ABSTRACT
The present study aimed to investigate the beneficial of prepared black rice anthocyanins nano-composite (An-AgNps) against hepatotoxicity induced by methotrexate (MTX) in rats. Anthocyanins nano-composite was prepared by silver as the metallic ion reduction and were characterized by IR and SEM. The rats in our experiment were divided into five groups. Serum lipid profile, serum transaminases (ALT and AST), ALP, LDH, TBA, GSH and SOD were examined. The results show that SEM of An-AgNps has average particle size from 70 to 130nm. In the group treated with MTX; TC, TG, LDL-c, ALT, AST, ALP, LDH and TBA levels were significantly (P≤0.05) increased than NC, while, HDL-c, SOD and GSH levels were significantly (P≤0.05) decreased. On the other hand, An-AgNps + MTX treated groups were reversed the levels of all biomarkers similar to NC. In conclusion, the results show that An-AgNps has a protective effect on MTX-induced hepatotoxicity and oxidative stress.
O presente estudo teve como objetivo investigar o benefício de nanocompósito de antocianinas de arroz preto preparado (An-AgNps) contra a hepatotoxicidade induzida por metotrexato (MTX) em ratos. O nanocompósito de antocianinas foi preparado a partir da prata por meio da redução do íon metálico e caracterizado por IR e SEM. Os ratos em nosso experimento foram divididos em cinco grupos, e foram examinados o perfil lipídico sérico, as transaminases séricas (ALT e AST), ALP, LDH, TBA, GSH e SOD. Os resultados mostram que SEM de An-AgNps tem tamanho médio de partícula de 70 a 130 nm. ABSTRACT
BACKGROUND: Studies have suggested the chemopreventive effects of anthocyanins on breast cancer carcinogenesis. This systematic review and meta-analysis aimed to evaluate the effect of anthocyanins on triple-negative breast cancer cells (TNBC) cultured in vitro. METHODS: We searched for all relevant studies that evaluated the mechanisms of migration, invasion, Akt/mTOR and MAPK pathways, and apoptosis, using PubMed and Scopus. Means and standard deviation were used, and a randomized effects model was applied, with a confidence interval of 95%. Statistical heterogeneity between studies was assessed using the Chi2 test and I2 statistics. All analyses were performed using RevMan software (version 5.4). RESULTS: Eleven studies were included in the systematic review and ten in the meta-analysis, where the roles of anthocyanin-enriched extract or cyanidin-3-O-glucoside (C-3-O-G) on MDA-MB-231 and MDA-MB-453 cells were investigated. DISCUSSION: There was a significant reduction in invasion (mean difference: -98.64; 95% CI: -153.98, -43.3; p Ë 0.00001) and migration (mean difference: -90.13; 95% CI: -130.57, -49.68; p Ë 0.00001) in TNBC cells after anthocyanins treatment. Anthocyanins also downregulated Akt (mean difference: -0.63; 95% CI: -0.70, -0.57; p Ë 0.00001) and mTOR (mean difference: -0.93; 95% CI: -1.58, -0.29; p = 0.005), while JNK (mean difference: -0.06; 95% CI: -1.21, 1.09; p = 0.92) and p38 (mean difference: 0.05; 95% CI: -1.32, 1.41; p = 0.95) were not modulated. There was also an increase in cleaved caspase-3 (mean difference: 1.13; 95% CI: 0.11, 2.16; p = 0.03), cleaved caspase-8 (mean difference: 1.64; 95% CI: 0.05, 3.22; p = 0.04), and cleaved PARP (mean difference: 0.93; 95% CI: 0.54, 1.32). Although the difference between control and anthocyanin groups was not significant regarding apoptosis rate (mean difference: 3.63; 95% CI: -2.88, 10.14; p = 0.27), the analysis between subgroups showed that anthocyanins are more favorable in inducing overall apoptosis (p Ë 0.00001). CONCLUSION: The results show that anthocyanins hold promise in fighting against TNBC, but their effects should not be generalized. In addition, further primary studies should be conducted so that more accurate conclusions can be drawn.

