ABSTRACT

Rats and mice are the most used experimental models in research. Globally, animal houses are subjected to parasite outbreaks. In Brazil, the parasitological profile is used to reflect the sanitary condition of laboratory animals and must be monitored frequently. The present study developed an integrative review of scientific studies on the parasitological profile of rats and mice in animal facilities in Brazil. It identified the most prevalent parasite species reported in animal facilities from different geographic regions of the country, as well as factors contributing to the perpetuation of these parasites, and proposed measures to help prevent such infections. Based on the guiding question "which endoparasites and ectoparasites have already been identified in rat and mouse colonies in animal facilities in Brazil?" and considering inclusion and exclusion criteria, 28 studies published between 1974 and 2021 were selected in four scientific bases. These studies covered facilities in 12 Brazilian States, describing 16 genera of parasites in mice and 18 in rats. Most of the facilities in the selected studies were of the conventional type and with few sanitary barriers, and these conditions may favor the recurrence of parasites. Efforts are, therefore, necessary for institutions to have animal facilities in accordance with the legislation and practice optimal methodologies. The measures proposed in this article can contribute to change the panorama of parasites in the national animal facilities, aiming to safeguard the quality of scientific data and animal welfare.


Ratos e camundongos são os modelos experimentais mais utilizados em pesquisa. Globalmente, as instalações de animais estão sujeitas a surtos de parasitas. No Brasil, o perfil parasitológico é utilizado para refletir a condição sanitária dos animais de laboratório e deve ser monitorado frequentemente. O presente estudo teve como objetivo desenvolver uma revisão integrativa de estudos científicos sobre o perfil parasitológico de ratos e camundongos em biotérios no Brasil. A revisão identifica as espécies parasitárias mais prevalentes relatadas em biotérios de diferentes regiões geográficas do país, bem como os fatores que contribuem para a perpetuação desses parasitas, e propõe medidas para ajudar a prevenir tais infecções. Com base na questão norteadora "quais endoparasitas e ectoparasitas já foram identificados em colônias de ratos e camundongos em biotérios no Brasil?" e considerando os critérios de inclusão e exclusão, foram selecionados 28 estudos publicados entre 1974 e 2021 em quatro bases científicas. Esses estudos abrangeram instalações em 12 estados brasileiros, descrevendo 16 gêneros de parasitas em camundongos e 18 em ratos. A maioria das instalações dos estudos selecionados era do tipo convencional e com poucas barreiras sanitárias, e essas condições podem favorecer a recorrência de parasitas. Portanto, esforços são necessários para que as instituições tenham biotérios de acordo com a legislação e pratiquem metodologias otimizadas. As medidas propostas neste artigo podem contribuir para mudar o panorama das parasitoses nos biotérios nacionais, visando salvaguardar a qualidade dos dados científicos e o bem-estar animal.

Inbred species are useful resources for a variety of biomedical research applications. To create isogenic zebrafish, it is feasible to stop meiosis II (repeatedly) or mitosis (two times) in a haploid embryo by applying pressure or by delivering a heat shock, respectively. In this study, to improve the repeatability, we suggest a less complicated approach based on sperm ultraviolet-C (UV-C) exposure for a shorter period followed by heat shock at various temperatures, eliminating the use of pressure in meiotic therapy since heat shock is more accessible to laboratories. In this study, the survivability rates of meiotic (Mei) and mitotic (Mit) gynogenesis offspring produced by various combinations of irradiation (28.5, 105, and 210 mJ/cm2) and temperature (Mei: 40.40°C, 40.60°C, or 40.90°C; Mt: 41.40°C, 41.90°C, or 42.45°C) were compared with diploid (C) and haploid (H) controls. Our findings demonstrated that 40.60°C and 41.90°C were the most suitable temperatures to produce meiotic and mitotic gynogenesis, respectively, whereas 28.5 mJ/cm2 was more successful in ensuring haploid embryos. As a result, we deduced that meiotic gynogenesis produces more viable offspring than the mitotic approach and requires a lower temperature to maintain the second polar body.

