Background: Suicide is defined as the deliberate ending of one's own life or a deadly self-injurious act with evidence of the intention to die. Suicide among youths is a significant public mental health issue. Young individuals, particularly adolescents, are naturally susceptible to mental health problems. Methods: The present study has the cross-sectional study design based on simple random sampling technique. Initially, the questionnaire was self -administered to 4353 respondents. Questionnaire with incomplete responses was discarded, and finally, a sample of 4278 was used for the analysis. The study is based in the Kingdom of Saudi Arabia, and the data was collected between the months of January 2022 and April 2022. The survey instrument used in the present study comprised of two sections. Results: In our study, 79.5% of the respondents agreed to the statement "the ruling of suicide is the biggest factor to the low incidences of suicide in our community." Further, 66.6% of the respondents agreed that suicide is an idea or an act that a person can repel by religious practices such as prayer. Conclusion: A focus on suicidal behavior is a critical public health priority because although suicide is a prominent cause of mortality among teenagers; many more youths are at risk for suicide as a result of having suicidal ideation, creating suicide plans, and making an attempt. Trends in teenage suicide attempts increased overall and among numerous demographic categories.

Cinnamomum tamala (bay leaf) is widely used for culinary and medicinal purposes in South Asia. A leaf blight/spot disease was first discovered on nearly 90% of C. tamala plants with a mean severity of 48% to 74.4% in Gazipur and Bogura, Bangladesh, in 2019. The present study identified and characterized the causal organism and formulated the optimum growth conditions and effective fungicides for the chemical control of the pathogen. The characteristic symptoms on the infected leaves appeared circular to oval reddish-brown spots with raised margins and often developed in tear-stain patterns. Severe infection of C. tamala sapling resulted in dieback symptoms with leaf defoliation. A fungus with floccose, dense, white colonies with well-differentiated acervuli was isolated from the infected leaves. Combined cultural, morphological, and molecular characteristics identified the pathogen as Colletotrichum siamense. Inculcating healthy leaves and 1-year-old saplings of C. tamala with a conidial suspension of the fungus reproduced the same symptoms observed in the bay leaf orchard. The highest mycelial growth was recorded on V-8 Juice Agar media, while the maximum radial mycelial growth and level of sporulation of the fungus were significantly higher in incubation temperature 30°C. Fungicide trials showed that carbendazim 50 WP, azoxystrobin, mancozeb, and trifloxystrobin, either singly or in combination, successfully reduced fungal mycelial growth in vitro. Therefore, disease management strategies should be opted to halt the further spread of this issue. To our knowledge, this is the first study to document the incidence of Colletotrichum leaf blight on C. tamala in Bangladesh and even in the world.

RESUMEN

In this research study, we developed a voltammetric electrochemical sensor probe with a copolymer Nafion (Sulfonated Tetrafluoroethylene-based Fluoro-polymer) decorated with hydrothermally prepared sandwich-type CuO/ZnO nanospikes (NSs) onto a glassy carbon electrode (GCE) for reliable thiourea (TU) detection. The detailed characterizations in terms of structural morphology, binding energy, elemental compositions, grain size and crystallinity for synthesized NSs were performed by field emission scanning electron microscopy (FESEM), X-ray photoelectron spectroscopy (XPS), energy-dispersive X-ray spectroscopy (EDS) and X-ray diffraction (XRD) analysis, respectively. The differential pulse voltammetric (DPV) analysis for TU showed good linearity at current-versus-TU concentration on the calibration plot in the 0.15~1.20 mM range, which is defined as a dynamic detection range (LDR) of TU in a phosphate buffer solution. Considering the slope of LDR over the GCE-coated NSs surface area (0.0316 cm2), the TU sensor sensitivity (0.4122 µA µM-1 cm-2) was obtained. Besides this, the low limit (LOD) for TU detection was calculated and found to be 23.03 ± 1.15 µM. The fabricated Nafion/CuO/ZnO NSs/GCE sensor probe was created as a reliable sensor based on reproducibility, interference effect, stability and response time. Real bio-samples were investigated and the results confirm the anticipated reliability of the TU sensor probe. Thus, this is a noble way to develop enzyme-free electrochemical sensors that could be an alternative approach for the detection of chemicals in the field of enzyme-free biosensor development technology.

OBJECTIVE: To determine if viable virus could be isolated from the air within a car driven by a patient infected with SARS-CoV-2, and to assess the size range of the infectious particles. METHODS: We used a Sioutas personal cascade impactor sampler (PCIS) to screen for SARS-CoV-2 in a car driven by a COVID-19 patient. The patient, who had only mild illness without fever or cough and was not wearing a mask, drove the car for 15 min with the air conditioning turned on and windows closed. The PCIS was clipped to the sun-visor above the front passenger seat and was retrieved from the car two hours after completion of the drive. RESULTS: SARS-CoV-2 was detectable at all PCIS stages by PCR and was cultured from the section of the sampler collecting particles in the 0.25-0.50 µm size range. CONCLUSIONS: Our data highlight the potential risk of SARS-CoV-2 transmission by minimally symptomatic persons in the closed space inside of a car and suggest that a substantial component of that risk is via aerosolized virus.

