ABSTRACT

Several mechanisms have been suggested to explain the adverse effects of air pollutants on airway cells. One such explanation is the presence of high concentrations of oxidants and pro-oxidants in environmental pollutants. All animal and plant cells have developed several mechanisms to prevent damage by oxidative molecules. Among these, the peroxiredoxins (PRDXs) are of interest due to a high reactivity with reactive oxygen species (ROS) through the functioning of the thioredoxin/thioredoxin reductase system. This study aimed to verify the gene expression patterns of the PRDX family in bronchial epithelial airway cells (BEAS-2B) cells exposed to diesel exhaust particles (DEPs) at a concentration of 15 µg/mL for 1 or 2 h because this it is a major component of particulate matter in the atmosphere. There was a significant decrease in mRNA fold changes of PRDX2 (0.43 ± 0.34; *p = 0.0220), PRDX5 (0.43 ± 0.34; *p = 0.0220), and PRDX6 (0.33 ± 0.25; *p = 0.0069) after 1 h of exposure to DEPs. The reduction in mRNA levels may consequently lead to a decrease in the levels of PRDX proteins, increasing oxidative stress in bronchial epithelial cells BEAS-2B and thus, negatively affecting cellular functions.

ABSTRACT

A commercialprobioticcontaining Bacillussubtilis(109 CFU g-1) was evaluated incaged matrinxã,Bryconamazonicus,by measuring hematological parametersand macrophage activity after 42 and 84 days after feeding. The product wasadded tocommercial feed using 2%soybean oil as a protectant. A randomized three-treatmentexperiment was performed using fourreplicates per treatment. The groups included: (a) control without probiotic, (b) 5 g kg-1probiotic, and (c) 10 g kg-1probiotic. Forhematological analysis,eightfishper treatmentwere used to determinetotal cell count (RBC); thrombocytes, differential, and total leukocyte count (TLC); hematocrit (Htc); hemoglobin tax; mean corpuscular volume (MCV); and mean corpuscular hemoglobin concentration (MCHC). Furthermore, plasma cortisol and glucose levels were measured in blood samples. Macrophage phagocytic activity was evaluated by injecting Saccharomyces cerevisiae(11,000 cells in a 3 mLvolume) into the coelomic cavity incubating for 8hours.Addition of probiotics to the diet of caged matrinxã altered the Htc, RBC, MCV, MCHC, TLC, lymphocyte, and eosinophil values. We observed increased cortisol and glucose levels and phagocytic activity, but no increase in the phagocytic index. We thus conclude that supplementing caged Brycon amazonicuswith probiotics improves their non-specific immunity and alters blood profiles.

ABSTRACT

The toxic actions of acute exposition to different diesel exhaust particles (DEPA) fractions on the mucociliary epithelium are not yet fully understood due to different concentrations of organic and inorganic elements. These chemicals elements produce damage to the respiratory epithelium and exacerbate pre-existent diseases. In our study we showed these differences in two experimental studies. Study I (dose-response curve - DRCS): Forty frog-palates were exposed to the following dilutions: frog ringer, intact DEPA diluted in frog-ringer at 3mg/L, 6mg/L and 12mg/L. Study II (DEPF) (DEPA fractions diluted at 12mg/L): Fifty palates - Frog ringer, intact DEPA, DEPA treated with hexane, nitric acid and methanol. Variables analyzed: relative time of mucociliary transport (MCT), ciliary beating frequency (CBF) and morphometric analysis for mucin profile (neutral/acid) and vacuolization. The Results of DRCS: Group DEPA-12mg/L presented a significant increase in the MCT (p<0.05), proportional volume of acid mucus (p<0.05) and decreased proportional volume of neutral mucus and vacuoles (p<0.05). In relation of DEPF: A significant increase in the MCT associated to a decrease in the proportional volume of neutral mucus was founded in nitric acid group. In addition, a significant increase in the proportional volume of acid mucus was found in methanol group. We concluded that: 1) Increasing concentrations of intact DEPA can progressively increase MCT and promote an acidification of intra-epithelial mucins associated to a depletion of neutral mucus. 2) Intact DEPA seem to act as secretagogue substance, promoting mucus extrusion and consequently reducing epithelial thickness. 3) Organic fraction of low polarity seems to play a pivotal role on the acute toxicity to the mucociliary epithelium, by promoting a significant increase in the MCT associated to changes in the chemical profile of the intracellular mucins.

