RESUMEN

Background: Paraquat (1,1'-dimethyl-4-4'-bipyridinium dichloride) exposure is well-established as a neurotoxic agent capable of causing neurological deficits in offspring. This study aimed to investigate therapeutic effects of Arbutus unedo L. aqueous extract (AU) against paraquat (PQ) exposure. Methods: For that the phytoconstituents of AU was determined by LC/MS, and then its antioxidant potential was assessed by DPPH and ABTS assays. The assessment included its impact on cell viability and mitochondrial metabolism using N27 dopaminergic cells. Additionally, we evaluated the effects of prenatal PQ exposure on motor coordination, dopamine levels, trace element levels, and total antioxidant capacity (TAC) in rat progeny. Results: The phytochemical profile of AU extract revealed the presence of 35 compounds, primarily phenolic and organic acids, and flavonoids. This accounted for its strong in vitro antioxidant activities against DPPH and ABTS radicals, surpassing the activities of vitamin C. Our findings demonstrated that AU effectively inhibited PQ-induced loss of N27 rat dopaminergic neural cells and significantly enhanced their mitochondrial respiration. Furthermore, daily post-treatment with AU during the 21 days of the rat's pregnancy alleviated PQ-induced motor deficits and akinesia in rat progeny. These effects inhibited dopamine depletion and reduced iron levels in the striatal tissues. The observed outcomes appeared to be mediated by the robust antioxidant activity of AU, effectively counteracting the PQ-induced decrease in TAC in the blood plasma of rat progeny. These effects could be attributed to the bioactive compounds present in AU, including phenolic acids such as gallic acid and flavonoids such as quercetin, rutin, apigenin, glucuronide, and kaempferol, all known for their potent antioxidant capacity. Discussion: In conclusion, this preclinical study provided the first evidence of the therapeutic potential of AU extract against PQ-induced neurotoxicity. These findings emphasize the need for further exploration of the clinical applicability of AU in mitigating neurotoxin-induced brain damage.

The presence of persistent organic pollutants (POPs) in human milk is an indicator of the level of these substances in the environment and enables evaluation of the potential exposure of breastfed children. POPs concentrations in a Moroccan human milk pooled sample (59 donors) were determined for the first time by the WHO reference laboratory to provide a baseline and allow monitoring of the future trends for those persistent organohalogen compounds. Eighty-one different POPs, including organochlorine pesticides (OCPs), polychlorinated biphenyls (PCBs), polybrominated diphenyl ethers (PBDEs), and dioxins and furans (PCDD/Fs) were identified and quantified by using gas chromatography (GC) with an electron capture detector (ECD) and confirmed by gas chromatography-mass spectrometry (GC/LRMS). The obtained results showed that the sum of DDT (sum of o,p'-DDD, p,p'-DDD, o,p'-DDE, p,p'-DDE, o,p'-DDT, and p,p'-DDT) was 237.9 ng/g, representing 94.0% of all detected OCPs levels, with a dominance of p,p'-DDE, and p,p'-DDT. Cis-heptachlor epoxide, HCB, and HCH were found at much lower levels. PCB indicator level was 60.7 ng/g of lipid, with the dominance of PCB 138, PCB 153, and PCB 180, presenting 98.3% of the total. Among the 25 PBDE congeners quantified, BDE-47, BDE-153, BDE-197, and BDE-207 were the dominant congeners account for 15.4% of the total concentration of PBDE (1.3 pg/g lipid). The lipid-adjusted level of PCDD/Fs and dl-PCBs expressed in Toxic Equivalents (TEQs: 6.65 pg/g) were lower in comparison to those found in the other countries. In summary, detected POPs levels in Moroccan human milk were much lower compared to the other countries, reflecting the effectiveness of the compliance with Stockholm Convention requirements on eliminating or reducing emissions of selected POPs. Continued monitoring is needed for these compounds, for which this is the first data available, provided by the WHO/UNEP survey in Morocco.

The strawberry tree (Arbutus unedo L.), grown throughout the Mediterranean, produces edible fruit; as it is easily bruised, the sweet, reddish fruit is used mostly to prepare jams, marmalades and alcoholic beverages. As the genus is paraphyletic, phytochemical analysis can assist in defining the fruit composition with the species Arbutus unedo L. (A. unedo). Here we report on the carbohydrate, total sugar, protein, fat, fiber, ash, and mineral content of wild fruit, harvested from 45 specimens from five locations. The dominant nutrients were carbohydrates (78.2-84.8 g/100 g), total sugars (52.1-67.2 g/100 g) and dietary fiber (11.0-20.1 g/100 g). Other important nutrients supplied by A. unedo fruit include P, K, and Fe. The fruit was observed to contain health-promoting components providing 42 and 36%, of recommended daily allowance (RDA) for fiber and zinc, respectively, as well as iron and manganese, at levels exceeding minimum RDA. The free-sugar profile revealed high glucose followed by fructose content with minor amounts of sucrose (14, 11, and 6 g/100 g, respectively). Significant differences both between regions and within individuals were observed for several traits. The richness of fruit nutrients in A. unedo confers nutritional value and as such, a promising alternative fruit source.