Recently, growing demand for products enriched with natural compounds that support human health has been observed. Black rice, its by-products, and residues are known to have in their composition a large amount of these compounds with biological potential, mainly anthocyanins. These compounds have reported effects on anti-obesity, antidiabetic, antimicrobial, anticancer, neuroprotective, and cardiovascular disease. Therefore, the extract from black rice or its by-products have great potential for application as ingredients in functional foods, supplements, or pharmacological formulations. This overview summarizes the methods employed for the extraction of anthocyanins from both black rice and its by-products. In addition, trends in applications of these extracts are also evaluated regarding their biological potential. Commonly, the extraction methods used to recover anthocyanins are conventional (maceration) and some emerging technologies (Ultrasound-Assisted Extraction - UAE, and Microwave-Assisted Extraction - MAE). Anthocyanin-rich extracts from black rice have presented a biological potential for human health. In vitro and in vivo assays (in mice) showed these compounds mainly with anti-cancer properties. However, more clinical trials are still needed to prove these potential biological effects. Extracts from black rice and its by-products have great potential in applying functional products with beneficial characteristics to humans and reducing agro-industrial residues.

In Rio Grande do Sul, the main rice producer State in Brazil, low temperatures can occur during germination and seedling establishment, and in some cases, during the reproductive stage. When low temperatures occur in the early developmental stages cause delay in germination, resulting in a non-homogeneous growing. In reproductive stage, low temperatures cause spikelet sterility, directly interfering with plant yield. Researchers have shown that some phenolic compounds such as proanthocyanidins and anthocyanin are associated with low temperature tolerance in plants due to their antioxidant capacity. The red and black color in the seeds of some rice genotypes is conferred by the phenolic compounds proanthocyanidins and anthocyanin, respectively. Therefore, tis study aimed to verify whether rice genotypes with red or black seeds are more tolerant to low temperatures during germination. In this study, five rice genotypes were tested, two present seeds without pigmentation and with contrasting response to low temperature tolerance (BRS Bojuru - tolerant and BRS Pampeira - sensitive), two genotypes with red seeds (BRS 902, SCS 119 Rubi) and one black seed genotype (SCS 120 Ônix). As expected, the genotypes with pigmented seeds had a greater total phenolic compounds content. However, under low temperature conditions, the genotypes with pigmented seed showed a similar response to the sensitive genotype. Therefore, the presence of proanthocyanidins and anthocyanin in the seed of the studied genotypes does not provide tolerance to low temperatures during germination.(AU)
No Rio Grande do Sul, principal estado produtor de arroz do Brasil, baixas temperaturas podem ocorrer durante a germinação e estabelecimento de plântulas, e em alguns casos, durante o estádio reprodutivo. Quando baixas temperaturas ocorrem no estádio inicial de desenvolvimento causam atraso da germinação, resultando em um crescimento não homogêneo. No estádio reprodutivo, baixas temperaturas podem ocasionar esterilidade das espiguetas, interferindo diretamente na produtividade da planta. Pesquisas têm mostrado que alguns compostos fenólicos como as proantocianidinas e antocianinas estão associadas com tolerância a baixa temperatura em plantas devido sua capacidade antioxidante. A coloração vermelha e preta nas sementes de alguns genótipos de arroz é conferida pelos compostos fenólicos proantocianidinas e antocianinas, respectivamente. Portanto, o objetivo desse estudo foi verificar se os genótipos de arroz com sementes vermelhas ou pretas são mais tolerantes a baixas temperaturas durante a germinação. Neste trabalho, cinco genótipos foram testados, dois com sementes sem pigmentação e com resposta contrastante para tolerância a baixa temperatura (BRS Bojuru - tolerante e BRS Pampeira - sensível), dois genótipos com sementes vermelhas (BRS 902, SCS 119 Rubi) e um genótipo com sementes pretas (SCS 120 Ônix). Como esperado, os genótipos com sementes pigmentadas têm maior conteúdo de compostos fenólicos totais. Entretanto, sob condições de baixa temperatura, os genótipos com semente pigmentada mostraram resposta similar ao genótipo sensível. Dessa forma, a presença de proantocianidinas e antocianinas nas sementes dos genótipos estudados não confere tolerância a baixas temperaturas durante a germinação.(AU)

This study was conducted to evaluate the protective role of extracted natural antioxidants from black rice and their effect on kidney failure and renal cirrhosis caused by ethanol-induced toxicity. Antioxidant activity in terms of total phenol content, flavonoid compounds and anthocyanin, as well as antioxidant capacity, was determined in an extract of black rice. The findings noted that the black rice extract contained high amounts of antioxidant activity and capacity. Total phenolic compounds from black rice extract were fractionated using HPLC and the results showed that ferulic, sinapic, ascorbic, salicylic and coumaric acids were the highest in the extract. Biological experiments were performed on male albino adult rats (40 animals, 10 rats for each group), divided into four groups. After five weeks, kidney functions and protein fractions were assessed. In addition, superoxide dismutase (SOD), glutathione (GSH) and malondialdehyde (MDA) enzyme activities were determined in all groups. The results found that kidney function, total protein, albumin and globulin were affected by renal dysfunction and renal fibrosis in the positive control (PC), whereas groups 3 and 4 noted an improvement in renal function nearly or equal to the healthy rats which were fed on a basal diet. Furthermore, the PC group showed significantly decreased levels of enzymatic antioxidants, namely SOD and GSH with a concomitant elevated MDA level compared with those in the negative rats fed on a basal diet. Groups 3 and 4 also reported improvements in enzyme activity. These results were further supported by histopathological findings which revealed a curative effect in groups 3 and 4, which avoided renal dysfunction and renal fibrosis from ethanol-induced toxicity. From the results, it can be said that the black rice extract with the highest amounts of antioxidants led to improvements in all parameters, especially kidney function, total protein, albumin, and globulin, in addition to enzyme activity. Therefore, black rice can be recommended as a benefit to general health.
