ABSTRACT

INTRODUCTION: The 12-lead electrocardiogram (ECG-12) is a useful tool in the assessment of athletes. The prevalence of normal electrocardiographic findings in elite Brazilian football players (EBFP) of different ethnicities is not well described. PURPOSE: To compare the prevalence of normal ECG-12 findings in Caucasian, Mixed-race (MR), and Afro-Brazilian (AB) EBFP. METHODS: A multicenter cross-sectional descriptive study was conducted with exams performed on EBFP as a pre-participation assessment from February 18, 2002, through October 20, 2022. RESULTS: 4,363 EBFP from 51 professional clubs (from all five regions of Brazil; 15 states, and 21 cities), aged 15-35 years (median: 19 years; IQR: 16-23) were evaluated. 2,027 were Caucasians, 1,324 were MRs, and 1,012 were ABs. In general, we recorded 4,155 ECG-12 with normal variations (95.2%). Positive Sokolow-Lyon criteria for left ventricular hypertrophy was more prevalent in ABs (41.8%) than in Caucasians (33.4%), or MRs (32.7%). The prevalence of early repolarization in ABs (41.5%) was higher than in Caucasians (34.6%), or MRs (35.5%). The PR interval was longer in ABs (157.1 ms) than in Caucasians (151.4 ms), or MRs (153.3 ms), and the first-degree atrioventricular block was more frequent in ABs compared to MRs (4.2% vs. 2.3%, respectively). On the other hand, Caucasians had a higher prevalence of incomplete right bundle branch block (14.2%) than MRs (11.2%), or ABs (9.7%). In our sample, only three EBFP showed second-degree atrioventricular block Mobitz type I (0.07%; one Caucasian and two AB athletes). CONCLUSIONS: This is the first large study to compare the prevalence of ECG-12 findings in EBFP from different ethnic groups. Those of AB origin have a significantly higher prevalence of ECG-12 findings compatible with the athlete's heart than Caucasians and MRs.

BACKGROUND: Oral cancer represents the sixth most common cancer in the world and is associated with 40-50% survival at 5 years. Within oral malignancies, oral squamous cell carcinoma (OSCC) is commonly preceded by potentially malignant lesions, which, according to histopathological criteria, are referred to as oral dysplasia and their diagnosis are associated with higher rates of malignant transformation towards cancer. We recently reported that aberrant activation of the Wnt/ß­catenin pathway is due to overexpression of Wnt ligands in oral dysplasia. However, the expression of other regulators of this pathway, namely components of the ß-catenin destruction complex has not been explored in oral dysplasia. MATERIAL AND METHODS: Using immunohistochemical analyses, we evaluated nuclear expression of ß­catenin and its association with Wnt3a and Wnt5a. Likewise, components of the ß-catenin destruction complex, including Adenomatous Polyposis Coli (APC), Axin and Glycogen Synthase Kinase 3 beta (GSK-3ß) were also evaluated in oral dysplasia and OSCC biopsies. RESULTS: We found that moderate and severe dysplasia samples, which harbored increased expression of nuclear ß­catenin, depicted augmented cytoplasmic expression of GSK­3ß, Axin and APC, in comparison with OSCC samples. Also, GSK-3ß was found nuclear in mild dysplasia and OSCC samples, when compared with other study samples. CONCLUSIONS: Cytoplasmic levels of components of the ß-catenin destruction complex are increased in oral dysplasia and might be responsible of augmented nuclear ß­catenin.