ABSTRACT

The present study evaluated the water quality of a polluted pond through the analysis of in vitro mucociliary transport, hematological parameters, and biomarkers of cyto-genotoxicity in the Nile tilapia (Oreochromis niloticus). Blood and mucus samples were collected from ten specimens from the polluted pond and from ten specimens from a control area. The fish were anesthetized with 3% benzocaine, mucus was collected directly from the gills, and blood was drawn from the caudal artery. Blood smears were stained using the May-Grünwald Giemsa process for the differential leukocyte counts and to determine the frequency of leukocytes, thrombocytes, erythroblasts, micronuclei, and nuclear abnormalities. The results revealed low transportability in vitro, a high percentage of monocytes and eosinophils, and increased frequency of leukocytes and nuclear abnormalities in fish from the polluted pond. However, the frequency of thrombocytes and erythroblasts and the percentage of lymphocytes and neutrophils were significantly lower. It is possible to conclude that changes in fish are due to poor water quality and that these non-destructive biomarkers can be used for the biomonitoring of aquatic environments vulnerable to contamination.

ABSTRACT

Diesel exhaust particles (DEPs) from diesel engines produce adverse alterations in cells of the airways by activating intracellular signaling pathways and apoptotic gene overexpression, and also by influencing metabolism and cytoskeleton changes. This study used human bronchial epithelium cells (BEAS-2B) in culture and evaluates their exposure to DEPs (15ug/mL for 1 and 2 h) in order to determine changes to cell rheology (viscoelasticity) and gene expression of the enzymes involved in oxidative stress, apoptosis, and cytotoxicity. BEAS-2B cells exposed to DEPs were found to have a significant loss in stiffness, membrane stability, and mitochondrial activity. The genes involved in apoptosis [B cell lymphoma 2 (BCL-2 and caspase-3)] presented inversely proportional expressions (p = 0.05, p = 0.01, respectively), low expression of the genes involved in antioxidant responses [SOD1 (superoxide dismutase 1); SOD2 (superoxide dismutase 2), and GPx (glutathione peroxidase) (p = 0.01)], along with an increase in cytochrome P450, family 1, subfamily A, polypeptide 1 (CYP1A1) (p = 0.01). These results suggest that alterations in cell rheology and cytotoxicity could be associated with oxidative stress and imbalance between pro- and anti-apoptotic genes.

ABSTRACT

This study assessed the effects of the diesel exhaust particles on ERK and JNK MAPKs activation, cell rheology (viscoelasticity), and cytotoxicity in bronchial epithelial airway cells (BEAS-2B). Crude DEP and DEP after extraction with hexane (DEP/HEX) were utilized. The partial reduction of some DEP/HEX organics increased the biodisponibility of many metallic elements. JNK and ERK were activated simultaneously by crude DEP with no alterations in viscoelasticity of the cells. Mitochondrial activity, however, revealed a decrease through the MTT assay. DEP/HEX treatment increased viscoelasticity and cytotoxicity (membrane damage), and also activated JNK. Our data suggest that the greater bioavailability of metals could be involved in JNK activation and, consequently, in the reduction of fiber coherence and increase in the viscoelasticity and cytotoxicity of BEAS cells. The adverse findings detected after exposure to crude DEP and to DEP/HEX reflect the toxic potential of diesel compounds. Considering the fact that the cells of the respiratory epithelium are the first line of defense between the body and the environment, our data contribute to a better understanding of the pathways leading to respiratory cell injury and provide evidence for the onset of or worsening of respiratory diseases caused by inorganic compounds present in DEP.