Hard ticks pose a threat to animal and human health. Active life stages need to feed on a vertebrate host in order to complete their life cycle. To study processes such as tick-pathogen interactions or drug efficacy and pharmacokinetics, it is necessary to maintain tick colonies under defined laboratory conditions, typically using laboratory animals. The aim of this study was to test a membrane-based artificial feeding system (AFS) applicable for Amblyomma ticks using Amblyomma tonelliae as a biological model. Adult ticks from a laboratory colony were fed in a membrane-based AFS. For comparison, other A. tonelliae adults were fed on calf and rabbit. The proportions of attached (AFS: 76%; calf/rabbit: 100%) and engorged females (AFS: 47.4%; calf/rabbit: 100%) in the AFS were significantly lower compared to animal-based feeding (p = 0.0265). The engorgement weight of in vitro fed ticks (x¯ = 658 mg; SD ± 259.80) did not significantly differ from that of ticks fed on animals (p = 0.3272, respectively 0.0947). The proportion of females that oviposited was 100% for all three feeding methods. However, the incubation period of eggs (x¯ = 54 days; SD ± 7) was longer in the AFS compared to conventional animal-based feeding (p = 0.0014); x¯ = 45 days; SD ± 2 in the rabbit and (p = 0.0144). x¯ = 48 days; SD ± 2 in the calf). Egg cluster hatching (x¯ = 41%; SD ± 44.82) was lower in the AFS than in the other feeding methods (rabbit: x¯ = 74%; SD ± 20; p = 0.0529; calf: x¯ = 81%; SD ± 22; p = 0.0256). Although the attachment, development, and the hatching of AFS ticks were below those from animal-based feeding, the method may be useful in future experiments. Nevertheless, further experiments with a higher number of tick specimens (including immature life stages) and different attractant stimuli are required to confirm the preliminary results of this study and to evaluate the applicability of AFS for Amblyomma ticks as an alternative to animal-based feeding methods.

ABSTRACT

Animal research is considered a key element in advance of biomedical science. Although its use is controversial and raises ethical challenges, the contribution of animal models in medicine is essential for understanding the physiopathology and novel treatment alternatives for several animal and human diseases. Current pandemics' pathology, such as the 2019 Coronavirus disease, has been studied in primate, rodent, and porcine models to recognize infection routes and develop therapeutic protocols. Worldwide issues such as diabetes, obesity, neurological disorders, pain, rehabilitation medicine, and surgical techniques require studying the process in different animal species before testing them on humans. Due to their relevance, this article aims to discuss the importance of animal models in diverse lines of biomedical research by analyzing the contributions of the various species utilized in science over the past five years about key topics concerning human and animal health.

ABSTRACT

Every year several animal species are used in evaluation tests and quality control of biological products. Meeting the requirements of regulatory institutions and the quest to promote animal welfare is part of the challenges faced by quality control laboratories.Each species used in tests needs different raising and management. To promote the best habitat and ensure animal welfare, it is necessary to implement a welfare animals environmental refinement program, focused on the social, and physical environment, composed of sensory (auditory, visual, olfactory, and tactile) and nutritional stimuli. Such measures favor the individual to manifest their natural characteristics within the laboratory, reducing stress and promoting comfort to the animal. Therefore, the implementation of environmental enrichment must consider the biology of the animal, the space available in the cage, and the material availability. This paper is focused on housing changes, maintenance of social interaction, and implementation of environmental enrichment items that have been shown to positively affect the welfare of laboratory animals.


Anualmente diversas espécies animais são utilizadas em testes de avaliação e controle de qualidade de produtos biológicos. Atender as exigências dos órgãos regulamentares e a busca pela promoção do bem estar animal fazem parte dos desafios de um laboratório de controle de qualidade. Cada espécie utilizada em testes exige uma necessidade diferente de criação e manejo. Para promover o melhor ambiente e garantir o bem-estar animal faz-se necessário a implementação de um programa de refinamento ambiental bem elaborado, focado no ambiente social, físico, e composto por estímulos sensoriais (auditivos, visuais, olfativos e táteis) e nutricionais. O uso do enriquecimento ambiental favorece que o indivíduo manifeste suas características naturais dentro do laboratório, diminuindo o estresse e promovendo conforto ao animal. Para isso, a implementação do enriquecimento ambiental deve considerar a biologia do animal, o espaço disponível na gaiola e a disponibilidade do material. Este trabalho concentrou-se nas alterações de habitação, manutenção do convívio social e implementação de itens de enriquecimento ambiental que demonstraram afetar positivamente o bem-estar dos animais de laboratório.

