCR). Both anti-Mtb hits ZINC14541509 and ZINC63908257 had been previously identified using pharmacophore-based virtual screening and MM-GBSA binding free energy. In addition, the bacterial activity of both compounds on Mycobacterium bovis was evaluated to see whether or not there is an effect on other mycobacteria of the Mtb complex. Results: qRT-PCR experiments showed that the ctpF transcription level was significantly higher in the presence of both compounds, especially ZINC14541509, strongly suggesting that CtpF may be a specific target of the selected compound. Conclusions: ZINC14541509 should be considered as an alternative for the structural-based design of novel anti-TB drugs.

ABSTRACT

CtpF is a Ca2+ transporter P-type ATPase key to the response to stress conditions and to Mycobacterium tuberculosis virulence, therefore, an interesting target for the design of novel anti-Mtb compounds. In this work, molecular dynamics simulations of four previously identified CtpF inhibitors allowed recognizing the key protein-ligand (P-L) interactions, which were then used to perform a pharmacophore-based virtual screening (PBVS) of 22 million compounds from ZINCPharmer. The top-rated compounds were then subjected to molecular docking, and their scores were refined by MM-GBSA calculations. In vitro assays showed that ZINC04030361 (Compound 7) was the best promising candidate, showing a MIC of 25.0 µg/mL, inhibition of Ca2+-ATPase activity (IC50) of 3.3 µM, cytotoxic activity of 27.2 %, and hemolysis of red blood cells lower than 0.2 %. Interestingly, the ctpF gene is upregulated in the presence of compound 7, compared to other alkali/alkaline P-type ATPases coding genes, strongly suggesting that CtpF is a compound 7-specific target.

ABSTRACT

Analisar o perfil epidemiológico da sífilis congênita no Paraná entre os anos de 2012 a 2021. Estudo epidemiológico observacional descritivo, do tipo levantamento, a partir de dados do Sistema de Informação de Agravos de Notificação disponíveis no DataSUS. O número de casos da sífilis congênita teve aumento de 19,7 a cada 10 mil nascidos vivos em 2012 para 51,4 em 2020. Das 22 regionais, Paranaguá, Pato Branco e Metropolitana registraram maior prevalência. O diagnóstico prevaleceu em crianças com até 6 dias de idade, da raça branca e resultante na cura diante do tratamento. Das mães, prevaleceu aquelas com baixa escolaridade. A maior parte dos casos ocorreu entre gestantes que realizaram o pré-natal, porém com parceiros não tratados. Há necessidade de investimento na educação em saúde às gestantes e educação continuada aos profissionais de saúde para o diagnóstico, monitoramento e tratamento da sífilis em gestante e, especialmente, nos parceiros.


To analyze the epidemiological profile of congenital syphilis in Paraná from 2012 to 2021. Observational descriptive epidemiological study, survey type, based on data from the Information System of Notifiable Diseases available at DataSUS. The number of cases of congenital syphilis increased from 19.7 per 10,000 live births in 2012 to 51.4 in 2020. Of the 22 regional units, Paranaguá, Pato Branco, and Metropolitana reported the highest prevalence. The diagnosis prevailed in white children of up to 6 days old, resulting in cure after treatment. Of the mothers, those with low level of education prevailed. Most cases occurred among pregnant women who had prenatal care, but with untreated partners. There is a need for investment in health education for pregnant women and continued education for health professionals for the diagnosis, monitoring, and treatment of syphilis in pregnant women and especially in partners.

Estudio epidemiológico observacional, descriptivo, tipo encuesta, basado en datos del Sistema de Información de Enfermedades de Declaración Obligatoria disponible en DataSUS. El número de casos de sífilis congénita aumentó de 19,7 por 10.000 nacidos vivos en 2012 a 51,4 en 2020. De las 22 regiones, Paranaguá, Pato Branco y Metropolitana registraron la mayor prevalencia. El diagnóstico prevaleció en niños de hasta 6 días de vida, blanco y resultando en la cura después del tratamiento. Entre las madres, predominaron las de baja escolaridad. La mayoría de los casos ocurrió entre embarazadas que tuvieron control prenatal, pero con parejas no tratadas. Es necesario invertir en la educación sanitaria de las gestantes y en la formación continuada de los profesionales de salud para el diagnóstico, seguimiento y tratamiento de la sífilis en embarazadas y, especialmente, en las parejas.

