RESUMEN

Olibanum, a golden oleo-gum resin from species in the Boswellia genus (Burseraceae family), is a famous traditional herbal medicine widely used around the world. Previous phytochemical studies mainly focused on the non-polar fractions of olibanum. In this study, nine novel diterpenoids, boswellianols A-I (1-9), and three known compounds were isolated from the polar methanolic fraction of the oleo-gum resin of Boswellia carterii. Their structures were determined through comprehensive spectroscopic analysis as well as experimental and calculated electronic circular dichroism (ECD) data comparison. Compound 1 is a novel diterpenoid possessing an undescribed prenylmaaliane-type skeleton with a 6/6/3 tricyclic system. Compounds 2-4 were unusual prenylaromadendrane-type diterpenoids, and compounds 5-9 were new highly oxidized cembrane-type diterpenoids. Compounds 1 and 5 showed significant transforming growth factor ß (TGF-ß) inhibitory activity via inhibiting the TGF-ß-induced phosphorylation of Smad3 and the expression of fibronectin and N-cadherin (the biomarker of the epithelial-mesenchymal transition) in a dose-dependent manner in LX-2 human hepatic stellate cells, indicating that compounds 1 and 5 should be potential anti-fibrosis agents. These findings give a new insight into the chemical constituents of the polar fraction of olibanum and their inhibitory activities on the TGF-ß/Smad signaling pathway.

Aiming to enhance the ns-LIBS signal, in this work, we introduced orbital angular momentum to modulate the laser phase of the Gaussian beam into the vortex beam. Under similar incident laser energy, the vortex beam promoted more uniform ablation and more ablation mass compared to the Gaussian beam, leading to elevated temperature and electron density in the laser-induced plasma. Consequently, the intensity of the ns-LIBS signal was improved. The enhancement effects based on the laser phase modulation were investigated on both metallic and non-metallic samples. The results showed that laser phase modulation resulted in a maximum 1.26-times increase in the peak intensities and a maximum 1.25-times increase in the signal-to-background ratio (SBR) of the Cu spectral lines of pure copper for a laser energy of 10 mJ. The peak intensities of Si atomic spectral lines were enhanced by 1.58-1.94 times using the vortex beam. Throughout the plasma evolution process, the plasma induced by the vortex beam exhibited prolonged duration and a longer continuous background, accompanied by a noticeable reduction in the relative standard deviation (RSD). The experimental results demonstrated that modulation the laser phase based on orbital angular momentum is a promising approach to enhancing the ns-LIBS signal.

Ribisin A has been shown to have neurotrophic activity. The aim of this study was to evaluate the neuroprotective effect of ribisin A on injured PC12 cells and elucidate its mechanism. In this project, PC12 cells were induced by H2O2 to establish an injury model. After treatment with ribisin A, the neuroprotective mechanism of ribisin A was investigated by methyl tetrazolium (MTT) assay, Enzyme-linked immunosorbent assay (ELISA), flow cytometric analysis, fluorescent probe analysis, and western blot. We found that ribisin A decreased the rate of lactate dehydrogenase (LDH) release, increased cellular superoxide dismutase (SOD) level, decreased the levels of tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), Ca2+ expression and reactive oxygen species (ROS). Moreover, ribisin A significantly increased mitochondrial membrane potential (MMP) and inhibited apoptosis of PC12 cells. Meanwhile, ribisin A activated the phosphorylation of ERK1/2 and its downstream molecule CREB by upregulating the expression of Trk A and Trk B, the upstream molecules of the ERK signaling pathway.

Steroidal alkaloids are the main bioactive components of the bulbs of Fritillaria, which have been used as traditional Chinese medicine, known as "Beimu", for the treatment of cough for thousands of years in China. Cough and dyspnea are the most common symptoms observed in patients with pulmonary fibrosis. However, the antifibrotic activity of steroidal alkaloids has not been reported yet. In this study, two previously unreported cevanine-type steroidal alkaloids (1 and 2), four previously undescribed cevanine-type alkaloid glycosides (3-6), and 19 known steroidal alkaloids (7-25) were isolated from the bulbs of Fritillaria unibracteata var. wabuensis. The structures of these compounds were elucidated by comprehensive HRESIMS and NMR spectroscopic data analysis, as well as DP4+ NMR calculations. The biological evaluation showed that compounds 2, 7-10, 14, 15, and 17 downregulated fibrotic markers induced by transforming growth factor-ß (TGF-ß) in MRC-5 cells. Moreover, compounds 14 and 17 dose dependently inhibited TGF-ß-induced epithelial-mesenchymal transition in A549 cells, alleviated TGF-ß-induced migration and proliferation of fibroblasts, and decreased the expression of fibrotic markers, fibronectin, and N-cadherin in TGF-ß-induced MRC-5 cells. The research showed the potential of cevanine-type alkaloids as a class of natural antifibrotic agents.

