ABSTRACT

Here we present an original approach to generate 2D high detail riverbed based on a drone photogrammetric survey, and RTK bathymetry measurements for Mera river in the Italian Alps. The aim is to better represent macro-roughness and riverbed structure of the river, also extending it to an ungauged area. Specifically, we apply a step-by-step approach. I) Depth and average slope of the riverbed were calculated from bathymetry data. II) Thus, a trapezoidal channel with constant slope and variable width was defined using the drone images. III) Riffle-pool sequence was assessed as a function of river width and applied to the generated channel. IV) Finally, the semi-random Perlin Noise was added to recreate riverbed irregularities in the natural stream. HEC-RAS 2D hydraulic software was then implemented to assess spatialized water depth and velocity. The proposed methodology could be quite relevant in river hydraulics to decouple roughness coefficient from water submergence, and in Physical Habitat Simulation Model (PHABSIM), where the dependency of the output is not linear with hydraulic parameters (i.e. water depth and velocity). Indeed, we apply PHABSIM for a case study of a stretch of the river and results are compared with a previous environmental study for Mera river.

ABSTRACT

Erosion is a main form of soil degradation, with severe consequences on slope stability and productivity, and erosion studies are required to predict possible variations of such phenomena, also under climate change scenarios. Here we estimated distributed soil erosion within Valchiavenna valley in the Rhaetian Alps, drained by Mera river, and covering Italy, and Switzerland. We used a Dynamic-RUSLE (D-RUSLE) model, which provides spatially distributed estimates of soil erosion explicitly considering snow dynamic (accumulation/melting) and snow cover, and vegetation seasonality. The model was tuned here during 2010-2019, and validation was pursued using river turbidity data, used to assess riverine sediment transport. The model parameter R-factor for rainfall erosivity was estimated using a hydrological model Poli-Hydro, properly set up in the study area. C-factor for land cover was assessed against land cover maps, with seasonally variable Normalized Difference Vegetation Index from satellite images, to account for variable vegetation stage, and large leaf cover in summer. The K-factor related to erosion susceptibility was evaluated through soil texture and organic content. LS-factor depending on slope was assessed using a DTM. Poli-Hydro and D-RUSLE models were then used to project forward potential soil erosion under climate change scenarios until 2100. Climate series (temperature, precipitation) were generated using 4 shared socio-economic pathways (SSPs) of the Sixth Assessment Report of the IPCC, with 3 global circulation models, properly downscaled locally. We analysed expected soil erosion during 2051-2060, and 2091-2100. We found increase of potential soil erosion, with exception of the EC-Earth model for the SSP2.6. Erosion would especially increase in winter, in response to smaller snow accumulation, and larger liquid rainfall share thereby, and decrease in summer, as due to decreased precipitation. Our results suggest the need for adaptation strategies to counteract increasing soil loss in the future, and may highlight most critical areas of intervention.

INTRODUCTION: Despite extensive research, anastomotic leakage (AL) remains one of the most dreaded complications after colorectal surgery. Since butyrate enemas are known to enhance anastomotic healing, several administration routes have been explored in this study. METHODS: Three intraluminal approaches involving butyrate were investigated: (1) butyrin-elucidating patch, (2) a single injection of hyaluronan-butyrate (HA-But) prior to construction of the proximal anastomosis and (3) rectal hyaluronan-butyrate (HA-But) enemas designed for distal anastomoses. The main outcome was AL and secondary outcomes were bursting pressure, histological analysis of the anastomosis, zymography to detect MMP activity and qPCR for gene expression of MMP2, MMP9, MUC2 and TFF3. RESULTS: Neither the patches nor the injections led to a reduction of AL in experiments 1 and 2. In experiment 3, a significant reduction of AL was accomplished with the (HA-But) enema compared to the control group together with a higher bursting pressure. Histological analysis detected only an increased inflammation in experiment 2 in the hyaluronan injection group compared to the control group. No other differences were found regarding wound healing. Zymography identified a decreased proenzyme of MMP9 when HA-But was administered as a rectal enema. qPCR did not show any significant differences between groups in any experiment. CONCLUSION: Butyrate enemas are effective in the enhancement of colonic anastomosis. Enhanced butyrate-based approaches designed to reduce AL in animal models for both proximal and distal anastomoses were not more effective than were butyrate enemas alone. Further research should focus on how exogenous butyrate can improve anastomotic healing after gastrointestinal surgery.

