ABSTRACT
The present study aimed at isolating and characterizing Salmonella spp. from chicken cuts marketed in Francisco Beltrão, PR, and verify the resistance profile of the isolates against antimicrobials used in human therapy. Samples of chicken cuts (n=40) were purchased from supermarkets and submitted to microbiological analysis for the detection of Salmonella spp. The suspected colonies underwent biochemical testing for the identification of enterobacteria. Four colonies were selected from each sample positive for Salmonella spp., totaling 28 isolates that were tested for antimicrobial sensitivity. Colonies that showed resistance to ceftriaxone were subjected to extended-spectrum beta-lactamases (ESBL). Among the analyzed chicken samples, seven (17.5%) showed biochemical behavior characteristic of Salmonella spp. Among the 28 isolates, seventeen different resistance profiles were found, of which 46.42% (n=13) had a multi-resistance profile, and 21.4% (n=6) of the isolates had a phenotype for ESBL production. The strains of Salmonella spp. isolated from chicken cuts found in this study showed a high level of resistance to antimicrobials of different classes and of last generations, these data serve as a warning, as they put the human treatment of salmonellosis at risk.(AU)
A pesquisa objetivou isolar e caracterizar Salmonella spp., a partir de cortes de frangos comercializados na cidade de Francisco Beltrão - PR, bem como verificar o perfil de resistência dos isolados em relação aos antimicrobianos utilizados na terapêutica humana. Amostras de cortes de frango (n=40) foram adquiridas em supermercados e submetidas à análise microbiológica para detecção de Salmonella spp. As colônias suspeitas foram submetidas a provas bioquímicas para identificação de enterobactérias. Quatro colônias foram selecionadas de cada amostra positiva para Salmonella spp., totalizando 28 isolados, que foram testadas quanto à sensibilidade a antimicrobianos. As colônias que apresentaram resistência à ceftriaxona foram submetidas à pesquisa de betalactamases de espectro estendido (ESBL). Das amostras de frango analisadas, sete (17,5%) apresentaram comportamento bioquímico característico de Salmonella spp. Entre os 28 isolados, foram encontrados 17 perfis diferentes de resistência, tendo 46,42% (n=13) apresentado perfil de multirresistência e 21,4% (n=6) apresentado fenótipo para produção de ESBL. As cepas de Salmonella spp. isoladas de cortes de frango, encontradas neste estudo, apresentaram alto índice de resistência a antimicrobianos de diferentes classes e de últimas gerações. Esses dados servem de alerta, uma vez que coloca em risco o tratamento da salmonelose humana.(AU)

Brazil is the number one exporter of chicken meat, and this industry maintains constant microbiological vigilance. The objective of this study was to characterize the pathogenicity, antimicrobial resistance (AMR) and the profile of biofilm production of Escherchia coli strains isolated from raw refrigerated cuts of chicken meat sold in retail markets of the four largest poultry companies in Brazil. We collected 150 samples of chicken meat, in order to isolate E. coli and performed susceptibility tests (to amoxicillin associated with clavulanic acid, ceftiofur, enrofloxacin, gentamicin, and trimethoprim + sulfamethoxazole). In addition, the disc approximation test to detect extended spectrum beta-lactamases enzymes (ESBLs) producers was performed. E. coli ability to form biofilm was checked using polystyrene microplates. We also searched for ESBLs genes (blaCTY-M2, blaSHV-1, blaTEM-1, blaCTX-M2, blaOXA-1, blaPSE-1 and AmpC) and adhesion genes (sfa/foc, afa/draB, iha, hrla, fimC, tsh, papC, mat, cr1, felA, fimH and papG) in ESBL-E. coli producers and in those E. coli classified as strongly biofilm formers, respectively. The overall percentage of E. coli isolation was 58.66%, with brand A having the highest percentage (70%), followed by brands D, B and C (60, 53.3 and 50%, respectively). The highest resistance profile was observed for beta-lactams (39.5%), followed by sulfonamide associated to trimethoprim (36.9%) and polymyxin (33.4%). Of the isolates obtained, 77% were non-susceptible to at least one antimicrobial. Brand A showed the highest overall percentage of resistance with 95.23%, followed by brands C (80%), B (75%) and D (69.44%). Overall, 73.86% of the isolates were non susceptible to at least one antibiotic and 36.3% were multiresistants. A total of 17.04% of E. coli strains were identified as ESBLs producers and 70.44% were able to form biofilms (moderate-to-strong). The blaTEM-1 gene was the most prevalent (73.33%), followed by blaSHV-1 (46.66%) and blaCMY-2 (6%). Of the 31 strongly biofilm-forming strains, 26 (83.87%), 24 (77.41%) and 20 (64.51%) expressed fimC, papG and crl genes, respectively. Taken together, our results show that Brazilian chicken meat can be contaminated with E. coli that are non-susceptible to multiple antibiotics, able to form biofilm and showing a diverse repertoire of adhesins linked to pathogenicity depending on the brand evaluated.