ABSTRACT This investigation aims to evaluate the antibacterial activity of nanostructured hydroxyapatite based materials doped with silver and fluorine, to be used as a biomaterial with antibacterial activity. Four different formulations were prepared by combustion method: hydroxyapatite, hydroxyapatite-fluorine, hydroxyapatite-silver-fluorine and hydroxyapatite-silver, with 2% of the doping agents. X-ray diffraction technique was used to determine the mineralogy, identifying the presence of Ca5(PO4)3OH, Ca2P2O7, Ag3PO4, AgCa10(PO4)7 Ca5(PO4)3F and CaF2 phases for the studied samples. Scanning electron microscopy was used to study the morphological structure and it showed homogeneous crystallization of the hydroxyapatite and the inclusion of dopant agents. The antibacterial activity was determined using a modified inhibition test zone to observe if the bacteria (E. faecalis) was susceptible to the antimicrobial agent by the appearance of the zone of inhibition on the agar plate. Both the hydroxyapatite-silver and the hydroxyapatite-silver-fluorine materials generated an inhibition zone. It was possible to determine the minimum inhibitory concentration needed to kill most viable organisms after 48 hours of incubation using the broth microdilution method, resulting in 75 µg/ml and 200 µg/ml for the hydroxyapatite-silver and the hydroxyapatite-silver-fluorine formulation, respectively. These materials could be used for the development of new biomaterials that can be used in dental applications.


RESUMEN El objetivo de esta investigación es analizar la actividad antibacteriana de materiales nanoestructurados a base de hidroxiapatita con iones de flúor y plata que le confieran características particulares para que pueda ser utilizado como un biomaterial con actividad antimicrobiana. Se realizaron cuatro formulaciones distintas: hidroxiapatita, hidroxiapatita-flúor, hidroxiapatita-plata-flúor e hidroxiapatita-plata con un 2% de los agentes dopantes. La síntesis del material se realizó a través del método de combustión. La caracterización mineral se realizó a través de difracción de rayos X identificando las siguientes fases en las diversas formulaciones: Ca5(PO4)3OH, Ca2P2O7, Ag3PO4, AgCa10(PO4)7 Ca5(PO4)3F and CaF2. La estructura morfológica se analizó a través de microscopía electrónica de barrido que muestra la formación de estructuras compactas, presencia de cristales y la incrustación de flúor y plata. Se analizó la actividad antimicrobiana utilizando una prueba modificada para la observación del halo de inhibición, encontrándose solamente que los materiales que contenían plata-flúor y plata generaron dicho halo de inhibición. Por otra parte, usando la prueba de microdilución en pozo se encontró que la concentración mínima inhibitoria para el material de HA-Ag fue de 75 μg/ml y para el material de HA-Ag-F fue de 200 μg/ml, después de 48 horas de incubación utilizando E. faecalis.

ABSTRACT

Plastic particle occurrence in the digestive tracts of fishes from a tropical estuarine system in the Gulf of California was investigated. A total of 1095 fish were analysed, representing 15 species. In total 1384 particles of plastic debris were recovered from the gastrointestinal tracts of 552 specimens belonging to 13 species, and all consisted of threads, the majority of which were small microplastics (0.23 to 1.89), followed by large microplastics (2.07 to 4.49), and few mesoplastics (5.4 to 19.86). Plastic particles were identified using ATR-FTIR spectroscopy. The mean frequency of occurrence of plastics in the gastrointestinal tracts of fishes from this system was 50.5%, which is higher than frequencies reported in similar systems in other areas. The polymers identified by ATR-FTIR were polyamide (51.2%), polyethylene (36.6%), polypropylene (7.3%), and polyacrylic (4.9%). These results show the first evidence of plastic contamination for estuarine biota in the Gulf of California.

AIM: To determine the methylation pattern of TLR2 gene promoter and its association with the transcriptional regulation of periapical inflammatory and angiogenic responses in symptomatic and asymptomatic forms of apical periodontitis. METHODOLOGY: In this cross-sectional study, apical lesions were obtained from volunteers with asymptomatic apical periodontitis (AAP) (n = 17) and symptomatic apical periodontitis (SAP) (n = 17) scheduled for tooth extraction, and both total RNA and DNA were extracted. DNA was bisulfite-treated, a region of CpG island within the TLR2 gene was amplified by qPCR and the products were sequenced. Additionally, the mRNA expression of TLR2, TLR4, IL-6, IL-12, TNFalpha, IL-23, IL-10, TGFbeta, VEGFA and CDH5 was analysed by qPCR. The data were analysed with chi-square tests, Mann-Whitney or unpaired t-tests, and Spearman´s correlation; variable adjustments were performed using multiple linear regression (P < 0.05). RESULTS: TLR2 depicted a hypomethylated DNA profile at the CpG island in SAP when compared with AAP, along with upregulated expression of TLR2, with pro-inflammatory cytokines IL-6 and IL-23, and the angiogenesis marker CDH5 (P < 0.05). TLR2 methylation percentage negatively correlated with mRNA levels of IL-23 and CDH5 in apical periodontitis. Lower methylation frequencies of single CpG dinucleotides -8 and -10 localized in close proximity to nuclear factor κB (NFκB) binding within the TLR2 promoter were identified in SAP versus AAP (P < 0.05). Finally, unmethylated -10 and -8 single sites demonstrated up-regulation of IL-23, IL-10 and CDH5 transcripts compared to their methylated counterparts (P < 0.05). CONCLUSIONS: TLR2 gene promoter hypomethylation was linked to transcriptional activity of pro-inflammatory cytokines and angiogenic markers in exacerbated periapical inflammation. Moreover, unmethylated single sites in close proximity to NFκB binding were involved in active transcription of IL-23, IL-10 and CDH5.

