The effect of anesthesia/euthanasia with ethyl 3-aminobenzoate methanesulfonate (MS-222) on the oxidative status of Hyla arborea tadpoles was examined to determine whether the use of the anesthetic can confound the experimental results of the oxidative stress-based investigation. The experiment was conducted on two groups of tadpoles reared at different temperatures to produce differences in antioxidant capacity between the groups. After development at different temperatures (20 °C and 25 °C), the animals were exposed to different concentrations of MS-222 (0, 0.1, 1, and 5 g/L) for 15 min. The higher temperature decreased catalase activity, glutathione and protein carbonyl levels and increased glutathione reductase activity. The glutathione level and glutathione/thiol-related parameters were significantly changed after MS-222 exposure. However, individuals from the different temperature groups responded differently to the tested anesthetic, pointing to the possible influence of the initial levels of antioxidant capacity. The analysis of the interaction between the factors (temperature and MS-222) confirmed that the anesthetic can confound the results regarding the effects of temperature on the oxidative status parameters. The concentration of 0.1 g/L MS-222 had the lowest influence on the alterations in oxidative status and the results of the effect of temperature. A brief review of the current literature on the use of MS-222 in tadpoles made clear the absence of precise information on anesthetic concentration and exposure time. Similar studies should be repeated and extended to other amphibian species and other factors of interest to provide better guidance on tadpole anesthesia/euthanasia for future experiments that consider oxidative status parameters.

Amphibians and reptiles are the most threatened vertebrates by traffic, especially near ponds and wetlands. The main aim of this study was to examine seasonal and spatial patterns of amphibian and reptile road mortality in Special Nature Reserve "Obedska bara" (Serbia). We chose a road section of 4.2 km near the largest wetland in Serbia, with two different habitat types (forest-pond and agricultural area-pond). During 2018 (32 fieldwork days) and 2019-2020 (three control fieldwork days), we found 20,457 and 2231 road-killed animals, respectively. We recorded nine amphibian and eight reptile species. Amphibians (93%) were more vulnerable to traffic than reptiles (7%). The number of killed amphibians and reptiles varied between the seasons. Generally, amphibian roadkill was most frequent during the summer, whereas reptile roadkill was most frequent in spring and at the end of the summer. Furthermore, different roadkill patterns were observed during the examined months. In addition, we found that habitat type significantly affects the vulnerability of animals towards the roads. For both amphibians and reptiles, there were significant aggregations of roadkill, and most hotspots were located at the forest pond habitat type where mitigation measures must be applied (construction of tunnels and fences).

This study evaluated the potential of antitumor activity of snake venom from Vipera ammodytes and L-amino acid oxidase from Crotalus adamanteus on different colorectal cancer cell lines through determination of cytotoxic activity by MTT assay, pro-apoptotic activity by acridine orange/ethidium bromide staining, and concentrations of redox status parameters (superoxide, reduced glutathione, lipid peroxidation) by colorimetric methods. The expression of genes involved in the biotransformation process and metabolite efflux was determined by qPCR method, while protein expression of glutathione synthetase and P-glycoprotein were analysed by immunocytochemistry. The analysis of cell death shows that snake venom dominantly leads cells to necrosis. Induction of apoptosis by L-amino acid oxidase was in correlation with oxidative disbalance in cancer cells. Gene expression profile of membrane transporters and CYP genes were different in each cell line and in correlation with their sensitivity of treatment. Our results show that L-amino acid oxidase from snake venom is a potent cytotoxic substance with pronounced pro-apoptotic activity. The inhibition of P-glycoprotein suggests that L-amino acid oxidase is a good substance for furter research of antitumor effect, with unexpressed potential for occurrence of drug resistance in vitro.