Brazilian poultry industry generates large amounts of organic waste, such as chicken litter, which is often used in agriculture. Among the bacteria present in organic fertilizer are members of the Enterobacteriaceae family, such as Escherichia coli. The aim of this study was to analyze the profile of virulence factors and antimicrobial resistance of E. coli isolates from avian organic fertilizer. A total of 47 E. coli strains were tested through Polymerase chain reaction to detect virulence genes (hlyF, iss, ompT, iutA and iroN). Fourteen antimicrobials were used to test antimicrobial susceptibility in the strains. Genes characteristic of Avian Pathogenic E. coli (APEC) were reported among the strains, with the hlyF, iss and ompT genes being the most prevalent. The isolates showed high resistance (˃50%) to tetracycline, gentamicin, cefotaxime, nitrofurantoin, trimethoprim-sulfamethoxazole and ampicillin. Multidrug resistance was reported in a great number of strains (>70%). The results showed the presence of APEC cells with virulence genes and antimicrobial resistance after 15 days of the windrowing process in poultry houses, it means this process should be improved to eliminate these cells.(AU)
A indústria avícola brasileira gera grandes quantidades de resíduos orgânicos, como a cama de frango, utilizada frequentemente na agricultura. Entre as bactérias presentes neste fertilizante orgânico estão os membros da família Enterobacteriaceae, entre eles a Escherichia coli. O objetivo deste estudo foi analisar o perfil de fatores de virulência e resistência antimicrobiana de isolados de E. coli provenientes do fertilizante orgânico aviário. Um total de 47 cepas de E. coli foram testadas por meio da reação em cadeia da polimerase para detectar genes de virulência (hlyF, iss, ompT, iutA e iroN). Quatorze antimicrobianos foram utilizados para testar a susceptibilidade antimicrobiana nos isolados. Genes característicos de E. coli Patogênica Aviária (APEC) foram encontrados entre os isolados, sendo os genes hlyF, iss e ompT os mais prevalentes. Os isolados apresentaram alta resistência (˃50%) à tetraciclina, gentamicina, cefotaxima, nitrofurantoína, trimetoprima-sulfametoxazole e ampicilina. Múltipla resistência a drogas antimicrobianas foi encontrada em grande número de isolados (>70%). Os resultados obtidos mostraram a presença de células APEC portando genes de virulência e resistência a antimicrobianos após 15 dias de processo de empilhamento nas granjas, indicando que o processo necessita de um aperfeiçoamento para eliminar estas células.(AU)

O objetivo deste trabalho foi desenvolver e testar a técnica de separação imunomagnética (IMS) na detecção de C. jejuni em produtos de frango. Micropartículas magnéticas ligadas a anticorpos policlonais anti-C. jejuni foram utilizadas para concentrar C. jejuni antes da semeadura em ágar. O protocolo foi comparado com o método convencional. C. jejuni foi recuperado do alimento experimentalmente contaminado por ambos os métodos, entretanto, quando foi usada a IMS, a presença de microrganismos contaminantes nos meios de cultura foi menor. C. jejuni foi isolado de 7% das amostras de alimento naturalmente contaminadas, usando IMS, e de 3% pelo método convencional. C. coli foi isolado de uma amostra pelo método convencional, mas não foi detectado pelo protocolo com IMS. A técnica de IMS pode ser usada para isolamento de C. jejuni de alimentos, oferecendo a vantagem de detectar em amostras o microrganismo cujo isolamento não é obtido por meio do método convencional.(AU)

