ABSTRACT

Pelas características anatômicas e fisiológicas dos rins, a lesão renal aguda tem sua origem nefrotóxica pela alta circulação local, o que favorece a alta concentração de substâncias tóxicas e seus metabólitos no tecido. A lesão renal aguda é uma complicação comum em internações hospitalares e principalmente em internações em unidades de terapia intensiva. A ciclofosfamida, um quimioterápico utilizado no tratamento de doenças autoimunes e neoplasias sólidas, pode causar nefrotoxicidade com disfunção glomerular e tubular. O uso de plantas medicinais, pelas suas potentes ações antioxidantes, tem sido usado para prevenção ou tratamento de lesões celulares induzidas pelo desequilíbrio entre enzimas antioxidantes e oxidantes. Por esse motivo, o objetivo do experimento foi avaliar o potencial efeito protetor da Echinodorus grandiflorus na prevenção da nefrotoxidade induzida pela ciclofosfamida. Para isso, foi realizado o experimento com a utilização de 35 ratos machos, Wistar, divididos em seis grupos experimentais, sendo administrado a ciclofosfamida na dose de 150mg/kg nos grupos G2 a G6 e diferentes doses da Echinodorus grandiflorus, com posterior análise de parâmetros sanguíneos e histológicos. A administração de ciclofosfamida na dose de 150mg/kg de massa corporal, em dose única, foi capaz de induzir a nefrotoxicidade aguda em todos os ratos. O extrato bruto de Echinodorus grandiflorus apresentou potencial efeito renoprotetor ao uso da ciclofosfamida, na dose de 300mg/kg de massa corporal, sendo possível observar redução dos efeitos nefrotóxicos do quimioterápico, pela redução dos danos tubulares e pela diminuição dos espaços capsulas, nitidamente encontradas alterados no grupo que recebeu apenas ciclofosfamida, denotando resultados promissores para utilização desta planta medicinal na prevenção da nefrotoxicidade induzida pelo fármaco. Contudo, novos estudos dos efeitos renoprotetor do chapéu de couro, poderão elucidar os mecanismos envolvidos na ação do extrato bruto do chapéu de couro. A utilização de extrato bruto de plantas medicinais torna-se um adjuvante aos tratamentos pelo baixo custo e pela facilidade de acesso das diferentes populações as plantas desde que devidamente orientados pelos profissionais habilitados.


Due to the anatomical and physiological characteristics of the kidneys, acute kidney injury has its nephrotoxic origin due to the high local circulation, which favors the high concentration of toxic substances and their metabolites in the tissue. Acute kidney injury is a common complication in hospital admissions and especially in intensive care unit admissions. Cyclophosphamide, a chemotherapy drug used in the treatment of autoimmune diseases and solid neoplasms, can cause nephrotoxicity with glomerular and tubular dysfunction. The use of medicinal plants, due to their potent antioxidant actions, has been used for the prevention or treatment of cellular injuries induced by the imbalance between antioxidant and oxidant enzymes. For this reason, the aim of the experiment was to evaluate the potential protective effect of Echinodorus grandiflorus in preventing cyclophosphamide-induced nephrotoxicity. For this, the experiment was carried out with the use of 35 male Wistar rats, divided into six experimental groups, being administered cyclophosphamide at a dose of 150mg/kg in groups G2 to G6 and different doses of Echinodorus grandiflorus, with subsequent analysis of parameters blood and histology. The administration of cyclophosphamide at a dose of 150mg/kg of body weight, in a single dose, was able to induce acute nephrotoxicity in all rats. The crude extract of Echinodorus grandiflorus showed a potential renoprotective effect with the use of cyclophosphamide, at a dose of 300mg/kg of body mass, and it was possible to observe a reduction in the nephrotoxic effects of the chemotherapy, due to the reduction of tubular damage and the reduction of capsule spaces, clearly found altered in the group that received only cyclophosphamide, showing promising results for the use of this medicinal plant in the prevention of drug-induced nephrotoxicity. However, further studies of the renoprotective effects of the leather hat may elucidate the mechanisms involved in the action of the crude extract of the leather hat. The use of raw extract of medicinal plants becomes an adjuvant to treatments due to the low cost and ease of access of different populations to plants, provided that they are properly guided by qualified professionals.


