Count variables are often positively skewed and may include many zero observations, requiring specific statistical approaches. Interpreting abiotic factor changes in insect populations of crop pests, under this condition, can be difficult. The analysis becomes even more complicated because of possible temporal or spatial correlation, irregularly spaced data, heterogeneity over time, and zero inflation. Generalized additive models (GAM) are important tools to evaluate abiotic factors. Moreover, Markov chain Monte Carlo (MCMC) techniques can be used to fit a model that contains a temporal correlation structure, based on Bayesian statistics (BGAM). We compared methods of modeling the effects of temperature, precipitation, and time for the Brevicoryne brassicae (L.) population in Uberlândia, Brasil. We applied the proposed BGAM to the data, comparing this to the GAM model with and without autocorrelation for time, using the statistical programming language R. Analysis of deviance identified significant effects of the smoothers for precipitation and time on the frequentist models. With BGAM, the problem in variance estimations for precipitation and temperature from the previous models was solved. Furthermore, trace and density plots for population-level effects for all parameters converged well. The estimated smoothing curves showed a linear effect with an increase of precipitation, where lower precipitation indicated no presence of the aphid. The average temperature did not affect the aphid incidence. Autocorrelation was solved with ARMA structures, and the excess of zero was solved with zero-inflation models. The example of B. brassicae incidence showed how well abiotic (and biotic) factors can be modeled and analyzed using BGAM.

Aphidius colemani (Viereck) was reported in Brazil before the Biological Control Program of Wheat Aphids (BCPWA) when Mediterranean genotypes were introduced from France and Israel. This species was re-described as a complex called A. colemani group composed of three species. Consequently, uncertainty remains about which parasitoid of the group is occurring in southern Brazil. This study has two main objectives: (i) re-examine the species status of A. colemani group collected during the introduction of parasitoids and from a 10-year (2009-2018) monitoring program in wheat fields in northern Rio Grande do Sul (RS), Brazil; (ii) describe the variation in the population density of parasitoids and its association with meteorological factors during this period. We examined 116 specimens from the Embrapa Wheat entomological collection, and those collected in Moericke traps in Coxilha, RS. All the parasitoids of the A. colemani group from the BCPWA period were identified as Aphidius platensis (Brèthes). In traps, 6541 cereal aphid parasitoids were collected, of which 61.9% (n = 4047) were from A. colemani group and all those were identified as A. platensis. Temperature was the factor that effected population density with the highest number of parasitoids recorded in the winter months. Sex ratio changed between years varying from 0.50 to 0.97. The parasitoid A. platensis was the only species in the A. colemani group sampled during 10 years of monitoring.

The present study evaluated the use of sex-sorted sperm upon estrus detection (ED) or following timed artificial insemination (TAI) in lactating dairy cows. Additionally, the effect of the presence of a corpus luteum (CL) at the beginning of the TAI protocol was verified. Cows (539 crossbred Gir × Holstein and 87 Holstein) were classified according to the presence or absence of CL by ultrasonography exam. Cows with a CL were randomly assigned into one of two groups (CL-ED/AI or CL-TAI), and cows without a CL (NoCL-TAI) received TAI. Cows from the CL-ED/AI group received 500mg of cloprostenol intramuscularly and were inseminated 12h after ED in the following five days. Cows from the TAI groups (CL or NoCL) received TAI. Cows receiving CL-ED/AI had a lower (P<0.0001) service rate (45.1%, 101/224) than TAI groups (CL-TAI=94.2%, 180/191 and NoCL-TAI=97.2%, 205/211). However, cows receiving AI upon ED (CL-ED/AI=31.7%, 32/101) presented higher (P=0.03) pregnancy per AI (P/AI) than cows bred following TAI (CL-TAI=19.4%, 35/180 and NoCL-TAI=23.9%, 49/205). Despite the lower P/AI, cows receiving TAI presented greater (P=0.07) proportion of pregnant cows at the end of the reproductive program (CL-TAI=18.3%, 35/191 and NoCL-TAI=23.2%, 49/211) than those inseminated upon ED (14.3%, 32/224). There was no effect (P=0.45) of the presence of a CL at the beginning of the synchronization protocol on P/AI. Thus, the use of TAI programs, regardless of the presence of CL in the beginning of the synchronization protocol, increases the service and pregnancy rates but reduces the P/AI when compared to the use of sex-sorted sperm upon ED.

alpha-Hydroxynitrile lyase (HNL, acetone cyanohydrin lyase, EC 4.1.2.37) was purified to homogeneity from young leaves of the cyanogenic tropical crop plant cassava (Manihot esculenta Crantz). The purified protein is a homo-trimer with a subunit relative molecular mass of 28,500 on sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The active protein is not glycosylated and does not contain a flavin group. HNL exhibits complex kinetics which vary according to substrate concentration and may be related to aggregation of the enzyme. HNL activity against two natural substrates, acetone cyanohydrin and 2-butanone cyanohydrin, and one nonphysiological substrate, 2-pentanone cyanohydrin, was demonstrated. N-terminal and internal peptide sequences, obtained from HNL digested with the endoproteinase Glu-C, were used to design degenerate oligonucleotide primers for polymerase chain reaction with single-strand cDNA, using purified mRNA from cotyledons as template. The resulting DNA fragment was used to probe a cassava cotyledon cDNA library. Four cDNA clones were isolated, sequenced, and shown to contain derived amino acid sequences identical to those obtained from the purified protein.

RESUMEN

A colorimetric assay for alpha-hydroxynitrile lyase which utilizes acetone cyanohydrin as a substrate is described. The assay is based on measurement of the HCN formed when the lyase catalyzes the dissociation of acetone cyanohydrin. The procedure was devised for use with the optically inactive acetone cyanohydrin but will be applicable to enzymes utilizing other cyanohydrins.

Foi efetuado um estudo cruzado comparativo duplo-cego, randomizado, controlado com placebo, para avaliar a eficacia da nifedipina retard quando adicionada a terapeutica basal anti-hipertensiva que nao tenha conseguido baixar os niveis da pressao sanguinea diastolica ate valores normais.Trinta e seis pacientes de ambulatorio com hipertensao leve e moderada receberam nifedipina ou placebo durante 4 semanas apos um periodo de "washout" de 4 semanas. Este procedimento foi seguido por uma semana de placebo e 4 semanas de nifedipina ou placebo. Os pacientes foram avaliados semanalmente. Nifedipina retard foi administrada na dose de 20 mg, 2 vezes ao dia. O placebo foi administrado em regime identico. Em ambos os grupos, a nifedipina provocou uma reducao estatisticamente significativa (p < 0.01) nas pressoes sistolica e diastolica. Nao foram registradas alteracoes significantes na frequencia cardiaca com nifedipina nem alteracoes significantes da pressao sanguinea ou frequencia cardiaca quando se associou placebo a terapeutica basal. A tolerancia foi boa, porem houve casos de discreta cefaleia,edema maleolar e rubor, em alguns pacientes

Os autores realizaram 16 urografias em pacientes portadores de insufiencia renal aguda e cronica, com altas doses de contraste iodado, obtendo imagens radiograficas diagnosticas, sem no entanto, evidenciarem alteracoes na funcao renal, baseado na dosasgem sanguinea de creatinina pre e pos exame radiologico. Tambem nao foram notados efeitos colaterais do tipo hipersensibilidade
