ABSTRACT

OBJECTIVE: To explore the effect of Shixiao Huoxue Decoction on pain and tumor necrosis factor (TNF)-α and interleukin (IL)-8 levels in patients with adenomyosis. METHODS: A total of 65 patients with adenomyosis admitted to South District of Guang'anmen Hospital from January 2020 to December 2021 were divided into two groups according to the treatment methods. The control group was treated with pregnatrienone, and the study group was treated with additional Shixiaohuoxue decoction. The incidence of complications, treatment efficacy, levels of inflammatory factors, Traditional Chinese Medicine symptom score, dysmenorrhea score, menstrual volume score, dysmenorrhea symptom score, changes in uterine volume, level of insulin-like growth factor 1 (IGF-1), and changes in the level of carbohydrate antigen (CA125) were observed before and after treatment in both two groups. Univariate Logistic analysis showed that uterine volume, IGF-1, CA125, serum IL-8 and TNF-α were correlated with the short-term efficacy of Meixiaohuoxue Decoction in the treatment of uterine adenomyosis (P<0.05). RESULTS: The levels of IL-8 and TNF-α in the study group were significantly lower than those in the control group after treatment (P<0.05). The scores of dyspareunia and non-menstrual pelvic pain in the study group were significantly lower than those in the control group (P<0.05). The overall response rate in the study group (93.75%) was significantly higher than that in the control group (66.66%) (P<0.05). The scores of Traditional Chinese Medicine symptoms, dysmenorrhea, menstrual volume, and dysmenorrhea symptoms in the study group were significantly lower than those in the control group after treatment (P<0.05). The IGF-1 and CA125 levels in the study group were significantly lower than those in the control group after treatment (P<0.05). However, no significant difference in uterine volume was found between the two groups after treatment (P>0.05). CONCLUSION: Xiaoxiao Huoxue Decoction demonstrated a better treatment efficacy in patients with adenomyosis through improving dysmenorrhea and Traditional Chinese Medicine symptoms, as well as reducing the levels of body inflammatory factors, non-menstrual pelvic pain, and dyspareunia, thus contributing to early recovery of patients. Therefore, Xiaoxiao Huoxue Decoction is worthy of promotion in clinical treatment of adenomyosis.

ABSTRACT

The RH blood group system is the most complex with over 50 antigens. So far over hundreds of RhCE variant alleles have been described resulting in weakened and/or partial expression of RhCE antigens [1], some variant Rh phenotypes are caused by exchange of genetic material between the RHD and RHCE genes, resulting in many hybrid genes, other phenotypes result from missense mutations. Variant alleles encode altered phenotypes with either weakened antigens, lacked antigens, or unexpected antigens. Besides, the mutation of RH blood group genes may lead to the changes of Rh antigen epitopes. RHCE gene mutations or polymorphisms may bring about altered RH antigens in quality and quantity [2]. Serologic weaknesses or discrepancies are regularly faced by blood transfusion laboratories, and molecular background explaining this feature can be precisely characterized only by the molecular biological methods.

ABSTRACT

Deep eutectic solvents (DESs) have emerged as promising and eco-friendly solvents for the efficient extraction of lignin from biomass due to their low cost and environmental benefits. Nevertheless, the prevalent use of acidic DESs in lignin extraction often results in excessive depolymerization and recondensation of lignin, thereby impeding its downstream applications. In this study, we developed a range of alkaline DESs (ADESs), both pure and peroxide-containing, for the extraction of high-quality lignin from bamboo. Moreover, carbon dioxide (CO2) was employed for the precipitation and regeneration of the extracted lignin. The obtained lignin fractions were comprehensively characterized in terms of yield, purity, morphology, solubility, structural features, and anti-UV/oxidant activity. The results showed that the monoethanolamine-based ADES demonstrated superior performance among the pure ADESs. Structural analysis confirmed the well-preserved substructures of lignin fractions obtained using ADESs, with ß-O-4 bond retention ranging from 49.8 % to 68.4 %. The incorporation of a suitable amount of peroxide improved lignin yield, morphology, solubility, and anti-UV/oxidant activity. Additionally, the anti-UV/oxidant activity of lignin exhibited a positive correlation with its phenolic hydroxyl content. This study provides a valuable reference for the green and sustainable production and valorization of lignin within the existing biorefinery framework.

