RESUMEN

The spleen structurally and functionally belongs to the hematopoietic organs and is also an important component of the reticuloendothelial system, which is known to play a major role in host defense. The histological structure of the spleen was investigated in the ostrich, a non-flying bird, the kestrel, a raptor, and the osprey, a fish-eating bird of prey (fish eagle). For this purpose, Mallory's modified triple stain, methyl green-pyronin and silver stain were used. Germinal centers were not present in the spleen of the osprey. In the spleen of the kestrel, penicillar arterioles and the surrounding lymphoid tissue were markedly dense. Compared to the other two birds, the red and white pulps were clearly distinguishable in the spleen of the ostrich.

RESUMEN

In this study, therapeutic effects of Vitamin K2, Raloxifene and their co-administration on bone, uterus, blood and weight profiles were investigated with an ovariectomized rat model. Forty Wistar rats were divided into five groups (n=8): Raloxifene (R), Vitamin K2 (K), Raloxifene+Vitamin K2 (R+K), ovariectomized controls (OVX) and Sham-operated controls (Sham). Treatment began 3 months after ovariectomy. Vitamin K2 and Raloxifene were administered 30 and 1.5 mg/kg/day separately and in combination five times per week for 12 weeks. All treatment groups had significantly higher ultimate strength and energy absorption capacity (P<0.05) than ovariectomized controls in both femur and tibia. Histological results showed that treatment groups had healthy lumen structure, whereas OVX had degeneration. Adverse effects which were seen in individual treatments (myometrium weakening in K, endometrium weakening in R, and ALP increase in group R) were not observed in the R+K group implying a synergistic effect of these two agents when they are co-administered. According to blood analysis, ALP values were significantly high in Raloxifene-only group (P<0.0001). This effect is suppressed in the co-administered group. In summary, the groups R, K and R+K had significantly higher ultimate strength and less susceptibility to fracture than ovariectomized controls. In summation, Vitamin K2 treated groups (either in single or combined with Raloxifene) had considerable biomechanical performance and reproductive tissue profile indicating that this agent is prospectively effective in osteoporosis management.

We examined gazelle peripheral blood leucocytes using the alpha-Naphthyl acetate esterase (ANAE) staining technique (pH 5.8). Our purpose was to determine the percentage of ANAE positive lymphocytes. The proportion of ANAE positive T-lymphocytes was 72%. T-lymphocytes showed an ANAE positive reaction, but eosinophilic granulocytes and monocytes also showed a positive reaction. By contrast, no reaction was detected in B-lymphocytes, neutrophil granulocytes or platelets. The reaction observed in T-lymphocytes was a red-brown coloration, usually 1-2 granules, but enough granules to fill the cytoplasm were detected rarely. As a result of ANAE enzyme staining, we concluded that the staining technique can be used as a cytochemical marker for gazelle T-lymphocytes.

The morphology of the retinal pigment epithelium (RPE) and closely associated Bruch's membrane and choriocapillaris was investigated by light and transmission electron microscopy in the camel (Camelus dromedarius). The study showed that RPE is composed of a single layer of hexanocuboidal cells that were joined laterally by a series of apically located tight junctions. In addition, adjacent from internal side of cell membrane at the level of tight junctions, an undefined structure which resembled the myofibrillar organization of skeletal muscles in appearance was located. These cells displayed numerous short basal infoldings and abundant thin apical processes which enclosed the rod outer segments. The epithelial cell nuclei were large, vesicular and eccentrically located. Within the epithelial cells, smooth endoplasmic reticulum was very abundant, while rough endoplasmic reticulum was present only in small amounts. Polysomes were also numerous and the mitochondria often displayed a ring-shaped structure. Lipofuscin granules were plentiful in all locations. Bruch's membrane (complexus basalis) was typically pentalaminate throughout the retina. The endothelium of the choriocapillaris facing Bruch's membrane was extremely thin and heavily fenestrated. These fenestrations displayed typical single-layered diaphragm as noted in most species.