OBJECTIVE: To investigate the epidemiology of burns that occurred within homes during the COVID-19 pandemic in Brazil. DESIGN AND SETTINGS: This was a quantitative, descriptive, and cross-sectional study with a non-probabilistic sample. METHODS: Data were collected through the distribution of survey links on social networking sites and websites, and through email between December 2020 and February 2021. Participants were over 18 years of age, living in Brazil. Data analysis was performed using descriptive and dispersion statistics. RESULTS: A total of 939 adults (aged > 18 years) participated in this study. The mean age was 37.2 years (standard deviation [SD] = 12.5), 75.6% were female, 70.0% self-reported white skin color, 74% had completed higher education, and 28.1% had an income of 3 to 6 times the monthly minimum wage. A total of 21.6% suffered burns during the pandemic, 44.3% from a hot object. Approximately 49.3% never had access to a burn prevention campaign. CONCLUSION: It is necessary to develop burn prevention strategies that reach a wider population and to strengthen public policies to reduce the prevalence of domestic burns, especially during the pandemic.

Este estudo teve o objetivo de identificar a percepção sobre o envelhecimento dos idosos usuários de uma Unidade de Atenção ao Idoso de um município brasileiro e avaliar a relação entre características sociodemográficas e de autopercepção sobre o envelhecimento. Estudo de caráter quantitativo, analítico, observacional e transversal realizado com 228 idosos. Foram utilizados os seguintes instrumentos: questionário sociodemográfico, Mini Exame do Estado Mental, APQ (Aging Perception Questionnaire), WHOQOL-Bref, WHOQOL OLD e ASKAS. Foram realizadas análises bivariada e multivariada. Encontrou-se predomínio do sexo feminino, com média de idade de 78,86 anos. A autopercepção sobre o envelhecimento foi moderada. Idosos do sexo feminino e aqueles que residem com filhos demonstraram maior autopercepção do envelhecimento.


This study aimed to identify the perception of aging of elderly users of an Elderly Care Unit in a Brazilian city and to evaluate the relationship between sociodemographic characteristics and self-perception of aging. An observational and descriptive, cross-sectional study with 228 elderly people. The following instruments were used: sociodemographic questionnaire, Mini Mental State Examination, APQ (Aging Perception Questionnaire), WHOQOL-Bref, WHOQOL OLD and ASKAS. Bivariate and multivariate analyzes were performed. There was a predominance of females, with a mean age of 78.86 years. Self-perception of aging was moderate. Elderly females and those who live with children have greater self-perception of aging.

Soil-transmitted helminths, such as Ascaris lumbricoides, are the most prevalent parasites globally. Optimal anthelmintic treatment for A. lumbricoides in endemically infected communities is challenged by several host-related and environmental factors influencing infection acquisition. We assessed the risk of A. lumbricoides (re)infection after treatment in a Venezuelan rural community. Individual merthiolate-iodine-formaldehyde-fixed faecal samples were collected from 224 persons before a single-dose pyrantel treatment and at 1, 3, 6, 9 and 15 months after treatment. Effects of age, sex and socioeconomic status (SES) on A. lumbricoides prevalence, eggs/gram faeces (EPG) and infection (re)acquisition were assessed using both generalised linear mixed-effects models and survival analysis. Pre-treatment A. lumbricoides prevalence was 39.7%. Higher prevalence was associated with younger age and lower SES. Higher EPG values were observed among young children. Median time to A. lumbricoides infection was six months after treatment: at 1, 3, 6, 9 and 15 months post-treatment, cumulative incidence was 6.7%, 18.9%, 34.6%, 42.2%, and 52.6%, respectively. Younger age, lower SES, and pre-treatment A. lumbricoides infection status showed significantly elevated hazard ratios. Mass drug administration protocols would benefit from considering these factors in selective treatment strategies and possibly more than just annual or biannual treatments in the target population.

ABSTRACT: Accumulating evidence indicates that transient receptor potential (TRP) channels are involved in the pathophysiological process in the heart, and monoterpenes, such as carvacrol, are able to modulate these channels activity. In this article, our purpose was to evaluate the direct cardiac effect of carvacrol on the contractility of cardiomyocytes and isolated right atria from spontaneously hypertensive and Wistar Kyoto rats. In this way, in vitro experiments were used to evaluate the ventricular cardiomyocytes contractility and the Ca2+ transient measuring, in addition to heart rhythm in the right atria. The role of TRPM channels in carvacrol-mediated cardiac activities was also investigated. The results demonstrated that carvacrol induced a significant reduction in ventricular cell contractility, without changes in transient Ca2+. In addition, carvacrol promoted a significant negative chronotropic response in spontaneously hypertensive and Wistar Kyoto rats' atria. Selective blockage of TRPM channels suggests the involvement of TRP melastatin subfamily 2 (TRPM2), TRPM4, and TRPM7 in the carvacrol-mediated cardiac effects. In silico studies were conducted to further investigate the putative role of TRPM4 in carvacrol-mediated cardiac action. FTMap underscores a conserved pocket in both TRPM4 and TRPM7, revealing a potential carvacrol binding site, and morphological similarity analysis demonstrated that carvacrol shares a more than 85% similarity to 9-phenanthrol. Taken together, these results suggest that carvacrol has direct cardiac actions, leading to reduced cellular contractility and inducing a negative chronotropic effect, which may be related to TRPM7 and TRPM4 modulation.