Debido a las características anatómicas y fisiológicas de los riñones, la lesión renal aguda tiene su origen nefrotóxico por la elevada circulación local, que favorece la alta concentración de sustancias tóxicas y sus metabolitos en el tejido. La lesión renal aguda es una complicación frecuente en los ingresos hospitalarios y principalmente en las unidades de cuidados intensivos. La ciclofosfamida, un quimioterápico utilizado en el tratamiento de enfermedades autoinmunes y neoplasias sólidas, puede causar nefrotoxicidad con disfunción glomerular y tubular. El uso de plantas medicinales, debido a sus potentes acciones antioxidantes, se ha utilizado para la prevención o el tratamiento de lesiones celulares inducidas por el desequilibrio entre enzimas antioxidantes y oxidantes. Por este motivo, el objetivo del experimento era evaluar el posible efecto protector del Echinodorus grandiflorus en la prevención de la nefrotoxicidad inducida por la ciclofosfamida. Para ello, se realizó el experimento utilizando 35 ratas Wistar macho, divididas en seis grupos experimentales, administrándoseles ciclofosfamida a una dosis de 150mg/kg en los grupos G2 a G6 y diferentes dosis de Echinodorus grandiflorus, con posterior análisis de sangre y parámetros histológicos. La administración de ciclofosfamida a una dosis de 150mg/kg de masa corporal, en dosis única, fue capaz de inducir nefrotoxicidad aguda en todas las ratas. El extracto crudo de Echinodorus grandiflorus presentó un potencial efecto renoprotector al uso de ciclofosfamida, a una dosis de 300mg/kg de masa corporal, siendo posible observar una reducción de los efectos nefrotóxicos de la quimioterapia, por la reducción del daño tubular y por la disminución de los espacios capsulares, encontrándose claramente alterados en el grupo que recibió solamente ciclofosfamida, denotando resultados promisorios para el uso de esta planta medicinal en la prevención de la nefrotoxicidad inducida por el fármaco. Sin embargo, nuevos estudios sobre los efectos renoprotectores del sombrero de cuero podrían dilucidar los mecanismos implicados en la acción del extracto crudo de sombrero de cuero. El uso de extractos crudos de plantas medicinales se convierte en un coadyuvante de los tratamientos por su bajo coste y la facilidad de acceso de las diferentes poblaciones a las plantas desde que son guiadas adecuadamente por profesionales cualificados.

ABSTRACT

cistite hemorrágica e a cistite intersticial expressam uma etiologia variável, desde idiopática à provocada por fármacos, dentre eles a ciclofosfamida. A cistite apresenta tratamento multifatorial, e o potencial efeito satisfatório do uso da medicina complementar, vem ganhando espaço na prática médica. Assim o objetivo do presente estudo foi avaliar o efeito protetivo do extrato bruto de Echinodorus grandiflorus sobre a bexiga de ratos induzidos a cistite por ciclofosfamida. Utilizou-se neste estudo, 35 ratos, machos, Wistar, com peso médio de 321g, que foram submetidos a indução de cistite com uso de ciclofosfamida por via intraperitoneal e tratados com diferentes doses de extrato de Echinodorus grandiflorus (30, 100, 300mg) e o grupo controle com o fármaco Mesna. Todos os animais foram mortos no décimo sétimo dia e suas bexigas urinarias foram ressecadas para avaliação macro e microscópica, além da análise de hemograma e leucograma. A análise do sangue mostrou leucopenia com diferença significativa em todos os animais que receberam a ciclofosfamida. Observou-se que a dose de 300mg/kg do extrato bruto da planta, apresentou efeito protetivo no urotélio vesical, porém, inferior ao uso de Mesna. Diante dos resultados apresentados neste estudo sugere-se que o extrato de Echinodorus grandiflorus apresenta efeito protetivo no urotélio vesical na dose de 300mg/kg, porém estudos futuros quanto a dose e também a uma possível associação terapêutica ao Mesna devam ser realizados. Por se tratar de uma patologia com prevalência importante e ser muitas vezes desagradável e limitante à vida, faz-se necessário o empenho em métodos terapêuticos alternativos aos atuais, afim de, diminuírem os custos e efeitos colaterais dos métodos já documentados.


