RESUMEN

A total of 216 Cobb 500 broiler breeder hens were randomly distributed across 2 dietary treatments with 0 or 1% spray-dried plasma (SDP) resulting in 27 replications/treatment and 4 birds/replication. In addition, 36 roosters were divided between the same treatments and housed, in individual pens, being each bird considered a replicate. Experimental diets were fed from 26 wk until 65 wk of age. At 29, 45, and 63 wk of age, broiler breeder hens were inseminated, and eggs were incubated. Three progeny studies were performed and hatched birds were allocated in a randomized 2 × 2 factorial design (maternal diet: with or without inclusion of 1% SDP × progeny diet: with or without the inclusion of 2% SDP from 1 to 7 d of age). After 7 d of age, all birds received the same diet until 42 d. In all trials, birds were challenged with coccidiosis vaccine at 7 d of age. Furthermore, in the second experiment, heat stress was also included for 6 h a day during the whole trial. At 42-days posthatch in the first experiment, greater FI, BW, and BWG in chicks hatched from breeders fed 1% dietary SDP was observed. This effect did not extend to the other hatches. In the second trial, a decreased FCR in broilers fed the control diet from breeder hens fed 1 % of SDP was observed, as well as an interaction between the SDP groups, since broilers supplemented with SDP and originated from breeders fed SDP showed higher BW and BWG in comparison to other groups at 42 d. In the third trial, contrary to what was observed in the first study, SDP supplementation did not affect any of the performance indexes. In the 3 studies, no differences were found in carcass characteristics. SDP did not affect hen BW, egg production, fertility, or hatch of fertile. These results suggest that providing dietary SDP to broilers has some beneficial effects on broiler chickens.

An experiment was conducted to test the hypothesis that inclusion of spray dried plasma (SDP) in diets increases apparent total tract digestibility (ATTD) and/or the standardized total tract digestibility (STTD) of gross energy (GE) and nutrients in diets for young pigs, and that ATTD of energy and nutrients or STTD of P in individual ingredients are additive in diets containing SDP. Eighty barrows (body weight: 9.30 ± 0.97 kg) were housed in individual metabolism crates and allotted to 1 of 10 diets in a randomized complete block design with 8 replicate pigs per diet. Four diets were prepared without SDP and contained ingredients commonly used in the United States, Canada, the European Union, or Asia. Four additional diets were prepared by mixing 94% of the previous four diets and 6% SDP. A diet containing SDP as the sole source of P and a P-free diet were also formulated. The ATTD of GE and nutrients and the STTD of P were calculated in all diets except the P-free diet and for the four regional diets containing 6% SDP, values were also predicted from the digestibility obtained in SDP alone and the regional diets without SDP. Differences between measured and predicted values for digestibility of GE and nutrients were also calculated. An interaction was observed between SDP and region for the ATTD of soluble dietary fiber where the digestibility decreased (P < 0.05) for pigs fed the U.S. diet with 6% SDP compared with 0% SDP, but that was not the case for the other regional diets. There was no interaction for the ATTD of GE, N, insoluble dietary fiber (IDF), total dietary fiber (TDF), Ca, and P or the STTD of P, but the ATTD and STTD values were greater (P < 0.05) or tended to be greater (P < 0.10%) when 6% SDP was included in the diet compared with diets with 0% SDP. The ATTD of GE, IDF, TDF, and P, and the STTD of P was greater (P < 0.05) for the Asia diet compared with the other diets regardless of inclusion of SDP. The measured ATTD of IDF and TDF was greater (P < 0.05) than the predicted values for the U.S. and European Union diets, and the measured ATTD of GE, N, Ca, and P and the STTD of P was greater (P < 0.05) than the predicted values for the Asia diet. In conclusion, addition of 6% SDP to a diet will increase the ATTD of energy and nutrients and the STTD of P in diets for weanling pigs, and in some cases, the measured ATTD of energy and nutrients or the STTD of P by pigs fed diets containing SDP is greater than predicted from individual ingredients.