RESUMEN El embarazo cornual es un tipo de embarazo ectópico poco frecuente, caracterizado por la implantación del saco gestacional en los cuernos uterinos, cuyo diagnóstico es desafiante, ya que realizado de forma precoz disminuye considerablemente la morbimortalidad materna. En este trabajo se reportan dos casos de un embarazo cornual, ambos diagnosticados a través de una ecografía transvaginal: uno en una paciente asintomática, y el otro en una paciente cuyo síntoma principal fue metrorragia, ambos tratados exitosamente de forma quirúrgica. En la revisión describimos la incidencia, los métodos disponibles para su diagnóstico y distintas opciones de manejo del embarazo ectópico cornual de acuerdo al tiempo de evolución, además de su relación con la ley 21.030 de interrupción del embarazo en tres causales, promulgada el 2017.


ABSTRACT Cornual pregnancy is a non-frequent form of ectopic pregnancy, where the implantation of the gestational sac occurs in the uterine horns; its diagnosis is challenging, and an early diagnosis decreases the morbimortality of the mother. In this paper we report two cases of cornual pregnancy, both diagnosed with ultrasound, one of the cases is in an asymptomatic patient; the principal symptom in the second case was abnormal uterine bleeding, and both being successfully managed with surgery. In the literature review we describe the incidence, available diagnosis methods and different options for the treatment of cornual pregnancy accord to the evolution time; also its relationship with the law 21.030 of pregnancy interrumption on three grounds, published in 2017.

ABSTRACT

BACKGROUND: Exacerbated proliferation of cancer cells in nascent tumors leads to the genesis of a hypoxic microenvironment, which is associated with poor patient prognosis, because these stress conditions enhance migratory, invasive and metastatic capacities of tumor cells. These changes are associated with the induction of the hypoxia-inducible factors (HIFs, mainly HIF1α) and increased expression of target genes, including Caveolin-1 (CAV1). Results from our group have shown that CAV1 expression in metastatic cancer cells promotes cell migration/invasion in vitro and metastasis in vivo in a manner dependent on tyrosine-14 phosphorylation by src family kinases. Here, we evaluated whether hypoxia-induced expression of CAV1 was required for hypoxia-dependent migration and invasion in cancer cells. METHODS: B16-F10 murine melanoma and HT29(US) colon adenocarcinoma cells were exposed to hypoxia (1% O2). CAV1 expression was evaluated by western blotting. Endogenous CAV1 and HIF1α were knocked-down using different shRNA constructs. Cell migration and invasion were evaluated in Boyden Chamber and Matrigel assays, respectively. RESULTS: We observed that hypoxia increased CAV1 protein levels in a HIF1 α- dependent manner, in B16-F10 and HT29(US) cells. Importantly, hypoxia-dependent migration of both tumor cell lines was blocked upon CAV1 knock-down. Likewise, pharmacological inhibition of HIF prevented hypoxia-induced migration and invasion in B16-F10 cells. Finally, hypoxia-induced migration was also blocked by the src-family kinase inhibitor 4-amino-5-(4-chloro-phenyl)-7-(t-butyl) pyrazolo3,4-dpyrimidine (PP2), an inhibitor of CAV1 phosphorylation. CONCLUSION: Hypoxia induced migration and invasion of metastatic cancer cells require HIF1α-dependent induction of CAV1 expression and src family kinase activation.