ABSTRACT

In this study, light-colored lignin was extracted from bamboo shoot shells (BSS) using a hydrated deep eutectic solvent (DES) pretreatment. The hydrated DES used in pretreatment consist of formic acid, benzyl triethylammonium chloride (BTEAC) and water. The pretreatment using a hydrated DES containing 30% water (H30) demonstrate efficient delignification (82.9%). Additionally, the hydrated DES protected the ß-O-4 linkage from excessive cleavage and recondensation as well as keep the light-colored of lignin. Moreover, the hydrated DES extracted lignin exhibits superior antioxidant performance and tyrosinase inhibitory capacity compared to the control. Notably, incorporating 5% lignin of H30-extracted lignin into a commercial suncream led to a remarkable enhancement of the SPF value, elevating from 14.8 to 32.6. In summary, the proposed hydrated DES pretreatment method offers significant benefits for extracting light-colored lignin, thereby promoting the multifunctional application of lignin in cosmetics.

ABSTRACT

The developing of pretreatment method to break the biomass barrier of lignocellulosic is a challenging task for achieve high value utilization. A fast microwave-assisted choline chloride/1,2-propanediol/methyl isobutyl ketone biphasic system was constructed for pretreating Eucalyptus to the production of furfural and cellulose-rich residues and the extraction of lignin. Results showed that the combination of AlCl3·6H2O and HCl had the best catalytic ability for furfural production among the examined catalysts. Under the optimal conditions (140 °C, 15 min, 0.075 M AlCl3·6H2O, 0.05 M HCl), the furfural yield of 55.4 %, the glucose yield of 90.3 % and the delignification rate of 92.4 % could be achieved. Moreover, the extracted lignin samples with a low polydispersity (1.55-1.73) and molecular weight (1380-2040 g/mol) are promising to act as precursor for the value-add products processing. These findings demonstrated an ultrafast pretreatment process with excellent results in biomass fractionation and comprehensive utilization of biomass components.

ABSTRACT

Objective: Shenling Baizhu powder (SBP) has been shown to reverse the abnormal expression of the aromatic hydrocarbon receptor (AHR) mediated by air pollution. Our study aimed to understand the main ingredient of SBP and investigate its action mechanism in preventing polycystic ovary syndrome (POCS) and postmenopausal osteoporosis (PMO). Methods: The active ingredients of SBP with the highest binding affinity to AHR were screened using a Chinese medicine database, and their binding mechanism was simulated using molecular dynamics simulation (MDS). Rutin was utilized to treat ovarian granulosa cell lines and osteoblast cell lines. The cell lines were treated with a gradient of rutin concentration (0.01 mmol/L, 0.05 mmol/L and 0.1 mmol/L) to find the optimal drug dose. PCR was used to detect AHR and apoptosis-related proteins, and WB to detect the expression of AHR, caspase-3 and cleaved-caspase-3. Finally, the CCK-8 cell proliferation assay detected the proliferation of cells. Results: We obtained Rutin through the Chinese medicine database, and dynamics simulation determined its binding sites. Ovarian granulosa cell lines and osteoblast cell lines were treated with Rutin. RT-PCR and western blotting revealed that the expression of apoptosis-associated protein Bcl-2 was elevated, and the expression of AHR, Bax, caspase-3 and PARP were decreased. CCK-8 results showed accelerated proliferation in both cell types. Conclusion: Rutin, the main ingredient of SBP compound, works by binding to AHR, which can improve POCS and PMO by inhibiting cell apoptosis and by promoting cell proliferation.