Health monitoring programs in animals used as experimental models are essential, since only disease-free subjects are considered suitable for research purposes. In laboratory-kept animals, hemoplasmas have been described as an important confounding variable. Different hemoplasma species have been detected infecting non-human primates (NHP) from Brazil. However, the occurrence of hemoplasma species in laboratory-kept NHP in Brazil has not-yet been assessed. Accordingly, this study aimed (i) to screen laboratory-kept rhesus monkeys for hemoplasmas, (ii) to verify if any of the hemoplasma-positive animals demonstrate hematological abnormalities, and (iii) to assess the genotype diversity of hemoplasma species in NHP from Brazil. Five out of eight (62.5%; 95% CI: 3.05-8.63) rhesus monkeys tested positive for hemotropic Mycoplasma spp. by PCR. Sequencing, phylogenetic, distance, and genotype diversity analyses of partial 16S rRNA gene demonstrate that rhesus monkeys were infected by 'Candidatus Mycoplasma haematomacacae' (formerly 'Candidatus Mycoplasma haemomacaque'). Assessments of partial 16S rRNA diversity of hemoplasma species in NHP suggest that at least four genetically diverse groups may occur in Brazil. Although no hematological abnormalities were demonstrated in rhesus monkeys evaluated herein, future studies are needed to elucidate the influence of 'Ca. M. haematomacacae' as a confounding variable on research studies.

ABSTRACT

The Mexican axolotl (Ambystoma mexicanum) is one of the most important models in contemporary regeneration research and regenerative medicine. This is the result of the long history of the species as an experimental and laboratory bred animal. One of many research questions investigated in the axolotl is regeneration. The species' astonishing ability to regenerate tissues and entire body parts already became apparent shortly after the first 34 living axolotls had been brought from Mexico to Europe in 1864. In the context of their unclear status as larvae or adults and the mysterious transformation of some animals into an adult form, the Paris zoologist Auguste Duméril cut off the gills of several individuals in an attempt to artificially induce the metamorphosis. This produced the first reports on the animals' regenerative powers and led to sporadic but continuous investigations. But it remained just one of the many phenomena studied in axolotls. Only at the beginning of the 20th century, regeneration became a more prominent aspect in the experimental investigations of axolotls. In experimental embryology, regeneration in axolotls was used in three different ways: it was studied as a phenomenon in its own right: more importantly, it served as a macroscopic model for normal development and, together with other techniques like grafting, became a technical object in the experimental systems of embryologists. In my paper, I will look into how the axolotl became an experimental animal in regeneration research, the role of practices and infrastructures in this process and the ways in which regeneration in the axolotl oscillated between epistemic thing and technical object.

Abstract Background: There are divergences in the literature regarding the experimental model (Wistar-WIS or Wistar Kyoto-WKY) to be used as a Spontaneously Hypertensive Rat (SHR) control. The characterization of these models in terms of cardiovascular parameters provides researchers with important tools at the time of selection and application in scientific research. Objective: The aim of this study was to evaluate the use of WIS and WKY as a Spontaneously Hypertensive Rat (SHR) control by assessing the long-term behavior of blood pressure and cardiac structure and function in these strains. Methods: To this end, WIS, WKY, and SHR underwent longitudinal experiments. Blood pressure and body mass were measured every two weeks from the 8th to the 72nd. Echocardiographic analysis was performed in all groups with 16, 48, and 72 weeks of life. After having applied the normality test, the Two-Way ANOVA of repeated measures followed by the Tukey post hoc test was used. A significance level of 5% was established. Results: The WIS group showed higher body mass (p<0.05), while the WKY and SHR presented higher body mass variation over time (p<0.05). SHR exhibited increased values of systolic, diastolic, and mean blood pressure when compared to WKY and WIS, whereas the WKY generally showed higher values than WIS (p<0.05). Regarding the cardiac function, SHR showed reduced values, while the WKY presented an early decrease when compared to WIS with aging (p<0.05). Conclusion: WIS is a more suitable normotensive control for SHR than WKY in experiments to test blood pressure and cardiac structure and function.

