We hypothesized that the coasting time may be beneficial to the quality of cumulus-oocyte complexes recovered from live ewes, as reported in cattle. The present study assessed the effect of coasting times on the quantity and quality of cumulus-oocyte complexes (COCs) in sheep. All ewes were subjected to the "Day 0 protocol", followed by an ovarian stimulation (80 mg of pFSH in three decreasing doses), varying only the coasting time [12 (G12), 36 (G36), or 60 h (G60]. In Experiment 1, data regarding follicular population was assessed. In Experiment 2, the COC quality was checked by their morphology, brilliant cresyl blue (BCB) test, evaluation of chromatin condensation pattern, and oocyte diameter. In Experiment 3, genes related to oocyte developmental competence were evaluated in BCB + COCs. The oocytes in the G60 group had more (P < 0.05) large follicles than the other groups and oocytes with a greater diameter than the G12. Oocyte morphology was similar (P > 0.05) among groups, as well as the BCB + COCs quantity. The G60-oocytes presented a better (P < 0.05) configuration of chromatin condensation compared with the other groups and a greater (P < 0.05) gene expression of BMP15, MATER, ZAR1, and PTGS2 compared with G12, and PTGS2 and HAS2 compared with G36 group. In conclusion, 60 h of coasting time positively affects the quality of COCs recovered after subjecting ewes to the "Day 0 protocol" and ovarian superstimulation. Implementing the appropriate coasting time to a given protocol can positively impact the in vitro embryo production outcomes in sheep.

To facilitate transcervical artificial insemination in sheep, the effects of local treatment with α1-adrenergic receptor antagonists on cervix dilation and hemodynamics were evaluated. Ewes (n = 7) were subjected to oestrous synchronisation every 40 days and assigned to treatments in a Latin square experimental design (seven animals × seven periods) with a factorial treatment arrangement (A × B), Factors A (prazosin or tamsulosin) and B (1, 2, or 4 mg/animal). Ewes of the six treatment groups (P1, P2, P4, T1, T2, and T4) were administered α1-adrenergic receptor antagonists while those of the control group (CG) were administered only α1-adrenergic antagonist carrier agent. Distance that the transcervical catheter penetrated without cervical resistance, mean arterial pressure, and uterine artery dopplerfluxometry were evaluated before and after 30 min, 1, 2, 4, 8, and 10 h of treatment. Catheter penetration distance was greater in ewes of the T4 and P4 groups (P < 0.01), with there being a positive correlation between dose and distance (r = 0.243). The penetration distance was similar (P = 0.84) for treated groups, with the greatest penetration occurring 2, 4, and 6 h after treatment (P < 0.01). The passage into the uterine lumen was greater (P = 0.013) in ewes of the P4 (17.9 %) and T4 (19.6 %) groups. There were no effects on blood pressure or uterine blood flow (P> 0.05). These preliminary results indicate there are benefits of treatment with 4 mg/animal of tamsulosin or prazosin in catheter passage through the sheep cervix 2-6 h after administration without hemodynamic effects.

Considering the limited physiological information available on neotropical rodents and the importance of this information for pathophysiological and conservation studies of these species, the aim of this study was to evaluate the cardiac electric physiology of healthy captivity spotted pacas (Cuniculus paca) under chemical restraint, due to wild nature of these animals. Eleven adult female pacas were evaluated by blood count and biochemical dosage to rule out any associate disease. Each animal was evaluated in three periods every 15 days. After chemical restraint with intramuscular midazolam 0.5mg/kg and ketamine 25mg/kg, animals were subjected to a computerized electrocardiogram, where bipolar (DI, DII and DIII) and augmented unipolar leads (aVR, aVL, aVF) were obtained. Descriptive statistics were calculated for each parameter and built the confidence interval (CI) at significance level of 95%. The electrocardiographic examination was performed without difficult. ECG tracing in DII represents a QRS complex with positive polarity, preceded by a P wave of the same polarity and proceeded by T wave of variable polarity. Heart rate mean was 150±17 bpm, and cardiac electrical axis 33.4±21.9°. All animals showed sinusal rhythm. The ECG recording technique associated with chemical restraint was well tolerated, allowing quick acquisition of reliable ECG tracings with high repeatability, which produced sufficient results to determine the heart rhythm and suggest measures of ECG complexes duration and amplitude.(AU)