This study aimed to evaluate the effects of feeding spray-dried porcine plasma (SDPP) on the protection afforded by the BA71∆CD2 African swine fever virus (ASFV) vaccine prototype. Two groups of pigs acclimated to diets without or with 8% SDPP were intranasally inoculated with 105 plaque-forming units (PFU) of live attenuated ASFV strain BA71∆CD2 and, three weeks later, left in direct contact with pigs infected with the pandemic Georgia 2007/01 ASFV strain. During the post-exposure (pe) period, 2/6 from the conventional diet group showed a transient peak rectal temperature >40.5 °C before day 20 pe, and some tissue samples collected at 20 d pe from 5/6 were PCR+ for ASFV, albeit showing Ct values much higher than Trojan pigs. Interestingly, the SDPP group did not show fever, neither PCR+ in blood nor rectal swab at any time pe, and none of the postmortem collected tissue samples were PCR+ for ASFV. Differential serum cytokine profiles among groups at vaccination, and a higher number of ASFV-specific IFNϒ-secreting T cells in pigs fed with SDPP soon after the Georgia 2007/01 encounter, confirmed the relevance of Th1-like responses in ASF protection. We believe that our result shows that nutritional interventions might contribute to improving future ASF vaccination strategies.

The objective of this study was to evaluate the potential benefits of feeding spray-dried porcine plasma (SDPP) to pigs infected with African swine fever virus (ASFV). Two groups of twelve weaned pigs each were fed with CONVENTIONAL or 8% SDPP enriched diets. Two pigs (trojans)/group) were injected intramuscularly with the pandemic ASFV (Georgia 2007/01) and comingled with the rest of the pigs (1:5 trojan:naïve ratio) to simulate a natural route of transmission. Trojans developed ASF and died within the first week after inoculation, but contact pigs did not develop ASF, viremia, or seroconversion. Therefore, three more trojans per group were introduced to optimize the ASFV transmission (1:2 trojan:naïve ratio). Blood, nasal, and rectal swabs were weekly harvested, and at end of the study ASFV-target organs collected. After the second exposure, rectal temperature of conventionally fed contact pigs increased >40.5 °C while fever was delayed in the SDPP contact pigs. Additionally, PCR Ct values in blood, secretions, and tissue samples were significantly lower (p < 0.05) for CONVENTIONAL compared to SDPP contact pigs. Under these study conditions, contact exposed pigs fed SDPP had delayed ASFV transmission and reduced virus load, likely by enhanced specific T-cell priming after the first ASFV-exposure.

Spray dried plasma (SDP) is commonly used in phase 1 diets for weanling pigs, but it is unknown if SDP affects energy or nutrient digestibility of the subsequent diet. Therefore, two experiments were conducted to test the null-hypothesis that inclusion of SDP in a phase 1 diet fed to weanling pigs will not affect energy or nutrient digestibility of a phase 2 diet without SDP. In experiment 1, 16 newly weaned barrows with an initial body weight (BW) of 4.47 ± 0.35 kg were randomly allotted to a phase 1 diet without SDP or a diet including 6% SDP for 14 d. Both diets were fed on an ad libitum basis. All pigs (BW: 6.92 ± 0.42 kg) had a T-cannula surgically inserted in the distal ileum, moved to individual pens, and fed the common phase 2 diet for 10 d with ileal digesta collection on days 9 and 10. In experiments 2, 24 newly weaned barrows (initial BW: 6.60 ± 0.22 kg) were randomly allotted to phase 1 diets without SDP or a diet containing 6% SDP for 20 d. Both diets were provided on an ad libitum basis. All pigs (BW: 9.37 ± 1.40 kg) were then moved to individual metabolism crates and fed the common phase 2 diet for 14 d with the initial 5 d being the adaptation period to the diet followed by 7 d of fecal and urine collection according to the marker-to-marker procedure. The apparent ileal digestibility (AID) of starch, crude protein (CP), amino acids (AA), and acid hydrolyzed ether extract (AEE), was determined in experiment 1, and the apparent total tract digestibility (ATTD) of gross energy (GE), insoluble-, soluble-, and total-dietary fiber, Ca, and P, and the retention and biological value of N were determined in experiment 2. The statistical model included diet as fixed effect and block and pig within block as random effects. Results of experiment 1 indicated that the AID of starch, CP, AEE, and AA in phase 2 were not affected by phase 1 treatment. Results of experiment 2 indicated that the ATTD of GE, insoluble-, soluble-, and total-dietary fiber, Ca, and P and N retention and biological value in phase 2 were also not affected by phase 1 treatment. In conclusion, feeding weanling pigs a diet with 6% SDP in phase 1 did not affect the AID or ATTD of energy and nutrients in a phase 2 diet without SDP.