This microorganism is difficult to isolate and the conventional methods are often inefficient and may lead to erroneous results. This study aimed at developing and testing the technique of immunomagnetic separation (IMS) in the detection of C. jejuni in chicken products. Microparticles magnetically connected anti-C. jejuni polyclonal antibodies were used to concentrate C. jejuni before agar seeding. The protocol was compared with the conventional method. C. jejuni was recovered from experimentally contaminated food for both methods, however, when the IMS was used, the presence of contaminating microorganisms in the means of culture was smaller. C. jejuni was isolated from 7% of samples of food naturally contaminated, using IMS, and 3% by conventional method. C. coli was isolated from a sample by conventional method, but it was not detected by protocol with IMS. The IMS technique can be used for isolation of C. jejuni in food, offering the advantage of detecting the microorganism in samples from which the isolation is not obtained with the use of the conventional method.(AU)

1. The current consumer preference for healthier meat products is associated with less additives in manufacturing (so-called 'clean-label') or the addition of non-meat ingredients with functional properties, recognised as improving specific technological properties in meat products.2. This study evaluated the effect of the addition of alpha-cyclodextrin and wheat fibre to low-fat chicken frankfurters containing 35% mechanically deboned chicken meat on the technological and sensorial properties during refrigerated storage.3. The results showed that the addition of dietary fibres (alpha-cyclodextrin and wheat fibre) in low-fat chicken frankfurters improved emulsion stability, hardness, chewiness and reduced cohesiveness.4. Alpha-cyclodextrin helped the retention of fat globules in the microstructure and affected colour in the sensorial evaluation.5. The use of alpha-cyclodextrin, in combination with wheat fibre, as a new ingredient to substitute fat in emulsified meat products containing mechanically deboned chicken, improved emulsion stability and texture.6. Alpha-cyclodextrin and wheat fibre were effective in contributing to fat reduction without affecting the sensory properties of the product.

During the last years, Brazilian government control programs have detected an increase of Salmonella Heidelberg in poultry slaughterhouses a condition that poses a threat to human health However, the reasons remain unclear. Differences in genetic virulence profiles may be a possible justification. In addition, effective control of Salmonella is related to an efficient epidemiological surveillance system through genotyping techniques. In this context, the aim of this study was the detection of 24 virulence-associated genes in 126 S. Heidelberg isolates. We classified the isolates into 56 different genetic profiles. None of the isolates presented all the virulence genes. The prevalence of these genes was high in all tested samples as the lowest number of genes detected in one isolate was 10/24. The lpfA and csgA (fimbriae), invA and sivH (TTSS), and msgA and tolC (intracellular survival) genes were present in 100% of the isolates analyzed. Genes encoding effector proteins were detected in the majority of SH isolates. No single isolate had the sefA gene. The pefA gene was found in only four isolates. We have also performed a screening of genes associated with iron metabolism: 88.9% of isolates had the iroN geneand 79.4% the sitC gene . Although all the isolates belong to the same serotype, several genotypic profiles were observed. These findings suggest that there is a diversity of S. Heidelberg isolates in poultry products. The fact that a single predominant profile was not found in this study indicates the presence of variable sources of contamination caused by SH. The detection of genetic profiles of Salmonella strains can be used to determine the virulence patterns of SH isolates.