AbstractTheoretically, animals integrate intrinsic and extrinsic factors to respond appropriately to the wide range of stressors they encounter during their life span. We examined how stress response varies between sexes and among morphotypes in wild dice snakes (Natrix tessellata). We also considered reproductive and feeding status and antipredator behavior. We used two indicators of stress (glucose [GLUC] and corticosterone [CORT] levels) at eight sampling time intervals (immediately after capture, up to 17 h after) and a large sample size (N=113 snakes). Concentrations of both markers increased sharply after capture (an equivalent of predation). This acute phase occurred earlier for GLUC (30 min) compared to CORT (60 min). Then the values plateaued to very high levels without decline over time, indicating prolonged saturation of the hypothalamus-pituitary-adrenal axis. In contrast to our expectations, we found no effect of sex, morphotype, or reproductive status. Yet the CORT stress response of those individuals displaying death-feigning (DF) antipredator behavior was attenuated compared to those that did not. Low stress hormones levels may facilitate the expression of DF (high levels supporting fleeing behavior). The presence of partially digested material in the stomach was associated with higher blood GLUC during the plateau. Assaying blood GLUC requires very little blood but was as good as CORT at gauging acute stress response. The prolonged plateau suggests that captivity should be minimized during field studies.

In this study we examined possible differences in heavy metal accumulation and oxidative stress parameters in the liver and muscle of two semi-aquatic snakes: grass snake (Natrix natrix) and dice snake (N. tessellata), that inhabit the same environment but differ in prey diversity. The obtained results revealed some interspecies, inter-tissue, prey-snake and prey-prey differences in heavy metal concentrations. Grass snakes pray contained significantly higher concentrations of Al, Cr and Fe as compared to food of dice snakes. Both investigated snakes accumulated generally lower concentrations of metals than their prey, indicating that they are not at risk of contaminant biomagnification. A significant interspecies difference in accumulation was observed only for Cu and Mn concentrations. On the other hand, analysis of oxidative stress biomarkers showed clear differences between the investigated snake species and the two investigated tissues. The liver of grass snake had increased superoxide dismutase, glutathione reductase and glutathione-S-transferase activities in comparison to dice snake. In muscle, a reverse trend was observed for the activities of these three enzymes, as well as for glutathione peroxidase activity. The higher number of significant correlations observed between oxidative stress biomarkers and heavy metal concentrations in grass snake points to upregulation of the antioxidative system (AOS), which resulted in a lower TBARS concentration. Results show that while the investigated snake species did not differ significantly in the accumulated metals, their defense mechanisms were different. This reveals the complexity of the AOS and points to the cooperation of different AOS components in individuals from natural populations.

We investigated in the liver of dice snakes during pre- and post-hibernation changes in the following antioxidant parameters: total, manganese and copper zinc containing superoxide dismutases (Tot SOD, MnSOD, CuZn SOD, respectively), catalase (CAT), glutathione peroxidase (GSH-Px), glutathione reductase (GR) and the concentrations of total glutathione (GSH) and sulfhydryl groups (-SH). In addition, we examined the expression of phase I biotransformation enzyme cytochrome P4501A (CYP1A) and the activity of phase II biotransformation enzyme glutathioneS-transferase (GST), the level of lipid peroxidation (by measuring the thiobarbituric acid-reactive substances (TBARS)), cholinesterase activity (ChE) and metallothionein expression (MT). We also measured the concentrations of heavy metals, including Al, Cd, As, Co, Cr, Cu, Fe, Mn, Mo, Ni, Pb and Zn in the water and snake liver during both periods. During the post-hibernation period, the activities of Tot SOD, CuZn SOD and GST and the concentration of GSH were significantly decreased, while GSH-Px and GR activities, the concentrations of -SH groups and TBARS were significantly increased. The activities of Mn SOD, CAT and ChE, and the relative amounts of CYP1A and MT did not significantly change during the investigated periods. The observed differences in the examined parameters probably represent adaptive physiological responses to sudden changes in tissue oxygenation during arousal from hibernation. Our findings also indicate that the accumulated metals modulated the responses of the examined parameters during the investigated periods.

PURPOSE: Extremely low frequency (ELF) magnetic fields as essential ecological factors may induce specific responses in genetically different lines. The object of this study was to investigate the impact of the ELF magnetic field on fitness components and locomotor activity of five Drosophila subobscura isofemale (IF) lines. MATERIALS AND METHODS: Each D. subobscura IF line, arbitrarily named: B16/1, B24/4, B39/1, B57/2 and B69/5, was maintained in five full-sib inbreeding generations. Their genetic structures were defined based on the mitochondrial DNA variability. Egg-first instar larvae and 1-day-old flies were exposed to an ELF magnetic field (50 Hz, 0.5 mT, 48 h) and thereafter, fitness components and locomotor activity of males and females in an open field test were observed for each selected IF line, respectively. RESULTS: Exposure of egg-first instar larvae to an ELF magnetic field shortened developmental time, and did not affect the viability and sex ratio of D. subobscura IF lines. Exposure of 1-day-old males and females IF lines B16/1 and B24/4 to an ELF magnetic field significantly decreased their locomotor activity and this effect lasted longer in females than males. CONCLUSIONS: These results indicate various responses of D. subobscura IF lines to the applied ELF magnetic field depending on their genetic background.

