ABSTRACT

Recent advances in DNA nanotechnology allow for the assembly of nanocomponents with nanoscale precision, leading to the emergence of DNA-based material fabrication approaches. Yet, transferring these nano- and micron-scale structural arrangements to the macroscale morphologies remains a challenge, which limits the development of materials and devices based on DNA nanotechnology. Here, we demonstrate a materials fabrication approach that combines DNA-programmable assembly with actively driven processes controlled by acoustic fields. This combination provides a prescribed nanoscale order, as dictated by equilibrium assembly through DNA-encoded interactions, and field-shaped macroscale morphology, as regulated by out-of-equilibrium materials formation through specific acoustic stimulation. Using optical and electron microscopy imaging and x-ray scattering, we further revealed the nucleation processes, domain fusion, and crystal growth under different acoustically stimulated conditions. The developed approach provides a pathway for the fabrication of complexly shaped macroscale morphologies for DNA-programmable nanomaterials by controlling spatiotemporal characteristics of the acoustic fields.

ABSTRACT

Functional gastrointestinal disorders, which had an impact on the dentofacial system (pain, loose teeth and falling out of them) in patients who have had COVID-19, drew the close attention of specialists of different profiles. The pathogenesis of worsening post-COVID edentulism is insufficiently studied, as many issues of adequate therapy remain unsolved, in which the role of non-drug technologies in the treatment of dental patients who have suffered from COVID-19 is extremely high. OBJECTIVE: To describe the mechanism of action and clinical effectiveness of the developed combined physiotherapy method, including the induced technique of piracetam iontophoresis on the frontooccipital technic and acupuncture laser therapy in dental patients with complaints of edentulism progression after COVID-19 on the basis of the analysis of single studies on the post-COVID loss of teeth treatment. MATERIAL AND METHODS: A number of patients equal 120 who complained of tooth loss after COVID-19 during the past 6 months were examined. The following initial and end points were considered: dental bleeding and inflammation scores, vascular and endothelial dysfunction markers - levels of intercellular adhesion molecules and their receptors (SlCAM-1, SVCAM-1, VEGF-A, ET-1) before and after treatment. RESULTS: Negative correlation between VEGF-A (pg/ml) concentration in peripheral blood serum and sVCAM-1 (ng/ml) level in the examined patients (r=0.4830, p<0.05) and strong inverse correlation between slCAM-1 (ng/ml) level and sVCAM-1 (r=0.7696, p<0.01) have been established. More significant effects after application of the combined induced method on the head's structures and laser acupuncture have been noted than after acupuncture laser exposure and after inducing technique separately, namely in the form of dental inflammation score correction by 1.76 times (p<0.001), decrease of bleeding score by 2.6 (p<0.05), decrease of concentration of SVCAM-1 by 1.7 times and SlCAM-1 by 2 times (p<0.001), increase of endothelin level by 1.7 times as well as the initial low VEGF-A (pg/ml) by 1.5 times (p<0.01). CONCLUSION: The developed physiotherapeutic complex, which includes laser acupuncture physiotherapy and induced technique of 5% piracetam iontophoresis, can potentially be considered as a physioprophylactic and therapeutic model of post-COVID edentulism.

