ABSTRACT

Introduction: Evaluations of interspecific hybrids are limited, as classical genebank accession descriptors are semi-subjective, have qualitative traits and show complications when evaluating intermediate accessions. However, descriptors can be quantified using recognized phenomic traits. This digitalization can identify phenomic traits which correspond to the percentage of parental descriptors remaining expressed/visible/measurable in the particular interspecific hybrid. In this study, a line of P. vulgaris, P. acutifolius and P. parvifolius accessions and their crosses were sown in the mesh house according to CIAT seed regeneration procedures. Methodology: Three accessions and one derived breeding line originating from their interspecific crosses were characterized and classified by selected phenomic descriptors using multivariate and machine learning techniques. The phenomic proportions of the interspecific hybrid (line INB 47) with respect to its three parent accessions were determined using a random forest and a respective confusion matrix. Results: The seed and pod morphometric traits, physiological behavior and yield performance were evaluated. In the classification of the accession, the phenomic descriptors with highest prediction force were Fm', Fo', Fs', LTD, Chl, seed area, seed height, seed Major, seed MinFeret, seed Minor, pod AR, pod Feret, pod round, pod solidity, pod area, pod major, pod seed weight and pod weight. Physiological traits measured in the interspecific hybrid present 2.2% similarity with the P. acutifolius and 1% with the P. parvifolius accessions. In addition, in seed morphometric characteristics, the hybrid showed 4.5% similarity with the P. acutifolius accession. Conclusions: Here we were able to determine the phenomic proportions of individual parents in their interspecific hybrid accession. After some careful generalization the methodology can be used to: i) verify trait-of-interest transfer from P. acutifolius and P. parvifolius accessions into their hybrids; ii) confirm selected traits as "phenomic markers" which would allow conserving desired physiological traits of exotic parental accessions, without losing key seed characteristics from elite common bean accessions; and iii) propose a quantitative tool that helps genebank curators and breeders to make better-informed decisions based on quantitative analysis.

ABSTRACT

Phytophthora andina is a heterothallic plant pathogen of Andean solanaceous hosts and is an interspecific hybrid of P. infestans and an unknown Phytophthora species. The objective of this study was to estimate the nuclear DNA content of isolates in three clonal lineages of P. andina relative to P. infestans Twelve isolates of P. andina and six isolates of P. infestans were measured for nuclear DNA content by propidium iodide-stained flow cytometry. We found that the DNA content of P. andina was similar but slightly smaller, on average, than that of our sample of P. infestans isolates. This is consistent with P. andina being a homoploid hybrid rather than allopolyploid hybrid. Nuclear DNA content was more variable among a smaller sample of P. infestans isolates, including a putative triploid isolate from Mexico, but small differences in nuclear DNA content were also observed among P. andina isolates. Both species appear to be able to tolerate significant variation in genome size.

ABSTRACT

In the present study, we assessed for the first time the changes in the antioxidant capacity, total phenolic content, and polyphenolic composition of interspecific hybrid palm oil extracted from Elaeis oleifera × Elaeis guineensis (O × G, Coari × La Mé cultivar) during the fruit ripening process 18, 20, 22, and 24 weeks after anthesis. A progressive decrease (p < 0.05) of phenolic content occurred during fruit development together with marked changes in polyphenol profiles. Significant negative correlations were established between antioxidant activity measured by TEAC (R = -0.954; p < 0.05) and ORAC (R = -0.745; p < 0.05) and the fruit ripening stage, while a positive correlation between total phenolic content was found using either the TEAC assay or the ORAC assay. The highest DPPH radical scavenging activity was also obtained with oils extracted at 18 WAA. These results highlight that O × G fruits of early ripeness represent a better source of phenolic compounds and may provide extracts with higher antioxidant activities when hybrid palm oil is aimed to be used as a functional ingredient for the development of food or food products with antioxidant properties.

Pseudoplatystoma corruscans (Spix and Agassiz, 1829) and Pseudoplatystoma reticulatum (Eingenmann and Eigenmann, 1889) are large migratory catfishes of high biological importance and great commercial value in South America. Because fertile crossbreeds can be artificially produced in hatcheries, a high genetic proximity between these two Pimelodidae species is conceivable. Possible escape of crossbred specimens from pisciculture stations is a serious environmental concern. Despite their importance, knowledge of P. corruscans and P. reticulatum biology, ecology, population diversity and genetics is limited. In the present work, the genetic divergence between P. corruscans and P. reticulatum populations from the Paraná River Basin was analyzed on the basis of polymorphisms in ISSR fragments and in the hypervariable sequence of the mitochondrial DNA (mtDNA) control region. Estimates of intraspecific haplotype (h > 0.5) and nucleotide diversities (π < 0.01) indicate that P. corruscans and P. reticulatum have survived a historical population decline, followed by a demographic expansion. The interspecific polymorphisms within the mtDNA control region and ISSR fragments were suitable as diagnostic molecular markers and could be used to discriminate the two species. A unique Pseudoplatystoma specimen, captured in the Upper Paraná River Floodplain, was identified by these DNA diagnostic markers as a hybrid P. reticulatum x P. corruscans, which possibly escaped from pisciculture. The integrity of the natural population of P. corruscans in the Upper Paraná River is at risk of genetic introgression or homogenization due to the presence of hybrids and the transposition of P. reticulatum upstream through the Canal da Piracema at Itaipu Dam. ABSTRACT