ABSTRACT

Activated neutrophils (PMNs), the ROS/RNS released by PMNs and the derived inflammatory processes are involved in the pathogenesis and progression of human inflammatory airway diseases. Similar diseases are also present in horses which suffer from recurrent airway obstruction (RAO), exercise-induced pulmonary haemorrhage (EIPH) and inflammatory airway diseases (IAD). Hyaluronic acid (HA) plays numerous roles in modulating inflammatory processes. The aim of this study was to examine whether a preparation of HA (MW 900 000 Da) interferes with ROS/RNS during the course of equine PMN respiratory bursts, and to establish the lowest concentration at which it still has antioxidant activity by means of luminol-amplified chemiluminescence (LACL). Electron paramagnetic resonance (EPR) spectroscopy was also used to investigate the direct antiradical activity of HA. The hydroxyl radical was significantly scavenged in a concentration-dependent manner at HA concentrations ranging from 2.5 to 0.16 mg/mL. Superoxide anion, Tempol radical and the ABTS(•+) were significantly inhibited at concentrations ranging from 2.5 to 0.62 mg/mL. The LACL of stimulated equine neutrophils showed that HA induced a statistically significant concentration-effect reduction from 5 mg/mL to 1.25 mg/mL. These findings were confirmed also when l-Arg was added to investigate the inhibition of the resulting peroxynitrite anion. Our findings indicate that, in addition to the human use, HA can also be used to antagonize the oxidative stress generated by free radicals in horses peripheral blood mononuclear cells (PBMCs). In order to achieve therapeutic concentrations, a direct aerosol administration to horses with horse respiratory diseases can be considered, as this route of application is also recommended in human medicine.

ABSTRACT

Fluid flow advecting one substance while others are immobilized can generate an instability in a homogeneous steady state of a reaction-diffusion-advection system. This differential-flow instability leads to the formation of steady spatial patterns in a moving reference frame. We study the effects of shear flow on this instability by considering two layers of fluid moving independently from each other, but allowing the substances to diffuse along and across the layers. We find that shear flow can generate instabilities even if the average flow velocity is zero for both substances. These instabilities are strongly dependent on which substance is advected by the shear flow. We explain these effects using the results of Taylor dispersion, where an effective diffusivity is enhanced by shear flow.

In a prospective trial 151 recipients of renal transplants were randomly assigned to treatment with CsA alone (74 patients) and to low dose of AZA, prednisolone, and CsA (77 patients). At two years, graft survival was 84% for the monotherapy and 90% for the triple therapy. This difference was not statistically significant. The number of rejection episodes was similar in the two groups, but the severity of rejection was significantly worse among the patients on monotherapy. More kidneys were lost because of rejection (6 versus 3), and a higher number of methylprednisolone pulses was used for treating rejection (5.2 +/- 2.3 versus 4.3 +/- 2.9; P = 0.0077). CsA nephrotoxicity episodes were more frequent among patients on monotherapy (23 versus 7; P less than 0.02). Infectious episodes were equally distributed between the two groups. Creatinine clearance was poorer in the monotherapy-treated patients at the third month (42 +/- 16 ml/min versus 48 +/- 15 ml/min; P = 0.02), but no differences were observed between the two groups since the sixth month after transplantation. Many patients on monotherapy required changes in maintenance therapy. In fact, one patient was switched to conventional immunosuppression because of Cremophor-induced anaphylaxis. Another patient who developed Kaposi's sarcoma 4 months after surgery was switched to steroids alone. Excluding 5 patients who lost their grafts a few days after transplantation, only 30 of 74 patients (40%) could be kept without steroids. We conclude that both the therapeutic protocols can give good results in renal allotransplantation; however, monotherapy could create some problems in keeping the balance between drug toxicity and significant immunosuppression. On the contrary, triple therapy is easier to handle, especially in the early posttransplant period when the differential diagnosis between acute rejection and CsA-related nephrotoxicity can be difficult even for a skilled clinician.