Spontaneous polydactyly has been described in several species, but only one report about it in Swiss mice. The aim of the current study was to report the spontaneous occurrence of pre-axial polydactyly in Swiss mice. Clinical examination showed one extra toe laterally to the first digit, in the plantar region, alopecia in the back, altered face growth anatomy and changed perineal region anatomy. Pre-axial polydactyly in the tibial side, fused metatarsals and Y-shaped free phalanges were evidenced in the radiographic images. Pre-axial polydactyly observed in the plantar region differed from that in reports on albino Swiss mice with post-axial polydactyly (Po/Po+) phenotype featured by one extra toe in the ulnar side of one, or both, front limbs, which is the dominant feature. The observed findings highlight the importance of both clinical examinations and close attention by professionals involved in rodents’ breeding on physical changes resulting from different causes, including the genetic ones, since they reveal mutations and, sometimes, new biomodels.


A polidactilia espontânea foi descrita em várias espécies, mas há apenas um relato em camundongos Swiss. O objetivo do presente estudo foi relatar uma ocorrência de polidactilia pré-axial em camundongos Swiss. O exame clínico revelou um dedo extra lateralmente ao primeiro dedo na região plantar, alopecia dorsal, anatomias facial e da região perineal alteradas. Nas imagens radiográficas foram evidenciados polidactilia pré-axial no lado tibial, metatarsos fundidos e falanges livres em forma de Y. A polidactilia pré-axial observada diferiu do relato existente de polidactilia pós-axial (Po/Po+) caracterizado por um dedo extra no lado ulnar de um ou ambos os membros anteriores e de característica dominante. Os achados observados destacam a importância dos exames clínicos e da atenção cuidadosa dos profissionais envolvidos na criação de roedores sobre as alterações físicas decorrentes de diferentes causas, inclusive as genéticas, em revelar mutações e, às vezes, novos biomodelos.

ABSTRACT

O uso de animais é uma importante etapa no desenvolvimento científico e tecnológico. Porém, a maioria das pessoas desconhece que há conceitos éticos preconizados na ciência de animais de laboratório como Bem-estar Animal e o princípio dos 3Rs (do inglês: substituir, reduzir e aperfeiçoar). Desta forma, a divulgação desse conhecimento é necessária durante a formação educacional do estudante, tendo em vista os alunos como comunicadores científicos na sociedade. Assim, nosso estudo teve como objetivo avaliar o conhecimento de alunos do ensino médio de escolas particulares e públicas da região Centro-Leste do estado de São Paulo, através de formulário online sobre a temática do conhecimento básico sobre a ciência de animais de laboratório, princípios éticos, bem-estar animal, 3Rs e uso de animais em pesquisa científica. Em nosso estudo, 40 alunos responderam o questionário online. Destes 25% não souberam qual a finalidade do uso de animais em pesquisas, 90% nunca visitaram ou estiveram em um laboratório que trabalha com pesquisa biomédica, e 62,5% a maioria, nunca ouviram falar nos conceitos de Bem-estar animal e 3Rs. A maioria dos adolescentes acredita que a principal contribuição do uso de animais de laboratório se limita na elaboração de vacinas, medicamentos e soros. Sendo assim, concluímos que a inserção de conteúdos temáticos, participação dos alunos em projetos de extensão e debates sobre a ciência de animais de laboratório são extremamente necessárias na formação educacional, de modo a disseminar a compreensão sobre o assunto e evitar a propagação de falsos conhecimentos.


