RESUMEN

OBJECTIVE: Nasopharyngeal carcinoma (NPC) is one of the most ordinary malignant tumors. Recent studies have revealed that circular RNAs play an important role in the progression of tumorigenesis. This study aims to identify how circular RNA_LARP4 functions in the progression of NPC. PATIENTS AND METHODS: We performed Real Time quantitative-Polymerase Chain Reaction (RT-qPCR) in 58 paired NPC patients' tissue samples and cell lines to detect circular RNA_LARP4 expression. The function of circular RNA_LARP4 in the NPC proliferation was identified by performing proliferation assay and colony formation assay. Moreover, the function of circular RNA_LARP4 in NPC metastasis was measured by performing scratch wound assay and transwell assay in vitro. Furthermore, the underlying mechanism was explored through Western blot assay and RT-qPCR. RESULTS: Circular RNA_LARP4 expression was significantly lower in NPC tissues compared to that in adjacent samples. Cell proliferation of NPC was inhibited after overexpression of circular RNA_LARP4 in vitro. Cell migration and invasion of NPC was inhibited after overexpression of circular RNA_LARP4 in vitro. Furthermore, the results of further experiments revealed that Rho-associated kinase 1 (ROCK1) was downregulated via overexpression of circular RNA_LARP4 and was also a direct target of circular RNA_LARP4 in NPC. CONCLUSIONS: This study suggests that circular RNA_LARP4 inhibits NPC cell proliferation and metastasis via targeting ROCK1 in vitro.

OBJECTIVE: To elucidate the function of long non-coding RNA (lncRNA) SNHG1 in acute myeloid leukemia (AML) and to explore its biological mechanism. PATIENTS AND METHODS: Expression levels of lncRNA SNHG1 and miR-488-5p in AML cell lines pAML and THP-1 were determined by quantitative Real Time-Polymerase Chain Reaction (qRT-PCR). Proliferative potential and cell cycle progression of pAML and THP-1 cells were detected after SNHG1 knockdown. Potential binding sites of SNHG1 and miR-488-5p were predicted by bioinformatics. Through RNA Binding Protein Immunoprecipitation (RIP) assay and luciferase reporter gene assay, we evaluated the binding condition between SNHG1 and miR-488-5p. MiR-488-5p expression in pAML and THP-1 cells with SNHG1 knockdown was detected by qRT-PCR to further verify their interaction. Subsequently, binding sites of NUP205 and miR-488-5p were predicted. Both mRNA and protein levels of NUP205 in pAML and THP-1 cells were examined. The regulatory effects of overexpressed NUP205 on proliferative potential and cell cycle progression of pAML and THP-1 cells transfected with si-SNHG1 were explored by gain-of-function experiments. RESULTS: LncRNA SNHG1 was highly expressed in pAML and THP-1 cells, while miR-488-5p was lowly expressed. SNHG1 knockdown in pAML and THP-1 cells inhibited proliferative ability and arrested cell cycle in G0/G1 phase. Knockdown of SNHG1 markedly upregulated the miR-488-5p expression in pAML and THP-1 cells. Furthermore, RIP and luciferase reporter gene assay confirmed the binding between SNHG1 and miR-488-5p. NUP205 was highly expressed in pAML and THP-1 cells at both mRNA and protein levels. Moreover, luciferase reporter gene assay indicated that NUP205 could bind to miR-488-5p. More importantly, the overexpression of NUP205 in pAML and THP-1 cells reversed the inhibitory effect of SNHG1 knockdown on proliferative ability and cell cycle progression. CONCLUSIONS: LncRNA SNHG1 promotes the development of AML through miR-488-5p/NUP205 axis.

Soybean protein hydrolysates (SPHs) bind with calcium, forming soluble SPH-calcium complexes via the carboxyl groups of glutamic and aspartic acid residues. However, their effect on calcium uptake is still unclear. In this study, Caco-2 cells were used to estimate the effect of SPH-calcium complexes with different molecular weights on calcium uptake in vitro. The changes in intracellular calcium ion concentration were measured by Fura-2 loading and expressed in fluorescence intensity. SPH-calcium complexes could promote calcium uptake. Improved fluorescence intensity was significantly different in SPH-calcium complexes (10 to 30 kDa), SPH-calcium complexes (3 to 10 kDa), and SPH-calcium complexes (1 to 3 kDa). The maximum levels of relative fluorescence intensity (18.3) occurred with SPH-calcium complexes (10 to 30 kDa). The effect of SPH-calcium complexes (10 to 30 kDa) on Ca(2+) increase was determined to be concentration dependent in the range of 0.5 to 4 mg/mL. Our results indicate that soybean protein itself might be responsible for promoting calcium absorption.

The soluble complexes formed between hydrolyzed soybean protein and calcium at pH 7.4 were investigated using dialysis, gel chromatography, and Fourier transform infrared spectrometry (FTIR). The results demonstrate that the amount of calcium bound was significantly different for soybean protein hydrolysates obtained using the proteases neutrase, flavourzyme, protease M, and pepsin. Maximum levels of calcium binding (66.9 mg/g) occurred with hydrolysates produced using protease M. Peptide fragments exhibiting high calcium binding capacity had molecular weights of either 14.4 kDa or 8 to 9 kDa, and the calcium binding capacity was linearly correlated with carboxyl group content (R(2)= 0.8204). FTIR experiments revealed that upon binding calcium, the amide I band underwent a shift to lower wave numbers. A wide, intense Ca-O absorption band also appeared between 400 and 100 cm(-1) in the far-infrared spectrum. The width and intensity of this band increased after treatment of samples with glutaminase. The amount of bound calcium was related to both the molecular weight of the peptides and to the carboxyl group content, and the most likely sites for calcium binding are the carboxyl groups of Asp and Glu.