Microsporogenesis in an interspecific Brachiaria hybrid, grown in the field under natural environmental conditions in Brazilian savannas, was analyzed in three distinct years of collection. Several types of meiotic abnormalities were recorded during those three years, but varied in type and frequency depending on the year. The average temperature and rainfall 15 days before collection was unusually high in those years. The percentage of abnormal meiocytes recorded was 62% in 2001, 73% in 2004, and 77% in 2005. The abnormalities observed during microsporogenesis compromised pollen viability by generating unbalanced gametes or affecting nucleolus organization. The environmental conditions under which the hybrid was growing could have affected the genetic control of meiosis. More detailed studies, under controlled conditions, are necessary to better understand the effects of environmental factors on Brachiaria microsporogenesis hybrids.

ABSTRACT

The genetic monitoring of interspecific hybrids involves the application of methodologies able to provide an easy and indubitable genetic characterization of both parental and hybrid individuals. In the present work, cytogenetic techniques were used to identify a hybrid lineage of "Piaupara" in order to caracterize them in relation to the parental species, Leporinus macrocephalus (piauçu) and L. elongatus (piapara). The cytogenetic analysis revealed that L. macrocephalus presented 2n = 54 chromosomes and a nucleolar organizer regions (NOR) at the telomere of the long arm of the submetacentric chromosome pair 2. Analysis of constitutive heterochromatin (C-banding) revealed a conspicuous block at the pericentromeric region on the long arm of a submetacentric chromosome pair. L. elongatus presented the same diploid number, 2n = 54, and a karyotypic formula similar to that of L. macrocephalus. The NORs were also at the telomere of the long arm of the submetacentric pair 2, which was morphologically different from that of L. macrocephalus. Heterochromatic blocks were observed at both telomeres of a submetacentric chromosome pair. The hybrid "Piaupara" presented the same diploid number (2n = 54) and karyotypic formula as the parental species and there were no visible differences between parental and hybrid individuals. Differently from the Giemsa staining, NOR- and C-banding analysis showed marked differences which allowed the identification of the hybrids by the different morphology and/or size of the chromosomes carrying the NORs and patterns of heterochromatin distribution in their chromosomes. Such genetic studies are important for fish culture since they can provide tools for monitoring natural and artificial hybridization. They are also useful in biological conservation programmes and in the proper management of natural and reared fish stocks.

ABSTRACT

Microsporogenesis was analyzed in an interspecific hybrid between an artificially tetraploidized sexual accession of Brachiaria ruziziensis (2n=4x=36) and a natural apomictic tetraploid accession of B. decumbens. Syncytes involving a large number of cells were recorded in 15.4 percent of meiocytes. Meiosis progressed normally in syncytes during prophase I; in metaphase I, however, several nuclei were found fusioned, showing chromosome stickiness and several chromosome fragments. Meiosis was arrested in metaphase I and pycnotic nuclei and micronuclei were formed. Abnormal cytokinesis fractionated the syncyte into abnormal meiotic products that were covered by the pollen wall. Meiocytes in leptotene were recorded in all the slides prepared for both meiotic divisions, and abnormal "pollen grains" with well-developed pollen wall but containing leptotene nuclei were recorded in 9.18 percent of grains analyzed. These findings suggested that the meiocytes received the signal to enter meiosis but lacked the signal to proceed beyond leptotene. Despite the absence of the meiotic process, such cells were covered by pollen grain wall. Total pollen sterility resulted from these abnormalities combined with still others observed among meiocytes.


A microsporogênese de um híbrido interespecífico entre um acesso sexual tetraploidizado artificialmente de Brachiaria ruziziensis (2n=4x=36) e um acesso apomítico tetraplóide natural de B. decumbens (2n=4x=36) foi analisada. Sincícios envolvendo um grande número de células foram encontrados em 15,40 por cento dos meiócitos. A meiose progrediu normalmente nos sincícios durante a prófase I; em metáfase I, todavia, muitos núcleos fundiram-se, mostrando ainda aderências cromossômicas e inúmeros fragmentos. O processo meiótico foi interrompido na metáfase I, quando a cromatina formou núcleos picnóticos. Citocineses anormais fracionaram os sincícios em produtos meióticos anômalos que foram recobertos pela parede do grão de pólen. Meiócitos em leptóteno também foram observados durante todo o processo meiótico e grãos de pólen anormais com parede de pólen bem desenvolvida, mas contendo núcleos leptotênicos, foram observados em 9,18 por cento dos grãos de pólen analisados. Os resultados sugerem que os meiócitos receberam o sinal para entrar em meiose, mas não receberam o sinal para prosseguir além do leptóteno. Apesar da ausência de processo meiótico completo, os meiócitos foram cobertos pela parede do grão de pólen. Estas anormalidades, combinadas com outras causadas pela poliploidia, resultaram em total esterilidade de pólen.