ABSTRACT

Bacterial stress adaptive response is formed due to changes in the cell gene expression profile in response to alterations in environmental conditions through the functioning of regulatory networks. The mutual influence of network signaling molecules represented by cells' natural metabolites, including indole and second messengers (p) ppGpp and cAMP, is hitherto not well understood, being the aim of this study. E. coli parent strain BW25141 ((p) ppGpp+) and deletion knockout BW25141ΔrelAΔspoT which is unable to synthesize (p)ppGpp ((p)ppGpp0) were cultivated in M9 medium supplemented with different glucose concentrations (5.6 and 22.2 mM) in the presence of tryptophan as a substrate for indole synthesis and in its absence. The glucose content was determined with the glucose oxidase method; the indole content, by means of HPLC; and the cAMP concentration, by ELISA. The onset of an increase in initially low intracellular cAMP content coincided with the depletion of glucose in the medium. Maximum cAMP accumulation in the cells was proportional to the concentration of initially added glucose. At the same time, the (p) ppGpp0 mutant showed a decrease in maximum cAMP levels compared to the (p)ppGpp+ parent, which was the most pronounced in the medium with 22.2 mM glucose. So, (p)ppGpp was able to positively regulate cAMP formation. The promoter of the tryptophanase operon responsible for indole biosynthesis is known to be under the positive control of catabolic repression. Therefore, in the cells of the (p)ppGpp+ strain grown in the tryptophan-free medium that were characterized by a low rate of spontaneous indole formation, its synthesis significantly increased in response to the rising cAMP level just after glucose depletion. However, this was not observed in the (p)ppGpp0 mutant cells with reduced cAMP accumulation. When tryptophan was added to the medium, both of these strains demonstrated high indole production, which was accompanied by a decrease in cAMP accumulation compared to the tryptophan-free control. Thus, under glucose depletion, (p)ppGpp can positively regulate the accumulation of both cAMP and indole, while the latter, in its turn, has a negative effect on cAMP formation.

ABSTRACT

Polyamines and indole are small regulatory molecules that are involved in the adaptation to stress in bacteria, including the regulation of gene expression. Genes, the translation of which is under the regulatory effects of polyamines, form the polyamine modulon. Previously, we showed that polyamines upregulated the transcription of genes encoding the ribosome hibernation factors RMF, RaiA, SRA, EttA and RsfS in Escherichia coli. At the same time, indole affected the expression at the transcriptional level of only the raiA and rmf genes. Ribosome hibernation factors reversibly inhibit translation under stress conditions, including exposure to antibiotics, to avoid resource waste and to conserve ribosomes for a quick restoration of their functions when favorable conditions occur. In this work, we have studied the influence of indole on the expression of the raiA and rmf genes at the translational level and regulatory effects of the polyamines putrescine, cadaverine and spermidine on the translation of the rmf, raiA, sra, ettA and rsfS genes. We have analyzed the mRNA primary structures of the studied genes and the predicted mRNA secondary structures obtained by using the RNAfold program for the availability of polyamine modulon features. We have found that all of the studied genes contain specific features typical of the polyamine modulon. Furthermore, to investigate the influence of polyamines and indole on the translation of the studied genes, we have constructed the translational reporter lacZ-fusions by using the pRS552/λRS45 system. According to the results obtained, polyamines upregulated the expression of the rmf, raiA and sra genes, the highest expression of which was observed at the stationary phase, but did not affect the translation of the ettA and rsfS genes, the highest expression of which took place during the exponential phase. The stimulatory effects were polyamine-specific and observed at the stationary phase, when bacteria are under multiple stresses. In addition, the data obtained demonstrated that indole significantly inhibited translation of the raiA and rmf genes, despite the stimulatory effect on their transcrip- tion. This can suggest the activity of a posttranscriptional regulatory mechanism of indole on gene expression.

ABSTRACT

OBJECTIVE: To study visual event-related potentials (ERPs) associated with presentation of erotic pictures in backward masking condition in order to investigate perception, attention and information processing underlying the initiation and maintenance of abnormal sexual behavior in pedophilia. MATERIAL AND METHODS: Participants (58 males committed sexual offenses against children) were divided into the pedophilia group (n=38) and the control group without disorders of sexual preference (n=20). Three types of stimuli were used: neutral pictures of nature (S1), adult heterosexual erotic pictures (S2) and erotic pictures with children (S3). Posterior N1 (N140) and anterior N1 (N120) and P3 (P320) ERP components were included in analysis. RESULTS: In both groups, the perception of erotic stimuli was accompanied by a significant increase in the amplitude and prolongation of the latency of the N140 and P320 components (p<0.001). N140 changes were characterized by right-hemispheric bias in both groups, however, asymmetry was observed for amplitude in the control group, and for latency - in the pedophilia group; besides, reactivity to S3 stimuli was weaker than to S2 ones in the pedophilia group. N120 amplitude was elevated significantly in response to S2 stimuli in the control group, and no changes were found in the pedophilia group. Between-group differences were revealed for early negative components N140 and N120, which parameters were entered as predictors into classification model based on discriminant analysis. The obtained accuracy of model was 87.23%. CONCLUSION: The specificity of neurophysiological reactions to erotic pictures in pedophilic patients corroborates the concept of disease as general neurodevelopmental disorder.

