RESUMEN

Genes encoding subunits of SWI/SNF (BAF) chromatin­remodeling complexes are recurrently mutated in a broad array of tumor types, and among the subunits, ARID1A is the most frequent target with mutations. In the present study, it was reported that ARID1A inhibits the epithelial­mesenchymal transition (EMT) and stemness of ovarian cancer cells, accompanied by reduced cell viability, migration and colony formation, suggesting that ARID1A acts as a tumor suppressor in ovarian cancer. Mechanistically, ARID1A exerts its inhibitory effects on ovarian cancer cells by activating the Hippo signaling pathway. Conversely, the overexpression of a gain­of­function transcriptional co­activator with PDZ­binding motif (TAZ) mutant (TAZ­Ser89) effectively reverses the effects induced by ARID1A. In addition, activation of Hippo signaling apparently upregulates ARID1A protein expression, whereas ectopic expression of TAZ­Ser89 results in the markedly decreased ARID1A levels, indicating a feedback of ARID1A­TAZ in regulating ovarian cancer cell EMT and stemness. Thus, the present study uncovered the role of ARID1A through the Hippo/TAZ pathway in modulating EMT and stemness of ovarian cancer cells, and providing with evidence that TAZ inhibitors could effectively prevent initiation and metastasis of ovarian cancer cases where ARID1A is lost or mutated.

The Rab small GTPases are characterized by the distinct intracellular localization and modulate various endocytic, transcytic and exocytic transport pathways. Rab proteins function as scaffolds that connect signaling pathways and intracellular membrane trafficking processes through the recruitment of effectors, such as tethering factors, phosphatases, motors and kinases. In different cancers, Rabs play as either an onco-protein or a tumor suppressor role, highly dependending on the context. The molecular mechanistic research has revealed that Rab proteins are involved in cancer progression through influences on migration, invasion, metabolism, exosome secretion, autophagy, and drug resistance of cancer cells. Therefore, targeting Rab GTPases to recover the dysregulated vesicle transport systems may provide potential strategy to restrain cancer progression. In this review, we discuss the regulation of Rab protein level and activity in modulating pathways involved in tumor progression, and propose that Rab proteins may serve as a prognostic factor in different cancers.

Objective: A common assertion in the social media literature is that passive media use undermines affective wellbeing, and active media use enhances it. The present study investigated the effects of social media use on negative affective wellbeing during pandemic crises and examined the mechanism underlying these effects through perceived uncertainty. Methods: Three studies were conducted during the Delta variant phase in the post-peak period of the COVID-19 pandemic in China. Participants were recruited from the medium-high-risk infection areas in late August 2022. Study 1 used a cross-sectional survey to explore the relationships between social media use, uncertainty, and negative affect during the pandemic crisis. Study 2 employed a repeated-measures experiment to demonstrate how social media use and (un)certainty impact negative affect. Study 3 utilized a one-week experience sampling design to examine the role of uncertainty in the relationship between social media use and negative affect in real life. Results: Despite some inconsistencies regarding social media use's direct effect on negative affect, across the three studies, perceived uncertainty was critical in linking pandemic-related social media use to individuals' negative affect, particularly for passive use. Conclusions: The relationships between social media use and affective wellbeing are complex and dynamic. While the perception of uncertainty provided an underlying mechanism that links social media use to individuals' affective wellbeing, this mechanism may be further moderated by individual-level factors. More research is needed as we seek to understand how social media use impacts affective wellbeing in uncertain contexts.

