ABSTRACT

Bovine Respiratory Disease (BRD) poses a significant threat to cattle welfare worldwide, affecting their respiratory system and causing substantial economic losses. BRD is multifactorial in nature. This research aimed to investigate the serological profile of calves for the five main bovine respiratory pathogens. Serum samples were collected from dairy calves aged 7-12 months that had never been vaccinated against tested pathogens and had recently shown signs of BRD. A total of 4095 calves from 650 dairy herds located in south-western Poland were examined. The Commercial Indirect Respiratory ELISA Kit Multiplexed-Double well-BIO K 284/5 (Bio-X Diagnostics, Rochefort, Belgium) was used to determine the presence of antibodies against BVDV, BoAHV1, BRSV, BPIV3, and M. bovis. The presence of antibodies against at least one of the tested pathogens was found in 306 (47.08%) herds. The overall prevalence of antibodies was as follows: BoAHV1 21.54%, BVDV 32.0%, BRSV 34.15%, BPIV3 34.31%, and M. bovis 31.38%. The strongest correlation was between BRSV antibodies positive sera and BPIV3 antibodies positive sera. Among the five pathogens tested, antibodies to BVDV, BRSV, BPIV3, and M. bovis were most commonly detected simultaneously. The results of this study indicate the need for preventive measures against these pathogens in the studied region.

This study's objective was to evaluate whether adding a combination of phytoactive (microencapsulated essential oils, minerals, turmeric extract, tannin, prebiotic, and probiotic) to the feed of lactating Jersey cows positively affects the production, composition, and quality of milk, rumen environment, and animal health. Fourteen Jersey cows were divided into two groups (control and phytogenic) for an experiment with two lactation phases of 45 days each (early lactation and mid-lactation). During the experiment, milk production was higher at various times in cows that consumed phytoactive, and these animals had the best feed efficiency. In mid-lactation, phytoactive intake increased nutrient digestibility. The number of lymphocytes in the blood is reduced when cows consume phytoactive substances. Globulin levels increased in these cows fed with the additive, which may be related to a higher concentration of immunoglobulins, especially IgA. Cows fed phytoactives had lower ceruloplasmin and haptoglobin concentrations. Lower serum lipid peroxidation, associated with greater glutathione S-transferase activity, is a good health indicator in cows that consume phytoactive substances. The higher concentration of volatile fatty acids was due to the higher proportion of acetic acid in the ruminal fluid combined with lower butyric acid. Somatic cell counts in milk were lower in cows that consumed phytoactives during mid-lactation, as well as the effect of the treatment on Streptococcus spp. (lower in cows that consumed the additive). We conclude that consuming the additive benefits cows' health modulates rumen fermentation and nutrient digestibility, and positively affects milk production and quality.

ABSTRACT

This study systematically investigated the effects of tea polyphenols on methane (CH4) production and the rumen epithelial cell transport capability in cattle using both in vitro and animal experiments, employing multi-omics techniques. The in vitro results demonstrated that, compared to the control group, tea polyphenols significantly reduced CH4 production and the acetate/propionate ratio (p < 0.05). Tea polyphenols reduced CH4 production by inhibiting the relative abundance of unclassified_d_Archaea methanogens and the protozoa Pseudoentodinium and g__Balantioides. The animal experiments showed that tea polyphenols significantly increased the concentrations of T-AOC and GSH-PX in bovine blood (p < 0.05). In addition, microbial groups such as Rikenellaceae_RC9_gut_group, Ruminococcaceae_NK4A214_group, and Butyrivibrio_2 were significantly enriched in the ruminal fluid of the tea polyphenol group (p < 0.05). The proteomic results indicated significant upregulation of proteins such as COIII, S100A8, FABP1, SLC2A8, and SLC29A1 (p < 0.05) and downregulation of proteins including HBB, RAB4A, RBP4, LOC107131172, HBA, and ZFYVE19 (p < 0.05), with FABP1 showing a positive correlation with propionate concentration, and RAB4A had a negative correlation (p < 0.05). Overall, tea polyphenols modulate the microbial composition within the rumen, inhibiting CH4 production and enhancing the host's rumen epithelial cell transport capacity for volatile fatty acids.

