ABSTRACT

Rapid and accurate measurements of immune protein markers are essential for diagnosis and treatment in all clinical settings. The recent pandemic has revealed a stark need for developing new tools and assays that could be rapidly used in diverse settings and provide useful information to clinicians. Here, we describe the development and test application of a novel one-step CRP/IP-10 duplex assay for the LightDeck platform capable of delivering reproducible and accurate measurements in under eight minutes. We used the optimized assay to measure CRP and IP-10 levels in human blood and serum samples from healthy, SARS-CoV-2 (COVID-19) positive, and influenza-like illness (ILI) presenting patients. Our results agreed with previously published analyte levels and enabled us to make statistically significant comparisons relevant to multiple clinical parameters. Our duplex assay is a simple and powerful tool for aiding prognostic decision-making in diverse settings.

ABSTRACT

The previously reported complexes [Mo(N(t)Bu)(O)(2,6-Me(2)C(6)H(3)O)(2)(py)] () and [Mo(N(t)Bu)(O)(2,6-(i)Pr(2)C(6)H(3)O)(2)(py)] () have been shown to contain a nucleophilic oxo ligand. The reaction of and tert-butylacetyl chloride gave the novel complex [Mo(N(t)Bu)(2,6-(i)Pr(2)C(6)H(3)O)(2)(O(2)CCH(2)(t)Bu)Cl] () in 63% yield as the product of nucleophilic acyl substitution. Complex undergoes a clean reaction with triflic anhydride to give the novel complex [{Mo(N(t)Bu)(2,6-Me(2)C(6)H(3)O)(2)}(micro-O)(micro-OTf)(2)] () in 75% yield. Complex appears to be formed from nucleophilic attack of the oxo ligand on triflic anhydride, followed by nucleophilic attack of a second oxo moiety on the cationic intermediate. The solid-state structures of and have been determined by X-ray crystallography. These are rare examples of nucleophilic oxo ligands bound to a d(0) transition metal centre.

ABSTRACT

[reaction: see text] A protocol for the enantioselective nitro-Mannich coupling between alkyl, aryl, and heterocyclic p-methoxybenzylimines and trimethylsilylnitropropanate catalyzed by a chiral tBu-BOX Cu(II) catalyst is described. It uses the lowest reported loading of commercially available metal catalyst and chiral ligand, and gives the highest yields and selectivities for a broad substrate range including nonaromatic aldimines. The resultant beta-nitroamines are obtained in 70-94% enantiomeric excess in good yield and can be readily reduced to synthetically useful 1,2-diamines.