We investigated the phenolic composition, radical scavenging activity, and antimicrobial activity of grapevine leaf extracts from two red grape varieties, Vranac and Merlot (Vitis vinifera L.). The extracts were prepared from healthy grapevine leaves and those infected by Plasmopara viticola (downy mildew). The phenolic composition of the grapevine leaf extracts was determined using spectrophotometric assays and reverse-phase high performance liquid chromatography (RP-HPLC). The radical scavenging activity of grapevine leaf extracts was determined by the 2,2-diphenyl-1-picrylhydrazyl assay, and their antimicrobial activity was determined by microwell dilution tests. The total phenolic content was higher in healthy grapevine leaf extracts than in infected grapevine leaf extracts. The RP-HPLC analysis detected significant amounts of flavonols, phenolic acids, and flavan-3-ols, and small amounts of stilbenes in the grapevine leaf extracts. Compared with the infected grapevine leaf extracts, the healthy grapevine leaf extracts were richer in flavonols, phenolic acids, and flavan-3-ols, but had lower stilbenes contents. All extracts showed strong free radical scavenging activity, which was strongly correlated with the total phenolic content (R(2) = 0.978). The extracts showed a stronger antimicrobial activity towards Gram-positive bacterial strains than towards Gram-negative bacterial strains and yeast. The phenolic compounds in grapevine leaves were responsible for their strong radical scavenging and antimicrobial activities. Together, these results demonstrate that grapevine leaves have high nutritional value and can be used as a fresh food and to prepare extracts that can be used as additives in food and medicines.

BACKGROUND: A number of recent studies have shown that the pattern of mitochondrial DNA variation and evolution is at odds with a neutral equilibrium model. Theory has suggested that selection on mitonuclear genotypes can act to maintain stable mitonuclear polymorphism within populations. However, this effect largely relies upon selection being either sex-specific or frequency dependent. Here, we use mitonuclear introgression lines to assess differences in a series of key life-history traits (egg-to-adult developmental time, viability, offspring sex-ratio, adult longevity and resistance to desiccation) in Drosophila subobscura fruit flies carrying one of three different sympatric mtDNA haplotypes. RESULTS: We found functional differences between these sympatric mtDNA haplotypes, but these effects were contingent upon the nuclear genome with which they were co-expressed. Further, we demonstrate a significant mitonuclear genetic effect on adult sex ratio, as well as a sex × mtDNA × nuDNA interaction for adult longevity. CONCLUSIONS: The observed effects suggest that sex specific mitonuclear selection contributes to the maintenance of mtDNA polymorphism and to mitonuclear linkage disequilibrium in this model system.

Local adaptation to environmental stress at different levels of genetic polymorphism in various plants and animals has been documented through evolution of heavy metal tolerance. We used samples of Drosophila subobscura populations from two differently polluted environments to analyze the change of chromosomal inversion polymorphism as genetic marker during laboratory exposure to lead. Exposure to environmental contamination can affect the genetic content within a particular inversion and produce targets for selection in populations from different environments. The aims were to discover whether the inversion polymorphism is shaped by the local natural environments, and if lead as a selection pressure would cause adaptive divergence of two populations during the multigenerational laboratory experiment. The results showed that populations retain signatures from past contamination events, and that heavy metal pollution can cause adaptive changes in population. Differences in inversion polymorphism between the two populations increased over generations under lead contamination in the laboratory. The inversion polymorphism of population originating from the more polluted natural environment was more stable during the experiment, both under conditions with and without lead. Therefore, results showed that inversion polymorphism as a genetic marker reflects a strong signature of adaptation to the local environment, and that historical demographic events and selection are important for both prediction of evolutionary potential and long-term viability of natural populations.