Hemorrhagic cystitis and interstitial cystitis have a variable etiology, from idiopathic to drug-induced, including cyclophosphamide. Cystitis has a multifactorial treatment, and the potential satisfactory effect of the use of complementary medicine has been gaining ground in medical practice. Thus, the aim of the present study was to evaluate the protective effect of the crude extract of Echinodorus grandiflorus on the bladder of rats induced to cystitis by cyclophosphamide. In this study, 35 male Wistar rats, with an average weight of 321g, were submitted to cystitis induction with intraperitoneal use of cyclophosphamide and treated with different doses of Echinodorus grandiflorus extract (30, 100, 300mg) and the control group with the drug Mesna. All animals were killed on the seventeenth day and their urinary bladders were resected for macro and microscopic evaluation, in addition to the analysis of blood count and leukogram. Blood analysis showed leukopenia with a significant difference in all animals that received cyclophosphamide. It was observed that the dose of 300mg/kg of the crude extract of the plant had a protective effect on the vesical urothelium, however, it was inferior to the use of Mesna. In view of the results presented in this study, it is suggested that the Echinodorus grandiflorus extract has a protective effect on the vesical urothelium at a dose of 300mg/kg, but future studies regarding the dose and also a possible therapeutic association with Mesna should be carried out. Because it is a pathology with significant prevalence and is often unpleasant and life-limiting, it is necessary to commit to alternative therapeutic methods to the current ones, in order to reduce the costs and side effects of the methods already documented.

Así, el objetivo del presente estudio fue evaluar el efecto protector del extracto crudo de Echinodorus grandiflorus sobre la vejiga de ratas inducidas a cistitis por ciclofosfamida. En este estudio, 35 ratas Wistar macho, con un peso promedio de 321g, fueron sometidas a inducción de cistitis con uso intraperitoneal de ciclofosfamida y tratadas con diferentes dosis de extracto de Echinodorus grandiflorus (30, 100, 300mg) y el grupo control con el fármaco Mesna. Todos los animales fueron sacrificados al decimoséptimo día y sus vejigas urinarias fueron resecadas para evaluación macro y microscópica, además del análisis de hemograma y leucograma. El análisis de sangre mostró leucopenia con una diferencia significativa en todos los animales que recibieron ciclofosfamida. Se observó que la dosis de 300 mg/kg del extracto crudo de la planta tuvo un efecto protector sobre el urotelio vesical, sin embargo, fue inferior al uso de Mesna. En vista de los resultados presentados en este estudio, se sugiere que el extracto de Echinodorus grandiflorus tiene un efecto protector sobre el urotelio vesical a una dosis de 300 mg/kg, pero se deben realizar estudios futuros sobre la dosis y también una posible asociación terapéutica con Mesna. llevado a cabo. Vitex megapotamica (Spreng.) Moldenke is a plant with medicinal properties popularly used in Brazil to treat diabetes and obesity. Despite the promising use of this plant, are still incipient toxicology studies on prolonged periods of treatment. The aim of this study was to evaluate the repeated dose 90-day oral toxicity study of V. megapotamica methanolic extract (VMME) in male and female Wistar rats. Different groups of rats (n = 8) were treated daily with three different doses of VMME (100, 300, and 900 mg/kg) or vehicle (filtered water). Body weight, water, and feed consumption, and clinical and behavioral changes were monitored daily. At the end of the experimental period, blood samples were obtained for hematological and biochemical analyzes. After euthanasia, the vital organs were removed for the determination of relative weight and for histopathological analysis. No clinical signs of toxicity or mortality were found during the experimental period. Treatment with VMME did not induce any change in body weight gain, eating patterns, and behavior. We found no statistically significant changes in the different hematological and biochemical parameters evaluated. The relative weight of the organs and histopathological analysis did not show any significant change when compared to animals treated with the vehicle. The data obtained in this study allow us to conclude that the VMME obtained from V. megapotamica is safe after a repeated-dose 90-day oral toxicity study in male and female Wistar rats.

Vitex megapotamica (Spreng) Moldenke is commonly known as tarumã, it is an important medicinal and edible fruit plant. It is native to regions of tropical and subtropical climate in greater proportion than temperate zones and widely distributed in Central America, South America, Asia, and Africa. In Brazil, it is present in the Atlantic Forest and Cerrado biomes. Despite its widespread use, there are no minimum standards for quality control or information on genotoxicity. Therefore, the aim of this study was to present a detailed description of the short-term genotoxicity assays of V. megapotamica and to provide parameters of a preparation routinely used in traditional folk medicine. For genotoxicity assays, five groups were used with eight wistar rats in each group. For this, three doses of the V. megapotamica extract in doses (100, 300, and 900 mg/kg) or negative control (filtered water) were administered orally and positive control cyclophosphamide monohydrate (20 mg/kg; Sigma-Aldrich®) was applied by the intraperitoneal route after 24 h. At the end, whole blood was collected in a tube containing EDTA for the comet test and later the animals were euthanized. For the micronucleus test, femurs were removed, and bone marrow was collected. In the comet assay, V. megapotamica crude extract did not show significant DNA damage at all doses tested. The micronucleus assay showed no significant increase in the frequency of inducing micronuclei at any dose examined. It can be concluded that the safety parameters in genotoxicity studies reveal that V. megapotamica has no toxicity, which characterizes the important quality control of this plant species.

