RESUMEN

Macroalgal bloom events have been frequent in recent years. Eutrophication and overexploitation fishing may favor blooms through nutrient availability and capturing top predators. We aim to investigate the drivers of the macroalgae blooms and their consequences on the food web of the two tropical coastal ecosystems: Porto do Mangue (with high macroalgae production) and Baía Formosa (control environment, without macroalgae), both exploited by artisanal fisheries in northeastern Brazil. The food webs are modeled using the Ecopath with Ecosim (EwE) approach. Our results suggest that fishing did not favor macroalgae blooms but rather the high concentration of nutrients added to the semi-arid conditions. Furthermore, the macroalgae bloom showed low trophic impact, so much of their biomass is transferred into detritus. However, when it decomposes, this accumulation of matter alters the structure and functioning of the ecosystem, affecting its main fish resources: shrimp and piscivorous fish. Investigating blooms is key to management.

Coastal marine ecosystems have structural and functional features usually connected by the seasonal transfer of nutrients and organisms. These environments can utilize inter-ecosystem subsidies to increase resilience and maturity and support human activities like fishing. However, the importance of the connection and the role of the seasonal pulse of energy flows to enhance maturity are still poorly understood and reported. Our objective in this paper is to assess the effect of seasonal hydrological pulses on two tropical coastal interconnected ecosystems. Thus, we made four Ecopath models for estuarine and neritic environments considering the dry and rainy seasons, with a similar sampling design that allowed them to be compared. Our results provide evidence for the occurrence of the pulsed ecosystems since both environments seem driven by the river flow. Estuary presents more and more substantial differences (measured by ecosystem attributes) in both seasons because it is directly affected by river floods than the neritic environment. The neritic is affected indirectly by the movement of species from the estuary and by a weaker river flow. In the dry season, the differences between ecosystems are lower because the dry season trend to homogenize cycling, maturity, homeostasis, and resilience. We found that the seasonal river flow (pulse) forces the variability of biomass, flows, and ecosystem features, and this variance creates the required stability for both ecosystems. Still, these environments benefit through the exchange of components that relieve the pressures of predation on specific groups and maintain the energy flow necessary for the functioning of their trophic webs. The pulse by the rainfall favors connectivity and equalizes the two systems, increasing the connectivity between them and the exchange of subsidies that strengthens the trophic structures, contributing to the increase in maturity. In these ecosystems, seasonal changes become a key factor for exchanging flows that will promote sustainability, the accumulation of more biomass (growth), and the optimization of reserve energy (development) in both systems. This efficient joint strategy of perpetuation is what promotes resistance and resilience to these ecosystems, which together can reach different states of equilibrium, translated into maturity to withstand new environmental changes.

Dynamic Light Scattering (DLS) and Small-Angle Neutron Scattering (SANS) are two key tools to probe the dynamic and static structure factors, respectively, in soft matter. Usually, DLS and SANS measurements are performed separately, in different laboratories, on different samples, and at different times. However, this methodology has particular disadvantages for a large variety of soft materials, which exhibit a high sensitivity to small changes in fundamental parameters, such as waiting times, concentration, pH, and ionic strength. Here, we report on a new portable DLS-SANS apparatus that allows one to simultaneously measure both the microscopic dynamics (through DLS) and the static structure (through SANS) on the same sample. The apparatus has been constructed as a collaboration between two laboratories, each an expert in one of the scattering methods, and was commissioned on the LOQ and ZOOM SANS instruments at the ISIS Pulsed Neutron and Muon Source, U.K.

