ABSTRACT

Heterostructure engineering and oxygen vacancy engineering are the most promising modification strategies to reinforce the Zn2+ ion storage of vanadium oxides. Herein, a rare mixed-dimensional material (VOx), composed of V2O5 (2D), V3O7 (3D), and V6O13 (3D) heterostructures, rich in oxygen vacancies, was synthesized via thermal decomposition of layered ammonium vanadate. The VOx cathode provides an exceptional discharge capacity (411 mA h g-1 at 0.1 A g-1) and superior cycling stability (the capacity retention remains close to 100% after 800 cycles at 2 A g-1) for aqueous zinc-ion batteries (AZIBs). Ex situ characterizations confirm that the byproduct Zn3V2O7(OH)2·nH2O is generated/decomposed during discharge/charge processes. Furthermore, VOx demonstrates reversible intercalation/deintercalation of H+/Zn2+ ions, enabling efficient energy storage. Remarkably, a reversible crystal-to-amorphous transformation in the V2O5 phase of VOx during charge-discharge was observed. This investigation reveals that mixed-dimensional heterostructured vanadium oxide, with abundant oxygen vacancies, serves as a highly promising electrode material for AZIBs, further advancing the comprehension of the storage mechanism within vanadium-based cathode materials.

ABSTRACT

Hyaluronate lyase (HA lyase) has potential in the industrial processing of hyaluronan. In this study, HylP, an HA lyase from Streptococcus pyogenes phage (SPB) was successfully expressed in Bacillus subtilis. To improve the extracellular enzyme activity of HylP in B. subtilis, signal peptide engineering systematic optimization was carried out, and cultured it from shake flasks and fermenters, followed by purification, characterization, and analysis of degradation products. The results showed that the replacement of the signal peptide increased the extracellular enzyme activity of HylP from 1.0 × 104 U/mL to 1.86 × 104 U/mL in the shake flask assay, and using a 20 L fermenter in a batch fermentation process, the extracellular enzyme activity achieved the level of 1.07 × 105 U/mL. HylP exhibited significant thermal and pH stability in the temperature range of 40 °C and pH range of 4-8, respectively. The enzyme showed optimum activity at 40 °C and pH 6, with significant activity in the presence of Na+, Mg2+, and Co2+ ions. Degradation analysis showed that HylP efficiently degraded hyaluronan as an endonuclease, releasing unsaturated disaccharides. These comprehensive findings underscore the substantial industrial potential of HylP for hyaluronan processing applications, offering valuable insights into enzyme characterization and optimization of expression for potential industrial utilization.

ABSTRACT

You-Gui-Yin (YGY) is a classic prescription for warming up kidney-Yang and filling in kidney essence in traditional Chinese medicine, and has been used to treat osteonecrosis of the femoral head (ONFH) effectively. However, the underlying mechanisms are still unknown. This study is aimed at exploring the possible mechanisms of action of the YGY in the treatment of ONFH based on network pharmacology and molecular docking. TCMSP was used to screen the active components and targets of YGY. The disease targets of ONFH were collected in several public databases. The protein-protein interaction (PPI) Network was constructed using the STRING platform. The Metascape database platform was used for Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analyses. The key active components and core target proteins of YGY in the treatment of ONFH were verified by the molecular docking. 120 active components were obtained from YGY, among which 73 components were hit by the 117 drug-disease intersection targets. Key effective components included quercetin, kaempferol, beta-sitosterol, glycitein, beta-carotene, and so on. Core target proteins included ALB, AKT1, TNF, IL6, TP53, and so on. According to GO and KEGG analyses, there were 1762 biological processes, 94 cellular component, 138 molecular function and 187 signaling pathways involved. we selected the top 20 biological processes (BP), cellular components (CC), molecular functions (MF) and signaling pathways to draw the heat maps, showing that Lipid and atherosclerosis signaling pathway, IL-17 signaling pathway, HIF-1 signaling pathway, relaxin signaling pathway and MAPK signaling pathway and other pathways may play a key role in the treatment of ONFH by YGY. The results of molecular docking showed that key effective components and corresponding core target proteins exhibited the good binding activity. YGY can treat ONFH through multicomponents, multitargets, and multipathways, which provides a reference for the subsequent research, development of targeted drugs and clinical application.