ABSTRACT

In the Americas, 8 million people are infected with Chagas disease, and an additional 90 million people are at risk for infection. Little is known about the role bats play in the sylvatic transmission cycle of Trypanosoma cruzi, the parasite causing Chagas disease. Here, we captured bats in the villages of Palmiche, Pachacutec, Nuevo San Martin, and Mayuriaga located in the Datem del Marañon Province in Loreto, Peru. Venous blood samples were collected by cardiac puncture or from the upper extremities, and trypanosomatids were identified by microscopy and molecularly. We collected blood samples from 121 bats on filter paper for molecular studies and 111 slides for microscopic examination of thin and thick blood smears from 16 different bat species. The prevalence of trypanosomatids in all bats species was 34.7% (42/121) and the prevalence of T. cruzi was 4.1% (5/121). In hematophagous bat species, the prevalence of trypanosomatids and T. cruzi was 36.9% (27/73) and 2.7% (2/73), respectively. In non-hematophagous bats, the prevalences of trypanosomatids and T. cruzi were 31.2% (15/48) and 6.2% (3/48), respectively. Also, we confirm the presence of T. cruzi in salivary glands of hematophagous bats Diaemus youngi. These results suggest a sylvatic cycle of trypanosomatid transmission in which bats may harbor infectious T. cruzi parasites that could be transmitted to humans via hematophagous bat bites or salivary contamination by non-hematophagous bats of vegetables consumed by humans.

Resumen Los manglares son ecosistemas de importante productividad primaria, donde se establece un flujo de energía (nutrientes) con zonas adyacentes y su ambiente acuático, impulsado principalmente por los procesos de degradación. El objetivo del estudio fue estimar el coeficiente de degradación diario (k) de hoja de mangle por medio de bolsas de degradación, en relación con los factores físico químicos del suelo y el aporte de nutrientes (fósforo y nitrógeno) en sedimento del manglar de la Laguna Mecoacán, Golfo de México. El coeficiente de degradación se estimó por medio de bolsas de degradación de hojarasca en seis sitios de monitoreo mensual. Se identificó una rápida degradación durante el primer mes de hasta 51 % en Avicennia germinans (L.) Stearn asociado a procesos de lixiviación por condiciones de inundación. La degradación (k) de Rhizophora mangle L. (k= 0.0052 ± 0.0002) (F= 12.2 p<0.05 n= 216) y Laguncularia racemosa (L.) Gaertn (k= 0.005 ± 0.0003) (F= 3.7 p= 0.02 n= 108) difieren significativamente de A. germinans (k= 0.009 ± 0.0003) (F= 1.2 p= 0.02 n= 216). En relación al T50 de R. mangle y L. racemosa presentaron mayor tiempo de degradación (133 y 138 días respectivamente) comparado con A. germinans (74 días). Se registró una correlación significativa entre la materia orgánica y la humedad del suelo con la constante de descomposición de A. germinans (r= 0.65 p< 0.05 y r= 0.55 p< 0.05 respectivamente). El más alto contenido de nitrógeno total se dio en Pajaral (2 683 mg.Kg) y presentó alta correlación con el contenido de materia orgánica (r= 0.9 p= 0.03); en relación al fósforo total, el nivel más alto se presentó en Boca (2 031 mg.Kg) correlacionado de forma negativa con el pH (r= -0.61 p< 0.05). En conclusión, las diferencias en la velocidad de degradación de las hojas de mangle dependen de la especie (composición foliar), tiempo de exposición o inmersión en agua (patrón de inundación) y heterogeneidad del sedimento (i.e., textura, pH, contenido de humedad y densidad aparente).


