RESUMEN

Pseudomonas aeruginosa vaccine (PV) containing cell proteins with molecular weight (Mr) 20,000-100,000 and up to 0.08% (w/v) admixture of lipopolysaccharide was obtained by water-salt extraction and subsequent ultrafiltration. PV protects mice against experimental P. aeruginosa infection, stimulates production of specific protective antibodies in rabbit and does not provoke obvious toxicity in laboratory animals.

RESUMEN

After the injection of P. aeruginosa live culture under the burned skin of mice sepsis develops within the first 24 hours, finally leading to the death of the animals. The microorganisms can be isolated from the blood, liver, kidneys and mesenterial lymph nodes till day 3 and from the spleen till day 5. After the intraperitoneal injection of P. aeruginosa live culture into mice, sepsis also develops within 24 hours, and the culture can be isolated from the blood and parenchymatous organs till day 3. The LD50 of the culture is equal to 5.1 X 10(6) microbial cells when introduced intraperitoneally and to 30 microbial cells in experimental burn sepsis. Experimental burn sepsis clearly demonstrates the effectiveness of Pseudomonas acellular protein vaccine: its index of effectiveness exceeds 3,000.

RESUMEN

The electron microscopic study of 175 P. aeruginosa museum and newly isolated strains has been made and their virulence has been determined. In 32% of these strains the presence of pili has been established. However, among the museum strains those with pili have been found to constitute only 19.5%, while among newly isolated strains pili have been detected in 39.7% of cases (the difference is statistically significant at p less than 0.01). No correlation between the formation of pili and the virulence of P. aeruginosa has been established.

RESUMEN

Two variants of cell-free protein vaccine have been prepared from the mixture of 4 P. aeruginosa strains, serovars 02, 06, 07 and 011, and from a single P. aeruginosa strain, serovar 02. The preparation contains proteins with molecular weight ranging from 20,000 to 100,000 and the admixture of lipopolysaccharide in negligible amounts (not exceeding 0.08% of dry weight). The vaccine produces no signs of toxicosis in laboratory animals. The vaccine effectively protects mice challenged with P. aeruginosa of different O-serotypes and stimulates the formation of specific protective antibodies in rabbits.