ABSTRACT

To investigate the clinical effects of specific Unified Classification System B (UCS B)-lesser trochanter periprosthetic fractures and determine whether they occur only with non-cemented stems. A retrospective analysis of 28 patients with specific UCS B2 fractures who underwent two surgical treatments, longer stem revision and internal fixation (LSRIF) and open reduction and internal fixation (ORIF), was performed. The patients were assessed at 1, 3, 6, 12, and 24 months and annually thereafter. Fracture healing, complications, Harris Hip Score (HHS), and the Short Form Health Survey questionnaire (SF-36) quality of life score were assessed at each follow-up. At the time of the last follow-up, seven patients had been lost: three were lost to contact, two died, and two were hospitalised elsewhere and unavailable for follow-up. The remaining 21 patients were followed for an average of 49.3 ± 15.4 (range: 24-74.4) months. Their average fracture healing time was 13.5 ± 1.1 (12-15.4) weeks. Complications included three cases (10.71%) of thrombus, one (3.57%) of heart failure, and one (3.57%) of pulmonary infection. There were no revisions due to prosthesis loosening, subsidence, or infection. At the last follow-up, the HHS, SF-36 mental score, and SF-36 physical score were recorded, LSRIF vs. ORIF (82.9 ± 6.6 vs. 74.7 ± 3.9, p = 0.059; 50.9 ± 7.6 vs. 38 ± 1.4, p = 0.012, and 51.7 ± 8.4 vs. 39.7 ± 3.4, p = 0.032, respectively). Specific UCS B2 fractures mostly occur with non-cemented stems. LSRIF with cables is the main treatment, while ORIF is an option for those elderly in poor condition.

ABSTRACT

OBJECTIVE: The isolated second metacarpal base fracture-dislocations in adults are rare, and the traditional main treatments include closed reduction with the plaster or splint and open reduction with internal fixation (ORIF). However, closed reduction with the plaster or splint is not solid enough and ORIF can damage the surrounding important tissues. The purpose of this study was to explore the clinical outcomes of closed reduction and external fixation (CREF) for the treatment of isolated second metacarpal base fracture-dislocations. METHODS: Ten patients who suffered isolated second metacarpal base fracture-dislocations between January 1, 2010, and February 1, 2020, were reviewed. All of the patients were treated by CREF. Radiographs and computed tomography scans were performed regularly after the operation. The grip and pinch strength, visual analog scale (VAS) pain score, American Society for Surgery of the Hand total active movement (TAM) and total active flexion (TAF) scores, Cooney score, Sollerman hand function test (SHFT) score, QuickDASH score, and range of motion of the index finger were recorded at the last follow-up visit to evaluate functional recovery; the injured and healthy hands were compared and we also recorded postoperative complications. The paired samples t-test was used to compare the healthy and injured sides. RESULTS: The patients were followed up for a median of 29.50 ± 4.2 months. There were no significant differences in the grip strength, pinch strength, angle of proximal interphalangeal joint (PIPJ) flexion, or angle of distal interphalangeal joint (DIPJ) flexion between the injured and healthy sides at the final follow-up visit (all p > 0.05). The mean TAM (268.20 ± 4.21) and TAF scores (270.60 ± 4.17) on injured side were significantly lower than those on healthy side (all p < 0.05). The mean Cooney score (93.50 ± 7.47) and SHFT score (78.50 ± 1.08) on injured side were lower, while the mean QuickDASH score (7.05 ± 3.11) on injured side was higher than those on healthy side (all p < 0.05). The mean VAS pain score was 0.50 ± 0.53 on injured side. There were no significant postoperative complications except for traumatic arthritis in one patient without obvious clinical symptoms. CONCLUSION: CREF achieves the satisfactory curative effect, and the patients obtain the good functional recovery without significant postoperative complications. CREF is a safe and effective treatment for isolated second metacarpal base fracture-dislocations with satisfactory clinical outcomes.

