ABSTRACT

White tea, one of the six traditional teas in China, is made only through natural withering and low-temperature drying processes. It demonstrates diverse pharmacological and health-promoting effects, including antioxidant, antiviral, anticancer, and hypolipidemic activities. Despite the significance of polysaccharides in white tea leaves, their fine structure and physiological functions remain unexplored. In this study, the polysaccharide fragment WTP-80a with anticancer activity was isolated and purified from white tea through water extraction, alcohol precipitation, DEAE-52 ion exchange column chromatography, and sephacryl S-200 dextran gel column chromatography. WTP-80a exhibited a molecular weight of 1.14 × 105 Da and consisted of galactose (Gal), arabinose (Ara), rhamnose (Rha), and glucuronic acid (Glc-UA). The main chain skeleton of WTP-80a contained 3,6)-ß-Galp-(1→, 3)-α-Galp-(1→, 5)-α-Araf-(1 â†’ and 3)-α-Glcp-UA-(1→. Branch chains included α-Araf-(1 â†’ and ß-Rhap-(1 â†’ connected to the C3 and C6 positions of →3,6)-ß-Galp-(1→, respectively. In vitro anticancer experiments revealed that WTP-80a effectively hindered the proliferation, colony formation, migration, and invasion of B16F10 cells. Additionally, it induced apoptosis in B16F10 cells by blocking the G2/M phase, increasing active oxygen content, and reducing mitochondrial membrane potential. These findings provide a solid theoretical foundation for the application of white tea polysaccharides as anticancer products.

ABSTRACT

Cascade reservoirs construction has modified the nutrients dynamics and biogeochemical cycles, consequently affecting the composition and productivity of river ecosystems. The Jinsha River, as the predominant contributor to runoff, suspended sediment (SS), and nutrients production within the Yangtze River, is a typical cascade reservoir region with unclear transport patterns and retention mechanisms of nutrients (nitrogen and phosphorus). Furthermore, how to regulate nutrients delivery in the cascade reservoirs region is also an urgent issue for basin water environment study. Therefore, we monitored monthly variations in nitrogen and phosphorus concentrations from November 2021 to October 2022 in the cascade reservoirs of the Jinsha River. The results indicated that the concentrations and fluxes of total phosphorus (TP) and particulate phosphorus (PP) decreased along the cascade of reservoirs, primarily due to PP deposited with SS, while opposing trends for total nitrogen (TN) and dissolved total nitrogen (DTN), which might be the consequences of human inputs and the increase of dissolved inorganic nitrogen discharged from the bottom of the reservoirs. Moreover, the positive average annual retention ratios for TP and PP were 10% and 16%, respectively, in contrast to the negative averages of -8 % for TN and -11% for particulate nitrogen (PN). The variability in runoff-sediment and hydraulic retention time (HRT) of cascade reservoirs played crucial roles in the retention of TP and PP. A regulatory threshold of HRT = 5.3 days in the flood season was obtained for controlling the balance of TP based on the stronger relationship between HRT and TP retention ratio. Consequently, the HRT of these reservoirs could be managed to control nutrients delivery, which was of particular significance for basin government institutions. This study enhances our comprehension of how cascade reservoirs influence the distribution and transport patterns of nutrients, offering a fresh perspective on nutrients delivery regulation.

ABSTRACT

Grafting the wine grape variety Cabernet Sauvignon onto salinity-tolerant rootstocks can improve salinity tolerance and grape yields in regions with high salinity soils. In this experiment, the effects of different rootstocks and rootstock combinations on the saline-alkaline stress (modified Hoagland nutrient solution + 50 mmol L-1 (NaCl + NaHCO3)) of Cabernet Sauvignon were studied. Correlation and principal component analyses were conducted on several physiological indicators of saline-alkaline stress. Salinity limited biomass accumulation, induced damage to the plant membrane, reduced the chlorophyll content and photosynthetic capacity of plants, and increased the content of malondialdehyde, sodium (Na+)/potassium (K+) ratio, and antioxidant enzyme activities (superoxide dismutase, peroxidase, and catalase). Significant differences in several indicators were observed among the experimental groups. The results indicate that the saline-alkaline tolerance of Cabernet Sauvignon after grafting was the same as that of the rootstock, indicating that the increased resistance of Cabernet Sauvignon grapes to saline-alkaline stress stems from the transferability of the saline-alkaline stress resistance of the rootstock to the scion.