In livestock, temperature, humidity, and Temperature-Humidity Index (THI) affect the welfare, yields, health and viability of animals. This study aimed to develop optimal temperature, humidity, and THI thresholds for dairy farms in temperate climate regions using a fuzzy logic model. THI values were calculated using three different literature-derived equations, considering different temperature and humidity situations in dairy farms. The Mamdani-type fuzzy logic method was utilized to formulate linguistic expressions for temperature, humidity, and THI values. According to the THI thresholds, the areas below the Receiver Operating Characteristic (ROC) were found to be significant (p < 0.001) in all fuzzy algorithms. The study found 100% harmony with the THI thresholds of 66 and 72 for cattle in temperate climates, but only 73.6% harmony with the threshold of 74 for cattle adapted to tropical climate. Briefly, in temperate dairy farms, the fuzzy logic revealed that the optimal temperature, humidity and THI values should be between 14-18.5°C, 65-70% and 52.5-64.5, respectively. However, further research is required to understand the impact of thresholds determined by fuzzy logic on dairy cow production and welfare.

ABSTRACT

Third-generation cephalosporins such as ceftiofur are critically important antibiotics because human pathogens with resistance to these drugs contribute to high mortality rates. These antibiotics are also frequently given to dairy cattle for treating infections, emphasizing the critical role they play in both human and veterinary medicine. To investigate the impact of intramuscular ceftiofur treatment on the concentration of resistant bacteria in the gut, we focused on cows with metritis, a common bacterial infection that frequently requires antibiotic intervention. Twelve cows with metritis (cases) were enrolled and treated with intramuscular ceftiofur for 5 d along with 12 matched healthy cows that were not given ceftiofur (controls). Fecal samples were collected weekly from cows in both the case and control groups for 4 weeks, starting before the treatment of the case group. Five fecal samples per cow were used for analysis (n = 120 samples). The abundance of Gram-negative bacteria was quantified per sample after plating on MacConkey agar, which was also used to quantify the abundance of Gram-negative bacteria with resistance to ceftiofur, ampicillin, and tetracycline. Interestingly, the case cows with metritis had a greater abundance of Gram-negative bacteria than the control cows just before treatment, but no difference in abundance was observed between groups at wk 1-4. The abundance of ceftiofur-resistant Gram-negative bacteria was also similar between the case and control cows immediately before treatment of the cases. However, a significant increase in abundance of ceftiofur-resistant Gram-negative bacteria was observed in the case cows 1-week after treatment that persisted through wk 3. Although the recovery of ampicillin- and tetracycline-resistant bacteria was similar between the 2 groups post-treatment, cases had significantly higher levels of ampicillin-resistant bacteria before treatment. Collectively, these findings demonstrate that intramuscular ceftiofur treatment can affect the abundance of cultivable Gram-negative bacteria and select for ceftiofur-resistant populations that can persist for up to 3 weeks. Judicious use practices are needed to ensure that ceftiofur and other critically important antibiotics are administered only when necessary to minimize the spread of resistance and safeguard public and animal health.

ABSTRACT

We offer a perspective on the major challenges that are confronting the management of reproduction in sheep and cattle in Australia and New Zealand, over the next two decades. An important context is the dominance of grazing systems in which large flocks or herds are managed over large areas where it is challenging to manage reproduction with precision. Consequently, the variable forage supply usually dominates reproductive outcomes, a problem that will be exacerbated by global heating. Thus, in extensive grazing systems, there is a great need for technological solutions to improve the management of nutrition. Global heating will also exert direct effects on reproductive function. Therefore, for the foreseeable future, reproduction will remain a focus for industry. In addition, as the industries develop, we foresee continued societal pressure to reduce medication, mitigate environmental damage, and improve animal well-being. Management solutions for extensive grazing systems must involve minimal interventions with the animals and be applicable to diverse genotypes and environments. Clearly, genetics and breeding will be at the heart of solutions and elegant strategies will be needed that focus on developing animals that are robust, if perhaps a little less productive. A high rate of genetic gain is the main reason for pursuing reproductive technologies, but highly advanced reproductive technology is not likely to be the best fit in extensive management systems. Even for AI, the simplest technology, uptake is limited and lateral thinking is needed to find ways to improve the rates of genetic gain. We conclude that there are many opportunities for improving reproductive performance in sheep and cattle in Australia and New Zealand. As we gain deeper understanding of the processes involved, we should be able to make progress in fertility and fecundity, embryo survival, and postnatal survival. Improvements in reproductive performance will increase productivity, and should also be associated with significantly improved animal well-being and a reduction in methane emissions intensity. To capture these benefits, the development of new management options will require lateral thinking about reproductive technology for extensive grazing systems, and a transdisciplinary approach that brings together the systems biology of grazing animals with an understanding of the barriers to adoption by farmers.

