ABSTRACT

OBJECTIVE: This study was designed to determine the efficacy of minocycline hydrochloride ointment (MHO) combined with Vitapex paste on middle-aged and elderly patients with combined periodontal-endodontic lesions (CPELs) and its effect on the inflammatory factors. METHODS: The data of 88 elderly patients with CPELs treated in the First Affiliated Hospital of Gannan Medical University from March 2020 to March 2022 were analyzed retrospectively. Among them, the patients treated with MHO and iodoform zinc oxide clove oil paste were assigned into the control group (n = 42) and the rest of the patients treated by MHO and Vitapex paste were assigned to the study group (n = 46). The inflammatory factors, periodontal indexes and efficacy were determined and compared between the two groups. The MOS 36-Item Short-Form Health Survey (SF-36) was adopted to evaluate the quality of life (QoL) of patients before and after treatment. Additionally, the adverse reactions of the two groups during treatment were analyzed and compared. The prognosis of the two groups of patients was analysed, and factors impacting their prognosis was analysed through the Logistic regression analysis. RESULTS: Before treatment, the levels of interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α) and interleukin-1ß (IL-1ß) were not significantly different between the two groups (all P > 0.05), while after treatment, these levels in both groups decreased significantly, with notably lower levels in the study group than those in the control group (all P < 0.05). Before treatment, the gingival index (GI), plaque index (PLI), probing depth (PD) and bleeding index (BD) of the two groups were similar (all P > 0.05); however, after treatment, the levels of GI, PD, PLI and BI of both groups decreased significantly (all P < 0.05), with more notable decreases in the study group than those in the control group (all P < 0.05). The study group showed a significantly higher overall response rate than the control group (P < 0.05). Before treatment, the SF-36 scores of the two groups were not significantly different (P > 0.05), while after treatment, both groups had significantly increased SF-36 scores, and the score in study group was significantly higher than the control group (P < 0.05). In addition, the incidence of adverse reactions was not notably different between the two groups (P > 0.05). According to univariate analysis, age, dental lesion grade, course of disease, and persistent dull pain were the risk factors affecting the prognosis of patients. According to multivariate analysis, dental lesion grade was the independent risk factor affecting the prognosis. CONCLUSION: MHO combined with Vitapex paste is effective for middle-aged and elderly patients with CPELs, which can effectively inhibit the patients' inflammatory reaction and improve their periodontal condition and QoL, with a low adverse reaction rate, so it is worthy of clinical promotion.

ABSTRACT

The American Musculoskeletal Society updated the diagnostic criteria for periprosthetic joint infection (PJI) in 2011 and 2018. However, the overall incidence of PJI since the introduction of these new standards has not been assessed. In order to fill this knowledge gap, a single-group meta-analysis was conducted using articles obtained from several databases, focusing on the incidence of PJI after primary total hip arthroplasty (THA). Our study revealed a significant difference in the incidence of PJI reported by different national or regional databases. Moreover, most cases of PJI were found to be underestimated. This highlights the crucial need for standardized diagnostic criteria and monitoring methods to accurately identify and track cases of PJI. Furthermore, a bibliometric analysis was conducted to provide a comprehensive overview of the current state of research on PJI after THA. This analysis explored the most productive countries, organizations, journals, and individuals in this research area. Additionally, it identified the research trends and hotspots of the last decade, highlighting the advancements and areas of focus in this field. By conducting these analyses, the study aims to contribute to the understanding of PJI after THA and provide valuable insights for clinicians, researchers, and policymakers involved in the management of this condition.

ABSTRACT

The adsorption and desorption of 9 PFAS, including 3 perfluoroalkyl sulphonic and 6 perfluoroalkyl carboxylic acids, in artificial groundwater was investigated using 3 commercial adsorbents that comprised a powdered activated carbon (PAC), a surface-modified organoclay (NMC+n), and a carbonaceous organic amendment (ROAC). Sorption kinetics and isotherms of PFAS, as well as the effects of adsorbent dose, pH, index ion and ionic strength on PFAS adsorption and desorption were investigated. Sorption of multicomponent PFAS in the adsorbents was rapid, especially for NMC+n and ROAC, regardless of PFAS chain length. The sorption and (and especially) desorption of PFAS in the adsorbents was impacted by the pH, index ion, and ionic strength of simulated groundwater, especially for the short chain PFAS, with only minimal impacts on NMC+n and PAC compared to ROAC. Although the potential mineral and charged constituents of the adsorbents contributed to the adsorption of short chain PFAS through electrostatic interactions, these interactions were susceptible to variable groundwater chemistry. Hydrophobic interactions also played a major role in facilitating and increasing PFAS sorption, especially in adsorbents with aliphatic functional groups. The desorption of PFAS from the adsorbents was below 8 % when the aqueous phase was deionised water, with no measurable desorption for NMC+n. In contrast, the desorption of short chain PFAS in simulated groundwater increased substantially (30-100 %) in the adsorbents, especially in ROAC and NMC+n, but more so with ROAC. In general, the three adsorbents exhibited strong stability for the long chain PFAS, especially the perfluoroalkyl sulphonic acids, with minimal to no sorption reversibility under different pH and ionic composition of simulated groundwater. This study highlights the importance of understanding not only the sorption of PFAS in groundwater using adsorbents, but also the desorption of PFAS, which may be useful for decision making during the ex-situ and in-situ treatment of PFAS-contaminated groundwater.