Highly digestible and palatable ingredients are usually included in diets for newly weaned pigs to improve feed consumption and nutrient digestibility. Weaning typically causes a decrease in digestion and nutrient absorption, but inclusion of sprayed dried plasma (SDP) in phase 1 diets to weanling pigs may increase nutrient digestibility. However, data to demonstrate possible effects of including SDP to a phase 1 diet for newly weaned pigs on nutrient digestibility of a phase 2 diet without SDP are limited. Therefore, two experiments were conducted to test the hypothesis that inclusion of SDP in a phase 1 diet fed to weanling pigs does not affect the energy or nutrient digestibility of a phase 2 diet without SDP. Results of the two experiments demonstrated that weaned pigs fed a phase 1 diet with 6% SDP did not increase nutrient or energy digestibility of phase 2 compared with pigs fed diets without SDP, indicating that SDP had no carryover effects on nutrient digestibility. Therefore, it appears that feeding SDP in phase 1 diets to weanling pigs does not cause long-term intestinal changes that will affect digestibility of nutrients and energy in phase 2.

This survey was conducted to estimate the incidence and level of potential viral contamination in commercially collected porcine plasma. Samples of spray dried porcine plasma (SDPP) were collected over a 12- month period from eight spray drying facilities in Spain, England, Northern Ireland, Brazil, Canada, and the United States. In this survey, viral load for several porcine pathogens including SVA, TGEV, PRRSV (EU and US strains), PEDV, PCV-2, SIV, SDCoV and PPV were determined by qPCR. Regression of Ct on TCID50 of serial diluted stock solution of each virus allowed the estimate of potential viral level in SDPP and unprocessed liquid plasma (using typical solids content of commercially collected porcine plasma). In this survey SVA, TGEV or SDCoV were not detected in any of the SDPP samples. Brazil SDPP samples were free of PRRSV and PEDV. Samples of SDPP from North America primarily contained the PRRSV-US strain while the European samples contained the PRRSV-EU strain (except for one sample from each region containing a relatively low estimated level of the alternative PRRSV strain). Estimated viral level tended to be in the range from <1.0 log10 TCID50 to <2.5 log10 TCID50. Estimated level of SIV was the exception with a very low incidence rate but higher estimated viral load <3.9 log10 TCID50. In summary, the incidence of potential viral contamination in commercially collected porcine plasma was variable and estimated virus level in samples containing viral DNA/RNA was relatively low compared with that occurring at the peak viremia during an infection for all viruses or when considering the minimal infectious dose for each of them.

The study was conducted to evaluate the effects of spray-dried plasma (SDP) supplementation during late gestation and lactation on productive performance and immune responses of sows and their litters. Twelve sows (227.78 ± 2.16 kg average body weight; 2.0 average parity) were randomly allotted to two dietary treatments: a basal diet (CON) and the basal diet supplemented with 1% SDP. Sows were fed experimental diets from d 30 before farrowing to weaning of their piglets. Blood samples were collected from sows on d 1, 3, and 7 of lactation and from two randomly selected nursing pigs per litter on d 3 and 7 after birth, and d 1, 3, and 7 after weaning. Productive performance and immune responses of sows and their piglets were measured. There was a trend of less body weight loss in sows supplemented with SDP (p < 0.10) during the lactation period and a trend of greater (p < 0.10) average daily gain in SDP piglets compared to those in the CON group. Sows in the SDP group tended to have lower (p < 0.10) serum concentrations of tumor necrosis factor-α (TNF-α), transforming growth factor-ß1 (TGF-ß1), and cortisol on d 3 and lower serum concentration of TNF-α on d 7 compared with sows in CON group. In comparison with CON piglets, piglets from SDP sows tended to have lower (p < 0.10) serum concentrations of TNF-α, TGF-ß1, and cortisol on d 7 after birth, lower (p < 0.10) serum TNF-α and C-reactive protein on d 3 and 7 after weaning, and greater (p < 0.10) average daily gain after weaning. Moreover, weaned pigs from sows fed SDP had significantly lower (p < 0.05) serum concentrations of cortisol and TGF-ß1 on d 3 and 7 postweaning, respectively, than CON piglets. In conclusion, SDP supplementation in sow diets from late gestation to weaning improved the productive performance of sows and their offspring; the beneficial effects of SDP may be mediated in part through modulation of immune responses of both sows and piglets.