ABSTRACT

Introdução: O exame de velocidade de hemossedimentação (VHS) mede a distância em que os eritrócitos caem durante 60 minutos em um tubo vertical. Objetivo: Comparar dois métodos de realização do exame de VHS em um hospital oncológico. Material e métodos: Foram coletados, de 45 pacientes, 5mL a 6mL de sangue que foram transferidos para dois tubos com anticoagulante. O sangue de ambos os tubos foi transferido para uma pipeta de vidro com capacidade de 200mm e 2,5mm de diâmetro interno, que foi adaptada em suporte vertical, aguardando por uma hora. Os resultados foram analisados pelo programa SPSS versão 25. Resultados: O nível de sedimentação em milímetros (mm) do método referência de Westergren modificado utilizando citrato de sódio 3,2% é menor quando comparado com o método alternativo utilizando anticoagulante EDTA, com Z score: -5,29 (p = 0,01) e R = 0,79. Discussão: O International Council for Standardization in Haematology (ICSH) descreveu grandes diferenças entre os métodos de Westergren e os não baseados em Westergren. Conclusão: Sugere-se a utilização do método de Westergren modificado na realização do exame. Além disso, com a padronização do melhor método de realização do exame, gerará resultados mais confiáveis, refletindo de forma positiva na rotina hospitalar.


Introduction: The erythrocyte velocity test (ESR) the distance at which erythrocytes fall within 60 minutes in a vertical tube. Objective: to compare two methods of performing the ESR test in an oncology hospital. Material and methods: 5-6 mL of blood were collected from 45 patients transferred to two tubes with anticoagulant. The blood from both tubes was transferred to a glass pipette with a 200mm capacity and 2.5mm internal diameter, which was adapted on a vertical support, waiting for 1 hour. The results obtained by the SPSS version 25 program. Results: The level of sentiment in mm (mm) of the Westergren reference method modified using sodium citrate 3.2% is lower when compared to the alternative method using EDTA anticoagulant, with Z score: -5.29 (p = 0.01) and R = 0.79. Discussion: The International Council for Standardization in Hematology (ICSH), layouts that greatly differ between Westergren's methods and those not based on Westergren. Conclusion: It is suggested to use the modified Westergren method when performing the exam. In addition, with the standardization of the best method of conducting the exam, it will generate better known results, reflecting positively on the hospital routine.

Sucralose is a widely consumed non-nutritive sweetener (NNS). Studies have shown that some NNS can favor weight gain by altering the intestinal microbiota, satiety hormone production, or aspects related to glucose homeostasis. In this study, we investigated the effects of ad libitum sucralose consumption in mice fed with normal or high-fat diet (HFD) for an extended period (16 weeks). Weight gain, final body composition, energy expenditure, intestinal and pancreatic hormone production, and endotoxemia during a voracity test, as well as liver and skeletal muscles were evaluated after 16 weeks. We observed that sucralose supplementation reduced weight gain in HFD-fed mice but did not change weight gain in mice fed with normal diet. The evaluation of HFD mice showed that sucralose supplementation resulted in improvements in glycemic homeostasis, hepatic steatosis, and increased energy expenditure. Our results suggest that sucralose consumption promotes different outcomes in relation to weight gain when combined with different diets, which may explain the controversial data in previous studies, and can be considered in future clinical research aimed at clarifying the impact of NNS consumption on human health.

O climatério é uma fase natural da vida da mulher que ocorre entre os 40 e 65 anos de idade e é caracterizado pela transição entre a fase reprodutiva e não reprodutiva. Neste período, devido às alterações hormonais, ocorrem alterações biológicas, endócrinas e clínicas. Sintomas vasomotores são típicos do hipoestrogenismo e podem interferir negativamente na qualidade de vida das mulheres. Este estudo teve como objetivo revisar os resultados dos estudos de intervenção que utilizaram isoflavonas na sintomatologia de mulheres climatéricas não usuárias de Terapia de Reposição Hormonal (TRH). Realizou-se uma revisão sistemática de artigos publicados entre os anos de 2008 e 2019 na base de dados PubMed. Foram encontrados 169 estudos, e considerando os critérios de inclusão, 18 artigos foram selecionados, em que houve intervenção com isoflavonas por meio de cápsulas e/ou suplementos ou alimentos para tratamento da síndrome climatérica. Foram verificados resultados positivos nos sintomas globais, com destaque para sintomas vasomotores, em mais da metade dos estudos avaliados, em que doses entre 45 mg a 160 mg diárias de isoflavonas por pelo menos 12 semanas foram administradas, especificadamente nas mulheres no período da pós-menopausa.(AU)