The facile wet-chemical technique was used to prepare the low-dimensional nano-formulated porous mixed metal oxide nanomaterials (CuO.Mn2O3.NiO; CMNO NMs) in an alkaline medium at low temperature. Detailed structural, morphological, crystalline, and functional characterization of CMNO NMs were performed by X-ray photoelectron spectroscopy (XPS), powder X-ray diffraction (XRD), ultraviolet-visible spectroscopy (UV-vis), Fourier-transform infrared spectroscopy (FTIR), field emission scanning electron microscopy (FESEM), and energy-dispersive X-ray spectroscopy (EDS) analyses. An efficient and selective creatine (CA) sensor probe was fabricated by using CMNO NMs decorated onto glassy carbon electrode (GCE) as CMNO NMs/GCE by using Nafion adhesive (5% suspension in ethanol). The relation of current versus the concentration of CA was plotted to draw a calibration curve of the CMNO NMs/GCE sensor probe, which was found to have a very linear value (r2 = 0.9995) over a large dynamic range (LDR: 0.1 nM~0.1 mM) for selective CA detection. The slope of LDR by considering the active surface area of GCE (0.0316 cm2) was applied to estimate the sensor sensitivity (14.6308 µAµM-1 cm-2). Moreover, the detection limit (21.63 ± 0.05 pM) of CMNO MNs modified GCE was calculated from the signal/noise (S/N) ratio at 3. As a CA sensor probe, it exhibited long-term stability, good reproducibility, and fast response time in the detection of CA by electrochemical approach. Therefore, this research technique is introduced as a promising platform to develop an efficient sensor probe for cancer metabolic biomarker by using nano-formulated mixed metal oxides for biochemical as well as biomedical research for the safety of health care fields.

A complete one- and two-photon spectroscopic and photophysical characterization of three diketopyrrolopyrrole (DPP)-porphyrin conjugates is reported. The increased conjugation introduced by the incremental addition of one, two and four DPP units on the meso porphyrin positions strongly affects the optical properties of the systems. Ground and triplet excited state absorption spectra show a gradual broadening and bathochromic shift and a trend to lower energies is also observed for both fluorescence and phosphorescence emission. Interestingly, the fluorescence quantum yield increases along the series, leading to remarkable NIR emission properties for the larger derivatives. Unlike the model porphyrin, all derivatives exhibit high two-photon absorption activity. An increase in two-photon absorption cross-section in the regions 800-840 nm and 910-930 nm is observed moving from one DPP to two DPP appended units, with a value of the order of 4000 GM at 910 nm for the latter system. The four compounds show high efficiency in generating singlet oxygen, with yields ranging from 0.7 to 0.5, envisaging favourable applications in both one- and two-photon photodynamic therapies. A detailed theoretical exploration of both linear (absorption and emission) and non-linear (two-photon absorption) properties proposes an analysis of the experimental spectra and a comprehensive interpretation of the two-photon activity within the series of compounds.

The SP1/Krüppel-like Factor (SP1/KLF) family of transcription factors plays a role in diverse cellular processes, including proliferation, differentiation and control of gene transcription. The discovery of KLF1 (EKLF), a key regulator of HBB (ß-globin) gene expression, expanded our understanding of the role of KLFs in erythropoiesis. In this study, we investigated a mechanism of HBG (γ-globin) regulation by KLF4. siRNA-mediated gene silencing and enforced expression of KLF4 in K562 cells substantiated the ability of KLF4 to positively regulate endogenous HBG gene transcription. The physiological significance of this finding was confirmed in primary erythroid cells, where KLF4 knockdown at day 11 significantly attenuated HBG mRNA levels and enforced expression at day 28 stimulated the silenced HBG genes. In vitro binding characterization using the γ-CACCC and ß-CACCC probes demonstrated KLF4 preferentially binds the endogenous γ-CACCC, while CREB binding protein (CREBBP) binding was not selective. Co-immunoprecipitation studies confirmed protein-protein interaction between KLF4 and CREBBP. Furthermore, sequential chromatin immunoprecipitation assays showed co-localization of both factors in the γ-CACCC region. Subsequent luciferase reporter studies demonstrated that KLF4 trans-activated HBG promoter activity and that CREBBP enforced expression resulted in gene repression. Our data supports a model of antagonistic interaction of KLF4/CREBBP trans-factors in HBG regulation.