Interpenetrated polymer network microgels, composed of crosslinked networks of poly(N-isopropylacrylamide) and polyacrylic acid (PAAc), have been investigated through rheological measurements at four different amounts of PAAc. Both PAAc content and crosslinking degree modify particle dimensions, mass and softness, thereby strongly affecting the volume fraction and the system viscosity. Here the volume fraction is derived from the flow curves at low concentrations by fitting the zero-shear viscosity with the Einstein-Batchelor equation which provides a parameterkto shift weight concentration to volume fraction. We find that particles with higher PAAc content and crosslinker are characterized by a greater value ofkand therefore by larger volume fractions when compared to softer particles. The packing fractions obtained from rheological measurements are compared with those from static light scattering for two PAAc contents revealing a good agreement. Moreover, the behaviour of the viscosity as a function of packing fraction, at room temperature, has highlighted an Arrhenius dependence for microgels synthesized with low PAAc content and a Vogel-Fulcher-Tammann dependence for the highest investigated PAAc concentration. A comparison with the hard spheres behaviour indicates a steepest increase of the viscosity with decreasing particles softness. Finally, the volume fraction dependence of the viscosity at a fixed PAAc and at two different temperatures, below and above the volume phase transition, shows a quantitative agreement with the structural relaxation time measured through dynamic light scattering indicating that interpenetrated polymer network microgels softness can be tuned with PAAc and temperature and that, depending on particle softness, two different routes are followed.

Hyaluronan (HA) is among the most used biopolymers for viscosupplementation and dermocosmetics. However, the current injectable HA-based formulations present relevant limitations: I) unmodified HA is quickly degraded by endogenous hyaluronidases (HAase), resulting in short lasting properties; II) cross-linked HA, although shows enhanced stability against HAase, often contains toxic chemical cross-linkers. As such, herein, we present biocompatible self-assembled hyaluronan-cholesterol nanohydrogels (HA-CH NHs) able to bind to HAase and inhibit the enzyme activity in vitro, more efficiently than currently marketed HA-based cross-linked formulations (e.g. Jonexa™). HA-CH NHs inhibit HAase through a mixed mechanism, by which NHs bind to HAase with an affinity constant 7-fold higher than that of native HA. Similar NHs, based on gellan-CH, evidenced no binding to HAase, neither inhibition of the enzyme activity, suggesting this effect might be due to the specific binding of HA-CH to the active site of the enzyme. Therefore, HA-CH NHs were engineered into injectable hybrid HA mixtures or physical hydrogels, able to halt the enzymatic degradation of HA.

Pseudomonas syringae pv. syringae, a Gammaproteobacterium belonging to genomospecies 2 within the P. syringae complex, is distributed worldwide, and it is responsible for bacterial canker on >100 different hosts, including the grapevine. P. syringae pv. syringae induces necrotic lesions in the leaf blades, veins, petioles, shoots, rachis, and tendrils on grapevine cultivars in different areas. P. syringae pv. syringae has been associated with severe economic losses in different grape cultivars in Australia, where it causes inflorescence rot. In midsummer to late summer 2017, symptoms of berry rots differing from those caused by the common berry rots agents were observed in different cultivar Red Globe vineyards of Apulia (southern Italy). As proven by fulfillment of Koch's postulates, these symptoms were caused by a bacterium that, according to the results of biochemical, physiological, nutritional, antimicrobial activity, and pathogenicity tests and sequencing of 16S ribosomal DNA, gyrB, rpoB, and rpoD genes, was identified as P. syringae pv. syringae. This is the first report of Pseudomonas grapevine bunch rot.

Monilinia spp. are responsible for brown rot decay of stone and pome fruit in the field as well as in postharvest. Monilinia laxa and M. fructigena are considered indigenous to Europe, while M. fructicola is a quarantine pathogen in the European and Mediterranean Plant Protection Organization area included in the A2 List. In Italy, it was first reported in 2009 in Piedmont (northern Italy) and rapidly spread to central Italy. We carried out a monitoring program on the occurrence of Monilinia spp. in southern Italy and a comparative characterization of the three main fungal pathogens. Molecular assays based on direct polymerase chain reaction (PCR) and real-time quantitative PCR for molecular identification of Monilinia spp. from rotted fruit were set up, validated, and applied in a monitoring program. Of the tested 519 isolates from 26 orchards, 388 (74.8%) were identified as M. fructicola, 118 (22.7%) as M. laxa, 10 (1.9%) as M. fructigena, and 3 (0.6%) were M. polystroma. M. fructicola colonies grew faster and had a higher optimal temperature for growth (26°C) than M. laxa (23°C) and M. fructigena (20°C). No relevant difference in virulence could be observed on artificially inoculated apricot, cherry, and peach fruit. The fungal species showed different responses to fungicides, because M. fructicola was more sensitive than M. laxa, especially to cyflufenamid, and M. fructigena revealed a lower sensitivity to succinate dehydrogenase inhibitors (boscalid, fluopyram, and fluxapyroxad) and quinone outside inhibitors (mandestrobin). In summary, the two species M. fructicola and M. polystroma were first detected in southern Italy where M. fructicola has largely displaced the two indigenous pathogens M. laxa and M. fructigena; the relative proportions of the three pathogens in orchards should be considered when defining the management of brown rot of stone fruit due to differences in their responses to fungicides.