ABSTRACT

Background: Hypoxia plays a significant role in the pathogenesis of pancreatic cancer, but the effect of hypoxia-related genes in pancreatic cancer remains to be elucidated. This study aimed to identify hypoxia-related genes related to pancreatic cancer and construct a prognostic signature. Methods: Pancreatic cancer datasets were retrieved from TCGA database. Cox regression analyses were used to identify hypoxia-related genes and construct a prognostic signature. Datasets from International Cancer Genome Consortium and GEO databases were used as validated cohorts. The CIBERSORT method was applied to estimate the fractions of immune cell types. DNA methylation and protein levels of the genes in pancreatic cancer were examined. Results: Three hypoxia-related genes (TES, LDHA, and ANXA2) were identified as associated with patient survival and selected to construct a prognostic signature. Patients were divided into high- and low-risk groups based on the signature. Those in the high-risk group showed worse survival than those in the low-risk group. The signature was shown to be involved in the HIF-1 signaling pathway. The time-dependent ROC analyses of three independent validated cohorts further revealed that this signature had a better prognostic value in the prediction of the survival of pancreatic cancer patients. Immune cells analysis for three datasets demonstrated that high-risk signature was significantly associated with macrophages and T cells. DNA methylation and protein levels of the three genes validated their aberrant expression in pancreatic cancer. Conclusions: Our research provided a novel and reliable prognostic signature that composes of three hypoxia-related genes to estimate the prognosis of pancreatic cancer.

ABSTRACT

Bamboo processing residue, which is rich in parenchyma cells, was treated as huge waste in bamboo processing industry, such as reassemble bamboo and bamboo flooring. Herein, autohydrolysis and rapid different deep eutectic solvents (DES) delignification strategy were consecutively performed to remove hemicelluloses and lignin from bamboo processing residue. The xylooligosaccharides (XOS) with high yield (34.35%) was achieved in the autohydrolysis process. Results showed that alkaline DES pretreatment resulted in the highest glucose yield (88.22%) and relatively high delignification rate (83.75%) as well as well-preserved lignin structures. However, the lignin fractions obtained under acidic DES conditions were tending to assemble into lignin nanoparticles (LNPs) and having excellent antioxidant activity as compared to those obtained from alkaline DES system. In brief, the combination of autohydrolysis and rapid DES delignification can achieve orientated fractionation of the components from the industrialized bamboo.

ABSTRACT

Developing a biorefinery process for a highly integrated valorization and fractionation of lignocellulose is crucial for its utilization. Herein, a biphasic system comprising choline chloride/lactic acid and 2-methyltetrahydrofuran with Al2(SO4)3 and H2SO4 as catalysts was applied to pretreat Eucalyptus. Results showed that under the optimized conditions (150 °C, 30 min, 0.2 M Al2(SO4)3, 0.075 M H2SO4), the furfural yield and enzymatic hydrolysis efficiency could reach 54.7% and 97.0%, respectively. The efficient cellulose conversion was attributed to remarkable removal of lignin (91.0%) and hemicelluloses (100.0%), thereby causing the disruption of cell wall structure and enhancement of cellulose accessibility. Meanwhile, confocal Raman microscope and atomic force microscope displayed that the pretreatment resulted in the decreasing intensities of carbohydrates and lignin different regions of cell walls, and exposing of the embedded microfibers from noncellulosic polymers. Overall, the deep eutectic solvent-based biphasic system displayed high performance for effective utilization of carbohydrate components in lignocellulose.

ABSTRACT

The wrist is a complex joint that bridges the hand to the forearm. Patients with wrist disorders increasingly prefer minimally invasive procedures for wrist joint diagnosis and treatment. Wrist arthroscopy offers direct visualization of the structures of the joint anatomy and existing disease processes while causing minimal damage to surrounding soft tissue. However, it requires a high level of technical ability for wrist arthroscopy practitioners. Therefore, an improved focus on wrist arthroscopy training combining new educational media and traditional practice should aid in the development of novel wrist arthroscopy training mode. This article aims to describe the status of wrist training and evaluation systems and introduce a new progressive wrist training system.