RESUMEN

The existence of various types of circumferential waves, both predominantly shell or fluid borne, and the repulsion of their dispersion curves is discussed here for an infinite, thin elastic, circular-cylindrical shell immersed in a fluid and filled with another fluid. The study is based on an analytic calculation of the partial-wave resonances in the acoustic scattering amplitude of a normally incident plane wave. A large number of cases of repulsion are found in the phase-velocity dispersion curves of the various types of circumferential waves due to the shell-fluid coupling.

From January 1989 to June 1990, 13 institutions in our country studied jointly the application of a 20-item neonatal behavioral neurological assessment (NBNA) in asphyxiated babies. Of 145 cases, 125 were followed up to 1-2 years of age. The results showed that in predicting prognosis, the sensitivity and specificity of NBNA score at 7 days after birth were 88.9% and 82.6% and 12-14 days, 84.6% and 97.4% respectively. Its value in predicting prognosis is superior to Sarnat degree, cranial CT and B ultrasound. This method is practical, economical and reliable. It is worth popularizing.

103,753 (male 51,994, female 51,759) primary and middle school students aged 6-15 years in two districts in Beijing city were surveyed from October 1987 to April 1989. The heights of the students were measured. According to the height standard of northern cities in China, 202 students with heights below the 3rd percentile for age were requested for detailed history, physical examination, screening GH test bone age, T4, SGPT, chest X-ray, routine urine test and sex chromatin (in female). If GH less than 10 micrograms/L, two provocative tests (L-dopa or clonidine and insulin hypoglycaemia test) were done. Then the heights of the short students were observed for 1/2-2 years. GHD was diagnosed in 12 cases based on the GH peak levels less than 10 micrograms/L in two provocative tests, whose growth velocity was slower than that for students of the same age and sex. Of these subjects with GHD, total GHD (GH less than 5 micrograms/L) was present in 7 and partial GHD (GH = 5-9.9 micrograms/L) in 5. The 12 GHD students (male 9, female 3) aged 8.9-15.7 years accounted for 1/8,646 in the total surveyed students. The male and female GHD accounted for 1/5,777 and 1/17,253 in the total males and females respectively.


PIP: The prevalence of growth hormone (GH) deficiency in Beijing children was estimated by measuring the heights of 103,753 children aged 6-15 and assessing those below the 3rd percentile in height. The following data were collected from 202 students: history, physical examination results, bone age, T4, chest x-ray, liver function tests, urinalysis, and sex chromatin in the girls who had not menstruated. 13 short children were excluded because of organic disease. Those with GH 10 mcg/L were subjected to provocative tests using L-dopa, clonidine, and insulin. GH deficiency was diagnosed in 12 children based on GH levels, provocative testing, and growth velocity over the next 1.5 years. There was total GH deficiency (GH 5 mcg/L) in 7 children and partial GH deficiency (GH 5-9.5 mcg/L) in 5 others. These 9 boys and 3 girls accounted for 1 case of GH deficiency/8646 middle school students in Beijing. The prevalence is between that published for Newcastle and Edinburgh, UK. This information will be useful for estimating the amount of synthetic GH needed in China.

Based on the method of Brazelton and Amiel-Tison for behavioral neurological measurement in the newborn and our own experience, we formulated a 20-item neonatal behavioral neurological assessment (NBNA). For setting up a practical method and a normal range of its scores in China, we examined 714 normal newborns (male 369, female 345), with NBNA at the age of 2-3, 12-14, 26-28 days for a total of 2,142 times, in 12 provinces of China from May to October 1988. NBNA contains 5 clusters: behavior (6 items); passive tone (4 items); active tone (4 items); primary reflexes (3 items); and general assessment (3 items). Each item has 3 scales (0,1,2). Twenty items have a maximum of 40 scores. Among the 2,142 times examined, 90.4% of the 714 newborns had 39-40 scores, 97% greater than 37 and none below 35. Measurements of the first, second and third time revealed that the total scores increased with age. The difference was statistically significant. The abilities of auditory and visual orientations and active neck tone improved obviously with age. NBNA shows distinct stability and reliability. The score is not influenced by geographic location; the scoring system is simple and the manipulation takes only ten minutes.

This article reports the efficacy and safety of treatment with Somatonorm in 9 children with idiopathic growth hormone (GH) deficiency in China. The growth velocity increased from 2.6 +/- 0.2 cm/year before treatment to 10.7 +/- 0.5 cm/year at the end of 1-year treatment. The increment of human growth hormone (hGH) level and area under the response curve above the baseline level were larger after one injection of Somatonorm at the dose of 0.1 IU/kg sc than that of 0.17 IU/kg im. Increments of delayed bone age were not significant. There was no correlation between the increase of height growth velocities and the serum Somatomedin-C (SMC) levels during hGH therapy (r = -0.055). Blood sugar levels and blood alkaline phosphatase concentrations increased significantly after therapy. There were no significant side effects. Serum T4 levels of 3 patients dropped below normal after treatment. Serum anti-hGH antibody appeared at 3-12 month in 2 patients, but it did not attenuate the growth velocity.

Fifty-nine patients with idiopathic growth hormone deficiency were included in a Chinese multicentre trial of recombinant somatropin, 0.5-0.7 IU/kg/week s.c. given in six or seven divided doses. The height velocity increased from 2.8 +/- 1.0 cm/year to 13.1 +/- 2.5 cm/year during 1 year of treatment, and typical catch-up growth was observed. The increase in height SDS for chronological age was significant, but the increase in height SDS for bone age was not statistically significant. No adverse reactions to the treatment were recorded. Recombinant somatropin was shown to be very effective and safe during the first year of therapy in patients with growth hormone deficiency.