OBJECTIVES: To investigate the level of neuropsychological development in large for gestational age (LGA) infants at the age of 12 months. METHODS: The infants, aged 12 to <13 months, who attended the Outpatient Service of Child Care in the First Affiliated Hospital of Shandong First Medical University from December 2021 to June 2023, were enrolled as subjects. According to the gestational age and birth weight, they were divided into preterm appropriate for gestational age (AGA) group, preterm LGA group, early term AGA group, early term LGA group, full-term AGA group, and full-term LGA group. A modified Poisson regression analysis was used to investigate the association between LGA and neuropsychological development outcome at 12 months of age. RESULTS: After adjustment for confounding factors, compared with the full-term AGA group at the age of 12 months, the full-term LGA group had a significant increase in the risk of language deficit (RR=1.364, 95%CI: 1.063-1.750), the early term LGA group had significant increases in the risk of abnormal gross motor, fine motor, language, and the preterm LGA group had significant increases in the risk of abnormal language, social behavior, and total developmental quotient (P<0.05); also, the early term AGA group had higher risks of developmental delay across all five attributes and in total developmental quotient at the age of 12 months (P<0.05); except for the language attribute, the preterm AGA group had higher risks of developmental delay in the other 4 attributes (P<0.05). CONCLUSIONS: The neuropsychological development of LGA infants with different gestational ages lags behind that of full-term AGA infants at 12 months of age, and follow-up and early intervention of such infants should be taken seriously in clinical practice.

Five undescribed sesquiterpenoid dimers, aucklandiolides A-E (1-5), one new sesquiterpenoid glycoside, ß-cyclocostunolide-15-ß-D-glucopyranoside (6), and seventeen known analogues (7-23) were isolated from the roots of Aucklandia costus. Their structures were elucidated by comprehensive HRESIMS and NMR spectroscopic data analysis, and their configurations were confirmed by the computational calculations of ECD and NMR chemical shifts. Aucklandiolides A and B are the first examples of dimeric sesquiterpenoids with a unique 6/6/6/5/6/6 ring system originated from a proposed Diels-Alder cycloaddition between two eudesmane sesquiterpenoids. Besides, compounds 9-11, 20, and 22 showed significant inhibition of nitric oxide production in LPS-stimulated RAW 264.7 cells at a concentration of 20 µM.

The glioma boundary is difficult to identify during surgery due to the infiltrative characteristics of tumor cells. In order to ensure a full resection rate and increase the postoperative survival of patients, it is often necessary to make an expansion range resection, which may have harmful effects on the quality of the patient's survival. A full-Stokes laser-induced breakdown spectroscopy (FSLIBS) theory with a corresponding system is proposed to combine the elemental composition information and polarization information for glioma boundary detection. To verify the elemental content of brain tissues and provide an analytical basis, inductively coupled plasma mass spectrometry (ICP-MS) and LIBS are also applied to analyze the healthy, boundary, and glioma tissues. Totally, 42 fresh tissue samples are analyzed, and the Ca, Na, K elemental lines and CN, C2 molecular fragmental bands are proved to take an important role in the different tissue identification. The FSLIBS provides complete polarization information and elemental information than conventional LIBS elemental analysis. The Stokes parameter spectra can significantly reduce the under-fitting phenomenon of artificial intelligence identification models. Meanwhile, the FSLIBS spectral features within glioma samples are relatively more stable than boundary and healthy tissues. Other tissues may be affected obviously by individual differences in lesion positions and patients. In the future, the FSLIBS may be used for the precise identification of glioma boundaries based on polarization and elemental characterizing ability.

Fritillaria has a long history in China, and it can be consumed as medicine and food. Owing to the high cost of Fritillaria cirrhosa, traders sometimes mix it with the cheaper Fritillaria thunbergii powder to make profit. Herein, we proposed a laser-induced breakdown spectroscopy (LIBS) technique to test the adulteration present in the sample of Fritillaria cirrhosa powder. Experimental samples with different adulteration levels were prepared, and their LIBS spectra were obtained. Partial least squares regression (PLSR) was adopted as the quantitative analysis model to compare the effects of four data standardization methods, namely, mean centring, normalization by total area, standard normal variable, and normalization by the maximum, on the performance of the PLSR model. Principal component analysis and least absolute shrinkage and selection operator (LASSO) were utilized for feature extraction and feature selection, and the performance of the PLSR model was determined based on its quantitative analysis. Subsequently, the optimal number of features was determined. The residuals were corrected using support vector regression (SVR). The mean absolute error and root mean square error of prediction obtained from the quantitative analysis results of the combined LASSO-PLSR-SVR model for the test set data were 5.0396% and 7.2491%, respectively, and the coefficient of determination R2 was 0.9983. The results showed that the LIBS technique can be adopted to test adulteration in the sample of Fritillaria cirrhosa powder and has potential applications in drug quality control.

A novel styrylpyrone derivative, named phelliribsin B (1), as well as four biogenetically related known compounds, phellifuropyranone A (2), inoscavin C (3), inoscavin A (4), and inoscavin D (5) were separated and purified from the medicinal fungus Phellinus ribis. The structure of phelliribsin B was determined by spectroscopic analysis, and the absolute configuration was assigned by experimental and calculated ECD data. Additionally, the plausible biosynthetic pathway of 1 was also proposed. Compound 1 showed moderately cytotoxic activity against HepG2 and SKOV-3 tumor cell lines with IC50 values of 32.71 and 57.89 µM, respectively. Based on the results of cytotoxicity against HepG2 tumor cells, the structure-activity relationship of compounds 1-4 with similar skeletons was discussed. The styrylpyrone derivatives with similar skeletons have moderately cytotoxic activity and have the potential to play an important role in the anti-tumor treatment.