Spray-dried animal plasma (SDP) in feed for several animal species provides health benefits, but research about use of SDP in shrimp feed is very limited. The objectives of the present study were to investigate the effects of dietary SDP on growth performance, feed utilization, immune responses, and prevention of Vibrio parahaemolyticus infection in Pacific white shrimp (Litopenaeus vannamei). In Experiment 1, the post-larvae were divided into five groups (four tank/group and 80 shrimp/tank) and fed four times daily diets with porcine SDP at 0, 1.5, 3, 4.5, and 6% of the diet for 45 days. In Experiment 2, the surviving shrimp from Experiment 1 were redistributed into six groups: four SDP groups as in Experiment 1 plus the positive and negative controls (four tank/group and 30 shrimp/tank). They were then challenged with V. parahaemolyticus by immersion at 105 colony-forming units (CFU)/mL and were fed with the same diets for another 4 days. In Experiment 1, shrimp fed 4.5% or 6% SDP diets had significantly higher body weight, survival rate, and improved feed conversion ratio. The immune parameters (total hemocyte count and phagocytic, phenoloxidase, and superoxide dismutase activities) of the shrimp fed 3-6% SDP diets also showed significant enhancement compared to the control. In Experiment 2, the survival rates of the 3-6% SDP groups were significantly higher than the positive control at day 4 after the immersion challenge. Likewise, the histopathological study revealed milder signs of bacterial infection in the hepatopancreas of the 3-6% SDP groups compared to the challenged positive control and 1.5% SDP groups. In conclusion, shrimp fed diets with SDP, especially at 4.5-6% of the diet, showed significant improvement in overall health conditions and better resistance to V. parahaemolyticus infection.

The aim of this study was to see how spray-dried plasma (SDP) supplementation affected broiler chicken performance, intestinal permeability, and bone strength during persistent heat stress. One-day-old chicks (n = 480) were randomly assigned into twelve environmental corrals; four thermoneutral (TN-negative control, maintained at 24 °C from d 21-42); four heat stress (HS, exposed to 35 °C from d 21-42); and four heat stress treated with 2% SDP in the feed until d 28 followed by 1% SDP until d 42 (HS-SDP). The performance and serum levels of fluorescein isothiocyanate-dextran (FITC-d) were evaluated at d 21, 28, 35, and 42. The tibias strength was evaluated on d 21 and 42. The increment in chicken temperature (p < 0.05) was observed two h following the increase in environmental temperature in both HS groups and was associated with decreased performance parameters compared with the TN group. At d 42 of age, the chickens exposed to HS had an impaired gut permeability and decreased tibia strength compared to the TN group (p < 0.05). However, partially feeding SDP mitigated these adverse effects significantly. These findings imply that using SDP strategically during stressful times, such as prolonged heat stress, may help mitigate its negative consequences.