Este estudo foi conduzido para avaliar o papel protetor dos antioxidantes naturais extraídos do arroz-preto e seu efeito na insuficiência renal e cirrose renal causada pela toxicidade induzida pelo etanol. A atividade antioxidante em termos de teor de fenóis totais, compostos flavonoides e antocianinas, bem como a capacidade antioxidante, foi determinada em um extrato de arroz-preto. As descobertas observaram que o extrato de arroz-preto continha grandes quantidades de atividade e capacidade antioxidante. Os compostos fenólicos totais do extrato de arroz-preto foram fracionados por HPLC e os resultados mostraram que os ácidos ferúlico, sinápico, ascórbico, salicílico e cumárico foram os mais elevados no extrato. Experimentos biológicos foram realizados em ratos adultos albinos machos (40 animais, 10 ratos para cada grupo), divididos em quatro grupos. Após cinco semanas, as funções renais e frações proteicas foram avaliadas. Além disso, as atividades das enzimas superóxido dismutase (SOD), glutationa (GSH) e malondialdeído (MDA) foram determinadas em todos os grupos. Os resultados mostraram que a função renal, proteína total, albumina e globulina foram afetadas por disfunção renal e fibrose renal no controle positivo (CP), enquanto os grupos 3 e 4 observaram uma melhora na função renal quase ou igual aos ratos saudáveis ââque foram alimentados em uma dieta básica. Além disso, o grupo PC apresentou níveis significativamente diminuídos de antioxidantes enzimáticos, ou seja, SOD e GSH com um nível concomitante de MDA elevado em comparação com aqueles nos ratos negativos alimentados com uma dieta basal. Os grupos 3 e 4 também relataram melhorias na atividade enzimática. Esses resultados foram ainda apoiados por achados histopatológicos que revelaram um efeito curativo nos grupos 3 e 4, que evitou a disfunção renal e fibrose renal por toxicidade induzida pelo etanol. A partir dos resultados, pode-se dizer que o extrato de arroz-preto com maior quantidade de antioxidantes levou a melhorias em todos os parâmetros, principalmente função renal, proteína total, albumina e globulina, além da atividade enzimática. Portanto, o arroz-preto pode ser recomendado como um benefício para a saúde geral.

A robust and sustainable sensor for the detection of Al(III) ions in water was developed by immobilization of anthocyanin (AN) from black rice in a film formed by carboxymethylcellulose (CMC) and starch. Characterization of the films was performed using solubility, thickness, FTIR, and mechanical analysis. The film exhibited an irreversible color change from red to purple in response to the presence of Al(III). The best colorimetric response of the sensor was observed at pH 4.5 and a time of 60â¯min, achieving the detection of 3â¯mgâ¯L-1 of Al(III). For concentrations higher than 5â¯mgâ¯L-1, the sensor response time decreased to 20â¯min. The minimum Al(III) concentration detected with the naked eye was lower than the maximum permissible concentrations in aqueous effluents according to different legislations, indicating the potential of this study to develop sensors for the detection of Al(III).
ABSTRACT
The aim of this work was to study the phenolic compounds identified in rice, their antioxidant activity and their potential beneficial effects on health. In vitro and in vivo studies evaluating the rice grains with different pericarp colour (light brown, red and black) showed potential beneficial effects on health related to the polyphenol content of the grain, such as reduction of oxidative stress, aid in the prevention of cancer, cardiovascular diseases and complications of diabetes, among others.