Alterations in aquatic systems and changes in water levels, whether due to rains or dam-mediated control can cause changes in community structure, forcing the community to readjust to the new environment. This study tested the hypothesis that there is an increase in the richness and abundance of aquatic insects during the rainy season in the Serra da Mesa Reservoir, with the premise that increasing the reservoir level provides greater external material input and habitat diversity, and, therefore, conditions that promote colonization by more species. We used the paired t test to test the differences in richness, beta diversity, and abundance, and a Non-metric Multidimensional Scaling (NMDS) was performed to identify patterns in the community under study. Additionally, Pearson correlations were analyzed between the richness, abundance, and beta diversity and the level of the reservoir. We collected 35,028 aquatic insect larvae (9,513 in dry period and 25,515 in the rainy season), predominantly of the Chironomidae family, followed by orders Ephemeroptera, Trichoptera, and Odonata. Among the 33 families collected, only 12 occurred in the dry season, while all occurred in the rainy season. These families are common in lentic environments, and the dominance of Chironomidae was associated with its fast colonization, their behavior of living at high densities and the great tolerance to low levels of oxygen in the environment. The hypothesis was confirmed, as the richness, beta diversity, and abundance were positively affected by the increase in water levels due to the rainy season, which most likely led to greater external material input, greater heterogeneity of habitat, and better conditions for colonization by several families.

Alterações nos sistemas aquáticos, bem como variações nos níveis da água, sejam devido a chuvas ou pelo controle exercido pela barragem podem provocar mudanças na estruturação das comunidades fazendo com que as mesmas sejam obrigadas a se readequar às novas características do ambiente. Este estudo testou a hipótese de que ocorre aumento na riqueza e abundância de insetos aquáticos na estação chuvosa no reservatório de Serra da Mesa com a premissa que o aumento do nível do reservatório proporciona maior entrada de material alóctone, maior diversidade de habitats e, consequentemente, condições para a colonização de mais espécies. Para testar as diferenças na riqueza, diversidade beta e abundância foi utilizado o test t pareado e, além disso, foi feita uma NMDS para identificar padrões na comunidade em estudo. Foram coletados 35.028 larvas de insetos aquáticos (9513 no período seco e 25.515 no período de cheia) com predominância da família Chironomidae seguida das famílias Ephemeroptera, Trichoptera e Odonata. Dentre as 33 famílias coletadas, apenas 12 ocorreram no período de seca, enquanto todas ocorreram no período chuvoso. Essas famílias são bastante encontradas em ambientes lênticos e a dominância de Chironomidae está associada ao seu rápido processo de colonização, ao seu comportamento de viver em altas densidades e sua grande tolerância a baixos teores de oxigênio no ambiente. A hipótese testada foi corroborada visto que a riqueza, a diversidade beta e a abundância de insetos aquáticos foram influenciadas positivamente pelo aumento do nível da água, decorrente do período chuvoso, que, provavelmente, propiciou uma maior entrada de material alóctone, maior heterogeneidade de habitat e melhores condições para a colonização das diversas famílias.