ABSTRACT

Diesel exhaust particles (DEP) contain organic and inorganic elements that produce damage to the respiratory epithelium. The aim of this study was to determine the mucus profile of tracheal explants exposed to either crude diesel exhaust particles (DEP) or DEP treated with nitric acid (DEP/NA), with hexane (DEP/HEX), or with methanol (DEP/MET) at concentrations of 50 and 100 µg/ml for 30 and 60 min. Tracheal explants were subjected to morphometric analyses to study acidic (AB+), neutral (PAS+), and mixed (AB+/PAS+) mucus production and vacuolization (V). Incubation with 50 µg/ml crude DEP resulted in a rise in acid mucus production, an increase in vacuolization at 30 min, and reduction in neutral mucus at 30 and 60 min. Tracheas exposed to DEP/MET at 50 µg/ml for 30 or 60 min resulted in a significant decrease in neutral mucus production and an elevation in acid mucus production. DEP/HEX increased vacuolization at both 50 and 100 µg/ml at 30 and 60 min of exposure. Treatment with 50 µg/ml for 30 or 60 min significantly elevated mixed mucus levels. These results suggest that DEP appear to be more toxic when administered in combination with HEX or MET. DEP/MET modified the mucus profile of the epithelium, while DEP/HEX altered mucus extrusion, and these responses might be due to bioavailability of individual elements in DEP fractions.

Fish bioassays are valuable tools that can be used to elucidate the toxicological potential of numerous substances that are present in the aquatic environment. In this study, we assessed the antagonistic action of selenium (Se) against the toxicity of mercury (Hg) in fish (Oreochromis niloticus). Six experimental groups with six fish each were defined as follows: (1) control, (2) mercury (HgCl(2)), (3) sodium selenite (Na(2)Se(4)O(3)), (4) sodium selenate (Na(2)Se(6)O(4)), (5) mercury + sodium selenite (HgCl(2) + Na(2)Se(4)O(3)), and (6) mercury + sodium selenate (HgCl(2) + Na(2)Se(6)O(4)). Hematological parameters [red blood cells (RBC), white blood cells (WBC), and erythroblasts (ERB)] in combination with cytogenotoxicity biomarkers [nuclear abnormalities (NAs) and micronuclei (MN)] were examined after three, seven, ten, and fourteen days. After 7 days of exposure, cytogenotoxic effects and increased erythroblasts caused by mercury, leukocytosis triggered by mercury + sodium selenite, leukopenia associated with sodium selenate, and anemia triggered by mercury + sodium selenate were observed. Positive correlations that were independent of time were observed between WBC and RBC, ERB and MN, and NA and MN. The results suggest that short-term exposure to chemical contaminants elicited changes in blood parameters and produced cytogenotoxic effects. Moreover, NAs are the primary manifestations of MN formation and should be included in a class characterized as NA only. Lastly, the staining techniques used can be applied to both hematological characterization and the measurement of cytogenotoxicity biomarkers.

Particulate matter from diesel exhaust (DEP) has toxic properties and can activate intracellular signaling pathways and induce metabolic changes. This study was conducted to evaluate the activation of extracellular signal-regulated kinase (ERK) and c-Jun N-terminal kinase (JNK) and to analyze the mucin profile (acid (AB(+) ), neutral (PAS(+) ), or mixed (AB/PAS(+) ) mucus) and vacuolization (V) of tracheal explants after treatment with 50 or 100 µg/mL DEP for 30 or 60 min. Western blot analyses showed small increases in ERK1/2 and JNK phosphorylation after 30 min of 100 µg/mL DEP treatment compared with the control. An increase in JNK phosphorylation was observed after 60 min of treatment with 50 µg/mL DEP compared with the control. We did not observe any change in the level of ERK1/2 phosphorylation after treatment with 50 µg/mL DEP. Other groups of tracheas were subjected to histological sectioning and stained with periodic acid-Schiff (PAS) reagent and Alcian Blue (AB). The stained tissue sections were then subjected to morphometric analysis. The results obtained were compared using ANOVA. Treatment with 50 µg/mL DEP for 30 min or 60 min showed a significant increase (p < 0.001) in the amount of acid mucus, a reduction in neutral mucus, a significant reduction in mixed mucus, and greater vacuolization. Our results suggest that compounds found in DEPs are able to activate acid mucus production and enhance vacuolization and cell signaling pathways, which can lead to airway diseases.