Veterinarians play a significant role in the treatment and prevention of livestock diseases at the farm level, safeguarding public health and ensuring food safety. In sub-Saharan Africa, access to quality veterinary services is a major challenge for livestock farmers due to the low number of publicly employed veterinarians, underfunding and privatisation of veterinary services. Low investment in veterinary services and infrastructure, including a lack of laboratories for diagnosis, has made veterinarians rely on their experience and knowledge of cattle disease symptoms developed over years of practice to diagnose and treat cattle diseases. A cross-sectional survey using a role-play approach was used to collect data on knowledge regarding cattle diseases among veterinarians in veterinary clinics and private practices in Addis Ababa, Oromia and Adama regions in Ethiopia. Veterinarians were given a number of disease scenarios based on "fictive disease symptoms" that are commonly manifested in a sick cow and asked to identify the disease what personal biosecurity they would use, diagnostic tests they would perform, treatments they would prescribe, treatment costs, and additional services and inputs they would recommend to the farmer. The results show that veterinarians could identify endemic cattle diseases through symptoms. The majority of veterinarians did not find it important to report notifiable diseases, a behaviour which could hamper disease surveillance and outbreak response. The advice and services the veterinarians said they would offer and recommend to farmers included improvement in feeding, vaccination, use of artificial insemination, and adoption of farm biosecurity measures that can reduce disease prevalence, and improve food safety, animal health and welfare. Low use of personal protective equipment and other protective biosecurity measures among veterinarians could expose them to zoonotic diseases. The study concludes that there is a need for increased funding for continuous training, improved access to animal health-related information, and investment in infrastructure such as laboratories to enable veterinarians to deliver quality animal health services.

BACKGROUND: Dairy cattle feed supplies in the tropics, and particularly in Ethiopia, are mostly derived from base diets (natural and improved grasslands, agricultural wastes, fodder trees and shrubs), agro-industrial by-products and non-conventional feeds. The availability of major feed resource kinds, sources and dairy cattle feeding practises is primarily influenced by agro-ecology and season, as well as their interactions. OBJECTIVES: The research was carried out in the Chencha District of Gamo Zone, Southern Ethiopia, with the goals of assessing the available, feeding system, opportunity, limits and copying strategy of dairy cattle production, as well as estimating feed balance. METHODS: Data were gathered through primary and secondary data sources, field observations, key informant interviews, focus group talks and individual interviews. The survey data were stratified into altitudes, coded and analysed using the Statistical Package for Social Sciences version 20 and the general linear model approach. Cross-tabulation was used to examine the statistical variance of categorical data. RESULTS: The presence of different feed resource types, distinct agro-ecologies (p < 0.05) and crop residue accessibility were the major opportunities for dairy cattle production in the study district, but land scarcity, a lack of dry season forages and land degradation due to erosion were the major constraints for dairy cattle feed production. In the research district, the main feed resource-copying approaches were reducing the number of dairy animals, saving optional feeds and purchasing optional feeds. The total mean of dairy cow feed supply to tropical livestock unit in terms of dry matter (DM) production each year was 3.86 t of DM/HH/annual, with a negative feed balance of 8.09 t that varied with seasons and agro-ecology (p < 0.05). CONCLUSIONS: According to the findings of this study, the low production of dairy cattle in the district is clearly related to the scarcity of feed resources. To address these issues, alternative feed production technologies should be implemented, such as farmers practising forage development on their own crop land and collecting crop residues during crop harvesting times and storing them under shed; the nutritive value of different types of fodder trees and shrubs should be further determined in the future and feed storage methods, particularly hay and silage, should be used when there are an abundance of feed resources available.