BACKGROUND: Psychological disorders are often associated with diseases of the upper digestive tract. Although emotions can influence gastrointestinal function in healthy individuals, psychological setting in upper gastrointestinal patients are unclear. We evaluate the psychological alterations prevalence in outpatients submitted to upper endoscopy. MATERIALS AND METHODS: A total of 130 patients (50 males and 80 females; mean age 54±17 years) submitted to upper gastrointestinal endoscopy, were enrolled over the period May 2009 - September 2010. Subjects were asked to complete questionnaires before endoscopic examination. Alexithymia, anxiety, depression and coping style were assessed using the Toronto Alexithymia Scale, Spielberger Trait Anxiety Inventory, Beck Depression Inventory and Coping Inventory for Stressful Situations, respectively. RESULTS: Coping impairment, Alexithymia, Anxiety and Depression were found respectively in 80.3%, 25.4%, 24.6% and 17.2%, often in association. Task-oriented, emotion-oriented and avoidance-oriented alterations were found in 41.8%, 40% and 30.6%, respectively. No correlations were demonstrated between diagnosis of upper gastrointestinal disease and psychometric results. CONCLUSIONS: In our study, a high prevalence of psychometric alterations in gastrointestinal outpatients was unconnected with endoscopic findings, especially considering coping style alterations. This aspect should be taken into account in patients management and a long-term follow-up should clarify a possible role of these factors in patients prognosis and compliance.

The process of dynamical arrest, leading to formation of different arrested states such as glasses and gels, along with the closely related process of aging, is central for both basic research and technology. Here we report on a study of the time-dependent structural evolution of two aqueous Laponite clay suspensions at different weight concentrations. Neutron diffraction experiments have been performed with the near and intermediate range order diffractometer (NIMROD) that allows studies of the structure of liquids and disordered materials over a continuous length scale ranging from 1 to 300 Å, i.e., from the atomistic to the mesoscopic scales. NIMROD is presently a unique diffractometer, bridging the length scales traditionally investigated by small angle neutron scattering or small angle x-ray scattering with that accessible by traditional diffractometers for liquids. Interestingly, we have unveiled a signature of aging of both suspensions in the length scale region of NIMROD. This phenomenon, ascribed to sporadic contacts between Laponite platelets at long times, has been observed with the sample arrested as gel or as repulsive glass. Moreover, water molecules within the layers closest to Laponite platelets surface show orientational and translational order, which maps into the crystalline structure of Laponite.

Polyamines (PAs) are nitrogenous molecules which play a well-established role in most cellular processes during growth and development under physiological or biotic/abiotic stress conditions. The molecular mode(s) of PA action have only recently started to be unveiled, and comprehensive models for their molecular interactions have been proposed. Their multiple roles are exerted, at least partially, through signalling by hydrogen peroxide (H(2)O(2)), which is generated by the oxidation/back-conversion of PAs by copper amine oxidases and PA oxidases. Accumulating evidence suggests that in plants the cellular titres of PAs are affected by other nitrogenous compounds. Here, we discuss the state of the art on the possible nitrogen flow in PAs, their interconnection with nitrogen metabolism, as well as the signalling roles of PA-derived H(2)O(2) during some developmental processes and stress responses.

OBJECTIVES: Following a Chikungunya (CHIKV) outbreak in Italy, a cohort study was conducted to describe the infection long-term clinical course and outcome. METHODS: Persons identified through active and passive surveillance as confirmed or possible CHIKV cases during the outbreak were enrolled and interviewed by trained public health nurses, between 4-5 and 12-13 months following the acute stage. Patients reporting persistent clinical symptoms were evaluated by rheumatologists. Serum samples were obtained and anti-CHIKV specific IgG and IgM immune responses detected. Only confirmed cases who completed the follow-up were analysed. RESULTS: Out of 250 patients, 66.5% still reported myalgia, asthenia or arthralgia (most frequent sign) after 12 months. Functional ability, measured by the ROAD index, was more impaired for lower extremities (3.75; Inter Quartile Range - IQR 4.4), and the activities of daily living (average 4.2; IQR 5). Variables independently associated with the presence of joint pain at 12-13 months were increasing age, and history of rheumatologic diseases). Elderly, females, and persons with history of rheumatologic diseases had higher anti-CHIKV IgG titres at 12-13 months. CONCLUSIONS: This study confirms, in an unselected population, that the long-lasting burden of CHIKV infection is significant.