Considerando a limitada informação sobre a fisiologia cardíaca em roedores neotropicais e a importância desta informação para estudos fisiopatológicos e de conservação desta espécie, objetivou-se com este estudo avaliar a atividade elétrica cardíaca de pacas saudáveis mantidas em cativeiro, sob restrição química, devido à natureza selvagem destes animais. Onze fêmeas adultas foram avaliadas clinicamente e por meio do hemograma e testes bioquímicos no intuito de descartar qualquer co-morbidade associada. Cada animal foi submetido a três períodos experimentais a cada 15 dias. Após a restrição química com 25mg/kg de cetamina e 0,5mg/kg de midazolam pela via intramuscular, os animais eram submetidos a eletrocardiografia (ECG) computadorizada, aonde as derivações bipolares (DI, DII E DIII) e unipolares aumentadas (aVR, aVL, aVF) foram obtidas. A estatística descritiva foi calculada para cada parâmetro e o intervalo de confiança (CI), construído a um nível de significância de 95%. O exame eletrocardiográfico foi realizado sem dificuldade. O traçado eletrocardiográfico em DII apresentou QRS com polaridade positiva, precedido por onda P da mesma polaridade e seguida por onda T de polaridade variável. A frequência cardíaca média foi de 150±17 bpm e o eixo elétrico cardíaco de 33.4±21.9°. Todos os animais mostraram ritmo sinusal. A técnica eletrocardiográfica associada com restrição química foi bem tolerada permitindo a pratica aquisição de traçados de confiança e alta reprodutibilidade, que produziram resultados suficientes para determinar o ritmo cardíaco e sugerir medidas de duração e amplitude dos complexos de ECG.(AU)

The aim of this study was to compare different staining methods for the evaluation of sperm morphology by light microscopy and also to describe the morphometry of the entire sperm in collared peccaries (Pecari tajacu). Semen from 10 males was obtained by electroejaculation and evaluated for sperm motility, vigor, and concentration. Semen smears were prepared through three different staining methods: Bengal rose, brome-phenol blue, and eosin-nigrosin. Smears were evaluated under light microscopy and sperm morphologic alterations were determined in percentage. In addition, sperm morphometric analysis was conducted by light microscopy coupled to image analyzer software. The smears stained with Bengal Rose provide the best results for the visualization of the sperm tail, midpiece, and head. The use of eosin-nigrosin stain did not allow an adequate impregnation, and some sperm presented a few contrasts with the background. A higher incidence of bent coiled tails was verified in the use of brome-phenol blue staining (P<0.05). Through morphometric evaluation, it was observed that the tail occupies the greatest proportion (89%) of the sperm which presents a discretely elongated head. According to the results, the use of the Bengal Rose stain is recommended for the morphologic evaluation of the collared peccary sperm.


O objetivo deste estudo foi comparar diferentes métodos de coloração para avaliação da morfologia espermática por microscopia de luz e também descrever a morfometria completa de espermatozoides de catetos (Pecari tajacu). Sêmen de 10 machos foi obtido por eletroejaculação e avaliado quanto à motilidade espermática, vigor e concentração. Foram preparados por três diferentes métodos de coloração: Rosa de Bengala, Azul de Bromofenol e Eosina-Nigrosina. Os esfregaços foram avaliados por microscopia de luz, e determinado o percentual das alterações morfológicas. Ainda, a análise da morfometria espermática foi realizada por microscópio de luz acoplado a um softwere de análise de imagens. Os esfregaços corados com Rosa de Bengala apresentaram melhores resultados de visualização da cauda, peça intermediária e cabeça dos espermatozoides. O uso do corante Eosina-Nigrosina não permitiu uma adequada impregnação e alguns dos espermatozoides apresentaram pouco contraste com o fundo da lâmina. Uma maior incidência de cauda fortemente enrolada foi verificada com o uso do corante Azul de Bromofenol (P<0.05). Através da avaliação morfométrica foi observada que a cauda ocupa a maior proporção (89%) do espermatozoide, e a cabeça apresenta-se discretamente alongada. De acordo com os resultados, o uso do corante Rosa de Bengala é recomendado para a avaliação morfológica de espermatozoides de catetos.

Despite the wide geographical distribution of coati (Nasua nasua) from the south of Canada to the north of Argentina, studies regarding the reproductive characteristics of this species are extremely limited. The objective of this study was to describe the various characteristics of coati semen by morphometric and ultrastructural analysis. Five mature males were anesthetized and electroejaculated for the collection of semen. Semen was immediately evaluated for color, volume, pH, sperm motility, vigor, morphology, acrosomal integrity, percentage of live cells and hypo-osmotic response by light microscopy. Sperm cell morphometry and ultrastructural analyses were also performed. Observations of seminal characteristics determined by electroejaculation in captive coatis represent a valuable baseline dataset for establishing fertility standards and provide background information that may be useful for assisted breeding programmes in members of the Procyonidae family.