PURPOSE: Extremely low frequency (ELF) magnetic fields are essential ecological factors which may induce changes in many organisms. The aim of this study was to examine the effects in Drosophila subobscura exposed for 48 h to ELF magnetic field (50 Hz, 0.5 mT) at different developmental stages. MATERIALS AND METHODS: Egg-first instar larvae developmental stage of D. subobscura isofemale lines was exposed to ELF magnetic field, and fitness components (developmental time, developmental dynamics, viability and sex ratio) and locomotor activity of three-day-old males and females were monitored. Also, just eclosed D. subobscura isofemale adults were exposed to ELF magnetic field and their locomotor activity was monitored just after. RESULTS: ELF magnetic field shortens developmental time, increases viability and does not affect sex ratio of D. subobscura. No matter which developmental stage is exposed, ELF magnetic field significantly decreases locomotor activity of adult flies, but after exposure of just eclosed adults observed change lasts longer. CONCLUSIONS: Applied ELF magnetic field modifies fitness components and locomotor activity of D. subobscura. Observed effects can be attributed to the influence of magnetic field on different stages of development where the hormonal and nervous systems play important role in the control of examined parameters.

Differences in heavy metal tolerance among separate populations of the same species have often been interpreted as local adaptation. Persistence of differences after removing the stressor indicates that mechanisms responsible for the increased tolerance were genetically determined. Drosophila subobscura Collin (Diptera: Drosophilidae) populations were sampled from two localities with different history of heavy metal pollution, and reared for eight generations in the laboratory on a standard medium and on media with different concentrations of lead (Pb). To determine whether flies from different natural populations exposed to the Pb-contaminated media in the laboratory show population specific variability in fitness components over generations, experimental groups with different concentrations of lead were assayed in three generations (F2 , F5 , and F8 ) for fecundity, developmental time, and egg-to-adult viability. On the contaminated medium, fecundity was reduced in later generations and viability was increased, irrespective of the environmental origin of populations. For both populations, developmental time showed a tendency of slowing down on media with lead. Faster development was observed in later generations. Preadaptation to contamination, meaning higher fecundity, higher viability, and faster egg to adult development in all studied generations, was found in D. subobscura originating from the locality with a higher level of heavy metal pollution.

ETHNOPHARMACOLOGICAL RELEVANCE: Gentiana lutea L., the yellow gentian, is herb known for its pharmacological properties, with a long tradition of use for the treatment of a variety of diseases including the use as a remedy for digestion, also in food products and in bitter beverages. The aim of the present study is to evaluate, for the first time, genotoxicity of gentian alone, and its antigenotoxicity against methyl methanesulfonate (MMS). MATERIALS AND METHODS: The water infusion of the underground part of gentian were evaluated in vivo using the Drosophila wing spot test, at the dose commonly used in traditional medicine. For antigenotoxic study two types of treatment with gentian and MMS were performed: chronic co-treatment, as well as post-treatment with gentian after acute exposure with MMS. RESULTS: Water infusion of gentian alone did not exhibit genotoxicity. The results of co- and post-treatment experiments with gentian show that gentian enhanced the frequency of mutant clones over the values obtained with MMS alone, instead of reducing the genotoxicity of MMS, for 22.64% and 27.13% respectively. CONCLUSIONS: This result suggests a synergism of gentian with MMS, and indicates that water infusion of gentian used in traditional medicine may have particular effects with regard to genotoxicity indicating careful use.

Ixodes ricinus Linnaeus (Acari: Ixodidae) ticks are vectors of numerous infectious diseases in humans and animals. The allozyme variability of MDH and alpha-Gpdh was detected by native polyacrylamide gel electrophoresis in I. ricinus natural populations in three localities in Serbia. Four alleles of Mdh locus (MDH 1, MDH 2, MDH 3 and MDH X) and four alleles of alpha-Gpdh locus (VS, S, F and VF) were detected. Interpopulation differences in Mdh and alpha-Gpdh allele frequencies were statistically insignificant. Significant difference in alpha-Gpdh allele frequencies between males and females was recorded in the largest sample only. Differences in allele frequencies, detected between borreliae-infected and uninfected I. ricinus ticks, were close to the level of statistical significance, especially for alpha-Gpdh locus. Clear significant difference appeared in females when sexes were tested separatelly (P = 0.037). It is interesting that genotypes containing rarer alleles (MDH 1 and S) were infected in higher proportion in comparison to other genotypes. Our results point towards a possible role of Mdh and alpha-Gpdh loci in I. ricinus ticks in the determination of energy requirements for host seeking.