ABSTRACT

This study reveals the genetic and biochemical changes underlying the enhanced hyaluronan (HA) biosynthesis in Streptococcus zooepidemicus. After multiple rounds of atmospheric and room temperature plasma (ARTP) mutagenesis combined with novel bovine serum albumin/cetyltrimethylammonium bromide coupled high-throughput screening assay, the HA yield of the mutant was increased by 42.9% and reached 0.813 g L-1 with a molecular weight of 0.54 × 106 Da within 18 h by shaking flask culture. HA production was increased to 4.56 g L-1 by batch culture in 5-L fermenter. Transcriptome sequencing exhibits that distinct mutants have similar genetic changes. Regulation in direction of metabolic flow into the HA biosynthesis, by enhancing genes responsible for the biosynthesis of HA including hasB, glmU and glmM, weaking downstream gene (nagA and nagB) of UDP-GlcNAc and significantly down-regulating transcription of wall-synthesizing genes, resulting in the accumulation of precursors (UDP-GlcA and UDP-GlcNAc) increased by 39.74% and 119.22%, respectively. These associated regulatory genes may provide control point for engineering of the efficient HA-producing cell factory.

ABSTRACT

Spiders of the genus Pholcus were collected for the first time during an expedition to the Lüliang Mountains in Shanxi Province, North China. Phylogenetic analyses of DNA sequence data from COI, H3, wnt, and 28S genes allowed us to group them into nine well-supported clades. We used morphology and four methods of molecular species delimitation, namely Automatic Barcode Gap Discovery (ABGD), the Generalized Mixed Yule Coalescent (GMYC), Bayesian Poisson Tree Processes (bPTP), and Bayesian Phylogenetics and Phylogeography (BPP), to investigate species boundaries. These integrative taxonomic analyses identified the nine clades as nine distinct species, comprising Pholcus luya Peng & Zhang, 2013 and eight other species new to science: Pholcus jiaocheng sp. nov., Pholcus linfen sp. nov., Pholcus lishi sp. nov., Pholcus luliang sp. nov., Pholcus wenshui sp. nov., Pholcus xiangfen sp. nov., Pholcus xuanzhong sp. nov., and Pholcus zhongyang sp. nov. The species occur in geographic proximity and show many morphological similarities. All of them belong to the P. phungiformes species group. The records from the Lüliang Mountains represent the westernmost distribution limit of this species group.

ABSTRACT

It has long been found that hyaluronidases exist in a variety of organisms, playing their roles in various biological processes including infection, envenomation and metabolic regulation through degrading hyaluronan. However, exploiting them as a bioresource for specific applications had not been extensively studied until the latest decades. In recent years, new application scenarios have been developed, which extended the field of application, and emphasized the research value of hyaluronidase. This critical review comprehensively summarizes existing studies on hyaluronidase from different source, particularly in their structures, action patterns, and biological functions in human and mammals. Furthermore, we give in-depth insight into the resource mining and protein engineering process of hyaluronidase, as well as strategies for their high-level production, indicating that mixed strategies should be adopted to obtain well-performing hyaluronidase with efficiency. In addition, advances in application of hyaluronidase were summarized and discussed. Finally, prospects for future researches are proposed, highlighting the importance of further investigation into the characteristics of hyaluronidases, and the necessity of investigating their products for the development of their application value.

ABSTRACT

OBJECTIVE: To explore clinical effectiveness and safety of ultrasound-guided closed reduction and K-wires internal fixation in treating of Kilfoyle Ⅱand Ⅲ medial condylar fracture of humerus in children. METHODS: Clinical data of 32 children with medial condylar fracture of humerus treated with closed reduction and internal fixation with K-wires under the guidance of ultrasound were retrospectively analyzed from January 2014 to August 2019, including 23 males and 9 females, age ranged from 3.2 to 12.8 years old with an average of (8.3±2.1) years old;According to classification of Kilfoyle, 12 patients classified to typeⅡ and 20 patients were type Ⅲ;5 patients combined with elbow dislocation;the time from injury to operation ranged from 1 to 5 days with an average of (3.1±1.3) days. Radiological evaluation of treatment results and complications were observed. At the final follow up, Mayo elbow performance score(MEPS) was used to evaluate elbow function. And humerus-ulna angle on the affect side and healthy side were measured and compared. RESULTS: All patients were followed up from 8 to 26 months with an average of(19.3±5.5) months. All fractures were healed well, the healing time ranged from 4 to 6 weeks with an average of (4.5±0.5) weeks. No infection, vascular and nerve injury, bone nonunion, trochlear necrosis, cubitus varus or valgus deformity were occurred. According to Mayo scoring, all patients were assessed as excellent. There was no significant difference in angle of humerus-ulna between affectedside (9.5±3.6)° and healthy side (9.1±3.5)°, and no difference in MEPS scores between affected side(95.3±2.5) and healthy side(96.3±2.2)(P>0.05). CONCLUSION: For Kilfoyle typeⅡand Ⅲ medial condylar fracture of humerus in children, closed reduction and internal fixation with K-wire under ultrasound guidance is a safe and effective method, and could promote in further.