ABSTRACT

The biomonitoring of fish using biomarkers represents a useful tool for the assessment of aquatic pollution. This study evaluated the sublethal toxic effects of aquatic pollution on fish collected from a site contaminated by metals. Water and fish (Oreochromis niloticus) samples were collected from a pond in the Parque Ecológico do Tietê (PET) that lies along the Tietê River (São Paulo, Brazil), and from a control site (an experimental fish farm). The metal content of the water was evaluated, and fish were used to examine the properties of gill mucus and blood. The PET fish were evaluated for alterations in the in vitro transportability of mucus and changes in blood properties (e.g., cell volume, hemoglobin concentration, red blood cells, and white blood cell count). The results of the water analyzes indicated metal levels above the legal standards for Fe (0.71 mg/L), Ni (0.06 mg/L), Mn (0.11 mg/L), and Pb (0.48 mg/L). Compared to the controls, the hematologic parameter analyzes of PET fish revealed significantly higher numbers of erythrocytes (RBC), leukocytes (WBC), lymphocytes, erythroblasts, and Mean Corpuscular Volume (MCV); however, the hemoglobin content and Mean Corpuscular Hemoglobin Concentration (MCHC) values were significantly lower. The frequencies of nuclear abnormalities and micronuclei were significantly higher and the mucociliary transport was significantly lower in PET fish than in the controls. These results suggest that fish from the contaminated site exhibit a series of physiological responses, which probably indicate health disturbances. Furthermore, the results suggest that blood and mucus are promising, non-destructive targets for use in the monitoring of pollution.

Partículas da exaustão de motores a diesel (DEP) têm propriedades toxicológicas, devido às características físico-químicas. O DEP é capaz de ativar as vias de sinalização intracelular e induzir alterações metabólicas em células e tecidos do sistema respiratório. O objetivo desta tese foi: 1) avaliar o perfil das mucinas e alterações epiteliais em explantes de traqueia de camundongo BALB/c expostos ao DEP e DEP tratado com ácido nítrico e solventes orgânicos; e 2) em cultura de células de epitélio brônquico humano (BEAS-2B) expostas ao DEP e DEP tratado com hexano (DEP/HEX) para avaliar ativação de MAPK (ERK e JNK), citotoxidade, integridade de citoesqueleto, viscoelasticidade celular e expressão gênica de enzimas envolvidas no estresse oxidativo e apoptose. Os resultados mostraram que, em explantes de traqueia, o DEP causa aumento significativo em relação ao grupo controle na quantidade de muco ácido (p= 0,001), diminuição no muco neutro (p=0,001), diminuição de muco misto (p= 0,001), aumento de vacuolização (p= 0,001), aumento de apoptose (p=0,001), ora com aumento de pERK e diminuição de pJNK, e vice-versa. Os explantes submetidos à exposição ao DEP e DEP/MET aumentaram significativamente o muco ácido (p=0,01) e DEP/HEX provocou aumento da extrusão do muco (p=0,007), provavelmente devido à ação do enriquecimento inorgânico. Para as células BEAS-2B, nos resultados obtidos com células epiteliais expostas ao DEP e DEP/HEX, foram observadas alterações na membrana citoplasmática, mitocôndrias e citoesqueleto. As células expostas apenas ao DEP em baixas concentrações (15ug/mL) apresentaram alterações na expressão de genes envolvidos no apoptose (BCL-2 e Caspase-3 (p=0,05 e p=0,01) e estresse oxidativo [(SOD1 e SOD2 e GPx. p=0,01 )], e CYP1A1 ((p=0,01).