ABSTRACT: Coturniculture has been promising, progressing from a subsistence to a technical activity due to its quick production, low breeding investment, and rapid economic return. After the restriction of antimicrobials as growth promoters, some studies aimed to evaluate alternative products that would make the farming of healthy birds viable without impacting their performance, with commercial Macleaya cordata extract being one of these substitutes. The functions of the gastrointestinal tract are coordinated mainly by the enteric nervous system, and the myenteric plexus is responsible for the reflex control of contractile activities of the external muscles. Thus, this study located and demonstrated the distribution of the myenteric plexus, quantifing the total population of myenteric neurons (Giemsa+) and the subpopulation of myenteric nitrergic neurons (NADPH-d+), and evaluated the effects of commercial Macleaya cordata extract on these populations of quail jejunum neurons. A total of 240 one-day-old female laying quails were distributed into four treatments, with four repetitions of 15 birds each. The test groups (T1, T2, and T3) were treated with commercial Macleaya cordata extract throughout the experimental period using the following doses: T1 - test group, basal diet added with 150 ppm of the extract in the feed; T2- test group, basal diet added with100 ppm of the extract in the feed; T3 - test group, basal diet added with 50 ppm of the extract in the feed; and T4 - control group, basal diet with no added extract. The study included histological analysis, Giemsa+, and NADPH-d+ myenteric neuron staining. The results showed that the myenteric plexus is located between longitudinal layer fibers and in the transition region between the longitudinal and circular layers of the muscular tunic, with the myenteric population organized into ganglia and isolated in the region of neuronal fiber bundles. The commercial Macleaya cordata extract showed no quantitative changes in the myenteric Giemsa+ population and myenteric NADPH-d+ subpopulation, however, the groups that consumed the extract showed greater NADPH-d+ neuron activity compared to the control group, implying that the food remained longer in the intestinal lumen, therefore, enabling greater nutrient use and resulting in increased productive performance.


RESUMO: A coturnicultura tem apresentado características promissoras, deixando de ser uma atividade de subsistência e ocupando patamares tecnificados devido a sua precocidade produtiva, baixo investimento de criação e rápido retorno econômico. A partir da restrição da utilização de antimicrobianos como promotores de crescimento, estudos foram direcionados com o objetivo de se avaliar produtos alternativos que viabilizassem a criação de aves saudáveis, sem comprometer seu desempenho, sendo o extrato comercial da Macleaya cordata um destes substitutos. As funções do trato gastrintestinal são coordenadas principalmente pelo sistema nervoso entérico, sendo o plexo mioentérico responsável pelo controle reflexo das atividades contráteis da musculatura externa. Desta forma, o presente trabalho teve como objetivo localizar e demonstrar a distribuição do plexo mioentérico, quantificar a população total de neurônios mioentéricos (Giemsa+), e a subpopulação de neurônios mioentéricos nitrérgicos (NADPH-d+), além de avaliar os efeitos do extrato comercial da Macleaya cordata sobre estas populações de neurônios do jejuno de codornas. Foram alojadas 240 codornas de postura, fêmeas, com um dia de idade, distribuídas aleatoriamente em quatro tratamentos, com quatro repetições de 15 aves cada. Os grupos testes (T1, T2 e T3) foram tratados com extrato comercial de Macleaya cordata durante todo o período experimental conforme as doses indicadas, sendo: T1 - grupo teste, com dieta basal adicionada de 150 ppm do extrato na ração; T2 - grupo teste, com dieta basal adicionada de 100 ppm do extrato na ração; T3 - grupo teste, com dieta basal adicionada de 50 ppm do extrato na ração, e T4 - grupo controle, com dieta basal isenta do extrato. Foram realizadas análise histológica e a marcação dos neurônios mioentéricos Giemsa+ e NADPH-d+. Os resultados demonstraram que o plexo mioentérico está localizado entre as fibras do estrato longitudinal, e na região de transição entre os estratos longitudinal e circular da túnica muscular, estando a população mioentérica organizada em gânglios, e também isoladamente na região dos feixes das fibras neuronais. O extrato comercial da Macleaya cordata não alterou quantitativamente os neurônios da população mioentérica Giemsa+ e da subpopulação mioentérica NADPH-d+, mas os grupos que consumiram o extrato apresentaram maior atividade dos neurônios NADPH-d+ em relação ao grupo controle, permitindo inferir que o alimento permaneceu maior tempo no lúmen intestinal e, portanto, possibilitou um maior aproveitamento dos nutrientes, podendo refletir em melhor desempenho produtivo.