The use of animals is an important step in scientific and technological development in biomedical research. Animals kept in suitable conditions for scientific purposes can be considered laboratory animals. However, most people are unaware of ethical concepts used in laboratory animal science such as Animal Welfare and the principle of the 3Rs (replacement, reduction, and refinement). Dissemination of this knowledge is necessary during student education, considering the students as scientific communicators in society. The aim was to evaluate the knowledge of high school students from private and public schools in the East-Center region of São Paulo state. The analysis was performed using an online survey form about the laboratory animals science issue of basic knowledge, ethical principles, animal welfare, 3Rs, and the use of animals in scientific research. Forty students answered the online survey form. Twenty-five percent of adolescents did not know the purpose of using animals in research, 90% had never visited or been to a laboratory that works with biomedical research, and 62.5% had never heard of the concepts of animal welfare and the 3Rs. Most adolescents believe that laboratory animals' contribution is limited to the elaboration of vaccines, medicines, and serums. Therefore, we conclude that the insertion of thematic content, student participation in extension projects, and debates about the science of laboratory animals are extremely necessary for disseminating an understanding of the subject and avoiding the spread of false knowledge.

In digital curation, data are essential for actions, enabling better administrative management and greater efficiency in a sector or activity. In a research animal facility, the collection of management data associated with management systems or control sheets is the basis for ensuring quality information that subsidizes research and generates performance indicators and the needs and challenges to be transposed in the breeding and maintenance of animals. The inconsistency of data or incomplete records compromises the choices adopted in husbandry management practices or the interpretation of research results. For this reason, a survey was carried out about software available on the market that helps laboratory animal facilities with the management regarding software used in non-human primate breeding institutions (NHP) for scientific or conservation purposes. We identified 38 software that supports the handling of lab animals used in biomedical research; most of them with functionalities of environmental control (63.15%), control of animal files/tags (63.15%), and control of reproductive management (60%) and only two software related to the management of nonhuman primates kept under human care in research institutions. Regarding the tools used to manage the breeding of non-human primates in captivity in Brazilian institutions, five of the institutions questioned and reported that none currently uses software for colony management. In conclusion, there are several software for managing laboratory animals, but most of them are geared toward the management of rodent animal facilities. There is a lack of specific software on the market for use in animal facilities managing non-human primate breeding, which indicates the need for software developed to meet the management needs of animal facilities for these facilities.

ABSTRACT

This study describes an outbreak of alopecia in laboratory rats caused by the saprophytic fungi Scopulariopsis brevicaulis in a Brazilian lab animal facility. Initially, lesions were identified only in females during the lactation period, which showed circumscribed areas with alopecia without pruritus extensively at the abdomen and the medial side of the thighs. Later during the course of the infection, skin lesions affected both genders reaching about 10% of the colony. Skin scrapings, hair, and environmental samples from the facility were sent for analysis. The cultures were carried out in Sabouraud agar and over seven days fast-growing colonies varying in color from white to brown were observed. Micromorphology showed mycelium and characteristic ringworms of the fungus S. brevicaulis. There is a similar report of an outbreak of this fungus in Turkey. To date, this is the first report in the literature of the fungus S. brevicaulis causing disease in laboratory rats in Brazil.

ABSTRACT

Zebrafish (Danio rerio) is a tropical fish species widely used in research, worldwide. The development of genetically modified animals and the increasing number of zebrafish breeding facilities due to their emerging use in several research fields, opened room for new ethical challenges for research carried out with this species. It is necessary to raise the scientific community's awareness of the ethical standards and laws in force, on animal research. Thus, the aim of the current study is to describe 10 Rs ethical principles by using zebrafish as model system in research. The classical 3 Rs concerning animal welfare, namely replacement, reduction and refinement; and the added 7 Rs related to scientific (registration, reporting, robustness, reproducibility and relevance) and conduct principles (responsibility, and respect) in zebrafish research are herein presented and critically discussed. The use of these 10 Rs by researchers, institutions and the Animal Ethics Committee is recommended to support regulations, decision-making about and the promotion of zebrafish health and welfare in research.