ABSTRACT

OBJECTIVE: The neurophysiological objectification of intranosological differentiation of main clusters (schizoid/paranoid and hysterical/emotionally unstable) within the typology of personality disorders (PD), as well as the differentiation of schizoid/paranoid PD and schizophrenia. MATERIAL AND METHODS: Two groups of patients with PD participated in the study: PD1 - with schizoid/paranoid pattern (n=16), and PD2 - with hysterical/emotionally unstable pattern (n=18). The data were compared with the control group of healthy volunteers (n=86) and the group of schizophrenic patients (Sz, n=67). The analysis of four neurophysiological endophenotypes (measures of prepulse inhibition (PPI) of acoustic startle response and of P50 suppression, P300 amplitude, percentage of antisaccade errors) was carried out. RESULTS: The decrease of PPI and P50 suppression (measures of sensorimotor and sensory gating) relative to the control group was found both for Sz (p<0.01) and PD1 (p<0.05) groups. The highest P300 amplitude was observed in the control group, while the lowest values were observed in the Sz group (p<0.001); the values of PD1 and PD2 groups were intermediate. In all clinical groups percent of antisaccade errors was significantly higher compared to the control group (Sz - p<0.001; PD1 - p<0.05; PD2 - p<0.01). However, the level of the fronto-central cortical activation during antisaccade performance (estimated by contingent negative variation amplitude) was significantly reduced only in the Sz group (p<0.01), which is consistent with the concept of «hypofrontality¼ in schizophrenia. The changes associated with increased emotional reactivity were found in PD2 group only (P300 amplitude frontal maximum, asymmetry of error percent in antisaccade task). CONCLUSION: The study showed that two personality patterns in PD patients are related to the individual specificity of functional brain networks.

ABSTRACT

Nowadays, sliding is the least investigated mode of bacterial motility. Sliding is a process of passive movement on the surface of semi-liquid mediums which was originally described for mycobacteria and other bacterial species deprived of the organelles specialized for movement. Some mycobacteria are able to colonize surfaces, including tissues of macro-organisms, using glycopeptidolipids localized in the cell envelope for this aim. This is a serious problem for effective therapy of mycobacteriosis caused by nontuberculosis mycobacteria. Furthermore, animal tissues contain biogenic polyamines, which can increase tolerance of microorganisms to stresses, including antibiotics, and modulate cell motility. Therefore, studying mutual effects of biogenic polyamines and antibiotics on the expansion of mycobacteria is important for medicine. Mycobacterial strains, including the parent Mycolicibacterium smegmatis mc2 155 and strains containing single (ΔrelMsm) or double (ΔrelMsmΔrelZ) deletions, were used as the objects of this study. The content of glycopeptidolipids was determined using thin layer chromatography. Sliding motility was assessed by measuring the area of the sliding colony. The effectiveness of antibiotics was measured by comparison of the areas of sliding colonies in the presence of comparable concentrations of antibiotics. The polyamines spermidine and spermine had different effects on the sliding of mycobacteria through an increase or decrease in the colony areas. At the same time, polyamines had neither bactericidal nor bacteriostatic effects. The polyamines contained in the medium decreased the bactericidal effects of the antibiotics streptomycin or isoniazid, but enhanced the effects of DMNP, a synthetic analogue of the natural antibiotic erogorgiaene. Rifampicin was the most effective of all antibiotics investigated here. Moreover, we found that glycopeptidolipids are, apparently, not the only regulators of mycobacterial sliding.