DEPs are able to activate intracellular signaling pathways and induce metabolic changes to cells and tissues of the human respiratory system. This dissertation sought to evaluate: 1) the profile of mucins and the epithelial changes to the tracheal explants of BALB/c mice exposed to both DEP and DEP treated with nitric acid and organic solvents (50 and 100 ug/mL; and 2) human bronchial epithelial cells (BEAS-2B) in culture after their exposure to both DEP and DEP treated with hexane (DEP/HEX) at 100 ug/mL in order to determine MAPK (ERK/JNK) activation, cytotoxicity, cytoskeletal integrity, cell viscoelasticity and gene expression of the enzymes involved in oxidative stress and apoptosis. The results show that, in tracheal explants, DEP causes a significant increase (compared to the control) in the quantity of acidic mucus (p=0.001), a decrease in alkaline mucus (p=0.001), a decrease in mixed mucus (p=0.001), an increase in vacuolization (p=0.001), an increase in apoptosis (p=0.001), along with an increase in pERK and a decrease in pJNK, and vice versa. The explants that were exposed to DEP and DEP/MET were found to have significantly higher quantities of acidic mucus (p=0.01), and DEP/HEX caused an increase in mucus extrusion (p=0.007), which was likely due to inorganic enrichment. In the case of BEAS-2B cells, the results obtained from epithelial cells exposted to DEP and DEP/HEX revealed alterations in the cytoplasmic membrane, the mitochondria, and the cytoskeleton. The cells exposed to DEP alone at low concentrations (15 ug/mL) experienced alterations in the genes involved in apoptosis (BCL-2 and Caspase-3; p=0.05 and p=0.01, respectively), as well as oxidative stress [(SOD1, SOD2, and GPx; p=0.01 )], and changes to CYP1A1 (p=0.01).

Selenium (Se) is described as an essential micronutrient and participates in different biological functions, as the antioxidant defense systems maintenance and regulation. However, when in high concentrations, Se may cause toxic effects as well as hematological changes in fish. The aim of the present study was to determine the toxicity of selenium in the form of sodium selenate (Na2Se6+O4) in Oreochromis niloticus based on hematological parameters, after exposure to different concentrations (0.01, 0.14 and 1.4 mg Se6+ L-1). The erythrocytic and leukocytic series were examined over 14 days at intervals of 0, 3, 5, 7,10 and 14 days. The erythrocytic series showed significant alterations in the first 7 days, including the control group. Neutrophils and monocytes showed variations in the first 3 days at a concentration of 1.40 mgSe6+ L-1 characterizing an acute response. The total number of leukocytes was different in relation to time zero on all Se concentrations. The thrombocyte count also differed statistically from time zero and control in the first 3 days at 0.14 mgSe6+ L-1. These results indicate that different concentrations induce an acute response with diminution of total leukocytes, neutrophilia, monocytosis and thrombocytosis.

ABSTRACT

Selenium (Se) is described as an essential micronutrient and participates in different biological functions, as the antioxidant defense systems maintenance and regulation. However, when in high concentrations, Se may cause toxic effects as well as hematological changes in fish. The aim of the present study was to determine the toxicity of selenium in the form of sodium selenate (Na2Se6+O4) in Oreochromis niloticus based on hematological parameters, after exposure to different concentrations (0.01, 0. 14 and 1.4 mg Se 6+ L-1). The erythrocytic and leukocytic series were examined over 14 days at intervals of 0, 3, 5, 7,10 and 14 days. The erythrocytic series showed significant alterations in the first 7 days, including the control group. Neutrophils and monocytes showed variations in the first 3 days at a concentration of 1.40 mgSe6+ L-1 characterizing an acute response. The total number of leukocytes was different in relation to time zero on all Se concentrations. The thrombocyte count also differed statistically from time zero and control in the first 3 days at 0.14 mgSe6+ L-1. These results indicate that different concentrations induce an acute response with diminution of total leukocytes, neutrophilia, monocytosis and thrombocytosis.


O selênio (Se) é descrito como um micronutriente essencial e participa de diversas funções biológicas. No entanto, quando em concentrações elevadas, o Se pode causar efeitos tóxicos, bem como alterações hematológicas em peixes. O objetivo do presente estudo foi determinar a toxicidade do selênio na forma de Selenato de sódio (Na2Se6+ O4) em Oreochromis niloticus com base em parâmetros hematológicos, após a exposição a diferentes concentrações (0,01, 0,14 e 1,4 mg + Se6+ L-1). A série eritrocitária e leucocitária foram examinadas por 14 dias, em intervalos de 0, 3, 5, 7,10 e 14 dias. A série eritrocítica mostrou alterações significativas nos primeiros 7 dias, incluindo o grupo controle. Neutrófilos e monócitos apresentaram variações nos primeiros 3 dias na concentração de 1,40 mgSe6+ L-1, caracterizando uma resposta aguda. O número total de leucócitos foi diferente em relação ao tempo zero em todas as concentrações de Se. A contagem de trombócitos também diferiram estatisticamente entre o tempo zero e os primeiros 3 dias a 0,14 mgSe6+ L-1. O resultados indicam que diferentes concentrações induzem a resposta aguda com diminuição dos leucócitos totais, neutrofilia, monocitose e trombocitose.