BACKGROUND: Nutritional strategies for sows designed to reduce peripartum stress are suggested to support postpartum recovery and productivity. Spray-dried plasma (SDP) in sow feed has been reported to benefit sow and litter performance. Stressed animals fed diets with SDP have a more efficient immune response supporting animal recovery and health. The objectives of the present study using 452 sows (147 parity 1 sows, 148 parity 2 sows) were to determine if 0, 0.5 or 2.5% spray-dried porcine plasma (SDPP) in peripartum feed provided from entry in maternity through day 5 of lactation affects sow productivity and serological immune and oxidation status markers around parturition. Post-weaning sow productivity parameters including litter size at the next parturition was evaluated, but peripartum diets were only provided during the first parturition. RESULTS: In the first parturition, total born litter size was lower (P <  0.05) especially for sows allotted to the peripartum diet with 2.5% SDPP. Percentage of stillborn pigs decreased quadratically (P <  0.05) for sows fed 0.5% or 2.5% SDPP compared to 0% SDPP in peripartum feed and this result was not affected by total born litter size. Serum glutathione peroxidase activity linearly increased (P <  0.01) with increased dietary SDPP for both prepartum and postpartum sampling periods. In the next parturition, total born pigs from combined data of parity 1 and 2 sows linearly increased (P <  0.05) and live born pigs tended (P = 0.09) to linearly increase as level of SDPP increased and this result was not affected by total born litter size in the first parturition. The change in total and live born pigs from the first to the next parturition linearly (P <  0.01) increased as dietary SDPP increased for parity 1 and 2 sows. CONCLUSIONS: The reduced percentage of stillborn pigs and increased litter size of parity 1 and 2 sows in the next parturition was independent of total born litter size in the first parturition suggesting SDPP in peripartum sow feed may have merit for reducing stillborn pigs and benefit litter size in the next parturition for parity 1 and parity 2 sows.

Dietary supplementation with spray-dried porcine plasma (SDP) can modulate the immune response of gut-associated lymphoid tissue. SDP supplementation reduces acute mucosal inflammation, as well as chronic inflammation associated with aging. The aim of this study was to analyze if SDP supplementation could ameliorate colitis in a genetic mouse model of inflammatory bowel disease (IBD). Wild-type mice and Mdr1a knockout (KO) mice were administered a control diet or an SDP-supplemented diet from day 21 (weaning) until day 56. The histopathological index, epithelial barrier, and intestinal immune system were analyzed in the colonic mucosa. KO mice had higher epithelial permeability, increased Muc1 and Muc4 expression, and lower abundance of E-cadherin and Muc2 (all p < 0.001). SDP prevented these effects (all p < 0.05) and decreased the colonic inflammation observed in KO mice, reducing neutrophil and monocyte infiltration and activation and the percentage of activated T helper lymphocytes in the colonic mucosa (all p < 0.05). SDP also diminished proinflammatory cytokine expression and increased the anti-inflammatory IL-10 concentration in the colonic mucosa (all p < 0.05). In conclusion, dietary supplementation with SDP enhances colon barrier function and reduces mucosal inflammation in a mouse model of IBD.

RESUMEN

Semi-structured interviews were employed to (a) characterize LEOs' knowledge of ASD, (b) understand interactions between LEOs and individuals with ASD, and (c) identify training needs to prepare LEOs for interactions with the ASD community. Researchers utilized a constructivist grounded theory approach to analyze data from 17 participants: (a) six LEOs, (b) six adults with ASD, and (c) five caregivers. Common themes included the (a) potential for misinterpretations of behavior of individuals with ASD; (b) helpfulness of an identification system/symbol for ASD; (c) need for interactive, mandatory training unique to LEOs' needs; and, (d) importance of building community connections between LEOs and individuals with ASD. Findings are discussed within the context of previous research related to law enforcement and ASD.

RESUMEN

Spray-dried plasma (SDP) is a functional ingredient commonly utilized in swine production and calf milk replacers to improve performance, feed efficiency, and health. The improvements noted with SDP in animal production are more pronounced under commercial production conditions compared to cleaner research settings. Multiple modes of action of SDP have been proposed, including either directly influencing the immune inflammatory response locally or systemically, and/or through the indirect modification of beneficial microbial populations. Spray-dried plasma included at various dietary levels and duration of feeding in broilers has been evaluated in different production and challenging conditions with beneficial effects on broiler performance, as noted in other animals. The purpose of this review is to discuss research related to the modes of action of SDP on immunomodulation and improved intestinal health and specifically discuss research conducted utilizing SDP in feed for poultry. Collectively, the data available indicates that SDP improves early intestinal health and supports an efficient immune system response both locally at the intestine and systemically, thereby benefiting growth, feed efficiency, and survival of broilers in conventional commercial production and under challenging conditions such as disease or environmental stressors.