Infection with hepatitis B virus (HBV), hepatitis C virus (HCV), human immunodeficiency virus (HIV) and syphilis are serious complications of blood transfusion. These infections are routinely screened by blood banks; such tests are obligatory for transfusion safety in Turkey. The results of screening 1 737 943 blood donations from blood centres in Istanbul between 1 January 1987 and 31 December 2003 for HBV, HCV, HIV and syphilis were analysed retrospectively. Hepatitis B surface antigen rates fell from 5.98% in 1987 to 2.07% in 2003. Anti-HCV seropositivity was found to be approximately 0.5%, whereas anti-HIV seropositivity was approximately 0.001%. Rapid plasma reagin rates (test for syphilis) were 0.04% in 1987, and increased to 0.2% in 2002. The decreasing trends observed in data from the 17-year period studied indicate the value of safety measures taken, in particular the implementation of donor screening procedures in 1997.

RESUMEN

The Harderian gland of the ostrich (Struthio camelus) is a tubuloalveolar gland containing holocrine secreting epithelial cells. The gland epithelium is composed of two different cell types, which can be classified as type I and type II. These cells contain dense secretory vesicles in their cytoplasm and they are connected laterally with desmosomes. At the basal site of these cells, myoepithelial cells are present. Plasma cells are observed in the subepithelial region of the gland. In the interlobular trabeculae, forming the gland stroma, fibroblasts, blood vessels and nerve fibres are included. Another important finding in the ostrich Harderian gland is the presence of homogeneous material.

The fine structure of the retinal pigment epithelium (RPE), Bruch's membrane and choriocapillaris in the ostrich (Struthio camelus) was investigated by using light microscopy and transmission electron microscopy. In this species, the RPE consisted of a single layer of low columnar cells. The epithelial cells were joined laterally by two type junctions, zonulae occludentes and zonulae adherentes located in the midregion of the cells. These cells displayed numerous deep basal infoldings and thick extensive apical processes, which enclosed the outer segments of the rods. The epithelial cell nuclei were large, vesicular and located basally within the epithelial cells. Smooth endoplasmic reticulum was very abundant, while rough endoplasmic reticulum was scarce. Mitochondria of various shapes were abundant basally while polysomes were plentiful and widespread. In the light-adapted state melanosomes were located in the apical region and in apical processes of the epithelial cells. Myeloid bodies were large, numerous and often showed ribosomes on their outer surface. Bruch's membrane (complexus basalis) was typical pentalaminate throughout the retina, as noted in the majority of other vertebrates. The endothelium of the choriocapillaris facing Bruch's membrane was extremely thin but only moderately fenestrated. Some of the fenestrations displayed a double-layered diaphragm while the majority showed the more typical single-layered diaphragm noted in most species.

Pain is the most common and feared symptom for patients, especially those with cancer. Treatment of chronic pain with conventional ways of medication usually fails with increasing severity of the pain. New approaches enabling the prolonged provision of pain relievers are required. We designed a controlled release system of pain relievers, mainly for opioids (morphine, M, codeine, C, and hydromorphone, HM), and a local anesthetic (bupivacaine, BP) in the form of poly(L-lactide-co-glycolide) (PLGA) rods. The efficacy of these rods implanted alone or in combination in relieving chronic pain in rats caused by the ligation of the sciatic nerve of their right hind limbs was studied. The two most common tests for measuring analgesia, i.e. tail-flick tests, that show analgesia at sites other than the site of injury, were used to study the degree of systemic distribution of the drugs and paw-withdrawal tests were used to study the analgesia at the site of injury. Alleviation of this chronic and severe neuropathic pain could be obtained for about 3-4 days when rods for two drugs, 'dual drug' (analgesic-anesthetic), were used. This duration is decreased by half (2 days) with the single-drug rods. Also the dual-drug rods, though at half the dose of each single drug application, enhanced the degree of analgesia of the first day. These in vivo results are also consistent with the previous in vitro results as in the case with codeine which had a higher first-day analgesia than morphine, despite a lower potency due to the faster in vitro release rate. Similarly, slower release of hydromorphone from PLGA (85:15) rods resulted in less systemic analgesia than the more rapidly eroding PLGA (50:50) rods of the same drug.