OBJECTIVE: To explore the effect of Guijiaosan Shenque acupoint paste on the obesity, endocrine, and TCM symptom scores of obese polycystic ovary syndrome (PCOS) patients with phlegm-dampness block. METHODS: From January 2020 to December 2020, 80 PCOS patients with phlegm-dampness block were recruited as the study cohort and randomly divided into a research group (RG) or a control group (CG), with 40 cases in each group. The CG was treated with herbal decoctions according to syndrome differentiation, while the RG was administered Guijiaosan paste at the Shenque acupoint in addition to the treatment administered to the CG. The body weight indexes, endocrine function, insulin resistance indexes, TCM syndrome scores, and safety index changes were compared between the two groups before and after the treatment. RESULTS: Compared with the CG, the body weight indexes, endocrine function, insulin resistance indexes, and TCM syndrome scores in the RG were significantly improved (P<0.05), and there was no significant difference in the safety indexes between the two groups before the treatment (P>0.05). CONCLUSION: The application of Guijiaosan paste at the Shenque acupoint can effectively improve the obesity and endocrine functions of obese patients with PCOS and improve their clinical symptoms, so it is worthy of clinical promotion.

BACKGROUND: Cocksfoot grass (Dactylis glomerata L.) with high biomass yield and rich cellulose can be used to produce bioethanol as fuel additive. In view of this, ultrasonic and hydrothermal pretreatments followed by successive alkali extractions were assembled into an integrated biorefinery process applied on cocksfoot grass to improve its enzymatic hydrolysis. In this work, the effects of ultrasonic and hydrothermal pretreatments followed by sequential alkali extractions on the enzymatic hydrolysis of cocksfoot grass were investigated. In addition, since large amount of hemicelluloses were released during the hydrothermal pretreatment and alkali extraction process, the yields, structural characteristics and differentials of water- and alkali-soluble hemicellulosic fractions isolated from different treatments were also comparatively explored. RESULTS: The integrated treatment significantly removed amorphous hemicelluloses and lignin, resulting in increased crystallinity of the treated residues. A maximum saccharification rate of 95.1% was obtained from the cellulose-rich substrate after the integrated treatment. In addition, the considerable hemicelluloses (31.4% water-soluble hemicelluloses and 53.4% alkali-soluble hemicelluloses) were isolated during the integrated treatment. The released water-soluble hemicellulosic fractions were found to be more branched as compared with the alkali-soluble hemicellulosic fractions and all hemicellulosic fractions were mixed polysaccharides mainly composed of branched xylans and ß-glucans. CONCLUSION: The combination of ultrasonic and hydrothermal pretreatments followed by successive alkali extractions can dramatically increase the enzymatic saccharification rate of the substrates and produce considerable amounts of hemicelluloses. Detailed information about the enzymatic hydrolysis rates of the treated substrates and the structural characteristics of the co-produced hemicelluloses will help the synergistic utilization of cellulose and hemicellulose in cocksfoot grass.

ABSTRACT

It is believed that high-grade serous ovarian cancer (HGSOC) is a solid or multilocular-solid cancer. Here, we report the case of a 40-yr-old woman with a left ovarian unilocular cyst. Ultrasonography and computed tomographic examination confirmed that the cyst was thin-walled and homogenous in thickness without mural nodules. It was considered to be an endometriotic cyst. Left ovarian cyst excision specimens proved it to be HGSOC after pathologic examination. Therefore, the patient underwent radical surgery for HGSOC. Pathologic examination of radical resection specimens confirmed that the HGSOC was still in FIGO stage IA and no fallopian tube lesion was found. Considering that the patient had a history of breast cancer in both the breasts at a young age, it was hypothesized that the breast cancer susceptibility gene (BRCA) gene may have a germline mutation. Next-generation sequencing confirmed the BRCA1 (c.3770_3771delAG) germline mutation in this patient. Previous studies have reported the special morphological characteristics and growth pattern of HGSOC with BRCA mutation in the advanced stage. Our case demonstrates that HGSOC with the BRCA mutation can also be a unilocular cyst with a thin wall and uniform thickness without a mural nodule, and in the early stage, may have unique gross morphology.