ABSTRACT

This study aimed to determine the variables leukocytary, erythrocytary, frequency of micronuclei and nuclear abnormalities in peripheral erythrocytes of Prochilodus argenteus, Pimelodus maculatus and Myleus micans from São Francisco river basin, Minas Gerais State, during the summer and winter. Thrombocytes, hematocrit and leukocyte of P.argenteus series, was influenced by climatic conditions. In P. maculatus, not were significant differences for all leukocytes and thrombocytes. In M. micans, values are unprecedented in the literature, with MCV and absolute number of leukocytes found superior to other species of the family Characidae. The frequency of micronuclei and nuclear abnormalities in erythrocytes, the three species not showed seasonal differences between the sites. However, the highest values were found in summer. Furthermore, we observed a positive correlation between the increase in the percentage of micronuclei with nuclear abnormalities


O presente estudo teve como objetivo, determinar as variáveis leucocitárias, eritrocíticas, freqüência de micronúcleos e anomalias nucleares nos eritrócitos do sangue periférico de Prochilodus argenteus, Pimelodus maculatus e Myleus micans de um trecho da bacia do rio São Francisco, Minas Gerais, no verão e inverno. Trombóctios, hematócrito e a serie leucocítária de P.argenteus, foi influenciado pela sazonalidade. Em P. maculatus, não ocorreram diferenças significativas para todos os leucócitos e trombócitos. Em M. micans, os valores são inéditos na literatura, apresentando VCM e número absoluto de leucócitos superior ao encontrados para outras espécies da família Characidae. A freqüência de micronúcleos e de anomalias nucleares nos eritrócitos, nas três espécies não apresentou diferenças sazonais e entre os pontos amostrais. No entanto, os valores mais altos foram encontrados no verão. Além disso, foi possível observar correlação positiva entre o aumento da porcentagem de anomalias nucleares com micronúcleos.

In aquaculture, analysis of activation and increased macrophages migration are used in order to verify the ability of the nonspecific immune fish exposed to a challenge. This study aimed to determine the time of macrophages migration in matrinxã, Brycon amazonicus, through the technique of yeast Saccharomyces cerevisiae inoculation, verifying possible changes in hematological parameters. Thirty animals with average weight of 101.55 ± 24.50 g and average length of 19.75 ± 1.72 cm were employed. The experimental inoculation periods were 2, 4, 8 and 12 hours. Thereafter, animals were anesthetized and blood was withdrawn through a caudal puncture for the determination of total erythrocytes number, differential and total leukocyte counts and total thrombocytes count, hematocrit, hemoglobin concentration and calculation of the erythrocytes index. The results for the phagocytic capacity were not significantly different between experimental periods. In the phagocytic index, the period of 2 hours presented the highest rate of phagocytized cells, indicating that 2 hours of incubation was sufficient for the macrophages migration in B. amazonicus. The number of erythrocyte was the only parameter that presented significant difference among periods.

Centropomus parallelus, an important food resource for local populations, is a predator fish that feeds on small fishes and benthic invertebrates, thus being vulnerable to the bioconcentration and biomagnification processes. This study aimed to evaluate cytogenotoxic responses in erythrocytes from C. parallelus juveniles collected in the Cananéia and São Vicente estuaries, both in winter and in summer. After anesthesia, blood samples were collected by caudal puncture. Blood smears were prepared on glass slides and stained with May-Grünwald-Giemsa dye. Two thousand cells were analyzed per slide (1000x), and nuclear abnormalities (NA) and micronuclei (MN) were scored. The São Vicente sample showed MN and NA frequencies (%/1000 cells) of 0.325 and 3.575, in winter, and of 0.125 and 2.935 in summer respectively; the Cananéia sample showed frequencies of 0.0325 and 0.03, in winter, and of 0.065 and 0.355 in summer, respectively. The rates found in São Vicente were significantly higher than those found in Cananéia, evidencing that the levels of pollution in that estuary were high enough to induce genetic damages.