ABSTRACT

The outcomes of some breast cancer patients remain poor due to being susceptible to recurrence, metastasis and drug resistance, and lactate metabolism has been described as a hallmark of cancer and a contributor to cancer progression and immune escape. Hence, it is worthy of seeking potentially novel biomarkers from lactate metabolism relevant perspectives for this particular cohort of patients. In this context, 205 available lactate metabolism-related genes (LMGs) were obtained by a search of multiple genesets, and the landscape of somatic mutation, copy number variation, and mRNA expression levels was investigated among these genes. Crucially, 9 overall survival-related LMGs were identified through univariate Cox regression analysis in The Cancer Genome Atlas (TCGA) and Molecular Taxonomy of Breast Cancer International Consortium (METABRIC) databases. Subsequently, a prognostic signature, defined as Lactate Metabolism Index (LMI), was established with 5 OS-related LMGs using Least Absolute Shrinkage and Selection Operator (LASSO) Cox hazard regression analysis in TCGA training set, and then validated in two external cohorts (METABRIC and GSE96058). From the comprehensive results, breast cancer patients with high LMI had considerably poorer survival probability across all cohorts, and the degree of clinical features tended to be more severe as the LMI value increased. Furthermore, a prognostic nomogram incorporating LMI, age, and AJCC stage was constructed and demonstrated great prediction performance for OS of breast cancer patients, which was evaluated by the calibration plot and the decision curve analysis. Moreover, the potential effect of different LMI values on levels of immune checkpoints, tumor-infiltrating immune cells, and cytokines were explored ultimately, and patients with higher LMI values might gain an immunosuppressive tumor microenvironment that contributed to immune escape of breast cancer and inferior prognosis. Collectively, all findings in the study indicated the potential prognostic value of LMI in breast cancer, providing further implications for the role of lactate metabolism in breast cancer prognosis, tumor immune microenvironment, and immunotherapy.

ABSTRACT

Periodontitis (PD), also known as gum disease, is a condition causing inflammatory bone resorption and tooth loss. Regulatory T cells (Tregs) and regulatory B cells (Bregs) are vital in controlling the immune response and hence play a role in infections and peripheral tolerance adjustment. These cells have immunosuppressive and tissue-repairing capabilities that are important for periodontal health; however, in inflammatory circumstances, Tregs may become unstable and dysfunctional, accelerating tissue deterioration. In recent years, Regulatory cell-mediated immunotherapy has been shown to be effective in many inflammatory diseases. Considering the roles of Tregs and Bregs in shaping immune responses, this study aimed to review the published articles in this field to provide a comprehensive view of the existing knowledge about the role of regulatory T and B cells, as well as their therapeutic applications in PD.

ABSTRACT

Low pH is an important limiting factor for plant development in the south of China due to problems of acid red soil and boron (B) deficiency. Whereas, there is very limited information on the relationship between media pH and B distribution in plant, the physiological process changed by the interaction of pH and B in citrus growth also unclear. This experiment was conducted on trifoliate rootstock by employing two different concentrations of B (0 or 10 µM B) under three pH levels: pH 4, pH 5, and pH 6. Our results illustrate that low pH inhibite plant growth and cause oxidative stress in the roots, resulting in cell membrane injury. The increase of pH and B addition reduce the accumulation of ROS (O2.- and H2O2) by regulating the activity of Class Ⅲ peroxidases (CIII Prxs). Moreover, increased pH improves the internal circulation of B in plants and decrease the content of lignin and cellulose in cell wall (CW). In summary, our investigation demonstrated that the increase of pH in nutrient solution can accelerate the re-distribution of B by roots to promote citrus growth. The accumulation of B in roots can protect plants from the damage of ROS by regulating the activity of CIII Prxs as well as decrease the content of lignin and cellulose are to promotes roots elongation.