O peixe-zebra (Danio rerio) é um peixe tropical amplamente usado em pesquisas em todo o mundo. Devido ao seu uso emergente em várias áreas de pesquisa, o desenvolvimento de animais geneticamente modificados e o aumento das instalações de peixe-zebra, novos desafos éticos surgem na pesquisa com o peixe-zebra. Além disso, é necessária a conscientização da comunidade científica sobre as normas éticas e leis vigentes na pesquisa com animais. Assim, o presente estudo teve como objetivo descrever os princípios éticos de 10 Rs usando o peixe-zebra como sistema modelo em pesquisa. Os 3 Rs clássicos relativos ao bem-estar animal (substituição, redução e refinamento) e 7 Rs adicionais relacionados aos princípios científicos (registro, relatório, robustez, reprodutibilidade e relevância) e de conduta (responsabilidade e respeito) na pesquisa do peixe-zebra são apresentados e discutido criticamente. Recomendamos o uso desses 10 Rs pelos pesquisadores, instituições e Comitê de Ética Animal na regulamentação, decisão e promoção da saúde e bem-estar do peixe-zebra em pesquisas.

O objetivo do presente estudo foi relatar uma ocorrência de polidactilia pré-axial em camundongos Swiss. O exame clínico revelou um dedo extra lateralmente ao primeiro dedo na região plantar, alopecia dorsal, anatomias facial e da região perineal alteradas. Nas imagens radiográficas foram evidenciados polidactilia pré-axial no lado tibial, metatarsos fundidos e falanges livres em forma de Y. ABSTRACT: Spontaneous polydactyly has been described in several species, but only one report about it in Swiss mice. The aim of the current study was to report the spontaneous occurrence of pre-axial polydactyly in Swiss mice. Clinical examination showed one extra toe laterally to the first digit, in the plantar region, alopecia in the back, altered face growth anatomy and changed perineal region anatomy. Pre-axial polydactyly in the tibial side, fused metatarsals and Y-shaped free phalanges were evidenced in the radiographic images. ABSTRACT

Evaluating laboratory animals' health and thermostability are fundamental components of all experimental designs. Alterations in either one of these parameters have been shown to trigger physiological changes that can compromise the welfare of the species and the replicability and robustness of the results obtained. Due to the nature and complexity of evaluating and managing the species involved in research protocols, non-invasive tools such as infrared thermography (IRT) have been adopted to quantify these parameters without altering them or inducing stress responses in the animals. IRT technology makes it possible to quantify changes in surface temperatures that are derived from alterations in blood flow that can result from inflammatory, stressful, or pathological processes; changes can be measured in diverse regions, called thermal windows, according to their specific characteristics. The principal body regions that were employed for this purpose in laboratory animals were the orbital zone (regio orbitalis), auricular pavilion (regio auricularis), tail (cauda), and the interscapular area (regio scapularis). However, depending on the species and certain external factors, the sensitivity and specificity of these windows are still subject to controversy due to contradictory results published in the available literature. For these reasons, the objectives of the present review are to discuss the neurophysiological mechanisms involved in vasomotor responses and thermogenesis via BAT in laboratory animals and to evaluate the scientific usefulness of IRT and the thermal windows that are currently used in research involving laboratory animals.