ABSTRACT

OBJECTIVE: To optimize postoperative rehabilitation by applying low-intensity laser irradiation (LILI) with different wavelengths in the early postoperative period to prevent inflammatory complications. MATERIAL AND METHODS: After radiological examination and ultrasound diagnostics of the periodontal tissue vessels, surgical methods of orthognathic treatment were performed, after completion of which a course of LILI was carried out. The VEGF its receptors (sVEGF-R1; sVEGF-R2) content was measured by enzyme immunoassay using standard reagent kits. The laser therapy using 635 nm laser light was applied directly to the vestibular and oral surfaces of the gingival tissues and in the operation area, changeable by the scanning method, for 1.5 minutes (5 W power); the pulsed infrared laser therapy (PILT) with the 904 nm wavelength (light pulse duration 100 s, power 15 W, 1500 Hz) applied epicutaneously to the operation projective zones (four control points of the upper and lower jaw) through the skin of the cheek, in stable method (1.5 minutes) with a time range between red and infrared wavelength LILI not exceeding 100 s (1.5 minutes). RESULTS: Vascular and endothelial dysfunction after laser irradiation with different wavelengths is better controlled by increasing the microcapillary blood flow (66.7% gain; p<0.05) in arteriolar and 70.3% in venular sections of capillaries (p<0.01), which is associated by vasodilatation: diameter increased by 26.9% compared to that under the influence of red laser radiation (by 13.0%) and infrared laser radiation (by 7.2%); p<0.01). CONCLUSIONS: Early laser therapy using the low-intensity laser irradiation with different wavelengths improves vasoactive processes of hemoregulation in dental tissues associated with the elimination of vasospasm caused by operative stress, activation of arteriolodilatory effects, contributes to the prevention of development of inflammatory complications.

ABSTRACT

Oxidative stress caused by an overproduction of reactive oxygen species (ROS) is the key factor in developing a variety of pathological conditions. Recently various nanomaterials have attracted growing interest as nanoantioxidants with ROS-regulating ability. Here, for the first time, we report on high antioxidant behavior (enzyme-like activity) of GdYVO4:Eu3+nanoparticles (GdYVO NPs) revealed by spectroscopic methods both in cell-free and biological milieu using various ROS sensors. It was revealed that GdYVO NPs (d= 2 nm) effectively scavenge hydroxyl radicals·OH,superoxide anionsO2·-,hydrogen peroxideH2O2,peroxyl radicalsROO·,and remarkably reduce the lipopolysaccharide-induced ROS generation in rat leukocytes. The antioxidant activity of GdYVO NPs is ascribed to high amount of V4+and V3+ions in the structure of the NPs and the reversible switchingV3+↔V4+andV4+↔V5+vanadium oxidation states.

ABSTRACT

OBJECTIVE: To study the changes of evoked EEG oscillations in response to erotic visual stimuli to identify specific patterns in subjects with different variants of pedophilia. MATERIAL AND METHODS: Forty-eight right-handed male offenders who have committed sexual acts against children participated in the study. Seventeen subjects without paraphilic disorders were included in group 1, 17 subjects with heterosexual pedophilia were included in group 2, and 13 subjects with homosexual pedophilia were included in group 3. The stimuli were colored pictures of neutral, heterosexual normative, hetero- and homosexual pedophilic content shortly demonstrated with backward masking. Spectral power of delta, theta, alpha, and beta evoked oscillations (EO) was analyzed. Further, the number of significant changes of EO power in response to erotic stimuli relative to neutral ones was calculated, and topography changes of EO power in response to specific erotic pictures compared to neutral ones were analyzed for each group. RESULTS: The diminished number of significant reactions to specific stimuli was found in the pedophilia groups (18 and 4 in groups 2 and 3, respectively) in comparison with the group without paraphilia (25). The specific changes of EO power in response to specific erotic visual pictures were found for each group. Increased power of delta, theta and alpha EO in the 104-164 ms time window, and decreased power of alpha EO in the 344-920 ms time window were found in group 1. In group 2, specific stimulation elicited decline of delta and theta EO power, possibly related to impaired motivational and emotional aspects of erotic stimuli perception. The weak changes were found in group 3 - increased power of beta EO in the frontal area. CONCLUSION: The obtained frequency and spatial patterns of EO changes in response to erotic visual pictures are related to specificity of hetero- and homosexual pedophilia.

