RESUMEN

In the present work, a nondestructive device set up for a rapid and reliable freshness assessment of rainbow trout during 10 days of storage in ice was evaluated. The device was characterized by a vector network analyzer interfaced with an open coaxial probe to be placed in contact with the fish eye. The acquisition of the reflected scattering parameter (S11), which is the ratio between the amplitude of the reflected and the incident signal, was assessed in the 50 kHz-3 GHz spectral range. S11 is composed of a real part and an imaginary part, and both parts were used to predict quality index method for freshness evaluations. Partial least squares regression predictive models of the demerit scores related to fish eye attributes (eye pupil and eye shape) and the day of storage were set up. The main results showed that both the real and imaginary parts of the S11 decrease as a function of storage time. The combination with multivariate analysis allowed to set up predictive models of the storage time and the demerit scores with R2 values up to 0.946 (root mean square error [RMSE] = 0.88 days) and 0.942 (RMSE = 3.17 demerit scores related to the fish eyes attributes), respectively (external validation). According to our results, the proposed cheap solution appears a useful tool for the freshness assessment of rainbow trout. PRACTICAL APPLICATION: This work shows that dielectric properties have the potential to discriminate stored fish according to their freshness quality. A device based on this principle can play a significant role in the postharvest processes, contributing to higher product quality and safety and supporting producers and retailers during the qualitative inspections.

Early and significant results for a real-time, column-free miniaturized gas mass spectrometer in detecting target species with partial overlapping spectra are reported. The achievements have been made using both nanoscale holes as a nanofluidic sampling inlet system and a robust statistical technique. Even if the presented physical implementation could be used with gas chromatography columns, the aim of high miniaturization requires investigating its detection performance with no aid. As a study case, in the first experiment, dichloromethane (CH2Cl2) and cyclohexane (C6H12) with concentrations in the 6-93 ppm range in single and compound mixtures were used. The nano-orifice column-free approach acquired raw spectra in 60 s with correlation coefficients of 0.525 and 0.578 to the NIST reference database, respectively. Then, we built a calibration dataset on 320 raw spectra of 10 known different blends of these two compounds using partial least square regression (PLSR) for statistical data inference. The model showed a normalized full-scale root-mean-square deviation (NRMSD) accuracy of [Formula: see text] and [Formula: see text] for each species, respectively, even in combined mixtures. A second experiment was conducted on mixes containing two other gasses, Xylene and Limonene, acting as interferents. Further 256 spectra were acquired on 8 new mixes, from which two models were developed to predict CH2Cl2 and C6H12, obtaining NRMSD values of 6.4% and 13.9%, respectively.

RESUMEN

The simultaneous use of UVA light irradiation coupled with low energy cold plasma generated by a dielectric barrier discharge prototype, results in significant enhancement of efficiency of the integrated process with respect to the sole plasma treatment. This effect has been demonstrated both on microbial inactivation of a food-borne pathogen, i.e. Listeria monocytogenes, and on the degradation of a compound of biological origin such as phenylalanine. In the latter case, the analysis of its reaction intermediates and the spectroscopic identification and quantification of peroxynitrites, allowed to propose mechanistic hypotheses on the nature of the observed synergistic effects. Moreover, it has been demonstrated that the process does not affect the quality of trout fillets, indicating its suitability as a chlorine-free, green, and sustainable tool for the decontamination of fish processing water.

Non-thermal atmospheric plasma (NTAP) has gained attention as a decontamination and shelf-life extension technology. In this study its effect on psychrotrophic histamine-producing bacteria (HPB) and histamine formation in fish stored at 0-5°C was evaluated. Mackerel filets were artificially inoculated with Morganella psychrotolerans and Photobacterium phosphoreum and exposed to NTAP to evaluate its effect on their viability and the histidine decarboxylase (HDC) activity in broth cultures and the accumulation of histamine in fish samples, stored on melting ice or at fridge temperature (5°C). NTAP treatment was made under wet conditions for 30 min, using a dielectric barrier discharge (DBD) reactor. The voltage output was characterized by a peak-to-peak value of 13.8 kV (fundamental frequency around 12.7 KHz). This treatment resulted in a significant reduction of the number of M. psychrotolerans and P. phosphoreum (≈3 log cfu/cm2) on skin samples that have been prewashed with surfactant (SDS) or SDS and lactic acid. A marked reduction of their histamine-producing potential was also observed in HDC broth incubated at either 20 or 5°C. Lower accumulation of histamine was observed in NTAP-treated mackerel filets that have been inoculated with M. psychrotolerans or P. phosphoreum and pre-washed with either normal saline or SDS solution (0.05% w/v) and stored at 5°C for 10 days. Mean histamine level in treated and control groups for the samples inoculated with either M. psychrotolerans or P. phosphoreum (≈5 log cfu/g) varied from 7 to 32 and from 49 to 66 µg/g, respectively. No synergistic effect of SDS was observed in the challenge test on meat samples. Any detectable amount of histamine was produced in the meat samples held at melting ice temperature (0-2°C) for 7 days. The effects of NTAP on the quality properties of mackerel's filets were negligible, whereas its effect on the psychrotrophic HPB might be useful when time and environmental conditions are challenging for the cool-keeping capacity throughout the transport/storage period.