ABSTRACT

The present review has two objectives, the first is to investigate the differences in temperament between Bos indicus and Bos taurus breeds and determining the effects on production due to positive treatment and to compare this with negative HAR, by using the Five Domain Model as framework. The second objective is to discuss potential strategies to achieve better HAR when working with Bos indicus cattle. Bos indicus are more reactive and temperamental than Bos taurus cattle. When human animal relationships (HAR) are evaluated, Bos indicus cattle may react with greater intensity. They may be more likely to develop a negative emotional state, especially in extensively raised Bos indicus cattle that are handled only a few times each year. Bos indicus cattle can have positive emotional states when they have frequent positive interactions with people. Interactions with people, both positive and negative, would be in the fourth Domain of the Five Domains model. Cattle that are more reactive during handling may also have lower weight gain, even when they have abundant feed. This would be in the first Domain of Nutrition. When cattle are handled in races and corrals, injuries may be more likely to occur. Injuries and bruises would be in the third Domain of Health. Injuries could be caused by either poor handling practices by people or poor handling facilities. Yelling or electric prod use would be examples of poor HAR. Second Environmental Domain issues may be broken facilities or slick, slippery floors that are associated with falls.

ABSTRACT

Aging reduces the nutritional value of corn grain, which may be improved by fermentation prior to use. This study investigated the effects of replacing conventional corn (CC) with aging corn (AC) and fermented aging corn (FAC) in the diets of lactating Holstein cows. Six healthy third-parity Holstein cows were used in a replicated 3 × 3 Latin square experiment with 21-day periods. Cows were fed twice and milked twice daily. The cows were randomly divided into three treatment groups: (1) CC diet: a diet with 23.6% starch in diet DM containing 24.0% conventional corn; (2) AC diet: a diet with 23.5% starch in diet DM containing 24.0% aging corn replacing conventional corn; and (3) FAC diet: a diet with 23.2% starch in diet DM containing 24.6% fermented aging corn replacing conventional corn. The diets were formulated to be isonitrogenous and contained identical roughage. The FAC treatment increased the starch digestibility compared with AC. Feeding FAC increased the concentrations of total volatile fatty acid compared with CC and AC and decreased the molar proportion of acetate compared with AC. At the family level, the relative abundance of Prevotellaceae was higher on FAC than CC. The relative abundance of Succinivibrionaceae was lower on FAC than on CC and AC. Besides, at the genera level, the relative abundance of Succinivibrionaceae_UCG_002 in the rumen was higher on AC than CC and FAC. The relative abundance of Prevotella and Ruminococcus was higher on FAC than CC and AC. The relative abundance of Succinivibrionaceae_UCG_001 was lower on FAC than CC. The Simpson index was lower on FAC than CC and AC. The FAC treatment increased the milk yield (34.0, 33.7, and 35.2 kg/d for CC, AC, and FAC group, respectively) and protein yield, and thus, energy-corrected milk production was increased, and at the same time, decreased the somatic cell score compared with CC and AC. The AC treatment increased the malondialdehyde concentration in plasma compared with CC and FAC. The concentrations in plasma of triglyceride and malondialdehyde were lower on FAC than AC. The immunoglobulin G concentration in plasma was higher on FAC than CC and AC. Overall, feeding AC resulted in decreased plasma antioxidant capacity compared with CC, whereas feeding FAC altered the relative abundance of bacteria in the rumen and improved starch digestibility, ruminal bacterial diversity, lactation performance, plasma antioxidant capacity and immune competence compared with AC in dairy cows.

ABSTRACT

One suggested approach to improve the reproductive performance of dairy herds is through the targeted management of subgroups of biologically similar animals, such as those with similar probabilities of becoming pregnant, termed pregnancy risk. We aimed to use readily available farm data to develop predictive models of pregnancy risk in dairy cows. Data from a convenience sample of 108 dairy herds in the UK were collated and each herd was randomly allocated, at a ratio of 80:20, to either training or testing data sets. Following data cleaning, there were a total of 78 herds in the training data set and 20 herds in the testing data set. Data were further split by parity into nulliparous, primiparous, and multiparous subsets. An XGBoost model was trained to predict the insemination outcome in each parity subset, with predictors from farm records of breeding, calving and milk recording. Training data comprised 74,511 inseminations in 45,909 nulliparous animals, 86,420 inseminations in 39,439 primiparous animals, and 158,294 inseminations in 32,520 multiparous animals. The final models were evaluated by predicting with the testing data, comprising 31,740 inseminations in 19,647 nulliparous animals, 38,588 inseminations in 16,215 primiparous animals, and 65,049 inseminations in 12,439 multiparous animals. Model discrimination was assessed by calculating the area under receiver operating characteristic curves (AUC); model calibration was assessed by plotting calibration curves and compared across test herds by calculating the expected calibration error (ECE) in each test herd. The models were unable to discriminate between insemination outcomes with high accuracy, with an AUC of 0.63, 0.59 and 0.62 in the nulliparous, primiparous and multiparous subsets, respectively. The models were generally well-calibrated, meaning the model-predicted pregnancy risks were similar to the observed pregnancy risks. The mean (SD) ECE in the test herds was 0.038 (0.023), 0.028 (0.012) and 0.020 (0.008) in the nulliparous, primiparous and multiparous subsets respectively. The predictive models reported here could theoretically be used to identify subgroups of animals with similar pregnancy risk to facilitate targeted reproductive management; or provide information about cows' relative pregnancy risk compared with the herd average, which may support on-farm decision-making. Further research is needed to evaluate the generalizability of these predictive models and understand the source of variation in ECE between herds; however, this study demonstrates that it is possible to accurately predict pregnancy risk in dairy cows using readily available farm data.