ABSTRACT

Despite 250 years of taxonomic classification and over 1.2 million species already catalogued, known species diversity is only a small part of true species diversity on Earth, and thus, the known species are only the tip of iceberg. Here, we investigated the genus Pholcus Walckenaer, 1805 of the family Pholcidae C. L. Koch, 1850 in the Changbai Mountains, Northeast China, which provides an excellent case of high species diversity. Previously, only 14 endemic Pholcus spiders, all belonging to the P. phungiformes species group, and two introduced species P. manueli Gertsch, 1937 and P. zichyi Kulczynski, 1901 from the P. crypticolens species group, have been recorded from this area. Our study confirmed 11 new species of the P. phungiformes species group based on morphology and three methods of molecular species delimitation: P. gaizhou Yao & Li, sp. nov., P. guanshui Yao & Li, sp. nov., P. jiguanshan Yao & Li, sp. nov., P. longxigu Yao & Li, sp. nov., P. luoquanbei Yao & Li, sp. nov., P. shenshi Yao & Li, sp. nov., P. tianmenshan Yao & Li, sp. nov., P. wangjiang Yao & Li, sp. nov., P. xingqi Yao & Li, sp. nov., P. yaoshan Yao & Li, sp. nov., and P. yuhuangshan Yao & Li, sp. nov. This study brings the fauna of the P. phungiformes species group from the Changbai Mountains to 25 species, approximately two times more than previously known, which could indicate that species diversity in the area is underestimated for all arthropod fauna.

ABSTRACT

Owing to its outstanding water-retention ability, viscoelasticity, biocompatibility and non-immunogenicity, Hyaluronic acid (HA), a natural linear polymer alternating linked by d-glucuronic acid and N-acetylglucosamine, has been widely employed in cosmetic, medical and clinical applications. With the development of synthetic biology and bioprocessing optimization, HA production via microbial fermentation is an economical and sustainable alternative over traditional animal extraction methods. Indeed, recently Streptococci and other recombinant systems for HA synthesis has received increasing interests due to its technical advantages. This review summarizes the production of HA by microorganisms and demonstrates its synthesis mechanism, focusing on the current status in various production systems, as well as common synthetic biology strategies include driving more carbon flux into HA biosynthesis and regulating the molecular weight (MW), and finally discusses the major challenges and prospects.

The zero/negative thermal expansion (ZTE/NTE), which is an intriguing physical property of solids, has been observed in a few families of materials. ZTE materials possess practical applications in specific circumstances such as space-related applications, engineering structures and precision instrument. Generally, NTE materials are used as additives to form a composite of the ZTE material with positive thermal expansion material. It is still a tremendous challenge to design new families of ZTE/NTE materials. Herein, we presented a temperature-dependent single crystal structure analysis in 110-300 K for a layered (NH4)2V3O8, which crystallizes in a tetragonal space group P4bm and comprises mixed valence [V3O82-]∞ monolayers and NH4+ residual in the interlayer spaces. Along the c-axis, (NH4)2V3O8 demonstrated uniaxial expansion behaviors, i.e., ZTE with αc = -1.10 × 10-6 K-1 in 110-170 K and NTE with αc = -16.25 × 10-6 K-1 in 170-220 K. Along the a-axis, (NH4)2V3O8 exhibited ZTE with αa = + 2.06 × 10-6 K-1 in 240-300 K. The mechanisms of ZTE and NTE were explored using structural analysis. The conduction of NH4+ ions in the interlayer space was studied, indicating that the conductivity rapidly rises with the expansion of interlayer space at temperatures of >293 K. This study discloses that layered vanadates are promising ZTE/NTE materials.