ABSTRACT

Currently, a large number of consumers seek to include functional foods in their diets, aiming beyond the nutritional value prevent health problems, among these foods are probiotic products and vegetables containing bioactive compounds. The objective of this work was to develop and physical-chemically evaluate fresh Shanklish cheeses with the addition of kefir and turmeric extract in order to develop a functional cheese. Shanklish cheese was manufactured and submitted to three different treatments: with the addition of kefir, turmeric extract and both of them. The results obtained for cheese composition were close to the results found in the literature and the cheeses showed stable pH values during the 21 days of storage at 8ºC. It concluded that the addition of kefir and turmeric extract in Shanklish cheese is a way to develop a functional cheese.(AU)


Atualmente é grande o número de consumidores que buscam incluir alimentos funcionais em suas dietas visando além do valor nutricional a prevenção de problemas de saúde, entre esses alimentos encontram-se os produtos probióticos e os vegetais contendo compostos bioativos. O objetivo deste trabalho foi desenvolver e avaliar físico-quimicamente queijos Chancliche frescos com adição de kefir e extrato de cúrcuma buscando o desenvolvimento de um queijo funcional. Foram fabricados queijos Chancliche e submetidos a três tratamentos diferentes: com adição de kefir, extrato de cúrcuma e ambos. Os resultados obtidos para a composição dos queijos estavam próximos dos resultados encontrados na literatura e os queijos mostraram pH estável durante os 21 dias de armazenamento a 8ºC. Concluiu-se que a adição de kefir e extrato de cúrcuma em queijo Chancliche é uma maneira de desenvolver um queijo funcional.(AU)


Actualmente, una gran cantidad de consumidores buscan incluir alimentos funcionales en sus dietas, apuntando más allá del valor nutricional para prevenir problemas de salud, entre estos alimentos se encuentran los productos probióticos y los vegetales que contienen compuestos bioactivos. El objetivo de este trabajo fue desarrollar y evaluar químicamente quesos Chancliche frescos con la adición de kéfir y extracto de cúrcuma para desarrollar un queso funcional. Los quesos Chancliche fueron elaborados y sometidos a tres tratamientos diferentes: con la adición de kéfir, extracto de cúrcuma y ambos. Los resultados obtenidos para la composición de los quesos estaban cercanos a los resultados encontrados en la literatura y los quesos mostraron pH estable durante los 21 días de almacenamiento a 8ºC. Se concluyó que la adición de kéfir y extracto de cúrcuma en queso Chancliche es una forma de desarrollar un queso funcional.(AU)

ETHNOPHARMACOLOGICAL RELEVANCE: Baccharis trimera (Less.) DC is a perennial subshrub, popularly known as "carqueja," that belongs to the Asteraceae family. Ethnobotanical studies indicate that this species is used for the treatment of diabetes and digestive and liver diseases. However, studies that sought to validate its popular use were conducted using ethanolic extracts of the plant, which does not reflect the ethnomedicinal use of this species in humans. AIM OF THE STUDY: Non-alcoholic fatty liver disease (NAFLD) is characterized by triglyceride accumulation in the liver that can progress to cirrhosis and hepatocellular carcinoma. Because of the severity of this disease, less toxic and more effective therapeutic agents need to be developed. B. trimera may be a promising therapeutic alternative, but its activity against multiple risk factors for liver disease (e.g., smoking, dyslipidemia, and diabetes mellitus) has not been studied. The present study investigated the effects of an ethnomedicinal form of a B. trimera preparation in a rat model of NAFLD that is associated with multiple risk factors. MATERIAL AND METHODS: Phytochemical analysis of the ethanolic soluble fraction of B. trimera extract was performed using ultra-performance liquid chromatography coupled to high-resolution mass spectrometry. Streptozotocin was used to induce diabetes in male Wistar rats. The rats received a 0.5% cholesterol-enriched diet and were exposed to cigarette smoke (9 cigarettes/day, 5 days/week, for 4 weeks). In the last 2 weeks, the animals were orally treated with vehicle (negative control group), B. trimera extract (30, 100, and 300 mg/kg), or insulin + simvastatin. One group of rats that was not exposed to these risk factors was also evaluated. Blood was collected for glucose, alanine aminotransferase (ALT), and aspartate aminotransferase (AST) analysis. The liver and feces were collected for lipid quantification. The liver was additionally processed for histopathological analysis. RESULTS: The model successfully induced NAFLD and increased levels of glucose, AST, and ALT in the negative control group. Treatment with the B. trimera extract (30 and 100 mg/kg) and insulin + simvastatin decreased hepatic and fecal lipids. In contrast to insulin + simvastatin treatment, all three doses of B. trimera effectively reduced AST and ALT levels. CONCLUSION: B. trimera may be promising as a hepatoprotective agent against hepatic lesions that are caused by multiple risk factors.