The objective of this study was to evaluate the effectiveness of the SurePure Turbulator ultraviolet-C (UV-C, 254 nm wavelength) irradiation equipment on inactivation of different enveloped and non-enveloped viruses in commercially collected liquid animal plasma. Specifically, Pseudorabies virus (PRV), Porcine reproductive and respiratory syndrome virus (PRRSV), Porcine epidemic diarrhea virus (PEDV), Bovine viral diarrhea virus (BVDV), Classical swine fever virus (CSFV), Swine influenza virus (SIV) as enveloped viruses and Porcine parvovirus (PPV), Swine vesicular disease virus (SVDV), Porcine circovirus type 2 (PCV-2) and Senecavirus A (SVA) as non-enveloped viruses, were inoculated in bovine or porcine plasma and subjected to different UV-C irradiation doses (0, 750, 1500, 3000, 6000 and 9000 J/L) using an UV-C device developed for opaque liquid working under turbulent flow. The enveloped viruses tested were inactivated at < 3000 J/L of UV-C, being the dose needed to inactivate 4 log TCID50 (4D) of 1612 J/L for PRV,1004 J/L for PRRSV, 1953 J/L for PEDV, 1639 J/L for SIV, 1641 J/L for CSFV and 1943 J/L for BVDV. The non-enveloped viruses tended to have higher 4D values: 2161 J/L for PPV, 3223 J/L for SVA and 3708 J/L for SVDV. Because the initial viral concentration was <4.0 Log for PCV-2, it was not possible to calculate the 4D value for this virus. In conclusion, these results demonstrated that the SurePure Turbulator UV-C treatment system is capable of inactivating significant levels of swine viruses inoculated in commercially collected porcine or bovine plasma. It was concluded that irradiation with UV-C can provide an additional redundant biosafety feature in the manufacturing process of spray-dried animal plasma.

Experimental objectives of this study were to determine effects of formaldehyde treatment on the chemical composition of spray-dried plasma (SDP) and to test the hypothesis that growth performance of pigs fed formaldehyde-treated diets containing SDP or diets containing formaldehyde-treated SDP is not reduced compared with pigs fed untreated control diets. Sal CURB ASF liquid antimicrobial and CURB RM Extra liquid mold inhibitor (Kemin Industries, Des Moines, IA) were applied on SDP at 0.1% or 0.3% to determine effects of the products on chemical and functional properties of SDP. Regardless of product, there were no changes in SDP for analyzed protein, ash, pH, or moisture concentration, but IgG concentration in SDP was decreased 8% and 24%, respectively, for 0.1% and 0.3% inclusion of Sal CURB or CURB RM. Two feeding studies using weaned pigs were conducted to determine effects of formaldehyde applied at 0.3% to SDP (experiment 1) or 0.3% to a complete diet containing 5% SDP (experiment 2). Experiment 1 pigs (n = 265) were weaned at 20 ± 2 d of age and allotted to five treatment groups. Experiment 2 pigs (n = 135) were weaned in two groups at 20 ± 2 d of age and allotted to three treatments groups. In experiment 1, the untreated control diet contained soy protein concentrate (SPC) and test diets contained 2.5% or 5.0% SDP without or with formaldehyde treatment. In experiment 2, formaldehyde was applied to a diet containing 5% SDP and an untreated SPC control diet and an untreated diet containing 5% SDP were also included in the experiment. In experiment 1, linear increases (P < 0.05) in average daily gain (ADG), average daily feed intake (ADFI), and gain-to-feed ratio (G:F) were observed as SDP was included in the diets and the relative bioavailability of formaldehyde-treated SDP was 62% (P = 0.018) if calculations were based on ADG and 15% (P = 0.031) if calculations were based on ADFI. In experiment 2, pigs fed the SDP diet untreated or treated with formaldehyde had increased (P < 0.05) final body weight, ADG, ADFI, and G:F compared with pigs fed the control diet. However, formaldehyde treatment of the plasma-containing diet did not affect pig growth performance compared with pigs fed the untreated SDP diet. In conclusion, formaldehyde treatment applied directly on SDP affects analyzed concentrations of IgG and reduces growth rate of pigs. Treating a complete diet containing 5% SDP with formaldehyde did not affect pig growth performance, and pigs fed diets containing SDP had improved growth performance than those fed the control diet without SDP.

