RESUMEN

New members of the influenza A virus genus have been detected recently in bats from South America. By molecular investigations, using a generic real-time RT-PCR (RT-qPCR) that detects all previously known influenza A virus subtypes (H1-H16) and a newly developed RT-qPCR specific for the South American bat influenza-like virus of subtype H17, a total of 1571 samples obtained from 1369 individual bats of 26 species from Central Europe were examined. No evidence for the occurrence of such influenza viruses was found. Further attempts towards a more comprehensive evaluation of the role of bats in the ecology and epidemiology of influenza viruses should be based on more intense monitoring efforts. However, given the protected status of bats, not only in Europe, such activities need to be embedded into existing pathogen-monitoring programs.

While Pseudogymnoascus destructans has been responsible for mass bat mortalities from white-nose syndrome (WNS) in North America, its virulence in Europe has been questioned. To shed the light on the issue of host-pathogen interaction between European bats and P. destructans, we examined seventeen bats emerging from the fungus-positive underground hibernacula in the Czech Republic during early spring 2013. Dual wing-membrane biopsies were taken from Barbastella barbastellus (1), Myotis daubentonii (1), Myotis emarginatus (1), Myotis myotis (11), Myotis nattereri (1) and Plecotus auritus (2) for standard histopathology and transmission electron microscopy. Non-lethal collection of suspected WNS lesions was guided by trans-illumination of the wing membranes with ultraviolet light. All bats selected for the present study were PCR-positive for P. destructans and showed microscopic findings consistent with the histopathological criteria for WNS diagnosis. Ultramicroscopy revealed oedema of the connective tissue and derangement of the fibroblasts and elastic fibres associated with skin invasion by P. destructans. Extensive fungal infection induced a marked inflammatory infiltration by neutrophils at the interface between the damaged part of the wing membrane replaced by the fungus and membrane tissue not yet invaded by the pathogen. There was no sign of keratinolytic activity in the stratum corneum. Here, we show that lesions pathognomonic for WNS are common in European bats and may also include overwhelming full-thickness fungal growth through the wing membrane equal in severity to reports from North America. Inter-continental differences in the outcome of WNS in bats in terms of morbidity/mortality may therefore not be due to differences in the pathogen itself.

Mortality of wild aquatic birds has recently been attributed to cyanobacterial toxins. Despite this, no experimental studies on the effects of defined doses of microcystins administered orally to birds exist. In this experiment, four groups of male Japanese quails daily ingesting 10ml of Microcystis biomass containing 0.045, 0.459, 4.605 or 46.044mug of microcystins, respectively, for 10 and 30 days, showed no mortality. Histopathological hepatic changes in birds after the biomass exposure included cloudy swelling of hepatocytes, vacuolar dystrophy, steatosis and hyperplasia of lymphatic centres. On subcellular level, shrunken nuclei of hepatocytes containing ring-like nucleoli, cristolysis within mitochondria and vacuoles with pseudomyelin structures were present. Vacuolar degeneration of the testicular germinative epithelium was found in two exposed males. Statistically significant differences in biochemical parameters were on day 10 of exposure only. They comprised increased activities of lactate dehydrogenase and a drop in blood glucose in birds receiving the highest dose of the biomass. Principal component analysis revealed a pattern of responses in biochemical parameters on day 10 that clearly separated the two greatest exposure groups from the controls and lower exposures. The results indicate that diagnosis of microcystin intoxication solely based on clinical biochemical and haematological parameters is hardly possible in birds.