ABSTRACT

O cobre é um mineral essencial ao metabolismo e seu uso tem sido associado à melhora do desempenho zootécnico, aumento da resistência ao estresse e doença nas aves. A indústria avícola com o intuito de maximizar o desempenho produtivo formula normalmente as dietas com altas doses da fonte inorgânica de cobre, o que contribui para o desequilíbrio metabólico e como consequência aumenta a poluição ao meio ambiente. Dessa forma, o uso de fontes orgânicas de cobre tem sido estimulado. Sendo assim, o objetivo do presente trabalho foi revisar a utilização do cobre na dieta de frangos de corte com enfoque para os benefícios das fontes orgânicas. Conclui-se que fontes mais biodisponíveis de cobre são uma oportunidade para diminuir sua inclusão, mantendo o desempenho do animal, por meio da redução de agentes patogênicos, melhora na digestibilidade e redução ao impacto ambiental em dietas de frango de corte.(AU)


Copper is an essential mineral for the metabolism and its use has been associated with improved zootechnical performance and increased resistance to stress and diseases in broilers. In order to maximize the productive performance, poultry nutritionists usually formulate diets with high doses of copper inorganic sources, contributing thereby to favor metabolic imbalance and environmental pollution. The purpose of this study was to review the use of copper in broiler diets with a focus on the benefits of organic sources. It can be concluded that more bioavailable sources of copper represent an opportunity to reduce its inclusion without compromising animal performance. Moreover, proper management of copper in broiler diets can contribute to reduce pathogens, improve digestibility and reduce the environmental impact of the poultry industry.(AU)


El cobre es un mineral esencial al metabolismo y su uso se ha asociado a mejora del desempeño zootécnico, aumento de la resistencia al estrés y enfermedades en las aves. La industria avícola con el fin de maximizar el rendimiento productivo, formula normalmente las dietas con altas dosis de fuente inorgánica de cobre, lo que contribuye al desequilibrio metabólico y como consecuencia aumenta la contaminación para el medio ambiente. Por lo tanto, el uso de fuentes orgánicas de cobre ha sido estimulado. Así, el objetivo de este estudio fue revisar el uso de cobre en la dieta de pollos de engorde, con enfoque para los beneficios de las fuentes orgánicas. Se concluye que las fuentes más biodisponibles de cobre son una oportunidad para reducir su inclusión, manteniendo el rendimiento de los animales a través de la reducción de agentes patógenos, digestibilidad mejorada y reducción al impacto ambiental en dietas de pollo de engorde.(AU)

BACKGROUND: The mushroom Agaricus blazei has evoked considerable scientific and practical interest in several fields, especially those linked to its medicinal properties. This review aims to summarize and evaluate the past decade findings related to nutritional and therapeutic uses of A. blazei, with especial emphasis on the most recent discoveries regarding its chemical composition and clinical investigations. METHODS: The specialized literature was searched for basic and clinical studies. The main isolated and identified compounds or fractions are described and confronted with their corresponding bioactivities. RESULTS: Basic research of high quality using ex vivo and in vivo conditions are quite abundant in the specialized literature, but ony 17 clinical studies and two case reports were found. A great number of active molecules have been identified, and they can be divided into three categories, (1) hydrophilic small molecules (e.g., phenolics), (2) lipophilic or partially lipophilic small molecules (e.g., agarol) (3) and macromolecules (e.g., ß-glucans). At least the following bioactivities can be considered as being supported by experimental evidence: antioxidant activity (in aging or disease), immunomodulation and cell signaling, anti-inflammatory activity, antiparasitic actions, antimicrobial activity, anticancer effects and tumor growth inhibiting effects, antimutagenic activity, hepatoprotection against chemical or viral infection and antidiabetic activity. CONCLUSION: The amount and quality of the evidence that has been accumulating during the last decade strongly speaks in favor of the health benefits of the ingestion of A.blazei or derived products. However, there are many uncertainties and limitations when attempts are made to extrapolate or to demonstrate their biological effects in the human organism in health or disease. Clearly, more clinical trials, using reliable statistical methods and standardized preparations are needed to establish the efficacy of A. blazei as a therapeutic agent.