Abstract Mangroves are ecosystems with a high primary productivity that is mainly driven mainly by degradation processes. Energy (nutrients) flows from mangroves toward adjacent zones and the surrounding aquatic environment. The objective of the present study was to estimate the daily degradation coefficient (k) of mangrove leaves in relation to physical-chemical soil factors and in situ nutrient supply (phosphorus and nitrogen) in Mecoacán Lagoon, Gulf of Mexico. Leaf litter degradation bags were placed at six monthly monitoring sites to evaluate degradation and to calculate the corresponding degradation coefficients. A rapid degradation of up to 51 % was observed for Avicennia germinans (L.) Stearn during the first month in association with leaching resulting from flood conditions. The degradation of Rhizophora mangle (L.) (k= 0.0052±0.0002) (F= 12.2 p< 0.05 n= 216) and Laguncularia racemosa (L.) Gaertn (k= 0.005±0.0003) (F= 3.7 p= 0.2 n= 108), differed significantly from that of A. germinans (k= 0.009 ± 0.0003) (F= 1.2 p= 0.2 n= 216) did not present significant differences. To reach T50 degradation, R. mangle and L. racemosa required more time (133 and 138 days, respectively) than A. germinans (74 days). Organic matter and soil humidity were significantly correlated with the decay constant of A. germinans (r= 0.65 p< 0.05 and r= 0.55 p< 0.05, respectively). Total nitrogen content was highest in the Pajaral site (2 683 mg.kg) and was also highly correlated with organic matter content (r= 0.9 p= 0.003). Total phosphorus content was highest in the Boca site (2 031 mg.kg) and was also negatively correlated with pH (r= -0.61 p= 0.004). In conclusion, differences in the rate of mangrove leaf degradation depend on the involved species (leaf composition), time of exposure or immersion in water (flooding patterns) and sediment heterogeneity (i.e., texture, pH, humidity content and bulk density). Antioxidants have been widely used during in vitro production to decrease the negative effect of reactive oxygen species. It was reported that the complex resveratrol-methyl ß-cyclodextrin (RV-CD) improves resveratrol's stability and bioavailability and increases its antioxidant activity. This study evaluates the effect of RV-CD during in vitro oocyte maturation (IVM) or in vitro embryo culture (IVC) on developmental competence and quantitative changes in gene expression of developmental important genes. In experiment 1, RV-CD was added to IVM media and maturation level, embryo development and oocytes, cumulus cells, and blastocysts gene expression by RT-qPCR were examined. In experiment 2, presumptive zygotes were cultured in SOF supplemented with RV-CD and embryo development and blastocysts gene expression by RT-qPCR were studied. A group without RV-CD (control- ) and a group with cyclodextrin (control+ ) were included. No differences were found in cleavage rate or blastocyst yield between groups. However, the expression of LIPE was higher in blastocysts derived from oocytes treated with resveratrol compared with control groups (p < .05). Blastocysts produced by IVC with resveratrol showed that RV-CD could modify the expression of genes related to lipid metabolism (CYP51A1, PNPLA2 and MTORC1) compared with control groups (p < .05). RV-CD in the IVM and IVC media could reduce accumulated fat by increasing lipolysis and suppressing lipogenesis of blastocysts.

ABSTRACT

Wounds in the oral mucosa heal faster and more efficiently than those in the skin, although the mechanisms underlying these differences are not completely clear. In the last 10 years, a group of salivary peptides, the histatins, has gained attention on behalf of their ability to improve several phases of the wound-healing process. In addition to their roles as anti-microbial agents and in enamel maintenance, histatins elicit other biological effects, namely by promoting the migration of different cell types contained in the oral mucosa and in non-oral tissues. Histatins, and specifically histatin-1, promote cell adhesion and migration in oral keratinocytes, gingival and dermal fibroblasts, non-oral epithelial cells, and endothelial cells. This is particularly relevant, as histatin-1 promotes the re-epithelialization phase and the angiogenic responses by increasing epithelial and endothelial cell migration. Although the molecular mechanisms associated with histatin-dependent cell migration remain poorly understood, recent studies have pointed to the control of signaling endosomes and the balance of small GTPases. This review aimed to update the literature on the effects of histatins in cell migration, with a focus on wound healing. We will also discuss the consequences that this increasing field will have in disease and therapy design.