In this period, due to hormonal changes, biological, endocrine and clinical modifications also occur. Vasomotor symptoms are characteristic of hypoestrogenism and can negatively affect womens quality of life. This study aimed to review the results of intervention studies which used isoflavones to treat the symptoms of climacteric women who did not undergo Hormone Replacement Therapy (HRT). A systematic review of articles published between 2008 and 2019 in the PubMed database was carried out. 169 studies were found, and considering the inclusion criteria, 18 articles were selected, in which it was described isoflavones intervention with capsules and/ or supplements or foods for the treatment of climacteric syndrome. Positive results were observed regarding to global symptoms, with emphasis on vasomotor symptoms in more than half of the studies, in which daily doses of isoflavones, between 45 mg to 160 mg, for at least 12 weeks, were administered specifically in postmenopausal women.(AU)

O artigo busca contribuir com a discussão sobre residência multiprofissional e as condições de trabalho dos residentes. Trata-se de uma pesquisa de abordagem qualitativa, que totalizou seis entrevistas através de um roteiro semi-estrututurado, envolvendo diferentes núcleos profissionais, com caráter descritivo e exploratório, sendo os seus resultados apresentados em forma de narrativas, que se misturam com fatos reais e ficcionais, conectando a dureza com a leveza do viver, um processo de criação que expõe um ensaio narrativo. As narrativas expressam a percepção dos residentes a respeito da realidade vivenciada durante o percurso da residência. O que representa este trabalho? Ou qual a representatividade desse trabalho? Aqui se tem um produto da força de trabalho, com seus valores de uso, agregados em dois anos de dedicação exclusiva, no residir nosso de cada dia. A situação pandêmica não nos impediu de realizá-lo, ao contrário, proporcionou transbordar as necessidades vividas pelos residentes/trabalhadores. (AU)

ABSTRACT

Tropical forests have played an important role as a carbon sink over time. However, the carbon dynamics of Brazilian non-Amazon tropical forests are still not well understood. Here, we used data from 32 tropical seasonal forest sites, monitored from 1987 to 2020 (mean site monitoring length, ~15 years) to investigate their long-term trends in carbon stocks and sinks. Our results highlight a long-term decline in the net carbon sink (0.13 Mg C ha-1 year-1) caused by decreasing carbon gains (2.6% by year) and increasing carbon losses (3.4% by year). The driest and warmest sites are experiencing the most severe carbon sink decline and have already moved from carbon sinks to carbon sources. Because of the importance of the terrestrial carbon sink for the global climate, policies are needed to mitigate the emission of greenhouse gases and to restore and protect tropical seasonal forests.

ABSTRACT

The emergence of tuberculosis (TB) produced by multi-drug resistance (MDR) and extensively-drug resistance (XDR) Mycobacterium tuberculosis (Mtb), encourages the development of new antituberculous compounds, as well as the identification of novel drug targets. In this regard, plasma membrane P-type ATPases are interesting targets because they play a crucial role in ion homeostasis and mycobacterial survival. We focused on Mtb CtpF, a calcium P-type ATPase that responds to a broad number of intraphagosomal conditions, as a novel target. In this study, we evaluated the capacity of cyclopiazonic acid (CPA), a well-known inhibitor of the sarco-endoplasmic reticulum Ca2+-ATPase (SERCA), to inhibit the ATPase activity of CtpF and the Mtb growth demonstrating that CtpF is a druggable target. A homology modeling of CtpF was generated for molecular docking studies of CtpF with CPA and key pharmacophoric features were identified, which were used to perform a pharmacophore-based virtual screening of the ZINC database, and to identify CtpF inhibitor candidates. Molecular docking-based virtual screening and MM-BGSA calculations of candidates allowed identifying six compounds with the best binding energies. The compounds displayed in vitro minimum inhibitory concentrations (MIC) ranging from 50 to 100 µg/mL, growth inhibitions from 29.5 to 64.0% on Mtb, and inhibitions of Ca2+-dependent ATPase activity in Mtb membrane vesicles (IC50) ranging from 4.1 to 35.8 µM. The compound ZINC63908257 was the best candidate by displaying a MIC of 50 µg/mL and a Ca2+ P-type ATPase inhibition of 45% with IC50 = 4.4 µM. Overall, the results indicate that CtpF is a druggable target for designing new antituberculous compounds.