ABSTRACT

As espécies reativas ao oxigênio (EROS) são produzidas como mecanismo de defesa celular, participando dos processos de cicatrização celular. Entretanto, altos níveis de EROS podem causar danos como a peroxidação lipídica (PL). O presente estudo teve como objetivo, verificar os níveis de PL por meio da determinação das substâncias reativas ao ácido tiobarbitúrico (TBARS) no plasma de ratos com lesão tecidual induzida. Foram utilizados 32 ratos machos, Rattus norvegicus albinus da linhagem Wistar, os quais foram pesados e da média ± 10% do peso foram distribuídos em quatro grupos: A ­ controle negativo; B - Vetaglós®; C ­ hidrogel de poliamido de mandioca+ Vetaglós®; D ­ Hidrogel de poliamido de mandioca. Após 21 dias, todos os animais foram anestesiados com isoflurano e foi feita a coleta de sangue por punção cardíaca, e os plasmas foram obtidos após centrifugação, na sequência por superdosagem do anestésico foi realizada a eutanásia. Os níveis de PL nos plasmas dos ratos foram determinados pelo método do TBARS. Não houve diferença significativa entre os grupos em relação à PL, indicando um equilíbrio entre as defesas antioxidantes celulares e os níveis de EROS produzidos durante o processo de cicatrização celular. Essa ausência nos diferentes grupos experimentais, em relação à PL, deixa claro a importância de se contemplar estudos de parâmetros de bioindicadores de estresse oxidativo em protocolos experimentais.


Reactive oxygen species (ROS) are produced as a cellular defense mechanism, participating in the processes of cellular healing. However, high levels of ROS can cause damages such as lipid peroxidation (LPO). This study aimed to verify the levels of LPO through the determination of reactive substances to thiobarbituric acid (TBARS) in rat plasma with induced tissue injury. A total of 32 Rattus norvegicus albinus Wistar were used, with a mean weight ± 10%. They were divided into four groups: A ­ negative control; B - Vetaglós®; C - Polyamide cassava; D - Polyamide cassava + Vetaglós®. After 21 days, all animals were anesthetized with isoflurane and blood was collected by cardiac puncture. Plasma was obtained after centrifugation. Euthanasia was performed with administration of an overdose of inhalational anesthetic previously used. The LPO levels in rat plasma were determined using the TBARS method. There was no significant difference between the groups in relation to LPO, indicating a balance between antioxidant defenses and cellular levels of ROS produced during the cellular healing process. This absence in the different experimental groups in relation to LPO emphasizes the importance of further studies related to the bio-indicator parameters for oxidative stress in experimental protocols.


Las especies reactivas al oxígeno (EROS) se producen como mecanismo de defensa celular, que participan en los procesos de curación celulares. Sin embargo, los altos niveles de EROS pueden causar daños como la peroxidación lipídica (PL). Este estudio tuvo como objetivo verificar los niveles de peroxidación lipídica por sustancias reactivas al ácido tiobarbitúrico (TBARS) en el plasma de ratas con lesión tisular inducida. Se han utilizado 32 ratas machos, Rattus norvegicus albinus de linaje Wistar, que se pesaron y la media ± 10% en peso, y se dividieron en cuatro grupos: A ­ control negativo; B - Vetaglós®; C ­ hidrogel de poliamida de yuca + Vetaglós®; D ­ Hidrogel de poliamida de yuca. Después de 21 días, todos los animales fueron anestesiados con isoflurano y se hizo la extracción de sangre por punción cardiaca, y se obtuvieron los plasmas después de la centrifugación, enseguida con sobredosis de anestésico se realizó la eutanasia. Los niveles de PL en los plasmas de las ratas se determinaron por el método de TBARS. No hubo diferencia significativa entre los grupos en relación a la peroxidación lipídica, lo que indica un equilibrio entre las defensas antioxidantes celulares y los niveles de EROS producidos durante el proceso de curación celular. Esa ausencia en los diferentes grupos experimentales, en relación a la PL, pone de manifiesto la importancia de contemplarse estudios de parámetros de bioindicadores de estrés oxidativo en los protocolos experimentales.