This study evaluated whether dietary spray-dried plasma (SDP) can ameliorate inflammation, lethargic behaviors, and impairment of reproduction caused by lipopolysaccharide (LPS) challenge during late pregnancy. Two experiments were conducted with 125 mated female mice (C57BL/6 strain) in each experiment. All mice were shipped from a vendor on the gestation day (GD) 1 and arrived at the laboratory on GD 3. Mice were randomly assigned to dietary treatments with or without 8% SDP in the diet. On GD 17, mice determined pregnant by BW and abdomen shape were randomly assigned to intraperitoneal injections with or without 2 µg LPS. In experiment 1, 17 mice (26.7 ± 1.7 g BW) were identified pregnant and euthanized 6 h after the LPS challenge to measure inflammatory responses in uterus and placenta. In experiment 2, 44 mice (26.0 ± 1.6 g BW) were identified pregnant and euthanized 24 h after the LPS challenge to assess behavior and late-term pregnancy loss. Growth performance and reproductive responses, such as loss of pregnancy, percentage of fetal death, and etc., were measured in all pregnant mice. The LPS challenge increased (P < 0.05) uterine and placental tumor necrosis factor-α and interferon-γ, late-term pregnancy loss, and lethargy score, and decreased (P < 0.05) uterine transforming growth factor-ß1, moving time and number of rearing, and growth and feed intake. The SDP decreased (P < 0.05) concentrations of both pro-inflammatory and anti-inflammatory cytokines in one or both tissues, and the lethargy score, and increased (P < 0.05) moving time and number of rearing, growth of pregnant mice, and fetal weight. However, the SDP did not affect late-term pregnancy loss caused by the LPS challenge. Consequently, dietary SDP attenuated acute inflammation and lethargic behaviors of pregnant mice caused by the LPS challenge, but did not affect late-term pregnancy loss after the acute inflammation.

BACKGROUND: Stress causes inflammation that impairs intestinal barrier function. Dietary spray-dried plasma (SDP) has recognized anti-inflammatory effects and improvement of gut barrier function. Therefore, the purpose of this study was to investigate the effects of dietary SDP on intestinal morphology of mated female mice under stress condition. RESULTS: Villus height, width, and area of small intestines were low on gestation day (GD) 3 or 4 under stress conditions, and higher later (Time, P < 0.05). Crypt depth of colon was low on GD 4 and higher later (Time, P < 0.05). Meanwhile, the SDP treatments improved (P < 0.05) intestinal morphology, indicated by increased villus height, villus width, villus area, and ratio between villus height and crypt depth of small intestines and crypt depth of colon, and by decreased crypt depth of small intestines, compared with the control diet. The SDP treatments also increased (P < 0.05) the number of goblet cells in intestines compared with the control diet. There were no differences between different levels of SDP. CONCLUSION: Dietary SDP improves intestinal morphology of mated female mice under stress condition.

Dietary immunoglobulin concentrates prepared from animal plasma can modulate the immune response of gut-associated lymphoid tissue (GALT). Previous studies have revealed that supplementation with serum-derived bovine immunoglobulin/protein isolate (SBI) ameliorates colonic barrier alterations in the mdr1a-/- genetic mouse model of IBD. Here, we examine the effects of SBI on mucosal inflammation in mdr1a-/- mice that spontaneously develop colitis. Wild type (WT) mice and mice lacking the mdr1a gene (KO) were fed diets supplemented with either SBI (2% w/w) or milk proteins (Control diet), from day 21 (weaning) until day 56. Leucocytes in mesenteric lymph nodes (MLN) and in lamina propria were determined, as was mucosal cytokine production. Neutrophil recruitment and activation in MLN and lamina propria of KO mice were increased, but were significantly reduced in both by SBI supplementation (p < 0.05). The increased neutrophil recruitment and activation observed in KO mice correlated with increased colon oxidative stress (p < 0.05) and SBI supplementation reduced this variable (p < 0.05). The Tact/Treg lymphocyte ratios in MLN and lamina propria were also increased in KO animals, but SBI prevented these changes (both p < 0.05). In the colon of KO mice, there was an increased production of mucosal pro-inflammatory cytokines such as IL-2 (2-fold), IL-6 (26-fold) and IL-17 (19-fold), and of chemokines MIP-1ß (4.5-fold) and MCP-1 (7.2-fold). These effects were significantly prevented by SBI (p < 0.05). SBI also significantly increased TGF-ß secretion in the colon mucosa, suggesting a role of this anti-inflammatory cytokine in the modulation of GALT and the reduction of the severity of the inflammatory response during the onset of colitis.