ABSTRACT

OBJECTIVE: Amplitude and latency of evoked potential P300 are considered as major correlates of cognitive functions. At the same time the changes in the theta-band event-related oscillations that appears at the same time window as P300 wave are less studied. The purpose of this study was to provide the complex assessment and comparison of neurophysiological indices of cognitive processes in healthy people and patients with personality disorder, schizotypal disorder and schizophrenia with different levels of cognitive dysfunction. MATERIAL AND METHODS: The total number of participants was 124, including 44 healthy volunteers, 40 schizophrenic patients, 22 patients with personality disorder, 18 patients with schizotypal disorder. Patients were examined in standard two-stimulus oddball paradigm. The P300 amplitude and latency, and evoked theta-band power and coherence were identified for target and non-target stimuli. RESULTS AND CONCLUSION: The decrease in P300 amplitude, prolongation of P300 latency, and reduction in the evoked theta-band power and coherence were revealed in all groups of patients compared to healthy subjects. These changes were spatially generalized and the most pronounced in schizophrenic patients. In patients with personality and schizotypal disorders, the changes had a local character, and didn't differ significantly when the groups were compared. The results show the gradual cognitive decline as follows: healthy patients, patients with schizotypal disorder, patients with personality disorder, and patients with schizophrenia. Such cognitive decline might be in line with the reduction of critical and prognostic abilities.

ABSTRACT

AIM: To consider the patterns of stimuli analysis and task execution in conditions of voluntary and involuntary attention in people with schizotypal disorder from the psychopathological and psychological points of view. MATERIAL AND METHODS: A total of 40 subjects were examined including 16 patients diagnosed with schizotypal disorder, 13 with personality disorders and 11 without psychiatric pathology (controls). In the first part of the study, subjects looked upon various static images without any instructions, followed by a test with antisaccade task in vertical and horizontal directions; the last task duplicated the first, but the subjects were asked to remember the presented images as best as possible. RESULTS: In the group of patients with schizotypal disorder, an increase in average and maximum fixation variance, its greater variation between images, and a decrease in the scanpath length were found. These differences were associated with the intentional focus of attention and several indicators of the accomplish efficiency of the antisaccade task. CONCLUSION: The changes in fixative eye movements observed in individuals with schizotypal disorder indicate violations of information selectivity and construction of an image of future action. Assumptions about inadequacy of their perceptual strategy to the simultaneous one and the primacy of the sense-bearing (motivational) violations in the programming of saccadic activity were made.

ABSTRACT

We report on spectral range stabilization in a self-sweeping laser by adding a narrowband fiber Bragg grating (FBG) to the output mirror in the Michelson configuration. The effects of FBG reflectivity and optical path difference in the Michelson interferometer on the laser spectral dynamics are investigated. Optimization of the interferometer allows us to demonstrate broadband (over 16 nm) self-sweeping operation and reduction of the start and stop wavelength fluctuations by two orders and one order of magnitude (∼100 and 15 times) for start and stop bounds, respectively (down to several picometers). The proposed approaches significantly improve quality of the spectral dynamics and facilitate application of the self-sweeping lasers.

ABSTRACT

The aryl hydrocarbon receptor (AHR) is a ligand-dependent transcription factor involved in the metabolism of physiological substances and xenobiotics, representing an interesting target in both toxicology and pharmacology. In this study, we investigated the ligand-dependent conjunction of nuclear import of the human AHR in living cells and target gene induction. Our findings strengthen the theory that the AHR triggers a precisely defined and rapid reaction upon binding to endogenous ligands, while the xenobiotic ß-naphthoflavone only induces rather unspecific and slow effects. To better illuminate the ligand-mediated responses of the human AHR, we applied site-directed mutagenesis and identified histidine 291 as key residue for AHR functionality, essential for both nuclear import and target gene induction. Contrary, replacing histidine at position 291 by alanine did not affect nucleo-cytoplasmic shuttling, showing that permanent endogenous import and ligand-induced import of the AHR into the nucleus are two independent and differently regulated processes. Combining these observations with our structural investigations using a homology model of the AHR-PAS B domain, we suggest a dual role of histidine 291: (1) a major role for shaping the ligand binding site including direct interactions with ligands and, (2) an essential role for the conformational dynamics of a PAS B loop, which most likely influences the association of the AHR with the AHR nuclear translocator through interference with their protein-protein interface.