ABSTRACT

In this work, a functionalized polycaprolactone (PCL) composite fiber combining calf-type I collagen (CO) and natural drug usnic acid (UA) was prepared, in which UA was used as an antibacterial agent. Through 3D near-field electrospinning, the mixed solution was prepared into PCL/CO/UA composite fibers (PCUCF), which has a well-defined perfect arrangement structure. The influence of electrospinning process parameters on fiber diameter was investigated, the optimal electrospinning parameters were determined, and the electric field simulation was conducted to verify the optimal parameters. The addition of 20% collagen made the composite fiber have good hydrophilicity and water absorption property. In the presence of PCUCF, 1% UA content significantly inhibited the growth rate of Gram-positive and negative bacteria in the plate culture. The AC-PCUCF (after crosslinking PCUCF) prepared by crosslinking collagen with genipin showed stronger mechanical properties, water absorption property, thermal stability, and drug release performance. Cell proliferation experiments showed that PCUCF and AC-PCUCF had no cytotoxicity and could promote cell proliferation and adhesion. The results show that PCL/CO/UA composite fiber has potential application prospects in biomedical dressing.

Zirconium alginate/poly(N-isopropyl acrylamide) hydrogel beads with a semi-interpenetrating network (ZA/PNIPAM) were prepared by using the ionic crosslinking and radical polymerization method and investigated for phosphate removal from aqueous solutions. The effects on the adsorption performance of hydrogel beads, including initial pH, adsorbent dose, initial phosphate concentration, and co-existing anions, were evaluated systematically. Results showed that the ZA/PNIPAM could exhibit a maximum uptake capacity of phosphate at pH 2.The uptake capacity of the adsorbent increased with a decrease in the dose or an increase in the initial phosphate concentration. The presence of SO42- had a more negative effect on phosphate removal compared to Cl- and NO3-. The kinetics fitted a pseudo-second-order model and intraparticle diffusion model, suggesting the adsorption rate was mainly controlled by surface adsorption and diffusion into the interior of the hydrogel beads. The isotherm data could be described by the Freundlich model, indicating that the adsorption process was heterogeneous multilayer adsorption. The studies of FTIR, XPS, and zero point of charge with relevant adsorption data revealed that the phosphate adsorption mechanisms could be electrostatic attraction (physical adsorption) and ligand exchange reactions (chemical adsorption). After four cycles of regeneration, ZA/PNIPAM exhibited a stable uptake capacity, indicating favorable reusability.

ABSTRACT

OBJECTIVE: To study the radiobiological characteristics of a HepG2 cell line with mitochondrial DNA (mtDNA) deletion. METHODS: HepG2 cells were cultured in a medium containing ethidium bromide, acetylformic acid and uracil. The HepG2 cell line with mtDNA deletion (ρ(0)HepG2 cells) were acquired after 30 subcultures by limited dilution cloning. The cell survival was then observed in the absence of acetylformic acid and uracil, and the total mtDNA deletion in the cells was confirmed by PCR. The radiosensitivity of HepG2 and ρ(0)HepG2 cells was evaluated by exposure to gradient doses of 6 MV X ray irradiation. The cell apoptosis was assessed following a 2 Gy X-ray exposure with Hochest33342 staining, and the invasiveness of ρ(0)HepG2 cells was measured by Transwell assay. RESULTS: HepG2 cells could survive 30 subcultures in the presence of ethidium bromide, and massive cell death occurred after removal of acetylformic acid and uracil from the medium. PCR confirmed total mtDNA deletion from ρ(0)HepG2 cells, whose α/ß value was significantly lower than that of HepG2 cells. ρ(0)Hep-G2 cells showed an obviously lowered cell apoptosis rate following X-ray exposure with enhanced cell invasiveness. CONCLUSION: HepG2 cells can be induced by ethidium bromide into ρ(0)HepG2 cells with an increased radiation resistance, anti-apoptosis ability and cell invasiveness.

ABSTRACT

The standard treatment for adult T-Lymphoblastic lymphoma (T-LBL) has not been defined. This study was to analyze the efficiency of modified BFM-NHL-90 regimen in 36 adult patients with newly diagnosed T-LBL at the Sun Yat-Sen cancer center between August 2000 and December 2010. After the induction protocols, 34/36 (94%) of the patients achieved complete remission or unconfirmed complete remission. At the median follow-up of 36 months, 13 patients relapsed, but no relapses were observed in the CNS. The 3-year overall survival and 3-year event-free survival rates were 66.9% and 65.4%, respectively. Patients of female gender with hepatomegaly and hemoglobin less than 120 g/L, and the time interval exceeding 38 days between induction 1a and 1b, had inferior EFS and OS. The results was comparable to the previous regimens and the regimen could prevent CNS relapse with 4 high-dose MTX every 3 months during the maintenance phase.