ABSTRACT

Different rootstocks for grapes can significantly affect fruit color and quality, possibly by affecting hormone contents, related genetic pathways, and fruit coloring mechanisms in skin. 'Cabernet Sauvignon' was grafted to '5BB', 'SO4', '140R', 'CS', '3309M' and 'Vitis riparia' rootstocks, with self-rooting seedlings as the control (CS/CS), and sampled from the early stage of veraison to the ripening stage. The effects of rootstock on the contents of gibberellin (GA3), auxin (IAA), and abscisic acid (ABA) in grape skin were determined alongside the expression levels of eight anthocyanin synthesis related genes using real-time fluorescence quantitative PCR methods. The rootstock cultivars exhibited accelerated fruit color change, and the CS/140R combination resulted in grapes with more color than the control group in the same period. With the development of fruit, the IAA and GA3 contents in the skin of different rootstock combinations showed trends of increasing initially, then decreasing, while the ABA content decreased initially and then increased. During the verasion (28 July), the various 'Cabernet Sauvignon' rootstock combinations exhibited varying degrees of increases in GA3, ABA, and IAA contents; correlation analysis showed that, at the start of veraison, the expression levels of the anthocyanin synthesis-related genes VvCHS, VvDFR, and VvUFGT had strong positive correlations with hormone contents, which indicated they are key genes involved in the endogenous hormone responsive anthocyanin biosynthesis pathway. The results of this study showed that rootstock regulates the fruit coloring process by influencing the metabolism level of peel hormones in the 'Cabernet Sauvignon' grape.

ABSTRACT

Colon adenocarcinoma (COAD) is a malignant tumor with a high mortality rate. Angiogenesis plays a key role in the development and progression of cancer. However, in COAD, studies between angiogenesis and prognosis, immune cell infiltration, and personalized treatment guidance are currently lacking. In the present study, we comprehensively assessed 35 angiogenesis-related genes (ARG) and identified key ARGs affecting OS in COAD patients. The ARG Prognostic Index (ARGPI) was constructed based on a univariate Cox regression model and its prognostic value was evaluated in TCGA-COAD, GSE39582, GSE161158 and TRSJTUSM Cohort. We constructed ARGPI as an independent risk factor for OS in COAD patients and combined with clinical parameters to further construct an ARGPI-based nomogram, which showed a strong ability to predict overall survival in COAD patients. High ARGPI is associated with cancer-related and immune-related biological processes and signaling pathways; high TP53 mutation rate; high infiltration of MSC, pericytes, and stromal cells; and more CMS4 subtype. And low ARGPI benefited more from immune checkpoint inhibitor treatment. In addition, we also predicted the sensitivity of different ARGPI groups to common chemotherapeutic and targeted agents. In conclusion, this study constructed an ARGPI based on ARG, which robustly predicted the OS of COAD patients and provided a possible personalized treatment regime for COAD patients.

ABSTRACT

Background: Multicenter clinical research faces many challenges, including how to quantitatively evaluate the data contribution of each research center. However, few data pricing model meets the requirements to the scenario. Thus, a suitable mechanism to measure the data value for clinical research is required. Methods: Extensive documents were acquired and analyzed, including a rare disease list from the National Health Commission, data structures of the electronic medical records (EMR) system, diagnosis-related groups (DRGs) regulations from the Health Commission of Zhejiang Province, and the Clinical Service Price List of Zhejiang Province. Nine senior experts were invited as consultants from hospital and enterprises with professional field of clinical research, data governance, and health economics. After brainstorming and expert evaluation, seven data attributes were identified as the main factors affecting the value of medical data. Different weights were assigned for each attribute based on its influence on data value. Each attribute was quantized to an index based on proposed algorithms. The data value models for chronic diseases and other diseases were distinguished given the different sensitivity of data timeliness. A simulation system using blockchain and federated learning techniques was constructed to verify the data pricing model in the scenario of clinical research. Results: A comprehensive clinical data pricing model is proposed and the simulation of three research centers with 50 million real clinical data entries was conducted to verify its effectiveness. It demonstrates that the proposed model can compute medical data value quantitatively. Conclusions: Quantitative evaluation of the value of medical data for multicenter clinical research based on the proposed data pricing model works well in simulation. This model will be improved by real-world applications in the near future.