Tyre wear generates not only large pieces of microplastics but also airborne particle emissions, which have attracted considerable attention due to their adverse impacts on the environment, human health, and the water system. However, the study on tyre wear is scarce in real-world driving conditions. In the present study, the left-front and left-rear tyre wear in terms of volume lost in mm3 of 76 taxi cars was measured about every three months. This study covered 22 months from September 2019 to June 2021 and included more than 500 measurements in total. Some of the data was used to evaluate the effects of vehicle type and tyre type on tyre wear. In addition, a machine learning method (i.e., Extreme gradient boosting (XGBoost)) was used to probe the effect of driving behaviour on tyre wear by monitoring real-time driving behaviour. The current statistical results showed that, on average, the tyre wear was 72 mg veh-1 km-1 for a hybrid car and 53 mg veh-1 km-1 for a conventional internal combustion engine car. The average tyre wear measured for a taxi vehicle configuration featuring winter tyres was 160 mg veh-1 km-1, which was 1.4 and 3.0 times as much as those with all-season tyres and summer tyres, respectively. The wear rate of left-front tyres was 1.7 times higher than that of left-rear tyres. The XGBoost results indicated that compared to driving behaviour, tyre type and tyre position had more important effects on tyre wear. Among driving behaviours, braking and accelerating events presented the most considerable impact on tyre wear, followed by cornering manoeuvres and driving speed. Thus, it seems that limiting harsh braking and acceleration has the potential to reduce tyre wear significantly.

The outbreak of COVID-19 in China started at the end of December 2019. This led to a series of containment measurements to control the spread of COVID-19. Despite of the widely reported effects of these measures, inadequate attention has gone to their social impacts. The elderly, as one of the most susceptible populations, has experienced a considerable reduction in mobility. This paper explores the role mobility played and how the social environment influenced elderly mobility in the first 2 months of the COVID-19 outbreak. We surveyed 186 families with a total of 248 elderly people in Kunming. The results show that mobility improves the quality of daily living, such as access to grocery shopping, maintenance of outdoor activities for health cultivation and preserving social networks even during the pandemic. Four themes relating to social environment emerged from the data as elements influencing elderly mobility during the pandemic: social pressure, practice of the virtue of Xiao, the social norm of respecting the aged and the impacts of technological advances. Among them, the virtue of Xiao enabled the elderly to stay in place in the early phase of COVID-19 by fulfilling their needs for daily necessities and social interactions, whilst being less technology-savvy further excluded them socially by restraining them from restoring mobility after the lifting of travel restrictions.

Although many cross-sectional studies have confirmed the positive associations between greenspaces and physical activity, evidence from natural experiments is scarce, especially for large-scale greenspace interventions. In addition, it is unclear how the physical-activity-related benefits of a greenspace intervention vary with distance from residences to greenspaces. We used a natural experimental approach to explore the impact on physical activity of a large-scale greenway intervention, namely the East Lake greenway, in Wuhan, China. Two waves of survey data (before and after the intervention in 2016 and 2019, respectively) were collected from 1020 participants residing in 52 neighbourhoods at different distances (0-1, 1-2, 2-3, 3-4, and 4-5 km) from the 102-km-long greenway. The results obtained using difference-in-difference models indicated that the greenway intervention had positive effects on both moderate-to-vigorous physical activity (MVPA) and overall physical activity (MET-minutes/week) after controlling for individual and neighbourhood covariates. Furthermore, the physical activity benefits of the greenway intervention were found to decrease with increasing distance between the greenway and the participants' residences. Individuals living closer to this large-scale greenway accrued more substantial physical activity benefits. Our results, together with those of other natural experimental studies, suggest that large-scale greenspace interventions may provide long-term physical activity benefits to residents living in a wide geographic area.

Individuals have experienced various degrees of accessibility loss during the COVID-19 pandemic, which may consequently influence transport equity. However, conventional measurements of accessibility cannot capture individual experiences and perceptions of accessibility. Moreover, since many daily necessities and services can only be acquired online during the pandemic, the ease of using smartphone-based services play an essential role in people's everyday lives. Therefore, this paper investigates the relationship between the ease of using smartphone-based services, perceived accessibility, and perceived transport equity during the pandemic. Based on 186 family interviews, a panel survey with 569 respondents was conducted monthly from February to October 2020 in Kunming, China, and a three-wave cross-lagged panel model was developed to understand the causal relationship between the three constructs. The results indicate that the ease of using smartphone-based services dominantly influence transport equity in the early phase of the pandemic, but its effect faded after the lifting of travel restrictions. Perceived accessibility to services appears a sound indicator for transport equity in the new normal, but perceived accessibility and transport equity are not strongly associated when staying at home is perceived as desirable. Moreover, we found that contemporary practices of smartphone-based new mobility services only favour those who already have convenient access to services and have further excluded and marginalised disadvantaged populations, which urgently require policy interventions.