CLAVATA (CLV) system including CLV1-like kinase and CLE-peptides is the part of the AON (autoregulation of nodulation) that controls nodule number in legume plants. Moreover, CLV system plays a key role in meristems, where it regulates the expression of WOX genes in organizing centers. Recently, we found that WOX5 homolog in pea is also expressed in nodules and in tumors induced by Agrobacterium tumefaciens. Based on this, we hypothesized that both nodules and agrobacterial tumors may be regulated by and may trigger the same components of AON, including the same WOX and CLV genes. Here, we found that pea plants with agrobacterial tumors induced prior to rhizobial inoculation had reduced the number of nodules. This effect was absent in pea sym29 mutant defective in CLV1-like kinase, the key component of AON. That suggests that agrobacterial tumors may produce a signal activating CLV1-like kinase and thereby decrease the nodule number. Since CLE peptides are known to act upstream of CLV1-like kinase, expression analysis of CLE genes has been performed both in developing nodules and tumors. Overall, 45 CLE genes were identified, and among them nine nodulation-induced CLEs were found in pea. In agrobacterial tumors, no expression of nodule-specific CLE genes the homologues of which inhibit nodulation in other legumes was observed. However, increased expression of two other nodulation-induced CLE genes was observed in agrobacterial tumors, suggesting that CLE genes are expressed in tumors that may still contribute to autoregulatory processes suppressing nodulation.

ABSTRACT

Therapeutic monoclonal antibodies and recombinant proteins including cytokines are commonly used in the treatment of cancer and inflammatory diseases. In most cases, these protein-based drugs exhibit a high therapeutic efficacy, which is unfortunately frequently associated with a variety of side effects. We have investigated the in vitro and in vivo immunogenicity of recombinant antitumor protein lactaptin (RL2). Based on the qRT-PCR analysis, we have shown that, in MDA-MB-231 human breast adenocarcinoma cells, RL2 suppresses the NF-kB signaling cascade that regulates the reactions of innate immunity. RL2 inhibits the expression of the CXCL1 protein and apoptosis inhibitor A20 and enhances expression of IkB, NF-kB repressor. The ELISA method has been used to evaluate the antibody titer in the blood of mice, which received single and triple intravenous or intraperitoneal injections of RL2. The multiplex immunoassay of 23 cytokines in the mice blood has shown that the RL2 injections lead to a slight increase in the levels of systemic pro-inflammatory cytokine interleukin-5 (IL-5) and keratinocyte chemoattractant (KC), a homologue of human macrophage inflammatory protein-1 (MIP-1). These observations indicate the low immunogenicity of the recombinant lactaptin analog, which can be considered to be a potential molecular drug candidate for further clinical development.

ABSTRACT

We proposed and demonstrated an all-fiber scheme for optical spectrum measurement based on stimulated Brillouin scattering and frequency self-sweeping laser without external driver and frequency tunable elements. The resolution and measuring range of proposed analyzer is measured to be 23 MHz and 5 THz respectively. The ways for improvement of the device characteristics are discussed.

ABSTRACT

AIM: The study aimed at analyzing the spatial patterns of evoked event-related oscillations in patients with paranoid schizophrenia and their relationship with clinical symptoms of disease. MATERIAL AND METHODS: Evoked delta, theta, alpha, beta, and gamma rhythms in response to target auditory stimulus in an oddball paradigm were studied in 21 schizophrenic patients and 22 healthy subjects. The independent spatial patterns were revealed within each of the frequency range using the principal component analysis. Each spatial pattern was characterized by the specificity of intra- and inter-hemispheric relations. RESULTS AND CONCLUSION: The schizophrenic patients were characterized by a decrease in the evoked activity in the theta range with the most pronounced changes in the frontal-central areas of the right hemisphere and parietal-occipital areas bilaterally. Associations of the evoked rhythms with PANSS positive and negative symptoms were identified. The study demonstrated the high functional significance of evoked EEG rhythms changes for neurophysiological characteristics of patients with paranoid schizophrenia.