RESUMEN

Standard analytical methods for fish freshness assessment are based on the measurement of chemical and physical attributes related to fish appearance, color, meat elasticity or texture, odor, and taste. These methods have plenty of disadvantages, such as being destructive, expensive, and time consuming. All these techniques require highly skilled operators. In the last decade, rapid advances in the development of novel techniques for evaluating food quality attributes have led to the development of non-invasive and non-destructive instrumental techniques, such as biosensors, e-sensors, and spectroscopic methods. The available scientific reports demonstrate that all these new techniques provide a great deal of information with only one test, making them suitable for on-line and/or at-line process control. Moreover, these techniques often require little or no sample preparation and allow sample destruction to be avoided.

Dielectric barrier discharge plasma and photocatalysis have been proposed as tools for decontamination of process water, especially in food industry. The present investigation aims to redefine and identify the features of coupling the two technologies in terms of degradation efficiency of a model compound. Results show that, when the process is carried out in plasma activated water in the presence of irradiated TiO2, the efficiency of the integrated process is lower than the sum of the two processes acting separately. It is proposed that afterglow species, e.g. hydrogen peroxide and/or peroxynitrites could be activated by UVA light irradiation producing hydroxyl radicals in the liquid phase. Even if TiO2 limits this additional effect by acting as UVA screen barrier material, its decontamination efficiency under certain conditions results higher than that obtained with plasma systems. These results open the route to chlorine-free decontamination processes and redefine the application framework of this integrated approach.

This paper will show the electronic architecture of a portable and non-invasive soil moisture system based on an open rectangular waveguide. The spectral information, measured in the range of 1.5-2.7 GHz, is elaborated on by an embedded predictive model, based on a partial least squares (PLS) regression tool, for the estimation of the soil moisture (%) in a real environment. The proposed system is composed of a waveguide, containing Tx and Rx antennas, and an electronic circuit driven by a microcontroller (MCU). It will be shown how the system provides a useful and fast estimation of moisture on a silty clay loam soil characterized by a moisture range of about 9% to 32% and a soil temperature ranging from about 8 °C and 18 °C. Using the PLS approach, the moisture content can be predicted with an R2 value of 0.892, a root mean square error (RMSE) of 1.0%, and a residual prediction deviation (RPD) of 4.3. The results prove that it is possible to make accurate and rapid moisture assessments without the use of invasive electrodes, as currently employed by state-of-the-art approaches.

Pulsed electric field (PEF) treatment is a non-thermal technology that has shown good potential for microbial inactivation. However, in many cases, it cannot be sufficient to avoid microbial proliferation, and the combination with other stabilizing technologies is needed. In the framework of the hurdle concept, several researches have been focused on the use of PEF in combination with heat and/or antimicrobials to increase its efficacy. This study investigated the inactivation effect of PEF on a strain of Saccharomyces cerevisiae (isolated from spoiled beverages) in a model system (growth medium). The efficacy of PEF treatment was evaluated in relation to different variables, such as electric field strength (25 and 50 kV/cm), treatment time (from 1 to 5 s), initial inoculum level (4 or 6 log cfu/ml), preheating at 50°C, medium pH (4 or 6), and addition of citral at sublethal concentration (i.e., half of minimum inhibiting concentration). The data from plate counting, modeled with the Weibull equation, showed that one of the main factors affecting yeast inactivation was the preheating of the suspension at 50°C. Indeed, higher cell load reductions were obtained with heat-assisted PEF, especially in the presence of citral. The effect of initial cell load was negligible, while pH affected yeast inactivation only without preheating, with higher death kinetics at pH 6. Flow cytometry (FCM) analysis confirmed higher mortality under these conditions. However, the occurrence of injured cells, especially in samples treated at pH 4, was observed. The ability of these cells to recover from the damages induced by treatments was affected by both citral and preheating. The synergic effects of PEF, preheating, and citral were likely due to the increase of membrane permeability (especially at pH 6), as the primary target of electroporation, which favored the solubilization of citral in the cell membrane, enhancing the efficacy of the whole process. The multi-analytical approach (traditional plate counting and FCM) allowed defining parameters to increase PEF efficacy against S. cerevisiae. Moreover, FCM, able to discriminate different physiological states of the yeast population, was helpful to better clarify the action mechanism and the potential recovery of cells after treatment.