ABSTRACT

In this contribution, we apply a soft-template-assisted hydrothermal route using polyethylene-polypropylene glycol (F127) as soft-template agent and biomass batatas as carbon precursor to synthesis a novel hydrothermal mesoporous biochar (HMC-800) for adsorptive removal of tetracycline (TC) from wastewater. We use the biochar prepared without F127 and direct pyrolytic biochar for comparison. The physicochemical properties of all the studied biochar samples are measured using a suite of characterization techniques. Our results show that the HMC-800 displays the highest specific surface area (286.3 m2/g) and total pore volume (0.249 cm3/g), manifesting the introduction of F127 can result in formation of well-developed pore structures. Regarding adsorption properties, the HMC-800 outperforms other biochar samples for TC removal. Our finding shows that solution with near-neutral pH is favorable for TC removal, and the highest adsorption capacity is observed at initial solution pH value 7. In addition, our findings show that applying the pseudo-second-order kinetic and Freundlich isotherm equation closely models the recorded adsorption behavior. The maximum adsorption capacity is measured to be as much as 238.7 mg/g by Langmuir isotherm model. Pore filling, hydrogen-bonding and n-π interaction are suggested to be the prevailing adsorption mechanisms compared to the other mechanisms. Furthermore, the HMC-800 performs better in regeneration and reuse experiments, making it a promising adsorbent material for TC removal from wastewater.

The objective of this experiment was to study the effects of three irrigation methods, i.e., subsurface drip irrigation with a tank system (SDI) , plastic film mulched-drip irrigation (MDI), and conventional drip irrigation (DI) on the regulation between root and crown function of Vitis vinifera 'Cabernet Sauvignon' seedlings. The results showed that both the SDI and MDI systems promoted the growth of the grape seedlings compared with DI, with the SDI system promoting the root growth, and MDI system promoting the aboveground growth. Root area, root volume, and root activity and SOD enzyme activity in the SDI treatment were greater than those of MDI or DI treatment in the 20-60 cm soil layer. SDI treatment increased root penetration and physiological activity. Symptoms of drought stress appeared earlier in DI treatment than in either MDI or SDI treatment in the same watering schedule. Net photosynthetic rate (Pn) and stomatal conductance (g(s)) of leaves were higher in SDI and MDI treatments than in DI treatment. ΦPS II and qP at 12:00-14:00 were lower in the MDI treatment than in SDI treatment at 7 d after irrigation, suggesting that the degree of photoinhibition in the fluorescence process in MDI treatment was more than that in SDI treatment. The high biomass and physiological activity of roots in the 20-40 cm depth could increase both of total plant biomass and aboveground biomass. The regulation between root and crown function was better in SDI treatment than in MDI and DI treatments. Therefore, SDI could be used as an alternative technique of water-saving irrigation practices.

ABSTRACT

This study examined the performance of a novel hybrid system of forward osmosis (FO) combined with vacuum membrane distillation (VMD) for reclaiming water from shale gas drilling flow-back fluid (SGDF). In the hybrid FO-VMD system, water permeated through the FO membrane into a draw solution reservoir, and the VMD process was used for draw solute recovery and clean water production. Using a SGDF sample obtained from a drilling site in China, the hybrid system could achieve almost 90% water recovery. Quality of the reclaimed water was comparable to that of bottled water. In the hybrid FO-VMD system, FO functions as a pre-treatment step to remove most contaminants and constituents that may foul or scale the membrane distillation (MD) membrane, whereas MD produces high quality water. It is envisioned that the FO-VMD system can recover high quality water not only from SGDF but also other wastewaters with high salinity and complex compositions.

ABSTRACT

The insertion/deletion (I/D) polymorphism of the gene encoding angiotensin converting enzyme is a controversial risk factor for restenosis after percutaneous transluminal coronary angioplasties (PTCA) in patients. Genetic association studies can be problematic to reproduce due to insufficient power, phenotypic heterogeneity, population stratification, small effect of the variant and even publication biases. To derive a more precise estimation of the relationship as well as to quantify the between-study heterogeneity and potential bias, a meta-analysis including 11,193 patients from 33 published cohort studies was performed. In a combined analysis, the summary per-allele odds ratio for restenosis was 1.31 (95% CI: 1.08-1.58, P = 0.006), and 1.22 (95% CI: 0.95-1.56, P = 0.12), for PTCA-stent and PTCA-balloon, respectively. In the subgroup analysis by ethnicity, significantly increased restenosis risks after PTCA-stent were found in Asians for the polymorphism; whereas no significant associations were found among Caucasians. As for restenosis risks after PTCA-balloon, no evidence of any gene-disease association was obtained in the stratified analyses according to ethnicity and study size. In conclusion, this meta-analysis demonstrated that the DD homozygous of ACE I/D polymorphism was significantly associated with elevated restenosis susceptibility after PTCA-stent among Asian populations.