The geometry of milk liners may affect milking performance and cow comfort as the milk liner is the only part of the milking machine that comes into contact with the teat. To determine the effect of alternative shape of milk liners we compared square (SQR) vs. the conventional round (RND) teat cup liner on milking performance and comfort of dairy cows. Treatment milk liners were randomly allocated to clusters within each side of the 12 a side double up-herringbone dairy shed in a complete randomised block design over two periods. Milking performance data from a total of 10 065 (late stage of lactation and once-a-day milking frequency, LATE) and 18 048 (early stage of lactation and twice-a-day milking frequency, EARLY) milking events were automatically recorded by a DeLaval milk meter, and separately analysed for LATE and EARLY, respectively. In EARLY, cow comfort behaviour was also recorded during afternoon milking sessions. Across the two study periods, average milk flow rate, milk flow rate during 0-15, 15-30 and 30-60 s after cluster attachment, and milk flow rate at cluster take-off were higher in SQR compared to RND treatment. Proportion of time in a milking session with low milk flow rate and duration of milking session were less in SQR compared to RND treatment. However, effect of geometry of milk liner on peak milk flow rate was inconsistent across the two-study periods. Peak milk flow rate was higher (P < 0.001) in SQR than RND in LATE, but higher (P < 0.001) in RND than SQR in EARLY. Stomping and kicking behaviours of cows were similar between treatments. Results of this study suggest that square milk liners potentially improve milking performance, without adverse effect on cow comfort compared to conventional round liners. Long-term, multi-site studies are required to confirm potential teat-end health benefits associated with square milk liners and further verify these results.

AIMS: To assess whether a whole-herd lameness score on a New Zealand dairy farm in spring could predict lameness prevalence on the same farm in summer (and vice versa) and whether a single-herd lameness score could be used to determine whether herd lameness prevalence was < 5% in both spring and summer. METHODS: Prevalence data (proportion of the herd with lameness score ≥ 2 and with score 3; 0-3 scale) from a study where 120 dairy farms across New Zealand were scored in spring and in the following summer were analysed using limits-of-agreement analysis. In addition, farms were categorised as having either acceptable welfare (lameness prevalence < 5% in both spring and summer) or not (lameness prevalence ≥ 5% in either spring or summer or both). The accuracy and specificity of a single, whole-herd lameness score at identifying herds with acceptable welfare were then calculated. RESULTS: The limits-of-agreement analysis suggests that 95% of the time, the prevalence of lameness in summer would be expected to be between 0.23 and 4.3 times that of the prevalence in spring. The specificity and accuracy of identifying a farm as acceptable on both occasions from a single observation were, respectively, 74% and 92% in spring, and 59% and 87% in summer. CONCLUSIONS: A single, one-off, whole-herd lameness score does not accurately predict future lameness prevalence. Similarly, acceptable status (lameness prevalence < 5%) in one season is not sufficiently specific to be used to predict welfare status in subsequent seasons. CLINICAL RELEVANCE: Whole-herd lameness scoring should be used principally as a means of detecting lame cows for treatment. A single whole-herd lameness score by an independent assessor should not be used to determine a herd's welfare status.