ABSTRACT

The emerging organic ion plastic crystals (OIPCs) are the most promising candidates used as solid-state electrolytes in a range of ionic devices. To endow an OIPC with additional functionality may create a new type of material for multifunctional devices. Herein, we present an ion plastic crystal, [EMIm][Ni(mnt)2] (1; [EMIm]+ = 1-ethyl-3-methylimidazolium and mnt2- = maleonitriledithiolate), and its crystal consists of twin dimeric chains of [Ni(mnt)2]- anions, embraced by [EMIm]+ cations. A crystal-to-plastic crystal transformation with a large latent heat that occurred at ∼367/337 K on heating/cooling is confirmed by the differential scanning calorimetry (DSC) technique. The plastic crystal phase in 1, characterized by variable temperature powder X-ray diffraction (PXRD) and optical microscopy images, spans a broad temperature range with ΔT ∼123/153 K on heating/cooling (DSC measurement), and the wide ΔT is relevant to an extra stable anion chain owing to the strong antiferromagnetic (AFM) interactions protecting the chain from collapse in the plastic crystal state. 1 is a single-component ion plastic crystal with a record high ion conductivity, 0.21 S·cm-1, at 453 K. The crystal-to-plastic crystal transformation in 1 is coupled to a bistable magnetic transition to give a multi-in-one multifunctional material. This study provides a creative thought for the design of OIPCs with striking thermal, electrical, and magnetic multifunctionality.

ABSTRACT

As a class of anionic oxoclusters of early transition metals in their highest oxidation states, polyoxometalates (POMs) show considerable structural versatility and unique chemical and physical properties, making them promising multifunctional materials. In this study, a Keggin-type POM has been achieved, with a formula of [(H2O)0.3@K6(H2O)12]H4.45[PV7.45Mo4.55O40]·11H2O (1), and its microcrystals and nanocrystals have been obtained, respectively. This POM was characterized by elemental analysis for C, H and N, ICP-MS, TG, PXRD, SEM, X-band EPR and XPS techniques. Single crystal X-ray diffraction analysis demonstrated that 1 shows a rare extended structure with a high-connected three-dimensional (3D) all inorganic network of a Keggin-type POM, built from {Mo4.55V7.45PO40}10.45- polyoxoanions and {(H2O)0.3@K6(H2O)12}6+ clusters with CsCl-type crystal structure. In addition, to the best of our knowledge, 1 shows the highest ratio of vanadium to molybdenum among Keggin-type POMs reported thus far. Most interestingly, 1 exhibits intrinsic proton conduction below the freezing point of water, with a proton conductivity of 6.90 × 10-7 S cm-1 at 249 K and further reaching 3.36 × 10-6 S cm-1 at 272 K and Ea = 0.44 eV at 249-272 K.

ABSTRACT

A supramolecular crystal, built from a 1 : 1 molar ratio of potassium tetraphenylboron (KBPh4) with 1,4,7,10,13,16-hexaoxacyclooctadecane (18-Crown-6), contains an organometallic half-sandwich superstructure unit {K(18-Crown-6)(ηn-C6H5B(C6H5)3)} (1 with n = 1-6), and shows a reversible phase transition at ∼211 K. The analysis of crystal structures at 173 K in the low-temperature phase, and 293 and 423 K in the high-temperature phase revealed that the breaking-symmetry phase transition is associated with the order-disorder transformation of [K(18-Crown-6)]+ and the change of the coordination mode of the phenyl ring to K+. A dielectric anomaly appears at ca. 212 K and dielectric relaxation occurs above 375 K in 1. The dielectric and thermal anomaly temperatures are close to each other, and the dielectric relaxation is relevant to the relative displacement of [K(18-Crown-6)]+ and the tetraphenylboron anion.

Haloplumbate-based perovskites display promising functionalities for advanced photovoltaic, optoelectronic and other applications with high performances and low costs. Herein, we present a study of variable-temperature crystal structures, dielectrics and conductance at 153-513 K, and luminescence at ambient temperature for a one-dimensional organic-inorganic perovskite, [N-methyldabconium]PbI3 (1). Hybrid 1 shows extra thermo- and water-stability (thermal decomposition at ca. 653 K), switchable dielectric behaviour and conductance at around 348 K, owing to symmetry-breaking structure phase transition from the hexagonal space group P63/mmc in the high-temperature phase to the orthogonal space group Pcba in the low-temperature phase, and bright yellow-green emission at room temperature, originating from the electron transition within the semiconducting {PbI3}∞ chains. This study will broaden the scope of lead halide-based hybrid materials for practical application in optical and electrical devices.