BACKGROUND AND AIM: Phytoestrogens are traditionally used for cardiovascular risks but direct effects on the ischemic heart remain unclear. Plants with phytoestrogens are used for reducing menopausic symptoms and they could also be cardioprotectives. Here we investigated whether maca (Lepidium meyenii) contains isoflavones and prevents cardiac stunning, in comparison to soy isoflavones. EXPERIMENTAL PROCEDURE: Both products were orally and daily administered to rats during 1 week before exposing isolated hearts to ischemia/reperfusion (I/R). Young male (YM), female (YF) and aged female (AgF) rats treated with maca (MACA, 1 g/kg/day) or soy isoflavones (ISOF, 100 mg/kg/day) were compared to acute daidzein (DAZ, 5 mg/kg i.p.) and non-treated rat groups. Isolated ventricles were perfused inside a calorimeter to simultaneously measure contractile and calorimetrical signals before and during I/R. RESULTS AND CONCLUSIONS: Maca has genistein and daidzein. MACA and ISOF improved the post-ischemic contractile recovery (PICR) and muscle economy (P/Ht) in YM and YF hearts, but not in AgF hearts. DAZ improved PICR and P/Ht more in YM than in YF. The mKATP channels blockade reduced both PICR and P/Ht in DAZ-treated YM hearts, without affecting them in ISOF or MACA-treated YM hearts. In MACA treated YF hearts, the simultaneous blockade of NOS and mKATP channels, or the mNCX blockade reduced cardioprotection. Results show that subacute oral treatment with maca or with soy isoflavones was strongly preventive of cardiac ischemic dysfunction, more than the acute administration of a pure isoflavone (daidzein, genistein). Maca induced synergistic and complex mechanisms which prevented mitochondrial calcium overload.

Safety and quality of compound feed for experimental animals in Costa Rica is unknown. Some contaminants, such as Salmonella spp. and mycotoxins, could elicit confounding effects in laboratory animals used for biomedical research. In this study, different batches of extruded animal feed, intended for laboratory rodents in Costa Rica, were analyzed to determine mycotoxin and microbiological contamination (i.e., Salmonella spp., Escherichia coli, total coliform bacteria, and total yeast and molds enumeration). Two methods for Salmonella decontamination (UV light and thermal treatment) were assessed. Only n = 2 of the samples were negative (representing 12.50%) for the 26 mycotoxins tested. Enniatins and fumonisins were among the most frequent toxins found (with n = 4+ hits), but the level of contamination and the type of mycotoxins depended on the supplier. None of the indicator microorganisms, nor Salmonella, were found in any of the tested batches, and no mold contamination, nor Salmonella growth, occurs during storage (i.e., 2-6 months under laboratory conditions). However, mycotoxins, such as enniatins and fumonisins tend to decrease after the fourth month of storage, and Salmonella exhibited a lifespan of 64 days at 17 °C even in the presence of UV light. The D-values for Salmonella were between 65.58 ± 2.95 (65 °C) and 6.21 ± 0.11 (80 °C) min, and the thermal destruction time (z-value) was calculated at 15.62 °C. Results from this study suggest that laboratory rodents may be at risk of contamination from animal feed that could significantly affect the outcomes of biomedical experiments. Thus, improved quality controls and handling protocols for the product are suggested.

BACKGROUND: With the enactment of the Brazilian Law Arouca 11,794/2008 and Decree 6.899/2009, there has been an urgent need for changes in the processes related to animal experimentation in Brazil; in particular, there is a need for improvements in enhancements of the lab animal management. To improve the management capacity of the Lab animal facility of the Carlos Chagas Institute's Laboratory Animals Science (LAS), BioterC software was developed and implemented in 2014 for tracking mouse laboratory colonies. Five years after the implementation of this software, we sought to analyze the information in the database originated from BioterC using the Exploratory Analysis Data methodology (EDA). This article aims to identify animal breeding patterns using a data mining tool (Data Science) with Python programming language. RESULTS: The results show that from September 2014 to June 2019, under the license IACUC number LW- 6/17, 15.106 animals were produced. The C57BL/6, BALB/c and Swiss strains were the most frequently produced strains. The distribution of births due to crosses between these strains showed a median of 6 to 10 animals, depending on the genetic homozygosis and heterozygosis of the animal. The median number of days of mating was 35 days. In the sexing period, the records reported a median of 19 days. A total of 393 requests for animals from internal and external laboratories were registered. It was noted that approximately half of the animals produced to meet the demand for orders were discarded. Of the 15,106 animals, 38% were requested for animal experimentation, 58% were discarded and 4% did not have an outcome recorded in the data. CONCLUSIONS: This volume of data provides an initial view of the information retrieval capabilities contained in BioterC, allowing for unique breeding knowledge by installing laboratory animals.