S. Heidelberg é responsável por causar doença grave em humanos, e por apresentar alta resistência aos antimicrobianos, sendo considerada um problema emergente na saúde pública mundial. Objetivou-se determinar a resistência aos antimicrobianos em 67 cepas de S. Heidelberg isoladas na cadeia de produção avícola no ano de 2016. Utilizou-se a técnica de disco difusão frente ao ácido nalidíxico, amoxacilina/ácido clavulânico, ceftazidima, ceftiofur, ciprofloxacina, enrofloxacina, gentamicina, imipenem, sulfametoxazol/trimetoprim, sulfato de colistina, tetraciclina. Os maiores índices de resistência foram para o ácido nalidíxico e tetraciclina (100%), seguido do sulfato de colistina (98,5%).Todas as cepas foram classificadas como multirresistentes, alarmando para o perigo que representam para a saúde pública.(AU)

O estudo teve como objetivo avaliar o uso de pisos plásticos perfurados como substituto da cama de frango e a diminuição da carga microbiana. Os animais foram divididos em dois tratamentos: animais criados em cama (7 repetições) e animais criados no piso de plástico (7 repetições). Nos dias 4, 12, 25 e 40 de experimento, foram coletadas amostras de suabe de cloaca, suabe de arrasto do piso e amostras de cama para a contagem de coliformes totais, termotolerantes e Salmonella spp. Não foi possível encontrar diferença (p>0,0001) na contagem de coliformes entre os tratamentos. No teste presuntivo de Salmonella, até os 25 dias, os animais criados no piso tiveram as menores porcentagens de positivos. A utilização de pisos de plásticos e sua associação com protocolos higiênicos-sanitários pode ser uma estratégia para diminuir as contaminações bacterianas nos estabelecimentos avícolas comerciais e nas carcaças de frangos de corte.(AU)

Dentre as bactérias causadoras de Doenças Veiculadas por Alimentos (DVA) a Salmonella spp. é um dos principais patógenos na avicultura. Objetivou-se avaliar a presença de Salmonella spp. nas carcaças de frango de corte de um aviário de pequeno porte. Em dois lotes foi realizado formulário de acompanhamento das boas práticas de produção; aferição das temperaturas do ambiente e das carcaças; análises do pH e do cloro da água, e microbiológicas da bancada de evisceração, da depenadeira e das carcaças. A análise dos dados foi descritiva. Houve ausência do patógeno em todas as amostras. Assim, a implementação e o rigor no monitoramento dos padrões presentes nas Boas Práticas de Fabricação (BPF) e nos Procedimentos Padrão de Higiene Operacional (PPHO) além de controlar a propagação desse patógeno nas carcaças, asseguraram um produto final de qualidade ao consumidor.(AU)

O presente estudo tem como objetivo descrever a distribuição de probabilidades da temperatura de cada etapa da cadeia produtiva de carne de frango resfriada produzida sob inspeção federal no Brasil. Os resultados gerados geram subsídio para realização de uma Análise de Risco de salmonelose pelo consumo de carne de frango no país. Foram compilados registros de temperatura e os dados foram ajustados às distribuições de probabilidade utilizando o software @Risk. Nas etapas de abatedouro-frigorífico, centro de distribuição, serviço de alimentação e supermercado a média de temperatura foi inferior a 5,5 ºC, demostrando sucesso na manutenção da cadeia de frio. A distribuição de probabilidade Logistic demonstrou melhor ajuste para descrever as etapas de processamento. Os dados de distribuição de probabilidade gerados poderão ser utilizados em modelos matemáticos e de distribuição para predizer o crescimento microbiano em alimentos expostos a diferentes temperaturas.(AU)

Background: The genus Salmonella, associated with poultry products, is considered the leading cause of foodborne outbreaks in humans in many countries. In Brazil, Salmonella Enteritidis (SE) is the serovar remains as one most frequentlyisolated from humans, and it is also a major serovar found in animals, food, animal feed, and environmental samples, despiteall the efforts to control this pathogen. Also, the bacterium is able to form biofilms on different surfaces, protecting cellsfrom both cleaning and sanitizing procedures in the food industries. This study aimed to verify the ability of SalmonellaEnteritidis isolates to form biofilm on polystyrene at different incubation temperatures.Materials, Methods & Results: A total of 171 SE samples were isolated from foodborne outbreaks (foods and stool cultures) and poultry products between 2003 and 2010. The biofilm-forming ability of samples was measured at four differenttemperatures (3°C, 9ºC, 25ºC, and 36ºC), for 24 h, simulating temperatures usually found in poultry slaughterhouses. Later,200 μL of each bacterial suspension was inoculated, in triplicate, onto 96-well, flat-bottomed sterile polystyrene microtiterplates, washed, after that, the biofilm was fixed with methanol. The plates were dried at ambient temperature, stained with2% Huckers crystal violet. Afterwards, absorbance was read using an ELISA plate reader and the optical density (OD)of each isolate was obtained by the arithmetic mean of the absorbance of three wells and this value was compared withthe mean absorbance of negative controls (ODnc). The following classification was used for the determination of biofilmformation: no biofilm production, weak biofilm production, moderate biofilm production and strong biofilm production.Results demonstrated all isolates from stool cultures and foods involved in foodborne outbreaks, at least one of the fourtemperatures tested, were able to form biofilm...(AU)