ABSTRACT

A doença do refluxo gastroesofágico é causada pela redução da pressão do esfíncter esofágico inferior e pode ser agravado por fracionamento inadequado da dieta e pela produção de espécies reativas ao oxigênio. Os tratamentos medicamentosos e cirúrgicos se destacam, contudo, existem opções de terapêutica não farmacológica, como as vitaminas e minerais com funções antioxidantes, atuam como adjuvantes neste tipo de tratamento. Portanto, este trabalho teve como objetivo avaliar as funções das vitaminas e minerais suplementados junto à dieta fracionada, prescritas a uma paciente de cirurgia de hérnia de hiato, e a diminuição do quadro de esofagite. A paciente chegou com quadro de esofagite de refluxo, disfagia, dores na região gástrica e distensão abdominal. Como conduta foi adotada a suplementação com piridoxina 30mg, zinco quelato 20 mg e ácido pantotênico 20 mg e vitamina C revestida 200 mg, assim como aumento do fracionamento da dieta, refletindo na redução do volume das refeições para amenizar desconfortos gástricos. O fracionamento adequado da dieta, a redução do volume das refeições e a suplementação atuam de maneira eficaz na melhoria do quadro da DRGE. Cabe destacar que é papel do nutricionista garantir a adesão do paciente ao tratamento para não haver recidivas de sintomas.


The gastroesophageal reflux disease is caused by the reduction of the pressure on the lower esophageal sphincter and could be aggravated by inadequate division of the diet and by the production of species that are reactive to oxygen. Medical and surgical treatments are emphasized, but there are also non-pharmacological therapeutic options, such as vitamins and minerals with antioxidant function, acting as adjuvants in this kind of treatment. Therefore, this paper has the purpose of assessing the functions of supplemented vitamin and minerals with a fractionated diet, prescribed to a surgical patient with hiatal hernia, and the reduction of esophagitis. The patient arrived complaining of esophagitis reflux, dysphagia, pain in the gastric region and abdominal distension. Supplementation with 30mg pyridoxine, 20mg zinc chelate and 20mg pantothenic acid was prescribed as treatment, as well as 200mg coated vitamin C tablet, as well as increasing the diet division to reflect the reduction of meal volumes to sooth gastric discomforts. The appropriate fragmentation of the diet, the reduction of meal volumes and the supplementation were effective to improve the GERD condition. It is important to mention that the role of the nutritionist is to ensure the patient adheres to the treatment in order to prevent symptom relapse.

This study evaluated the effects of the supplementation with aqueous extract of Agaricus blazei Murrill (ABM) on biometric and blood parameters and quantitative morphology of the myenteric plexus and jejunal wall in aging Wistar rats. The animals were euthanized at 7 (C7), 12 (C12 and CA12), and 23 months of age (C23 and CA23). The CA12 and CA23 groups received a daily dose of ABM extract (26 mg/animal) via gavage, beginning at 7 months of age. A reduction in food intake was observed with aging, with increases in the Lee index, retroperitoneal fat, intestinal length, and levels of total cholesterol and total proteins. Aging led to a reduction of the total wall thickness, mucosa tunic, villus height, crypt depth, and number of goblet cells. In the myenteric plexus, aging quantitatively decreased the population of HuC/D(+) neuronal and S100(+) glial cells, with maintenance of the nNOS(+) nitrergic subpopulation and increase in the cell body area of these populations. Supplementation with the ABM extract preserved the myenteric plexus in old animals, in which no differences were detected in the density and cell body profile of neurons and glial cells in the CA12 and CA23 groups, compared with C7 group. The supplementation with the aqueous extract of ABM efficiently maintained myenteric plexus homeostasis, which positively influenced the physiology and prevented the death of the neurons and glial cells.

ABSTRACT

The fruit extracts of Citrus aurantium (bitter orange) are traditionally used as weight-loss products and as appetite supressants. An important fruit component is p-synephrine, which is structurally similar to the adrenergic agents. Weight-loss and adrenergic actions are always related to metabolic changes and this work was designed to investigate a possible action of the C. aurantium extract on liver metabolism. The isolated perfused rat liver was used to measure catabolic and anabolic pathways, including oxygen uptake and perfusion pressure. The C. aurantium extract and p-synephrine increased glycogenolysis, glycolysis, oxygen uptake and perfusion pressure. These changes were partly sensitive to α- and ß-adrenergic antagonists. p-Synephrine (200 µM) produced an increase in glucose output that was only 15% smaller than the increment caused by the extract containing 196 µM p-synephrine. At low concentrations the C. aurantium extract tended to increase gluconeogenesis, but at high concentrations it was inhibitory, opposite to what happened with p-synephrine. The action of the C. aurantium extract on liver metabolism is similar to the well known actions of adrenergic agents and can be partly attributed to its content in p-synephrine. Many of these actions are catabolic and compatible with the weight-loss effects usually attributed to C. aurantium.