ABSTRACT

Twenty three inbred HEXAPLOID wheat lines were evaluated. They were originated by selections made in populations submited to gamma radiation (27.5 krad), in the F4 generation, from the interespecific hybrid between BH-1146 (Triticum aestivum L.) and Anhinga "S" x Winged "S" (Triticum durum L.) and the cultivars BH-1146 and Yavaros "S" (T. durum L.). Nine trials were carried out with irrigation and in upland conditions. Several agronomic characteristics were assessed. Under laboratory conditions the genotypes were evaluated with respect to Al toxicity using nutrient solutions. Semidwarf lines 11 and 19, with moderate lodging resistance, medium cycle from emergence to flowering, tolerance to aluminum toxicity, were the most productive. Seven lines showed resistance to the causal agent of mildew, but all genotypes were susceptible to the causal agents of leaf spots. Some of the genotypes could be used in cross breeding as genetic sources for short plant, long head, large number of spikelets per head, large number of grains per head and per spikelet and for heavy grains. BH-1146 and seven from the 23 lines were considered the most tolerant to aluminum toxicity, presenting good regrowth after treatment in nutrient solutions with 10 nig/liter of Al+3.


Foram comparadas entre si e com dois controles vinte e três linhagens hexaplóides originárias de seleções realizadas em população submetida à radiação gama (27,5 krad), em geração F4, do cruzamento interespecífico entre BH-1146 (Triticum aestivum L.) e Anhinga "S" x Winged "S" (T. durum L.). Em nove ensaios em condições de irrigação e de sequeiro, analisou-se uma série de características agronômicas. Em laboratório efetuaram-se estudos da tolerância ao alumínio. As linhagens 11 e 19 de porte médio, com moderada resistência ao acamamento, tolerantes à toxicidade de alumínio destacaram-se quanto à produção de grãos. Sete linhagens apresentaram moderada resistência ao agente causai do oídio. Todos os genótipos foram suscetíveis aos agentes causais das manchas foliares. Alguns dos genótipos poderiam ser utilizados em programas de cruzamento como fontes genéticas para plantas de porte baixo, de espiga comprida, de grande numero de espiguetas por espiga, maior número de grãos por espiga, e maior número de grãos por espigueta e de grãos mais pesados. BH-1146 e sete das linhagens foram considerados os genótipos mais tolerantes à toxicidade de alumínio por por exibirem crescimento radicular após serem cultivados em soluções nutritivas contendo 10 mg/litro de Al+3.

Under laboratory conditions the genotypes were evaluated with respect to Al toxicity using nutrient solutions. Semidwarf lines 11 and 19, with moderate lodging resistance, medium cycle from emergence to flowering, tolerance to aluminum toxicity, were the most productive. Seven lines showed resistance to the causal agent of mildew, but all genotypes were susceptible to the causal agents of leaf spots. Some of the genotypes could be used in cross breeding as genetic sources for short plant, long head, large number of spikelets per head, large number of grains per head and per spikelet and for heavy grains. BH-1146 and seven from the 23 lines were considered the most tolerant to aluminum toxicity, presenting good regrowth after treatment in nutrient solutions with 10 nig/liter of Al+3.


Foram comparadas entre si e com dois controles vinte e três linhagens hexaplóides originárias de seleções realizadas em população submetida à radiação gama (27,5 krad), em geração F4, do cruzamento interespecífico entre BH-1146 (Triticum aestivum L.) e Anhinga "S" x Winged "S" (T. durum L.). Em nove ensaios em condições de irrigação e de sequeiro, analisou-se uma série de características agronômicas. Em laboratório efetuaram-se estudos da tolerância ao alumínio. As linhagens 11 e 19 de porte médio, com moderada resistência ao acamamento, tolerantes à toxicidade de alumínio destacaram-se quanto à produção de grãos. Sete linhagens apresentaram moderada resistência ao agente causai do oídio. Todos os genótipos foram suscetíveis aos agentes causais das manchas foliares. Alguns dos genótipos poderiam ser utilizados em programas de cruzamento como fontes genéticas para plantas de porte baixo, de espiga comprida, de grande numero de espiguetas por espiga, maior número de grãos por espiga, e maior número de grãos por espigueta e de grãos mais pesados. BH-1146 e sete das linhagens foram considerados os genótipos mais tolerantes à toxicidade de alumínio por por exibirem crescimento radicular após serem cultivados em soluções nutritivas contendo 10 mg/litro de Al+3.
