RESUMEN

The present study describes the development of a laboratory-scale waste stabilisation pond (WSP) system, undertaken in order to investigate the effects of hydraulic, physicochemical, microbial and physical parameters on wastewater treatment. Previous studies have focused predominantly on hydraulic characteristics. This system was engineered at a scale much smaller than had previously been seen in the literature. The scale of the model used here allows for improved optimisation at a shorter time scale that would be seen for larger pilot-scale systems. Additionally, with the addition of viruses, a smaller scale model allows for more control over viral concentration used. Once constructed, the system was dosed with wastewater from a wastewater treatment plant and both the influent and effluent were monitored using common testing methods as well as direct viral analysis. Successful wastewater treatment was seen in terms of reduction of indicator bacteria and virus, as determined by culture-based methods. This treatment and the associated stabilisation of physicochemical parameters such as dissolved oxygen and pH, indicates the successful development of a microbial community within the laboratory-scale WSP.

RESUMEN

Global environmental change has degraded ecosystems. Challenges such as climate change, resource depletion (with its huge implications for human health and wellbeing), and persistent social inequalities in health have been identified as global public health issues with implications for both communicable and noncommunicable diseases. This contributes to pressure on healthcare systems, as well as societal systems that affect health. A novel strategy to tackle these multiple, interacting and interdependent drivers of change is required to protect the population's health. Public health professionals have found that building strong, enduring interdisciplinary partnerships across disciplines can address environment and health complexities, and that developing Environmental and Public Health Tracking (EPHT) systems has been an effective tool. EPHT aims to merge, integrate, analyse and interpret environmental hazards, exposure and health data. In this article, we explain that public health decision-makers can use EPHT insights to drive public health actions, reduce exposure and prevent the occurrence of disease more precisely in efficient and cost-effective ways. An international network exists for practitioners and researchers to monitor and use environmental health intelligence, and to support countries and local areas toward sustainable and healthy development. A global network of EPHT programs and professionals has the potential to advance global health by implementing and sharing experience, to magnify the impact of local efforts and to pursue data knowledge improvement strategies, aiming to recognise and support best practices. EPHT can help increase the understanding of environmental public health and global health, improve comparability of risks between different areas of the world including Low and Middle-Income Countries (LMICs), enable transparency and trust among citizens, institutions and the private sector, and inform preventive decision making consistent with sustainable and healthy development. This shows how EPHT advances global health efforts by sharing recent global EPHT activities and resources with those working in this field. Experiences from the US, Europe, Asia and Australasia are outlined for operating successful tracking systems to advance global health.

Internationally several phthalates are subject to regulatory control regarding maximum allowable concentrations in children's toys. Such regulation is not in place in New Zealand. Phthalates have been associated with developmental toxicity and endocrine disruption. We determined the concentration of seven phthalates in children's toys purchased in Christchurch, New Zealand. These results provided data for an exposure assessment deriving Hazard Indices (HI) for oral and dermal exposure routes in children, based on the concentration of mixtures of phthalates shown by the EU to produce either reproductive/developmental or hepatotoxic effects. Of the 49 toys analyzed, 65% contained at least one phthalate at a concentration of >0.1% by mass; and 35% contained multiple-phthalates at individual concentrations of >0.1%. A HI of 3.4 was derived for the combined exposures to the four phthalates associated with reproductive and developmental effects. A HI of 0.3 was derived for the group of phthalates associated with hepatotoxic effects. Five phthalates were detected at levels exceeding the EU regulatory limit of 0.1% by mass. Risk assessment calculations indicate that, using realistic exposure scenarios, the worst-case combined exposure to phthalates associated with developmental toxicity exceeded a HI of 1 so may cause adverse developmental effects.

Cold-induced sweetening (CIS) is a serious post-harvest problem for potato tubers, which need to be stored cold to prevent sprouting and pathogenesis in order to maintain supply throughout the year. During storage at cold temperatures (below 10 °C), many cultivars accumulate free reducing sugars derived from a breakdown of starch to sucrose that is ultimately cleaved by acid invertase to produce glucose and fructose. When affected tubers are processed by frying or roasting, these reducing sugars react with free asparagine by the Maillard reaction, resulting in unacceptably dark-coloured and bitter-tasting product and generating the probable carcinogen acrylamide as a by-product. We have previously identified a vacuolar invertase inhibitor (INH2) whose expression correlates both with low acid invertase activity and with resistance to CIS. Here we show that, during cold storage, overexpression of the INH2 vacuolar invertase inhibitor gene in CIS-susceptible potato tubers reduced acid invertase activity, the accumulation of reducing sugars and the generation of acrylamide in subsequent fry tests. Conversely, suppression of vacuolar invertase inhibitor expression in a CIS-resistant line increased susceptibility to CIS. The results show that post-translational regulation of acid invertase by the vacuolar invertase inhibitor is an important component of resistance to CIS.