Using experiments, theory and simulations, we show that the arrested state observed in a colloidal clay at high concentrations is stabilized by screened Coulomb repulsion (Wigner glass). Dilution experiments allow us to distinguish this disconnected state, which melts upon addition of water, from a low-concentration gel state, which does not melt. Theoretical modeling and simulations at high concentrations reproduce the measured small angle x-ray scattering static structure factors and confirm the long-range electrostatic nature of the arrested structure. These findings are attributed to the different time scales controlling the competing attractive and repulsive interactions.

In 2007, the first chikungunya outbreak ever occurring in a temperate area of the Northern Hemisphere was reported in Emilia-Romagna, Italy. The present study aims to evaluate knowledge, attitudes and practices regarding chikungunya infection. Information was collected by standardised telephone questionnaire, administered to 325 people during a seroprevalence survey performed in the outbreak area. Most people (61%) knew that Chikungunya virus is transmitted through mosquito bites. A significant proportion of respondents perceived chikungunya infection as a high-risk disease (49.8%) and declared their intention to use mosquito repellents in the future (47.4%). Willingness to use skin repellents was more often declared by women, residents in households with children and people who knew infection transmission routes, while perception of high risk related to Chikungunya did not influence claimed future behaviour. Knowledge of the mechanisms of infection transmission was inversely related to risk perception: elderly people were less likely to know the disease than young people, but ranked the risk highest compared to younger age groups. Less educated residents were much more likely to perceive a high risk. After a chikungunya infection outbreak in a temperate area, where mosquito-borne infections have not been previously reported, only half of the study participants declared their intention to use individual protection in the future. Anticipated behaviour was not explained by the level of risk perception, but was associated with knowledge of the disease and demographic characteristics.

The phase diagram of alpha -cyclodextrin/water/4-methylpyridine solutions, a system undergoing inverse melting, has been studied by differential scanning calorimetry, rheological methods, and x-ray diffraction. Two different fluid phases separated by a solid region have been observed in the high alpha -cyclodextrin concentration range (c > or =150 mg/ml) . Decreasing c , the temperature interval where the solid phase exists decreases and eventually disappears, and a first-order phase transition is observed between the two different fluid phases.

The aging of a charged colloidal system has been studied by small-angle x-ray scattering, in the exchanged momentum range Q=0.03-5 nm(-1) , and by dynamic light scattering, at different clay concentrations (C(w)=0.6-2.8%) . The static structure factor S(Q) has been determined as a function of both aging time and concentration. This is the direct experimental evidence of the existence and evolution with aging time of two different arrested states in a single system simply obtained only by changing its volume fraction: an inhomogeneous state is reached at low concentrations, while a homogeneous one is found at high concentrations.

BACKGROUND: Chikungunya virus (CHIKV), which is transmitted by Aedes spp mosquitoes, has recently caused several outbreaks on islands in the Indian Ocean and on the Indian subcontinent. We report on an outbreak in Italy. METHODS: After reports of a large number of cases of febrile illness of unknown origin in two contiguous villages in northeastern Italy, an outbreak investigation was done to identify the primary source of infection and modes of transmission. An active surveillance system was also implemented. The clinical case definition was presentation with fever and joint pain. Blood samples were gathered and analysed by PCR and serological assays to identify the causal agent. Locally captured mosquitoes were also tested by PCR. Phylogenetic analysis of the CHIKV E1 region was done. FINDINGS: Analysis of samples from human beings and from mosquitoes showed that the outbreak was caused by CHIKV. We identified 205 cases of infection with CHIKV between July 4 and Sept 27, 2007. The presumed index case was a man from India who developed symptoms while visiting relatives in one of the villages. Phylogenetic analysis showed a high similarity between the strains found in Italy and those identified during an earlier outbreak on islands in the Indian Ocean. The disease was fairly mild in nearly all cases, with only one reported death. INTERPRETATION: This outbreak of CHIKV disease in a non-tropical area was to some extent unexpected and emphasises the need for preparedness and response to emerging infectious threats in the era of globalisation.
