RESUMEN

Development of a fast and accurate pesticide analysis system is a challenging task, as a large amount of commonly used pesticide has negative effects on humans' health. Detection of pesticide residues is crucial for food safety management and environmental protection. Aptamers─short single-stranded oligonucleotides (RNA or DNA) selected by aptamer selection method SELEX─can selectively bind to their target pesticide molecules with high affinity. Thus, in the present study, we developed an electrochemical biosensor based on aptamers to detect the commonly used pesticide, glyphosate. Carbon fibers were used as the platform to assemble polyelectrolyte layers with the incorporated aptamers selectively binding with glyphosate molecules for electrochemical detection. The best limit of detection of 0.3 µM was achieved at open-circuit potential measurements, which is comparable to the current need in detection of glyphosate. The developed method can be implemented into existing systems for the determination of pesticides on farms to control residual concentrations of glyphosate in soil and water.

ABSTRACT: The cell walls of cereals are rich sources of polysaccharide ß-glucan. In this study, the ß-glucan was extracted from oat bran using the hot-water extraction method and dried in a pure powder form. The concentration of the ß-glucan in the extract was determined using the l-cysteine sulfuric acid method. The results showed that the yield of ß-glucan using the hot-water extraction method is the highest compared to its yield achieved by enzymatic, acid, and alkaline methods. In this paper, the usage of the ß-glucan as a coating material for a water-insoluble carotenoid is considered. This study demonstrates for the first time the encapsulation of ß-carotene with modified octanoic acid ß-glucan. It implements to obtain a stable encapsulated polysaccharide-carotenoid system, which has been studied by a set of physicochemical methods and a cytotoxic analysis was performed on the HCT-116 cell line. The SEM image of the resulting encapsulated system is perfectly correlated with the DLS data, which has determined the size of MG capsules at 200 nm. The cytotoxic analysis demonstrates that the cell viability was more than 70%, which indicates its potential using in the food industry.

BACKGROUND: Ovarian cancer is one of the most common diseases of the female reproductive system. The aim of this study was the investigation of the antitumor cisplatin effect on ascitic fluid tumor cells in the ovarian cancer experimental model by digital cytomorphometry and cell bioinformatic analysis. METHODS: Female Wistar rats were inoculated by ovarian cancer strain. After ovarian cancer transplantation rats were divided into two groups: control group-without cisplatin treatment, the experimental group-under cisplatin treatment. The ascitic fluid was taken on the 9-th day after tumor cell inoculation. Digital cytomorphometric and cytobioinformatic analysis were used to study ascitic fluid cancer cell morphofunctional changes under cisplatin treatment. RESULTS: Digital cytomorphometric characteristics of rat ovarian cancer ascitic cells were obtained. Tumor cells were classified into four groups according to their geometric size: small, medium, large, "gigantic". The algorithm and the computer program based on tumor cell morphometric characteristics were created to calculate such cell bioinformatic characteristic as information redundancy coefficient R. Three parameters were determined as the criteria of cisplatin effect on cancer cells: cell number, nuclear/cytoplasmic ratio, R-value. CONCLUSIONS: Data obtained suggest that cisplatin reduces the total cell number, the nuclear/cytoplasmic ratio and R-value thus indicates a decrease in cellular resistance and adaptive potential. The digital cytomorphometry and bioinformatics could be recommended as a testing system in the experimental or clinical study.

BACKGROUND: There is some evidence that Benign Prostatic Hyperplasia (BPH) may increase the risk of developing prostate cancer, so conducting research on effective BPH inhibitors is important. OBJECTIVE: This research studied the inhibitory effect of Iodized Serum Milk Protein (ISMP) on BPH in rats. ISMP is a concentrate of lactic protein containing 2.2% iodine. METHODS: Male Wistar rats, aged 18 months, were used. In the intact control group, sunflower oil was administered intragastrically by gavage. In 36 rats, BPH was induced by surgical castration, followed by subcutaneous injections of prolonged testosterone - omnadren, 25mg/kg every other day (7 administrations). One group of rats served as BPH-control. ISMP and finasteride (positive control), dissolved in sunflower oil, were administered to rats intragastrically daily at a dose of 200µg/kg and 5mg/kg, respectively, for 4 weeks starting immediately after castration. RESULTS: ISMP inhibited the development of BPH in rats, significantly reducing the mass of the prostate and its parts (except for the anterior lobes) by 1.1-1.3 times and the prostatic index (the ratio of prostate weight to the body weight) - by 1.3-1.4 times. Finasteride inhibited the development of BPH, and its activity was higher (by 1.1-1.3 times) than in ISMP. Histological analysis of the prostate showed fewer pronounced morphological hyperplasia signs in animals treated with ISMP or finasteride. CONCLUSION: The iodine-containing preparation ISMP has the ability to inhibit the development of BPH in rats although its activity is somewhat lower than that of finasteride.

A functional food is any food that has (a) specific nutrient(s) added to it for a specific functional purpose. Selenium (Se) is a metalloid that belongs to group 16 of the periodic table. It may be obtained from myriad sources like soil, water, and living organisms. Se is required to sustain proper health in both animals and humans due to its linkage with various biological functions in the immune system. Nature has made it impossible for us to obtain sufficient Se from the diet since some regions across the globe have been designated as Se-deficient while others are becoming Se-toxic. Se deficiency is associated with a compromised immune system and increased susceptibility to various diseases. Therefore, designing Se functional foods and supplements is the way forward in curbing the menace mentioned above, since geographical location will not have any effect on the Se content of these foods. Brewing yeast has the necessary enzymes to biotransform inorganic Se (Na2SeO3) to its bioactive organic form, which is incorporated in the aged selenized beer. S. cerevisiae was found to have better biotransformation efficiency than other yeast species. A traditional Slavic beverage, selenized kvass, was brewed using rye grains soaked and germinated in solutions of Na2SeO3. Fruit yeasts (Lesaffre) and Evitalia were utilized as the starter cultures. A Se enriched solution was extracted from Se-biofortified pak-choi cabbage and incorporated into the wort before fermentation. Selenized yeast and algae can be taken as supplements or as food additives. There have been some reports about microcystins (MCs) in Spirulina. Therefore, the safety of Se-algae is not guaranteed. Pasteurized dried selenized supplements (yeast and algae) were proposed in formulating Selenized enriched Tom-brown. This review seeks to propose some possible foods that could be enriched with Se. A large portion of the population adequately consumes these proposed foods on a regular basis hence the target goal would be a success.