Bovine neosporosis is a widespread parasitic disease associated with significant economic losses. Its effects on the reproductive performance of cows have resulted in losses that run into the hundreds of millions of US dollars in dairy industries in various countries (Reichel et al., Int J Parasitol 43:133-142, 2013). Due to outdated and scant information on the occurrence of Neospora caninum infection in South Africa, the study aimed to determine the seroprevalence and risk factors associated with infection in dairy cattle in South Africa. A total of 1401 blood samples were randomly collected from cattle on 48 dairy farms in seven of the nine provinces in South Africa. A close-ended questionnaire was used in a cross-sectional study to obtain farm-level and animal-level data. Serological testing was done using a commercial IDvet Screen® Neospora caninum Indirect ELISA. An overall seroprevalence, adjusted for test sensitivity and specificity, of 2.3% (95% CI, 1.3-4.1) was detected and 48% (23/48) of sampled farms had at least one animal testing positive. The highest seroprevalence of N. caninum was in the KwaZulu-Natal province with 7.5% (95% CI, 3.8-14.3), and the lowest in Western Cape with 0.1% (95% CI, 0-1.2). The highest within-farm seroprevalence of 25% was detected on a farm in the North West Province. In a multivariable logistic regression model, the odds of N. caninum seropositivity were higher in Holstein-Friesian cattle when compared to other breeds. Good hygiene was identified as a protective factor. Cattle left out on pasture had increased odds of testing positive for N. caninum compared to those that were penned. The odds of testing seropositive for N. caninum was higher on farms that practised segregation of cattle into different age groups. The purchase of replacement animals was a significant risk factor, as open herds had increased odds of N. caninum seropositivity. Cattle on farms that did not have a specific calving location were more likely to be seropositive. This is the first such study in South Africa and shows that N. caninum is widely distributed in the country at a low seroprevalence, but it may be a cause of concern on certain farms.

ABSTRACT

BACKGROUND: Pharmacological activity of intramammary drugs depends on adequate drug concentrations within the cistern, but sampling is often limited. Insight into the active drug concentration within the mammary cistern may assist in determining effective and appropriate therapeutic decisions for cows being treated for mastitis. OBJECTIVE: Evaluate the disposition of ceftiofur hydrochloride administered intramammary in diseased and nondiseased quarters. Whole milk and ultrafiltrate sampling techniques were compared. ANIMALS: Ten mature, late lactation Holstein (n = 9) and Jersey (n = 1) dairy cows (422-670 kg) with naturally occurring clinical mastitis, producing between 1.4 and 15.9 kg/day of milk. METHODS: Ultrafiltration probes were placed in both mastitic and healthy quarters. Each quarter was treated with 2 doses of 125 mg ceftiofur hydrochloride suspension, and whole milk and milk ultrafiltrate samples were collected. Ceftiofur concentrations in composite whole milk and milk ultrafiltrate were analyzed. RESULTS: The maximum concentration of ceftiofur was higher in ultrafiltrate samples, but no differences were identified in healthy or mastitic quarters. The use of ultrafiltration probes provides a novel technique for free drug concentrations within the mastitic and healthy bovine mammary gland. CONCLUSIONS AND CLINICAL IMPORTANCE: Significant inter- and intracow variability and lower daily milk weights may overestimate ceftiofur concentrations available within the cistern. The pharmacokinetic (PK) parameters reported in milk ultrafiltrate will help establish a link between the PK and the corresponding drug effect, potentially providing a meaningful rationale for the selection of a safe and effective dose in cows with mastitis.

ABSTRACT

Heat stress (HS) is one of the key factors affecting an animal's immune system and productivity, as a result of a physiological reaction combined with environmental factors. This study examined the short-term effects of heat stress on cow behavior, as recorded by innovative technologies, and its impact on blood gas parameters, using 56 of the 1070 cows clinically evaluated during the second and subsequent lactations within the first 30 days postpartum. Throughout the experiment (from 4 June 2024 until 1 July 2024), cow behavior parameters (rumination time min/d. (RT), body temperature (°C), reticulorumen pH, water consumption (L/day), cow activity (h/day)) were monitored using specialized SmaXtec boluses and employing a blood gas analyzer (Siemens Healthineers, 1200 Courtneypark Dr E Mississauga, L5T 1P2, Canada). During the study period, the temperature-humidity index (THI), based on ambient temperature and humidity, was recorded and used to calculate THI and to categorize the data into four THI classes as follows: 1-THI 60-63 (4 June 2024-12 June 2024); 2-THI 65-69 (13 June 2024-18 June 2024); 3-THI 73-75 (19 June 2024-25 June 2024); and 4-THI 73-78 (26 June 2024-1 July 2024). The results showed that heat stress significantly reduced rumination time by up to 70% in cows within the highest THI class (73 to 78) and increased body temperature by 2%. It also caused a 12.6% decrease in partial carbon dioxide pressure (pCO2) and a 32% increase in partial oxygen pressure (pO2), also decreasing plasma sodium by 1.36% and potassium by 6%, while increasing chloride by 3%. The findings underscore the critical need for continuous monitoring, early detection, and proactive management to mitigate the adverse impacts of heat stress on dairy cow health and productivity. Recommendations include the use of advanced monitoring technologies and specific blood gas parameter tracking to detect the early signs of heat stress and implement more timely interventions.