Resumen Los manglares se distribuyen a lo largo de gradientes topográficos en zonas costeras, donde las variaciones en salinidad y períodos de inundación influyen en la composición y productividad primaria. En este estudio se evaluaron los atributos estructurales y producción de hojarasca de los manglares de la Laguna Mecoacán, Golfo de México. En seis sitios de monitoreo se determinó durante la segunda semana de julio 2014, la composición florística, el diámetro a la altura del pecho (DAP), altura, densidad del mangle adulto, juveniles, reclutas y plántulas. Además, se calcularon el área basal, índice de valor de importancia (IVI) y cobertura. Se tomaron muestras mensuales (noviembre 2014 a octubre 2015) de producción de hojarasca de las especies de mangle mediante trampas de captura, mientras que los parámetros físico-químicos de agua superficial, intersticial y subterránea fueron determinados mediante piezómetros. Los sitios de estudio en la Laguna Mecoacan se clasificaron en dos tipos fisonómicos: Borde (Boca, Aspoquero, Arrastradero y Pajaral) y cuenca (Cerros y Mojarrero). La densidad promedio del manglar fue de 2 870 árboles. ha-1. El área basal más alta se encontró en Pajaral (41.7 m-2. ha). Avicennia germinans (L.) Stearn presentó los valores más altos de IVI en todos los sitios. Se registró una cobertura media de 17.5 m2. Las especies mostraron zonación con los niveles de salinidad de agua intersticial; A. germinans con mayor densidad en áreas de alta salinidad (hipersalinas), mientras Laguncularia racemosa (L.) Gaertn y Rhizophora mangle L. en áreas de menor salinidad. La producción de hojarasca fue de 5.15 ton. ha-1. año-1, mostró una correlación positiva con la precipitación y alta correlación negativa con los niveles de salinidad y ORP intersticial y subterránea. Los resultados indican que la variabilidad en la composición, la estructura y la producción de hojarasca en la Laguna Mecoacán está determinada localmente por la capacidad de adaptación a las condiciones ambientales (precipitación, salinidad, potencial redox) de las especies presentes. Se recomienda establecer los sitios como parcelas permanentes de monitoreo y estimar la biomasa en pie y debajo del suelo para entender mejor la dinámica de nutrientes como una de las principales fuentes de energía in situ y exportado a zonas adyacentes.


Abstract Mangrove forests are distributed along the topographical gradients of coastal areas, where variations in salinity and flooding influence the composition and primary production of these forests. This study evaluated the structural attributes and production of leaf litter of mangrove species in Mecoacán Lagoon. We established six monitoring sites and studied the floristic composition, diameter at breast height (DBH), and density of adult, juvenile and recruited trees, as well as seedling density; additionally, the basal area, importance value index (IVI), and coverage were also calculated. Monthly samples (from November 2014 to October 2015) of the leaf litter production of different mangrove species, were collected using litter traps, and the physicalchemical parameters of surface, interstitial, and subterranean water were determined using piezometers. The study sites were classified into two physiognomic types: fringe (Boca, Aspoquero, Arrastradero, and Pajaral) and basin (Cerros and Mojarrero). The average density of mangrove trees was 2 870 trees. ha-1. The highest basal area was found in Pajaral (41.7 m2. ha-1). Avicennia germinans presented the highest IVI value in all sites. An average coverage of 17.5 m2 was recorded. The species were zoned with respect to interstitial salinity levels. A. germinans had a greater density in areas with high salinity (hypersaline) and Laguncularia racemosa and Rhizophora mangle in areas of lower salinity. The leaf litter production was 5.15 ton. ha-1. year-1 and showed a positive correlation with rainfall and a highly negative correlation with salinity level as well as with interstitial and subterranean oxidation redox potential (ORP). The results indicated that variability in the composition, structure, and production of leaf litter in Laguna Mecoacán is locally determined by the species capacity to adapt to environmental conditions (rainfall, salinity, redox potential). Several recommendations include the establishment of the sites evaluated in this study, as permanent monitoring areas and to estimate the above and belowground biomass, in order to understand the nutrient dynamics, as mangroves provide sources of energy both in situ and that may be exported to neighboring areas.