Atmospheric double-barrier discharge (DBD) plasma technology is a promising tool in the food industry as an alternative to traditional food preservation methods. However, the effect of the reactive species generated during the treatment on the content of bioactive compounds in food is still little studied, and there are no data concerning potential deleterious effects of DBD-treated foods on human cells. Some functional properties of DBD-treated minimally processed Pink Lady apples were evaluated in comparison with untreated samples through different in vitro and ex vivo tests. Plasma treatment caused only a slight reduction of antioxidant content and antioxidant capacity (up to 10%), mainly limited to the amphiphilic fraction. Noteworthy, treated apple polyphenol extracts did not reduce cell viability and did not suppress the beneficial physiological cell response to oxidative stress in terms of reactive oxygen species production and phase II enzyme activation in human cultured colonocytes.

Barley (Hordeum vulgare L.) is a cereal crop that has been cultivated since ancient times. However, its interest as nutritional food and as food ingredient is relatively new. Thus, in this study, the phenolic compounds of eighteen different varieties of barley (4 waxy and 14 non-waxy) grown under the same agronomic conditions in the same experimental field have been determined by HPLC-DAD-MS. Two new methodologies were developed using new generation superficially porous HPLC columns with different stationary phases: C18 and pentafluorophenyl (PFP). Twelve free phenolic compounds and eight bound phenolic compounds could be identified in barley samples in less than 22min. The study of different method parameters showed that C18 column was more suitable for the analysis of phenolic compounds of barley. Hierarchical cluster analysis (HCA) was conducted in order to assess the different ability of the two different core shell HPLC columns in the discrimination between "waxy" and "non-waxy" varieties, and only HCA of C18 column could separate waxy and non-waxy genotypes. Significant differences in the content of phenolic compounds between waxy and non-waxy samples were found, being waxy barley samples the ones which presented higher content of free and bound phenolic compounds. Once the best discriminant HPLC column was established, principal component analysis (PCA) was applied and it was able to discriminate between "waxy" and "non-waxy" varieties; however it discriminated the barley samples based only in free phenolic compounds. Because of that, partial least squares discriminant analysis (PLS-DA) and Artificial Neural Networks (ANN) were carried out. PLS-DA and ANN permitted the classification of waxy and non-waxy genotypes from both free and bound phenolic compounds.

The main aim of the present research is to assess the possibility of quickly screening fatty acid alkyl esters (FAAE) in olive oils using time domain reflectometry (TDR) and partial least-squares (PLS) multivariate statistical analysis. Eighteen virgin olive oil samples with fatty acid alkyl ester contents and fatty acid ethyl ester/methyl ester ratios (FAEE/FAME) ranging from 3 to 100 mg kg(-1) and from 0.3 to 2.6, respectively, were submitted to tests with time domain resolution of 1 ps. The results obtained in test set validation demonstrated that this new and fast analytical approach is able to predict FAME, FAEE, and FAME + FAEE contents with R(2) values of 0.905, 0.923, and 0.927, respectively. Further measurements on mixtures between olive oil and FAAE standards confirmed that the prediction is based on a direct influence of fatty acid alkyl esters on the TDR signal. The suggested technique appeared potentially suitable for monitoring one of the most important quality attribute of the olive oil in the extraction process.

UNLABELLED: The nondestructive assessment of apricot fruit quality (Bora cultivar) was carried out by means of FT-NIR reflectance spectroscopy in the wavenumber range 12000 to 4000 cm⁻¹. Samples were harvested at four different ripening stages and scanned by a fiber optical probe immediately after harvesting and after a storage of 3 d (2 d at 4 °C and 1 d at 18 °C); the flesh firmness (FF), the soluble solids content (SSC), the acidity (A), and the titratable acidity (malic and citric acids) were then measured by destructive methods. Soft independent modeling of class analogy (SIMCA) analysis was used to classify spectra according to the ripening stage and the storage: partial least squares regression (PLS) models to predict FF, SSC, A, and the titratable acidity were also set-up for both just harvested and stored apricots. Spectral pretreatments and wavenumber selections were conducted on the basis of explorative principal component analysis (PCA). Apricot spectra were correctly classified in the right class with a mean classification rate of 87% (range: 80% to 100%). Test set validations of PLS models showed R2 values up to 0.620, 0.863, 0.842, and 0.369 for FF, SSC, A, and the titratable acidity, respectively. The best models were obtained for the SSC and A and are suitable for rough screening; a lower power prediction emerged for the other maturity indices and the relative predictive models are not recommended. PRACTICAL APPLICATION: The results of the study could be used as a tool for the assessment of the ripening stage during the harvest and the quality during the postharvest storage of apricot fruits.