Shiga toxin-producing and Enteropathogenic Escherichia coli are foodborne pathogens commonly associated with diarrheal disease in humans. This study investigated the presence of STEC and EPEC in 771 dairy cattle fecal samples which were collected from 5 abattoirs and 9 dairy farms in South Africa. STEC and EPEC were detected, isolated and identified using culture and PCR. Furthermore, 339 STEC and 136 EPEC isolates were characterized by serotype and major virulence genes including stx1, stx2, eaeA and hlyA and the presence of eaeA and bfpA in EPEC. PCR screening of bacterial sweeps which were grown from fecal samples revealed that 42.2% and 23.3% were STEC and EPEC positive, respectively. PCR serotyping of 339 STEC and 136 EPEC isolates revealed 53 different STEC and 19 EPEC serotypes, respectively. The three most frequent STEC serotypes were O82:H8, OgX18:H2, and O157:H7. Only 10% of the isolates were classified as "Top 7" STEC serotypes: O26:H2, 0.3%; O26:H11, 3.2%; O103:H8, 0.6%; and O157:H7, 5.9%. The three most frequent EPEC serotypes were O10:H2, OgN9:H28, and O26:H11. The distribution of major virulence genes among the 339 STEC isolates was as follows: stx1, 72.9%; stx2, 85.7%; eaeA, 13.6% and hlyA, 69.9%. All the 136 EPEC isolates were eaeA-positive but bfpA-negative, while 46.5% carried hlyA. This study revealed that dairy cattle are a major reservoir of STEC and EPEC in South Africa. ABSTRACT

Bovine respiratory disease (BRD) is a common global health problem in dairy cattle. The definitive diagnosis of BRD is complex because its etiology involves several predisposing and determining factors. This report describes the etiology of a BRD outbreak in a dairy herd in the mesoregion of Central Eastern Paraná, which simultaneously affected young (calves and heifers) and adult (cows) Holstein-Friesian cattle. Nine biological samples, consisting of five lung samples from two cows and three suckling calves, and four nasal swab samples from heifers, were used for etiological diagnosis. The nucleic acids extracted from lung fragments and nasal swabs were subjected to PCR and RT-PCR assays for partial amplification of the genes of five viruses [bovine viral diarrhea virus (BVDV), bovine alphaherpesvirus 1 (BoAHV1), bovine respiratory syncytial virus (BRSV), bovine parainfluenza virus 3 (BPIV-3), and bovine coronavirus (BCoV)] and four bacteria (Mycoplasma bovis, Mannheimia haemolytica, Pasteurella multocida, and Histophilus somni) involved in the etiology of BRD. All nine biological samples from the animals with BRD tested negative for BoAHV1, BRSV, BPIV-3, BCoV, and H. somni. Therefore, the involvement of these microorganisms in the etiology of BRD outbreak can be ruled out. It was possible to identify the presence of BVDV and M. bovis in singular and mixed infections of the lower respiratory tract in cattle. BVDV was also identified in two nasal swabs: one as a single etiological agent and the other in association with two bacteria (P. multocida and M. haemolytica). The phylogenetic analysis conducted in the nucleotide sequence of the 5'UTR region and Npro gene of the BVDV amplicons demonstrated that the BVDV field strains of this BRD outbreak belong to subgenotype 2b. To the best of our knowledge, this is the first report of BVDV-2b involvement in the etiology of BRD in Brazil. Finally, it is necessary to highlight that the cattle were obtained from an open dairy herd with biannual vaccinations for BVDV-1a and - 2a.