ABSTRACT

Enterovirus-71 (EV71) was first isolated in California, United States in 1969, belongs to the genus Enterovirus, family Picornaviridae. Although infection normally causes mild, often undiagnosed illness, it can cause central nervous system infections that could turn fatal. Based on VP1 gene analysis, EV71 has been classified into six separate genotypes. Although the molecular epidemiology of EV71 has been well described via studies originating from Asia and Europe, it is mostly unknown in South America. From our study, four EV71 isolates from Peru were characterized using phylogenetic methods to determine their relationship with known reference strains. These four Peruvian EV71 isolates from between 2006 and 2009 were analyzed by RT-PCR using primers capable of amplifying the entire VP1 gene. Reference strains representing all six known genotypes were used to determine any recognizable phylogenetic relationships. In fact, all of our isolates clustered together within the genotype C1 lineage- separate from Asian, European, North American, and Australian strains. We present evidence that EV71 genotype C1 exists in Peru, and this is the first such report documenting EV71 genotype C1 circulating in South America. Gathering additional isolates will help elucidate a more complete global epidemiological picture of EV71 infections.

ABSTRACT

OBJECTIVES: To determine the molecular epidemiology of seven coxsackievirus A16 (CVA16) strains previously reported by this research group. METHODS: Full-length VP1 and VP4 sequences were obtained and phylogenetic analyses were performed. RESULTS: Six strains were classified as genotype C. Moreover, one divergent strain not clustered in any of the three currently reported genotypes was found. CONCLUSION: This is the first report of CVA16 in Peru and provides valuable baseline data about its potential distribution in South America, as well as evidence of a potential divergent genotype that has never before been reported.

BACKGROUND: Identification of the best embryos to transfer is a key element for success in assisted reproduction. In the last decade, several morphological criteria of oocytes and embryos were evaluated with regard to their potential for predicting embryo viability. The introduction of polarization light microscopy systems has allowed the visualization of the meiotic spindle and the different layers of the zona pellucida in human oocytes on the basis of birefringence in a non-destructive way. Conflicting results have been reported regarding the predictive value in ICSI cycles. OBJECTIVE: To assess the predictive ability of meiotic spindle and zona pellucida of human oocytes to implant by polarized microscopy in ICSI cycles. MATERIAL AND METHODS: Prospective and observational clinical study. 903 oocytes from 94 ICSI cycles were analyzed with polarized microscopy. Meiotic spindle visualization and zona pellucida birefringence values by polarized microscopy were correlated with ICSI cycles results. RESULTS: Meiotic spindle visualization and birefringence values of zona pellucida decreased in a direct basis with increasing age. In patients aged over the 35 years, the percentage of a visible spindle and mean zona pellucida birefringence was lower than in younger patients. Fertilization rate were higher in oocytes with visible meiotic spindle (81.3% vs. 64%; p < 0.0001), as well as embryo quality (47.4% vs. 39%; p=0.01). Fertilization rate was higher in oocytes with positive values of birefringence (77.5 % vs. 68.5% p=0.005) with similar embryo quality. Conception cycles showed oocytes with higher mean value of zona birefringence and visible spindle vs. no-conception cycles (p<0.05). CONCLUSIONS: Polarized light microscopy improves oocyte selection, which significantly impacts in the development of embryos with greater implantation potential. The use of polarized light microscopy with sperm selection methods, blastocyst culture and deferred embryo transfers will contribute to transfer fewer embryos without diminishing rates of live birth and single embryo transfer will be more feasible.

ABSTRACT

The aim of this study was to design a robust parameter identification algorithm to characterize the effect of gene deletion on Escherichia coli (E. coli) MG1655. Two genes (pta and poxB) in the competitive pathways were deleted from this microorganism to inhibit pyruvate consumption. This condition deviated the E. coli metabolism toward the Krebs cycle. As a consequence, the biomass, substrate (glucose), lactic, and acetate acids as well as ethanol concentrations were modified. A hybrid model was proposed to consider the effect of gene deletion on the metabolism of E. coli. The model parameters were estimated by the application of a least mean square method based on the instrument variable technique. To evaluate the parametric identifier method, a set of robust exact differentiators, based on the super-twisting algorithm, was implemented. The hybrid model was successfully characterized by the parameters obtained from experimental information of E. coli MG1655. The significant difference between parameters obtained with wild-type strain and the modified (with deleted genes) justifies the application of the parametric identification algorithm. This characterization can be used to optimize the production of different byproducts of commercial interest.