The study presented here was performed to determine the rates of immunity to tetanus in a cross section of the Turkish population and to assess the impact of a single-dose tetanus toxoid in previously unvaccinated individuals. Among 18-year-old participants who received their last tetanus booster 4 years prior to the study and 22-year-old participants who received the booster 8 years previously, certain durable protective antibody (>1 IU/ml) was detected in 100%. However, only 3.3% of 20-year-old participants who did not know whether they had been vaccinated as children or young adults had a protective antibody level. After administering a single dose of tetanus vaccine to this group, the rate of protective antibody increased from 3.3% to 96.7%. For the prevention of tetanus, the optimal policy should focus on improving rates of complete primary immunization in adults and children and giving a single booster every 10 years, or at least at age 40 or 50 years.

In the present study, we determined and compared the distribution and mean diameters of fiber in the cremaster muscles (CM) of boys with either inguinal hernia (IH) or undescended testis (UT). Samples of CM were obtained from 20 patients (10 boys with IH, and 10 boys with UT) of similar age. The CM muscles of two boys each, without inguinal pathology, were sampled during autopsy. Sections were stained for oxidative and glycolytic enzymes, as well as for ATP-ase reactions after acid (pH: 4.6) and alkaline (pH: 10.6) preincubations. Specimens were also analyzed morphometrically using a KONTRON 400 computerized image analysis system. The Mann- Whitney U test was applied to compare the percentages of fiber types and mean diameters of fibers according to the types of the CM of boys with IH or UT. In boys, the CM is mainly composed of type 1 fibers. The CMs of patients with UT reveal alterations of neurogenic origin. Although both type 1 and type 2 fibers reveal alterations, type 2 fibers appear to be affected more profoundly and characterized by significantly decreased mean diameters. Significantly decreased mean diameters of type 2 fibers in CM may support disuse, lack of sensitivity to the hormonal influences, or an alteration in the corticospinal tracts of boys with UT.

In all locations, the RPE consisted of a single layer of low cuboidal cells. The epithelial cells were joined laterally by apically located tight junctions. These cells displayed numerous basal infoldings and abundant thin apical processes which enclosed the rod outer segments. The epithelial cell nuclei were large and located basally. Within the epithelial cells, smooth endoplasmic reticulum was very abundant, while rough endoplasmic reticulum was scarce, polysomes and mitochondria, which often display a ring-shaped structure, were abundant. Melanosomes were abundant in the non-tapetal area but absent in the tapetal area. Bruch's membrane was pentalaminate throughout the retina. The endothelium of the choriocapillaris was heavily fenestrated.

The purpose of this study was to investigate the electron microscopic structure of conjunctiva associated lymphoid tissue (CALT) and to determine the uptake of macromolecules from follicle associated epithelium (FAE) of Angora goats. The sample tissues taken from lower and upper eyelids of ten 5-6 month-old healthy Angora goats were used in this study. The conjunctiva associated lymphoid tissue of Angora goats was formed by solitary or aggregate lymphoid follicle and follicle associated epithelium which cover these follicle. FAE was formed by flattened epithelial cells, lymphocytes, monocytes and polymorph nuclear leukocytes but no goblet cells. Ferritin particles were seen on the apical surface, in the invaginations, vesicles, vacuoles of flattened epithelial cells. It was concluded that these epithelial cells were specialized to uptake macromolecules.

The purpose of this study was to determine the percentage of peripheral blood T lymphocytes and the localization of ANAE enzyme at the electron microscopic level in Van cats by using an alpha-naphtyl acetate esterase procedure. Peripheral blood samples taken from 20 Van cats were used. The percentage of ANAE positive lymphocytes was 83.0%. Neutrophilic granulocytes gave a negative reaction, whereas monocytes, eosinophilic granulocytes showed a diffuse granular positivity. In the electron microscopic examination, ANAE positive reactions were seen in lysosomal granules found in lymphocytes.