ABSTRACT

Antimicrobial resistance (AMR) is one of the greatest threats to global health worldwide and is threatening not only humans, but also animal production systems, including dairy farms. The objective of this paper was to describe risks factors associated with AMR on dairy farms in Québec, Canada. This observational cross-sectional study included 101 commercial dairy farms and took place between the springs of 2017 and 2018 for a one-year period. We explored risk factors such as farm practices and producer's knowledge (measured using a questionnaire), antimicrobial use (quantified using veterinary invoices), and the presence of Salmonella Dublin (tested by serology). We evaluated AMR with fecal Escherichia coli retrieved from pre-weaned calves and lactating cows using the following outcomes: the presence of extended-spectrum-ß-lactamase/AmpC resistance and the number of resistances to antimicrobial classes. We used logistic regression models to evaluate the association between each risk factor and the 2 outcomes for the 2 types of samples (pre-weaned calves and lactating cows). Furthermore, we explored the relationships between these risk factors utilizing data dimensionality reduction and hierarchical clustering. Outputs of these analyses were used as regressors for AMR in regression models. While the results for univariate analyses were ambiguous, the unsupervised analysis naturally categorized the sample of farms according to their health/treatment status (dimension 1, explaining 12.9% of the variance) and herd size (dimension 2, explaining 7.8%). Three clusters of farms were identified (cluster 1: mainly healthy herds and low ceftiofur users, cluster 2: relatively high ceftiofur users, cluster 3: farms with a higher incidence of diseases and higher antimicrobial treatment rates). Dimension 1 and cluster membership were statistically associated with the presence of extended-spectrum-ß-lactamase/AmpC resistance in lactating cows and in pre-weaned calves. Dimension 1 was also statistically associated with the number of resistances in lactating cows and in pre-weaned calves. This study highlights the complexity of analyzing risk factors associated with AMR. Our results suggest that the herd health status and the AMU-related practices used are associated with AMR in dairy farms. However, prospective studies are needed to confirm a causal relation.

ABSTRACT

The present study investigates the susceptibility of two imported dairy cattle breeds to Algerian local climatic conditions, with a primary focus on heat stress (HS) and its repercussions on fertility traits. The dataset comprises 20,926 artificial insemination records involving 6,191 Prim'Holstein and 5,279 Montbéliarde cows. The animals originated from three distinct agro-ecological regions: littoral (L), semi-arid (SA), and arid (Ar), characterized by average annual Temperature-Humidity Index (THI) values of 75.2, 69.53, and 74.75, respectively. Logistic and linear regression models were performed to analyze the relationship between the THI on the AI day, season, and agro-ecological origin of the animals with the Conception Rate at 1st Artificial Insemination (CR 1stAI), Conception Risk (CR), Services per Conception (SPC), and reproductive period (RP). The results demonstrated a significant negative impact (P < 0.001) of THI > 72 compared to THI ≤ 72 on CR1st AI and CR for both cattle breeds (Prim'Holstein: -49.7% and - 17%, respectively; Montbéliarde: -20.7% and - 15%, respectively). Seasonal effects revealed a notably higher CR1stAI in winter and spring (≈ 25%) for Prim'Holstein and Montbéliarde cows compared to summer (19.41%) and autumn (19.12%), respectively. Furthermore, a reduced likelihood of conception at 1stAI and subsequent AI was observed during summer (0.839) and autumn (0.818) compared to winter for the Montbéliarde cows. Taking into account the littoral region as a reference, the likelihood of 1stAI success increased for both breeds in the SA region and decreased for the Ar region (P < 0.001). SPC increased for both breeds in THI > 72 categories (Prim'Holstein: 6.3%, Montbéliarde: 7.1%, P < 0.01), in the Ar region (Prim'Holstein: 30.9%, Montbéliarde: 26%, P < 0.001), and in the SA region (4%, P < 0.05) compared to the L region No significant seasonal effect on SPC was observed for either breed (P > 0.05). The RP increased in the THI > 72 category (Prim'Holstein: 4.1%, Montbéliarde: 7.4%, P < 0.001) and in the Ar region (Prim'Holstein: 122%, Montbéliarde: 73.4%) for both breeds. RP decreased in autumn compared to winter (Prim'Holstein: 15.3%, Montbéliarde: 8.4%). This study underscores the adverse impact of mild to severe heat stress (HS) and related factors (season, region) on fertility of Prim'Holstein and Montbéliarde cows under Algerian conditions, emphasizing the necessity for heat stress mitigation strategies, especially in adverse littoral humid and Saharan-arid environmental conditions.