ABSTRACT

Carbon-to-nitrogen ratio (CNR) has shown to be a relevant factor in microorganisms growth and metabolites production. It is usual that this factor compromises the productivity yield of different microorganisms. However, CNR has been rarely modeled and therefore the nature of its specific influence on metabolites production has not been understood clearly. This paper describes a parametric characterization of the CNR effect on the Escherichia coli metabolism. A set of parameters was proposed to introduce a mathematical model that considers the biomass, substrate and several byproducts dynamical behavior under batch regimen and CNR influence. Identification algorithm used to calculate the parameters considers a novel least mean square strategy that formalizes the CNR influence in E. coli metabolism. This scheme produced a step-by-step method that was suitable for obtaining the set of parameters that describes the model. This method was evaluated under two scenarios: (a) using the data from a set of numerical simulations where the model was tested under the presence of artificial noises and (b) the information obtained from a set of experiments under different CNR. In both cases, a leave-one-experiment-out cross-validation study was considered to evaluate the model prediction capabilities. Feasibility of the parametric identification method was proven in both considered scenarios.

El continuo desarrollo del sector salud hace cada vez más evidente la necesidad de evaluación de procesos y actividades de mejora continua, buscando siempre la calidad en la atención en salud para reducir los riesgos de usuarios y personal asistencial. La auditoría como proceso de evaluación permite recopilar información de cumplimiento en temas de normativa para habilitación y/o acreditación nacional en salud, pero los instrumentos para llevarla a cabo resultan ser poco eficientes. Por lo tanto es preciso disponer de una herramienta de auditoría que permita evaluar cumplimento, entregar resultados y de acuerdo a estos establecer planes de mejora, llevando finalmente a la obtención de un proceso más eficiente. En este trabajo se presenta el desarrollo de un prototipo de herramienta de auditoría que se sustenta en las normas vigentes de habilitación y acreditación, el cual por medio del desarrollo de una plantilla en Excel permite calificar estándares de gestión de tecnología, entregar gráficos específicos y generales para a partir de estos realizar en menor tiempo la recopilación de información que permitirá adoptar planes de mejora.


The continued development of the health sector makes increasingly evident the need for evaluation of processes and continuous improvement activities, always looking for quality in health care to reduce the risks of users and caregivers. The audit as evaluation process allows gathering information on issues of compliance rules to enable and/or reaching national health accreditation, but the instruments to implement it turn out to be inefficient. Therefore it is necessary to have an audit tool to assess compliance, and deliver results according to these, establishing improvement plans, ultimately leading to obtaining a more efficient process. In this paper, the development of a prototype audit tool that is based on the existing rules of licensing and accreditation is introduced, which through the development of a template in Excel allows qualifying technology standards management, delivering specific and general graphics which help to gather information in less time which will help to adopt improvement plans.


O desenvolvimento contínuo do setor de saúde torna cada vez mais evidente a necessidade de avaliação de processos e atividades de melhoria contínua, sempre à procura de cuidados de saúde de qualidade para reduzir os riscos de usuários e cuidadores. O processo de auditoria e avaliação permite a coleta de informações sobre questões de regras de conformidade permitem e / ou acreditação nacional de saúde, mas os instrumentos para implementá-lo são ineficientes. Por isso, é necessário ter uma ferramenta de auditoria para avaliar o cumprimento, e entregar resultados de acordo com estes estabelecer planos de melhoria, levando à obtenção de um processo mais eficiente. Neste trabalho, é apresentado o desenvolvimento de uma ferramenta protótipo de auditoria que se baseia nas regras existentes de licenciamento e acreditação, que através do desenvolvimento de um modelo no Excel permite avaliar o gerenciamento de padrões de gestão tecnológica, oferecendo gráficos específicos e gerais para a partir destes fazer em menos tempo a coleta de informações que permitam adotar planos de melhoria.