Objective: To investigate the feasibility and effectiveness of a novel remnant-preserving anterior cruciate ligament (ACL) reconstruction with bidirectional barbed suture. Methods: Between February 2014 and January 2016, 96 patients (96 knees) with ACL injury who met the inclusion criteria were recruited and randomly divided into 2 groups ( n=48). All patients underwent ACL reconstruction with autologous tendon. The tibial remnant was fixed with PDS-Ⅱ suture (control group) and bidirectional barbed suture (trial group). There was no significant difference in age, gender, injury side, the interval from injury to operation, and preoperative knee stability (KT-1000 test), International Knee Documentation Committee (IKDC) score, and Lysholm score ( P>0.05). The knee stability (KT-1000 test), IKDC score, Lysholm score, proprioception, and Cyclops lesion after operation were recorded. Results: All incisions healed by first intention in both groups. Forty-four patients in control group were followed up 26-47 months (mean, 36.6 months), and 45 patients in trial group were followed up 26-48 months (mean, 35.6 months). At last follow-up, the IKDC score, Lysholm score, and KT-1000 test were significantly improved when compared with preoperative ones in both groups ( P<0.05); but no significant difference was found between 2 groups ( P>0.05). There was no significant difference in proprioception between 2 groups at 1 and 2 years after operation, and between 1 year after operation and 2 years after operation ( P>0.05) in the same group. The incidences of Cyclops lesion at 6 months, 1 year and 2 years after operation were 0, 4.44% (2/45), and 4.44% (2/45) in trial group, and 13.6% (6/44), 13.6% (6/44), 20.5% (9/44) in control group, showing significant differences between 2 groups at 6 months and at 2 years after operation ( P=0.012; P=0.022) and no significant difference at 1 year after operation ( P=0.157). Conclusion: The remnant-preserving ACL reconstruction with bidirectional barbed suture can decrease the incidence of Cyclops lesions. However, this technique can not improve the effectiveness in terms of the clinical function and stability compared with the traditional technique.

Adverse lifestyle-associated health outcomes, and stroke in particular, have been aggravated in transitional countries under high-speed urbanisation. Against this backdrop, deciphering the nexus between built environments (BEs) and lifestyle-associated health outcomes is of importance for crafting proactive interventions. The existing literature on this topic, however, fails to sufficiently capture the multiplicity of health-related BEs and, in turn, the complexity of such a nexus, largely challenging the applicability of established frameworks and the reliability of relevant findings. Looking at the case of stroke in Wuhan, China, this research aims to flesh out the understanding of the nexus between multidimensional BEs and lifestyle-associated health outcomes in transitional cities, with regards to conceptual framework and empirical evidence. To this end, we clarified stroke-related BE elements and integrated them into one conceptual framework. We then visualised stroke risk and examined its BE determinants using the Bayesian conditional autoregressive model. The visualisation results showed that stroke risks exhibited significant clustering in the high-density urban core. The statistical analysis found that, after the data were controlled for sociodemographic characteristics, net population density and building density were positively associated with stroke risk. In contrast, an abundance of public parks and institutional land use and access to medical care facilities have presented negative correlations with stroke risk, regardless of urban density. Our research reveals that compact urban developments might not be a silver bullet for health promotion in transitional cities, calling for an urgent need to scrutinise their applicability. To offset the adverse effects of increasingly dense urban environments, more efforts should also be made to provide better access to the identified salubrious resources. Furthermore, we argue that the establishment of comprehensive conceptual frameworks that connect BEs and lifestyle-associated health outcomes deserves to be highlighted in further research, planning intervention schemes, and health impact assessment projects.