Chicken feet have become an important commodity in the international market, representing a significant portion of poultry products exported by countries such as Brazil and the USA. However, the presence of pododermatitis in the footpad is an important barrier to exportation, since importing countries do not accept injured feet or allow the use of automatic equipments to remove the affected tissue. The objective of this research was to evaluate the impact of using an automatic equipment to remove injuries of pododermatitis on histological and microbiological traits of broiler feet processed according to commercial practices. A total of 240 broiler feet obtained from a commercial processing plant was visually classified according to the degree of pododermatitis and distributed in a 4 × 2 factorial arrangement, totalizing eight treatments with 30 replications. Factors were feet classification (1 to 4) and injury removal (yes or no). Feet were sampled for microbiological and histological analysis before and after the mechanical removal of pododermatitis injuries by an automatic machine that promoted footpad epidermal scarification. No significant interaction between feet classification and injury removal was detected for any of the analyzed variables. Also, no significant effect of feet classification was detected on aerobic plate counts, total coliforms and Escherichia coli. Feet inflammation score tended to increase (P = 0.06) according to the downgrading of feet classification, but the mechanical removal of pododermatitis injuries reduced feet inflammation score (P < 0.01), total coliform counts (P = 0.01), and E. coli (P = 0.01) independently of feet classification. Together, these results demonstrate the efficacy of the automatic equipment in removing both the inflammatory tissue and its associated microbiota in broiler feet affected by pododermatitis. Therefore, in addition to the already authorized use of blades, the use of automatic equipments for epidermal scarification in the processing of broiler feet deserves further consideration by the regulatory agencies.

Campylobacter is regarded as the most common bacterial cause of gastroenteritis throughout the world and most cases of human campylobacteriosis can be traced back to the consumption of poultry meat. In Brazil, few studies evaluated the genetic relatedness among Campylobacter isolates. The aim of this research was to evaluate the genetic diversity of Campylobacter spp. isolated from poultry meat products sold on the retail market in Southern Brazil. The presumptive identification of Campylobacter was performed using traditional microbiological analysis, followed by molecular confirmation by PCR. The genetic diversity of isolates was analyzed by pulsed-field gel electrophoresis (PFGE). Campylobacter spp. was isolated from 91.7% (33/36) of the samples, totaling 48 isolates. Campylobacter jejuni was the most prevalent species isolated (90.8%). PFGE data revealed 26 pulsotypes and 18 PFGE patterns composed of only 1 isolate. Campylobacter isolates exhibited high genetic diversity; however, some clones were recurrent in the poultry meat products sold on the retail market. As the south region of Brazil is an important producer and exporter of chicken meat, our results highlight the need to control this pathogen in the food chain in this area of the world to reduce the risks of exposing consumers to campylobacteriosis.

In this study we report the characterization of plasmid-mediated CMY-2-producing Salmonella Heidelberg recovered from food, poultry, and poultry environment in Brazil, between 2014 and 2016. The blaCMY-2 resistance gene was allocated in large (90-148 kb) IncI1 type transferable plasmids. Salmonella Heidelberg isolates were genetically related, indicating the dissemination of closely related isolates among food, poultry, and its environment. This is the first report of IncI1 replicon-types of plasmids encoding the blaCMY-2 resistance gene in Salmonella Heidelberg isolates in Brazil, the world's biggest exporter of chickens.