ABSTRACT

Esta investigação teve como objetivo avaliar a qualidade de sedimentos do estuário do Rio Itanhaém, utilizando teste de toxicidade de sedimento integral com anfípodo escavador Tiburonella viscana e análise da comunidade bentônica. Os resultados mostraram que os sedimentos são principalmente arenosos, com baixa ou nenhuma toxicidade sobre T. viscana. A comunidade bentônica foi composta principalmente por organismos oportunistas, incluindo poliquetos capitelídeos, tendo sido considerada pobre. Sua estrutura foi controlada principalmente pelas variáveis ambientais, como a granulometria e as propriedades físico-químicas da água, porém uma leve participação da toxicidade pôde ser detectada. A qualidade dos sedimentos do estuário variou entre moderada e boa.


This work aimed to assess the quality of sediments from the estuary of the Itanhaém River using whole sediment toxicity tests with the burrowing amphipod Tiburonella viscana; and the analysis of the benthic community. Results showed that the sedimentsare preponderantly sandy, with low or absence of toxicity to T. viscana. The benthic community was poor, composed mainly by opportunistic organisms, including capitelid polychaetes. Its structure was mainly controlled by the environmental variables, as sediment grain size and water properties, but some influence of the toxicity was also observed. The quality of the sediments from the estuary ranged from moderate to good.


Este trabajo pretendió evaluar la calidad de sedimentos del estuario del Río Itanhaém usando pruebas de toxicidad de sedimento integral con anfípodo escavador Tiburonella viscana y el análisis de la comunidad bentónica. Los resultados mostraron que los sedimentos son preponderantemente arenosos, con baja o ninguna toxicidad para T. viscana. La comunidad béntica era pobre, formada principalmente por organismos oportunistas, incluso poliquetos capitelídeos. Su estructura fue controlada principalmente por las variables ambientales, como lo son el tamaño de los granos de sedimento y propiedades del agua, pero un poco de influencia de la toxicidad también fue observado. La calidad de los sedimentos del estuario varió de moderado a bueno.

ABSTRACT

O estuário do Rio Itanhaém situa-se ao sul da Baixada Santista do Estado de São Paulo (23° 50’, 24°15’ S; 46° 35’, 47° 00’ W) em uma áreasob intensa pressão de ocupação humana. Embora esse estuário ainda se encontre relativamente pouco degradado, o lançamento de poluentes nos seuscorpos hídricos, coloca em risco a vida aquática local, bem como a exploração dos seus recursos pesqueiros. Sabendo-se que os contaminantes possuemtempo de residência limitado na coluna d’água, e que tendem a precipitar no fundo, o presente estudo teve como objetivo avaliar a qualidade ambientaldeste estuário, através de testes de toxicidade com água e sedimento integral, análises físico-químicas e estrutura da comunidade bentônica. A partirdestes resultados pretende-se fornecer informações que possam servir como base para a educação ambiental e demonstrar as ameaças representadaspela poluição e de que forma ela pode influenciar sobre o bem estar das populações locais. Foram definidos três pontos amostrais, na região inferior doestuário. A toxicidade e a ausência de fauna no sedimento do Ponto 1 foram atribuídas à granulometria grosseira (falso-positivo), enquanto no Ponto3 houve indícios de enriquecimento orgânico natural. No sedimento do Ponto 2 observou-se evidências de degradação, pela ocorrência de toxicidade.Quanto à coluna d’água, os parâmetros físico-químicos e os testes de toxicidade indicaram condições satisfatórias. No entanto, a baixa sobrevivência deNitocra sp no Ponto 1 pode ser um indício de toxicidade, indicando a necessidade de estudos confirmatórios mais detalhados. Além disso, estes resultadospodem ser acrescentados como mais um dos argumentos para a educação ambiental na região.