BACKGROUND: Conventional vaccination programs using a single injection of a combined vaccine against porcine circovirus type 2 (PCV2) and Mycoplasma hyopneumoniae (MHYO) can promote a strong immune response that reduces feed intake for 24 to 48 h post injection. Often such vaccines are given around the time of weaning during a critical stress period in which feed intake is already compromised. Spray dried porcine plasma (SDPP) is a protein source used in starter diets that increases post-weaning feed intake of pigs. The objectives of this study were to determine the effects of a conventional vaccination program along with feeding SDPP in a starter diet on antibody development and wean to finish performance of pigs. RESULTS: Pigs fed the starter diet with SDPP had improved body weight, average daily weight gain and average daily feed intake during the initial 14 d after weaning along with improved feed efficiency during the initial 7 d after weaning and these responses were independent of vaccination. Vaccination at 3 d after weaning had no significant effect on performance during the initial 14 d after weaning. Cumulative mortality was reduced for pigs fed the starter diet with SDPP, while vaccinated pigs had reduced mortality from d 48 to 145. Both vaccinated pigs and those fed the starter diet with SDPP had heavier carcass weight. One pig per pen was challenged with PCV2 at d 63. A higher percentage of vaccinated pigs were sero-positive for antibodies against PCV2 and MHYO at d 35, 63 and 78. Antibody values against PCV2 were higher for vaccinated pigs at d 35 and 63, but lower at d 146. Percentage of positive samples for PCV2 genome in serum was reduced for vaccinated pigs at d 117 and 146. Antibody values against MHYO were increased for vaccinated pigs at d 35, 63 and 78. CONCLUSIONS: Vaccination supported a long term antibody response against PCV2 and a moderate but weaker antibody response against MHYO for early finishing pigs challenged with PCV2. Using SDPP in the starter diet along with vaccination supported the best long-term beneficial effects on survival to market and carcass weight.

A novel ultraviolet light irradiation (UV-C, 254 nm) process was designed as an additional safety feature for manufacturing of spray dried porcine plasma (SDPP). In Exp. 1, three 10-L batches of bovine plasma were inoculated with 10(5.2 ± 0.12) tissue culture infectious dose 50 (TCID50) of porcine parvovirus (PPV) per mL of plasma and subjected to UV-C ranging from 0 to 9180 J/L. No viable PPV was detected in bovine plasma by micro-titer assay in SK6 cell culture after UV-C at 2295 J/L. In Exp. 2, porcine plasma was subjected to UV-C (3672 J/L), then spray dried and mixed in complete mash diets. Diets were a control without SDPP (Control), UV-C SDPP either at 3% (UVSDPP3) or 6% (UVSDPP6) and non-UV-C SDPP at 3% (SDPP3) or 6% (SDPP6). Diets were fed ad libitum to 320 weaned pigs (26 d of age; 16 pens/diet; 4 pigs/pen) for 14 d after weaning and a common diet was fed d 15 to 28. During d 0 to 14, pigs fed UVSDPP3, UVSDPP6, or SDPP6 had higher (P < 0.05) weight gain and feed intake than control. During d 0 to 28, pigs fed UVSDPP3 and UVSDPP6 had higher (P < 0.05) weight gain and feed intake than control and SDPP3, and SDPP6 had higher (P < 0.05) feed intake than control. Also, pigs fed UVSDPP had higher (P < 0.05) weight gain than pigs fed SDPP. In conclusion, UV-C inactivated PPV in liquid plasma and UVSDPP used in pig feed had no detrimental effects on pig performance.
